Sulfated polysaccharides (SPs) are abundant in seaweed and have several industrial and biomedical applications, but their renal effects remain unclear. This study evaluated the effects of total sulfated polysaccharides (TSPs) from Gracilaria cornea using an isolated rat kidney perfusion model. TSP at 3 µg/mL increased perfusion pressure and renal vascular resistance at 90-120 min, while 4.5 µg/mL induced earlier and more pronounced changes (from 60 min). Urinary flow decreased at 1 µg/mL (90 min) but increased at 4.5 µg/mL (90-120 min). Sodium transport was reduced at all concentrations, where

Malaria infections are increasingly resisting drug therapy, indicating the need to address and provide more affordable alternative raw materials for medicines to overcome parasite resistance. In this context, seaweeds are one of the potential maritime resources that can be explored. Therefore, this study aimed to gain an overview of the Gracilaria sp. ethanol extract and its constituents in suppressing Plasmodium parasites in vitro. Seaweed was dried, powdered, and underwent ethanol extraction, then screening for compound content was achieved with a phytochemical method, infrared spectroscopy,

Enormous marine biodiversity offers an endless reservoir of chemicals for many applications. In this scenario, the extraction of seaweeds represents an interesting source of compounds displaying antimicrobial activity. In particular, among the different red algae, Gracilaria gracilis plays an important role due to the presence of important bioactives in its composition. In spite of these features, an efficient culture system is still absent. In the present study, a novel algal culture method was developed and compared to another more common cultural practice, widely reported in literature. A h

Bone remodeling, a physiological process characterized by bone formation by osteoblasts and bone resorption by osteoclasts, is important for the maintenance of healthy bone in adult humans. Osteoclasts play a critical role in bone erosion and osteoporosis and are bone-specific multinucleated cells generated through differentiation of monocyte/macrophage lineage precursors. Receptor activator of NF-κB ligand (RANKL) has been reported to induce osteoclast differentiation. In this study, we explored whether Gracilaria verrucosa extracts (GE) could affect RANKL-mediated osteoclast differentiation.

The invasive success of Gracilaria vermiculophylla has been attributed to its wide tolerance range to different abiotic factors, but its response to ultraviolet radiation (UVR) is yet to be investigated. In the laboratory, carpospores and vegetative thalli of an Atlantic population were exposed to different radiation treatments consisting of high PAR (photosynthetically active radiation) only (P), PAR+UV-A (PA) and PAR+UV-A+UV-B (PAB). Photosynthesis of carpospores was photoinhibited under different radiation treatments but photosystem II (PSII) function was restored after 12 h under dim white

The red seaweed Gracilaria birdiae (GB) is farmed and used as food in northeast Brazil. However, the economic potential of this seaweed has been explored little. To enable direct consumption and/or product diversification from GB, it is necessary to evaluate its effect in vivo. In this study, the food of mice was improved with the addition of GB. After 21 days, the consumption of seaweed reduced the weight gain and blood glucose levels in mice. In addition, it increased the trolox equivalent antioxidant capacity and glutathione reductase and catalase levels compared to those of the control gro

Gracilaria bailinae polysaccharides have limited applications due to their high viscosity, large molecular weight, and poor solubility. Microbial fermentation emerges as an efficient strategy to enhance the bioactivity of plant polysaccharides. This study aimed to prepare fermented water-soluble crude polysaccharide (GBV) from G. bailinae via fermentation with Bacillus velezensis, using water-extracted polysaccharide (GBP) as a control. The physicochemical properties, immunomodulatory activity, and trained immunity effects of GBV were systematically investigated. A significant decrease in mole

Marine macroalga are frequently exposed to environmental stresses impairing their overall physiology and growth potential. Among these, Gracilaria cornea (Rhodophyta) is a valuable red seaweed rich in protein and polysaccharides. To investigate its physiological responses under controlled conditions, we cultivated Gracilaria cornea in an indoor culture system at three different salinity levels (30, 40 and 50 ppt), employing continuous aeration, blue and white LED illumination (12:12 light: dark cycle), and exogenous addition of nitrogen and phosphorus. Physiological changes associated with pro

