[Gram-negative bacteria folliculitis].

Gram-negative folliculitis was first described in 1968 and had since given rise to numerous publications. To our knowledge, no case has yet been published in France, although we observed 6 of them in 12 months. Between March, 1985 and March, 1986, samples of pus for bacteriological examination were obtained from all our patients with acnea vulgaris resistant to the standard treatment (i.e. oral tetracycline combined with topical applications of tretinoin or benzoyl peroxide), thus enabling us to detect patients who had a Gram-negative bacillus (GNB) in at least one pustule. Inflammatory and pa

[Carbapenemases of gram-negative bacteria].

Carbapenems are often antibiotics of last resort for the treatment of severe infections. They are stable to most beta-lactamases produced by gram-negative bacteria. However, bacterial enzymes named carbapenemases can efficiently hydrolyze carbapenems. They are produced most frequently by Enterobacteriaceae and non-fermentative bacteria such as Pseudomonas aeruginosa and Acinetobacter baumannnii. They belong to group A (KPC, SME, IMI, NMC), B (VIM, IMP, SPM, GIM, NDM, SIM, DIM, AIM) and D (OXA-23, OXA-24, OXA-48, OXA-58, OXA-143). Non-fermentative Gram-negative bacteria.

Over the past decade, non-fermenting Gram-negative bacteria have emerged as important opportunistic pathogens in the increasing population of patients who are immunocompromised by their disease or medical treatment. These bacteria are assisted by their ubiquitous distribution in the environment and have a propensity for multiple, intrinsic or acquired drug resistance. The infections that they cause now pose significant problems in terms of treatment and infection control, whilst the commonly observed rapid emergence of bacterial resistance to new antimicrobial compounds raises concerns regardi

Resistance mechanisms in Gram-negative bacteria.

Enterobacterales resistant to carbapenems or producing extended-spectrum β-lactamases (ESBL) and non-fermenters resistant to carbapenems present resistance to many of the antimicrobials commonly used in clinical practice, and have been recognized by the World Health Organization as a critical priority for the development of new antimicrobials. In this review, the main mechanisms of resistance of Enterobacterales, Pseudomonas aeruginosa, Acinetobacter baumannii and Stenotrophomonas maltophilia to β-lactams, quinolones, aminoglycosides and polymyxins will be addressed. Updated information will b

Resistance in gram-negative bacteria: enterobacteriaceae.

The emergence and spread of resistance in Enterobacteriaceae are complicating the treatment of serious nosocomial infections and threatening to create species resistant to all currently available agents. Approximately 20% of Klebsiella pneumoniae infections and 31% of Enterobacter spp infections in intensive care units in the United States now involve strains not susceptible to third-generation cephalosporins. Such resistance in K pneumoniae to third-generation cephalosporins is typically caused by the acquisition of plasmids containing genes that encode for extended-spectrum beta-lactamases (

Gram-negative bacteria and B27 disease.

The role of infection in ankylosing spondylitis is speculative and different studies by several groups have yielded conflicting results. The role of infection, however, in reactive arthritis and Reiter's syndrome is well established. The Grampian region has experienced three outbreaks of gastroenteritis, two due to Campylobacter jejuni and one due to Salmonella typhimurium. These outbreaks have allowed a study of reactive arthritis in the affected population following these infections. These studies do not allow accurate estimate of the frequency of reactive arthritis, largely because of diffi

Corneal rings with gram-negative bacteria.

Corneal rings have been described with corneal ulcerations caused by Gram-negative bacteria. Corneal rings were produced by intracorneal injections of viable Pseudomonas aeruginosa as well as heat-inactivated suspensions of Gram-negative bacteria (P aeruginosa and Escherichia coli) but not Gram-positive bacteria (Staphylococcus aureus) or Freund's adjuvant. It is suggested that endotoxin is the factor responsible for their production since purified endotoxin produced corneal rings after intracorneal injection. Histopathological examination of the areas corresponding to the corneal rings disclo

Isolating DNA from Gram-Negative Bacteria.

The isolation of DNA from bacteria, described in this protocol, relies upon the use of sodium dodecyl sulfate and proteinase K to lyse the cells. High-molecular-weight DNA is then sheared (to reduce its viscosity and make it more manageable), extracted with phenol:chloroform, and precipitated with isopropanol. DNA isolated according to this procedure ranges from 30 to 80 kb in length.

Increasing threat of Gram-negative bacteria.

