Genetic Analysis of the Red Anther Trait Introgressed into Upland Cotton (Gossypium hirsutum) from Gossypium armourianum.

Upland cotton (Gossypium hirsutum, AD1) is the most important fiber crop for the world's textile industry. The genetic base of Upland cotton is narrow, and the introgression of germplasm from other species into Upland cotton can broaden its genetic diversity, including the introduction of new qualitative traits. The objectives of this study were to determine the inheritance, allelism, and linkage relationships of the red anther trait introduced into Upland from the red anther donor G. armourianum (D2-1) through crosses involving a synthetic tetraploid A1D2-1. After two generations of backcross

Rapid evolutionary divergence of diploid and allotetraploid Gossypium mitochondrial genomes.

BACKGROUND: Cotton (Gossypium spp.) is commonly grouped into eight diploid genomic groups and an allotetraploid genomic group, AD. The mitochondrial genomes supply new information to understand both the evolution process and the mechanism of cytoplasmic male sterility. Based on previously released mitochondrial genomes of G. hirsutum (AD1), G. barbadense (AD2), G. raimondii (D5) and G. arboreum (A2), together with data of six other mitochondrial genomes, to elucidate the evolution and diversity of mitochondrial genomes within Gossypium. RESULTS: Six Gossypium mitochondrial genomes, including

Genetic Diversity Analysis of 96 Gossypium hirsutum-Gossypium barbadense Introgression Lines and Early Maturing Northern China Cotton Lines Using a 40K Liquid-Phase Chip.

Background: Genetic diversity and genetic differentiation between Gossypium hirsutum-Gossypium barbadense introgression lines (ILs) and early-maturing upland cotton lines are critical for resolving the core breeding contradiction in Xinjiang cotton region: narrow genetic basis of early-maturing cultivars and late maturity of ILs with superior fiber quality. Xinjiang is one of the major cotton-producing regions in China, and breeding high-quality early-maturing upland cotton adapted to local ecological conditions is essential for improving cotton yield and quality. However, the genetic relation

MOLECULAR EVIDENCE FOR HOMOPLOID RETICULATE EVOLUTION AMONG AUSTRALIAN SPECIES OF GOSSYPIUM.

Interspecific hybridization and introgression are important evolutionary processes in plants, but their full significance with respect to speciation at the diploid level remains unresolved. In this study, molecular markers from the plastid and nuclear genomes were used to document an unusual evolutionary history of Gossypium bickii Prokh. (Malvaceae). This species is one of three morphologically similar Australian cottons (along with G. austrate F. Muell. and G. nelsonii Fryx.) in section Hibiscoidea. In contrast to expectations based on previous morphological data, cladistic analysis of mater

Genome-Wide Analysis of Multidrug and Toxic Compound Extrusion (MATE) Family in Gossypium raimondii and Gossypium arboreum and Its Expression Analysis Under Salt, Cadmium, and Drought Stress.

The extrusion of toxins and substances at a cellular level is a vital life process in plants under abiotic stress. The multidrug and toxic compound extrusion (MATE) gene family plays a large role in the exportation of toxins and other substrates. We carried out a genome-wide analysis of MATE gene families in Gossypium raimondii and Gossypium arboreum and assessed their expression levels under salt, cadmium and drought stresses. We identified 70 and 68 MATE genes in G. raimondii and G. arboreum, respectively. The majority of the genes were predicted to be localized within the plasma membrane, w

A modified protocol for rapid DNA isolation from cotton (Gossypium spp.).

Extraction of high-quality DNA from Gossypium (Cotton) species is notoriously difficult due to high contents of polysaccharides, quinones and polyphenols other secondary metabolites. Here, we describe a simple, rapid and modified procedure for high-quality DNA extraction from cotton, which is amenable for downstream analyses. In contrast to other CTAB methods, the described procedure is rapid, omits the use of liquid nitrogen, phenol, CsCl gradient ultracentrifugation, uses inexpensive and less hazardous reagents, and requires only ordinary laboratory equipment. The procedure employed the high

Comparative Proteomic Analysis of Gossypium thurberi in Response to Verticillium dahliae Inoculation.