ETHNOPHARMACOLOGICAL RELEVANCE: The use of marine seaweeds as a source of natural compounds with medicinal purposes is increasing in Western countries in the last decades, becoming an important alternative in the traditional medicine of many developing countries, where diarrhea still remains a severe public health problem, with high rates of mortality and morbidity. Sulfated polysaccharides (PLS) extracted from red seaweeds can exhibit therapeutic effects for the treatment of gastrointestinal disorders. Thus, the pharmacological properties of the PLS from Gracilaria cervicornis, an endemic sea

A quantitative NMR assay was utilized to identify methylglyoxal-scavenging natural products from Rhodophytes, Chondrus crispus and Gracilaria vermiculophylla. This revealed the activity of guanylurea-containing amino acid derivatives, gongrine and gigartinine. The molecules share structural features with the frontline blood glucose-lowering drug and plant natural product derivative, metformin, and scavenge methylglyoxal via the same mechanism, resulting in an imidazole-containing Advanced Glycation Endproduct or AGE. The protective effect of the molecules reported here was evaluated in a cell-

Food allergy has a significant impact on the health and well-being of individuals, affecting both their physical and mental states. Research on natural bioactive compounds, such as polysaccharides extracted from seaweeds, holds great promise in the treatment of food allergies. In this study, fermented Gracilaria lemaneiformis polysaccharides (F-GLSP) were prepared using probiotic fermentation. Probiotic fermentation of Gracilaria lemaneiformis reduces the particle size of polysaccharides. To compare the anti-allergic activity of F-GLSP with unfermented Gracilaria lemaneiformis polysaccharides

Diarrhoea is a significant health problem for children in developing countries that causes more than 1 million deaths annually. This study aimed to evaluate the antidiarrhoeal effect of sulphated polysaccharide (PLS) from the alga Gracilaria caudata in rodents. For the evaluation, acute diarrhoea was induced in Wistar rats (150-200 g) by administration of castor oil (10 mg/kg). Then, different parameters, including enteropooling and gastrointestinal transit and its pharmacological modulation by opioid and cholinergic pathways, were assessed using activated charcoal in Swiss Mice (25-30 g). Marine algae are valuable sources of bioactive compounds that have the potential to be used in the management of various pathologies. Despite the increasing prevalence of NAFLD, the absence of an approved effective pharmacological treatment with demonstrable effectiveness persists. In this context, the aim of the present study is to assess the effect of Gracilaria vermiculophylla red seaweed dietary supplementation on hepatic lipid accumulation, as well as on oxidative stress, inflammation and fibrosis- related markers on obese fa/fa Zucker rats fed with a standard diet, supplemented or not wi

Selenium (Se) is a beneficial element for plants, and can be used to mitigate the toxicity of heavy metals. However, the detoxification of Se in macroalgae, a crucial part of aquatic ecosystem productivity, has rarely been reported. In the present study, a red macroalga Gracilaria lemaneiformis was exposed to non-essential metal cadmium (Cd) or essential metal copper (Cu) with addition of different levels of Se. We then examined the changes in growth rate, metal accumulation, metal uptake rate, subcellular distribution, as well as thiol compound induction in this alga. Se addition alleviated C

The health effects of polysaccharides have attracted lots of attention, but the exact mechanism remains unclear. This study indicated that polysaccharides from Gracilaria lemaneiformis (GLPs) tolerated the conditions of mouth, stomach, and small intestine, and it reached the colon integrally, where it increased the production of short chain fatty acids, altered the gut microbiota, and especially increased the level of Bacteroides. To explore the underlying mechanism, hundreds of Bacteroides strains were isolated from the human feces and identified by MALDI-TOF/MS. It showed that Bacteroides sp

This study aimed to develop and evaluate a liposomal nano-delivery system for bioactive peptide fractions (PFs) derived from Gracilaria corticata algae proteins. The peptides were obtained by enzymatic hydrolysis using alcalase, pancreatin, and trypsin. The resulting PF-loaded nanoliposomes were characterized in terms of degree of hydrolysis (DH), particle size, ζ-potential, encapsulation efficiency (EE), and antioxidant capacity (DPPH and ABTS assays). Among the tested enzymes, alcalase produced hydrolysates with the highest DH (33%) and EE (84%). The particle size of nanoliposomes ranged fro
