Repurposing Avasimibe to Inhibit Bacterial Glycosyltransferases.

We are interested in identifying and characterizing small molecule inhibitors of bacterial virulence factors for their potential use as anti-virulence inhibitors. We identified from high-throughput screening assays a potential activity for avasimibe, a previously characterized acyl-coenzyme A: cholesterol acyltransferase inhibitor, in inhibiting the NleB and SseK arginine glycosyltransferases from Escherichia coli and Salmonella enterica, respectively. Avasimibe inhibited the activity of the Citrobacter rodentium NleB, E. coli NleB1, and S. enterica SseK1 enzymes, without affecting the activit

Glycosyltransferases: Capturing catalytic states and conformational gating.

Glycosyltransferases (GTs) install and remodel glycans that regulate protein function and shape extracellular matrices and microbial envelopes. Recent high-resolution structures, enabled by cryo-EM and X-ray crystallography, and increasingly complemented by AI-assisted modeling, now capture GTs in mechanistically informative states, including donor-acceptor complexes, gated conformations, and membrane polymerases engaged with nascent chains. In parallel, molecular simulations, including MD and QM/MM methodologies, are being used in selected GTs to map free-energy landscapes and resolve how act

Carcinoma-related alterations of glycosyltransferases in human tissues.

The glycosyltransferases responsible for catalyzing additions of A, B, and H sugars to cellular acceptors were studied in 23 cases of primary carcinoma. The carcinomas were derived from mouth, tongue, larynx, lung, cervix, esophagus, stomach, and colon. Comparisons of A, B, and H enzymes were made between mucosal extracts from tumor and from normal adjacent tissue and, in the case of gastrointestinal tract, extracts derived from mucosae of individuals free of disease. The most prevalent finding was that of alpha-2-fucosyltransferase (H enzyme) deficiency in tumor extracts from Group A, B, and

Halophytic C‑Glycosyltransferases Enable C‑Glycosylation in Organic Solvents.

C-glycosyltransferases from glycosyltransferase family 1 transfer sugar moieties to carbon atoms in the substituted aromatic rings of various small molecules. They are coveted biocatalysts for the synthesis of high-value glycosides since the resulting β-C-glycosidic linkage is usually more stable in vivo and in vitro than its O-glycosidic counterpart. One of the main bottlenecks in the biocatalytic glycosylation processes of small molecules is the low aqueous solubility of the acceptor substrate, drastically limiting product yields. One solution is to conduct the reaction in organic solvents p

Substrate-induced dimerization of elaiophylin glycosyltransferase reveals a novel self-activating form of glycosyltransferase for symmetric glycosylation.

Elaiophylin (Ela), a unique 16-membered symmetric macrodiolide antibiotic, displays broad biological activity. Two rare 2-deoxy-L-fucose moieties at the ends of Ela are critical for its activity. Previously, elaiophylin glycosyltransferase (ElaGT) was identified as the enzyme that is responsible for the symmetric glycosylation of Ela, acting as a potential enzymatic tool for enhancing the diversity and activity of Ela. However, a symmetric catalytic mechanism has never been reported for a glycosyltransferase (GT). To explore the catalytic mechanism, the structure of ElaGT was determined in fou

Glycosyltransferases as versatile tools to study the biology of glycans.

All cells are decorated with complex carbohydrate structures called glycans that serve as ligands for glycan-binding proteins (GBPs) to mediate a wide range of biological processes. Understanding the specific functions of glycans is key to advancing an understanding of human health and disease. However, the lack of convenient and accessible tools to study glycan-based interactions has been a defining challenge in glycobiology. Thus, the development of chemical and biochemical strategies to address these limitations has been a rapidly growing area of research. In this review, we describe the us

The roles of glycosyltransferases in shaping flavonoid diversity in plants.

Covering: up to 2026Flavonoids belong to one of the largest families of specialized metabolites in plants and play pivotal roles in plant adaptation and human health. In their biosynthesis, the formation of core aromatic structures is followed by various chemical modifications that further decorate these scaffolds, shaping their chemical diversity, biological functions, and physicochemical properties. Among these modifications, glycosylation is the most prevalent, transferring monosaccharides from activated nucleotide sugars to acceptor molecules to form glycosidic bonds. This reaction is cata

Glycoproteins, Glycolipids, or Both: Why do Glycosyltransferases Recognize Different Acceptors?

Glycosylation is an important post-translational modification catalyzed by glycosyltransferases (GTs), which comprise a large group of enzymes with diversified specificity. Most GTs show high specificity toward donor and acceptor molecules, but some can recognize several donor or acceptor molecules and/or create novel glycosidic linkage types. This promiscuity has profound implications for cellular processes, influencing signaling pathways, protein stability, and disease progression. GT promiscuity may result from the structural peculiarities of the enzyme and acceptor, modulation by a ligand,

Human Proteoglycan Linkage Region Glycosyltransferases are Dimeric and Show Unexpected Specificities.