Verticillium wilt is threatening cotton productivity globally. This disease is caused by soil-borne Verticillium dahliae which directly infects cotton roots, and exclusively colonizes and occludes xylem vessels, finally resulting in necrosis, defoliation, and most severely, plant death. For the first time, iTRAQ (isobaric tags for relative and absolute quantification) was applied to screen the differentially expressed proteins of Gossypium thurberi inoculated with V. dahliae. A total of 6533 proteins were identified from the roots of G. thurberi after inoculation with V. dahliae, and 396 showe

Use of meiotic FISH for identification of a new monosome in Gossypium hirsutum L.

The extensive use of molecular cytogenetics in human genetics and clinical diagnostics indicates that analogous applications in plants are highly feasible. One sort of application would be the identification of new aneuploids, which traditionally involves either direct karyotypic identification, which is feasible in only a few plant species, or tests with markers (cytogenetic, genetic, or molecular), which require sexual hybridization and at least one subsequent seed or plant generation. We have used meiotic fluorescence in situ hybridization (FISH) to analyze a new monosome of cotton (Gossypi

Genome-wide investigation and transcriptome analysis of the WRKY gene family in Gossypium.

WRKY transcription factors play important roles in various stress responses in diverse plant species. In cotton, this family has not been well studied, especially in relation to fiber development. Here, the genomes and transcriptomes of Gossypium raimondii and Gossypium arboreum were investigated to identify fiber development related WRKY genes. This represents the first comprehensive comparative study of WRKY transcription factors in both diploid A and D cotton species. In total, 112 G. raimondii and 109 G. arboreum WRKY genes were identified. No significant gene structure or domain alteratio

Morphology and metal binding characteristics of a natural polymer-kondagogu (Cochlospermum gossypium) gum.

Kondagogu (Cochlospermum gossypium) gum (KG), a natural tree exudate, was investigated for its morphological, adsorption and metal interaction behavior with various toxic heavy metals (Pb, Cd, Ni, Cr and Fe). SEM, AFM and TEM techniques were used to study the morphological changes occurring after metal adsorption onto the biopolymer structure. The degree of biosorption of metals on KG biopolymer surfaces was assessed by small-angle X-ray scattering analysis. EDXA spectrum revealed that the ion-exchange mechanism plays a major role in the binding process between KG and metal ions. The higher el

The conditions required to isolate and maintain viable cotton (Gossypium hirsutum L.) microspores.

Cultural systems developed for the successful production of haploid plants from anther and microspore culture of a number of species are lethal to cotton microspores. Components of these systems were examined individually and in simple combinations to determine their specific effect on cotton microspore viability during isolation and culture. An organically buffered pH of 7.0 was critical for survival. The addition of potassium salts, near standard concentrations used for other species, enhanced survival and the cytoplasmic appearance of isolated microspores. Severe toxicity resulted from ammo

Genomic characterization of a MAGIC population containing introgressions from four tetraploid Gossypium species.

One strategy for increasing the genetic diversity of upland cotton (Gossypium hirsutum) involves the introgression of beneficial alleles from related and wild tetraploid species. Progress in this area has been limited due to undesirable outcomes associated with whole genome interspecific hybridization. To minimize genetic influences from other chromosomes, chromosome substitution lines (CSLs) were developed in upland cotton TM-1 using Gossypium barbadense (GB), Gossypium mustelinum (GM), and Gossypium tomentosum (GT) as donor species. A multiparent advanced generation intercross (MAGIC) popula

AmCBF1 Transcription Factor Regulates Plant Architecture by Repressing GhPP2C1 or GhPP2C2 in Gossypium hirsutum.

Dwarfism is a beneficial trait in many crops. Dwarf crops hold certain advantages over taller crops in lodging resistance, fertilizer tolerance, and yield. Overexpression of CBF/DREB transcription factors can lead to dwarfing in many plant species, but the molecular mechanism of plant dwarfing caused by overexpression of CBF/DREB in upland cotton (Gossypium hirsutum) remains unclear. In this study, we observed that overexpression of the Ammopiptanthus mongolicus AmCBF1 transcription factor in upland cotton R15 reduced plant height, whereas virus-induced gene silencing of AmCBF1 in the derived

[Cloning and functional analysis of chitinase gene GbCHI from sea-island cotton (Gossypium barbadense)].

Chitinase is one of the important pathogenesis-related (PR) proteins in plants. By comparative proteomics study, a novel pathogen-responsive chitinase (known as GbCHI) has been identified from sea-island cotton (Gossypium barbadense). The GbCHI cDNA was cloned from wilt-resistant sea-island cotton and the anti-fungal activity of the gene product was investigated. qRT-PCR analysis indicated that GbCHI was expressed constitutively in root, stem, leaf, flower, and ovule of cotton plant, and the expression could be induced by Verticillium dahliae and plant hormone SA, ACC, and JA. A DNA Extraction Method for Nondestructive Testing and Evaluation of Cotton Seeds (Gossypium L.).

Kernels of cotton provide lint and linter for textiles, oil and protein for food and feed. Cotton seed is formed following fertilization between an ovule and a pollen grain. The seed coat is maternal in origin, whereas the embryo and attached cotyledonary leaves are hybrids of parental lines. The extraction of genomic DNA from an ungerminated whole, a portion or mixed seeds are prerequisite in genetic and genomic studies of cotton. As far as our knowledge, there is only one method of nondescriptive DNA extraction from ungerminated cotton seeds without affecting the seed germination capability,

Soil microbial activity is affected by Roundup WeatherMax and pesticides applied to cotton (Gossypium hirsutum).

Adoption of glyphosate-based weed control systems has led to increased use of the herbicide with continued use of additional pesticides. Combinations of pesticides may affect soil microbial activity differently than pesticides applied alone. Research was conducted to evaluate the influence of glyphosate-based cotton pest management systems on soil microbial activity. Soil was treated with commercial formulations of trifluralin, aldicarb, and mefenoxam + pentachloronitrobenzene (PCNB) with or without glyphosate (applied as Roundup WeatherMax). The soil microbial activity was measured by quantif

Genotyping by Sequencing Revealed QTL Hotspots for Trichome-Based Plant Defense in Gossypium hirsutum.

Cotton possesses certain physical features, including leaf and stem trichomes that help plants deter damage caused by insect pests, and to some extent, from abiotic factors as well. Among those features, trichomes (pubescence) hold a special place as a first line of defense and a managemental tool against sucking insect pests of cotton. Different insect pests of cotton (whiteflies, aphids, jassids, and boll weevil) severely damage the yield and quality of the crop. Likewise, whiteflies, aphids, jassids, and other insect pests are considered as potential carriers for cotton leaf curl viruses an

Effects of GhWUS from upland cotton (Gossypium hirsutum L.) on somatic embryogenesis and shoot regeneration.

The WUSCHEL (WUS) gene encodes a plant-specific homeodomain-containing transcriptional regulator, which plays important roles during embryogenesis, as well as in the formation of shoot and flower meristems. Here, we isolated two homologues of Arabidopsis thaliana WUS (AtWUS), GhWUS1a_At and GhWUS1b_At, from upland cotton (Gossypium hirsutum). Domain analysis suggested that the two putative GhWUS proteins contained a highly conserved DNA-binding HOX domain and a WUS-box. Expression profile analysis showed that GhWUSs were predominantly expressed during the embryoid stage. Ectopic expression of

Transcriptome profiling of Gossypium barbadense inoculated with Verticillium dahliae provides a resource for cotton improvement.

BACKGROUND: Verticillium wilt, caused by the fungal pathogen Verticillium dahliae, is the most severe disease in cotton (Gossypium spp.), causing great lint losses worldwide. Disease management could be achieved in the field if genetically improved, resistant plants were used. However, the interaction between V. dahliae and cotton is a complicated process, and its molecular mechanism remains obscure. To understand better the defense response to this pathogen as a means for obtaining more tolerant cultivars, we monitored the transcriptome profiles of roots from resistant plants of G. barbadense