We selected the N,O-glycosylated proteoglycan bikunin as a model to establish a chemoenzymatic approach to defined proteoglycans using native chemical ligation. Overexpression of the human linkage region glycosyltransferases B4GalT7, B3GalT6 and B3GlcAT-1 as N-terminal SUMO-fusions gave high yields of soluble and active enzymes in E. coli. When starting with xylosylated bikunin peptides the transferases performed well in enzymatic cascade reactions and provided the desired linkage region tetrasaccharide glycopeptides. B3GalT6 and B3GlcAT-1 led to side products with N,O-glycosylated bikunin pep

[Chondrocyte glycosyltransferases: new pharmacological targets for degenerative diseases of articular cartilage?].

Arthritis, osteoarthritis and other degenerative diseases characterized by cartilage deterioration are the most prevalent chronic human health disorders. Despite their major socioeconomic impact there is still no satisfactory treatment. Their frequency is increasing with the lengthening of life expectancy, creating a major public health challenge for coming years. It is important to diagnose such diseases at an early stage and to develop new effective therapies. We are attempting to develop new therapeutic approaches in this context, keeping in mind that cartilage is one of the few human tissu

Evaluation of deoxygenated oligosaccharide acceptor analogs as specific inhibitors of glycosyltransferases.

The glycosyltransferases controlling the biosynthesis of cell-surface complex carbohydrates transfer glycosyl residues from sugar nucleotides to specific hydroxyl groups of acceptor oligosaccharides. These enzymes represent prime targets for the design of glycosylation inhibitors with the potential to specifically alter the structures of cell-surface glycoconjugates. With the aim of producing such inhibitors, synthetic oligosaccharide substrates were prepared for eight different glycosyltransferases. The enzymes investigated were: A, alpha(1----2, porcine submaxillary gland); B, alpha(1----3/4

Glycosyltransferase Activity Assay Using Colorimetric Methods.

The glycosyltransferases (GTs) are an important subclass of enzymes that catalyze the biosynthesis of glycosidic bonds in oligosaccharides, polysaccharides and glycoconjugates by transferring a sugar residue from a donor substrate to an acceptor substrate. The membrane-associated GTs play a vital role in the biosynthesis of bacterial cell-wall polysaccharides. Characterization and quantification of GT activities is important for studies of biosynthesis of polysaccharides, drug target development, and production of bacterial products. In this chapter, colorimetric assays for the measurement of

Mechanism of the effect of glycosyltransferase GLT8D2 on fatty liver.

BACKGROUND: Recent studies have shown that some glycosyltransferases are involved in the development of nonalcoholic fatty liver disease (NAFLD). The objective of this study was to explore the effect and mechanism of glycosyltransferase GLT8D2 on fatty liver. METHODS: Rat model of NAFLD was established by induction with high-fat-diet. The GLT8D2 expression in rat liver was examined using immunohistochemistry. Oil Red O staining and triglyceride assay were used to measure the effect of abnormal GLT8D2 expression on lipid accumulation in HepG2 cells. The expression levels of lipid metabolism-re

Creation and characterization of glycosyltransferase mutants of Trypanosoma brucei.

The survival strategies of protozoan parasites frequently involve the participation of glycoconjugates. Trypanosoma brucei expresses complex glycoproteins throughout its life cycle and a review of its repertoire of glycosidic linkages suggests a minimum of 38 glycosyltransferase activities. Here we describe a functional characterization workflow in which we create glycosyltransferase null or conditional null mutants in both the bloodstream and procyclic life-cycle forms of the parasite. Subsequently, we characterize the biochemical phenotype of the mutant strains generated and assign precise f

Glycosyltransferase activities in human meningiomas. Preliminary results.

The biosynthesis of a given glycosphingolipid is under the control of specific glycosyltransferases, while its catabolism is catalyzed by step-wise action of glycosidases. The net amount of glycolipids apparently result from the difference between these two processes. However, other parameters should be taken into consideration, such as intracellular recycling of catabolic products, membrane insertion, and membrane turnover. In order to establish a possible correlation between ganglioside expression in brain tumor and the activities of the enzymes involved in their metabolism, we analyzed the

Diversity of sugar-diphospholipid-utilizing glycosyltransferase families.

Peptidoglycan polymerases, enterobacterial common antigen polymerases, O-antigen ligases, and other bacterial polysaccharide polymerases (BP-Pols) are glycosyltransferases (GTs) that build bacterial surface polysaccharides. These integral membrane enzymes share the particularity of using diphospholipid-activated sugars and were previously missing in the carbohydrate-active enzymes database (CAZy; www.cazy.org ). While the first three classes formed well-defined families of similar proteins, the sequences of BP-Pols were so diverse that a single family could not be built. To address this, we de

Structural details of the glycosyltransferase step of peptidoglycan assembly.

The importance of peptidoglycan in forming the basis of the bacterial cell wall has led to many studies investigating its synthesis. The step of cross-linkage via transpeptidation, and its inhibition by penicillins, has been extremely well characterized yet knowledge of the preceding glycosyltransfer reaction remained elusive until recently. The structures of two glycosyltransferase enzymes, catalyzing membrane-based polymerization of the lipid II monomer unit, have presented a means of elucidating the molecular details of this highly desirable antibiotic target. Evidence acquired before the p