The widespread use of broad-spectrum antibiotics has led to emergence of antibiotic-resistant strains of many Gram-negative organisms. This problem is particularly serious in critically ill patients, especially those with ventilator-associated pneumonia. Extensive antibiotic resistance has developed in Gram-negative bacteria, due both to innate resistance in some species and the fact that they are highly adept at acquiring antibiotic-resistant determinants from each other. Antibiotic resistance develops through the following three basic mechanisms: alteration of the drug target, prevention of

Impermeability to quinolones in gram-positive and gram-negative bacteria.

The initial step in the accumulation of fluoroquinolone antimicrobial agents is binding to cell surface components reduced by lowered pH and divalent cations. Uptake into gram-negative and gram-positive bacteria is by simple diffusion. Entry through the outer membrane occurs preferentially for most agents by the porin route but a second process using the self-promoted uptake pathway is active especially for more hydrophobic agents. Fluoroquinolones bind to vesicles of phospholipid which may be the initiating step in cross-cytoplasmic membrane diffusion. An active efflux system has been describ

[The rise of resistant gram-negative bacteria].

During the past few years there has been a global spread of resistant gram-negative bacteria that are insensitive to cephalosporins and carbapenems. Extended spectrum beta-lactamase (ESBL)-producing bacteria are capable of inactivating the newest generation of cephalosporins. It is notable that ESBL-producing bacteria are found predominantly outside the hospital situation in the environment, in food and in meat products, which leads to the presumption that the food chain is contributing to the rapid spread of these bacteria. Several types of carbapenemase-producing bacteria have been distingui

[Gram-negative bacteriae with resistance to carbapenems].

Clinically-significant Gram-negative species remain mostly Enterobacteriaceae, Pseudomonas aeruginosa and Acinetobacter baumannii. Carbapenem molecules are often the last resort for treating infections due to multidrug resistant isolates. In Enterobacteriaceae, resistance to carbapenems may result from combined mechanisms of resistance associating b-lactamases with weak (if any) intrinsic carbapenemase activity and decreased outer membrane permeability, or from true carbapenemases. KPC-type enzymes (partially inhibited by clavulanic acid) have been identified mostly in Klebsiella pneumoniae, f

Alternative drugs against multiresistant Gram-negative bacteria.

OBJECTIVES: Enterobacterales and other non-fermenting Gram-negative bacteria have become a threat worldwide owing to the frequency of multidrug resistance in these pathogens. On the other hand, efficacious therapeutic options are quickly diminishing. The aims of this study were to describe the susceptibility of 50 multiresistant Gram-negative bacteria, mostly pan-resistant, against old and less-used antimicrobial drugs and to investigate the presence of antimicrobial resistance genes. METHODS: A total of 50 genetically distinct isolates were included in this study, including 14 Acinetobacter

A Ratiometric pH Sensor for Gram-Positive and Gram-Negative Bacteria.

Fluctuating environments can lead to phenotypic heterogeneity within a monoclonal bacterial population, especially in response to antibiotics or the human immune system. Methods are required to analyze the physiology of single cells to understand how individual cells interact with their environment and adapt to pH stress. We report a ratiometric, fluorescent probe to sense cytoplasmic pH in bacteria. Our probes are based on hemicyanine dyes and are taken up into both Gram-positive and Gram-negative bacteria. The probes are selective under a broad range of biologically relevant conditions. The

Mechanism of chlorpromazine binding by gram-positive and gram-negative bacteria.

Chlorpromazine forms charge-transfer complexes with xanthene dyes in bacteria. These complexes permit the differentiation of Gram-positive and Gram-negative bacteria in both light and polarization microscopy. The birefringence induced by the charge-transfer complex might explain the molecular basis of bacterial staining. The charge-transfer complexes formed between chlorpromazine and xanthene dyes accumulate in the bacterial cell, mainly inside the bacterial cell wall. The complexes give the cells a color, which depends on the chemical composition of the staining structure, and in particular t

Activity of Specialized Biomolecules against Gram-Positive and Gram-Negative Bacteria.

The increased resistance of bacteria against conventional pharmaceutical solutions, the antibiotics, has raised serious health concerns. This has stimulated interest in the development of bio-based therapeutics with limited resistance, namely, essential oils (EOs) or antimicrobial peptides (AMPs). This study envisaged the evaluation of the antimicrobial efficacy of selected biomolecules, namely LL37, pexiganan, tea tree oil (TTO), cinnamon leaf oil (CLO) and niaouli oil (NO), against four bacteria commonly associated to nosocomial infections: Staphylococcus aureus, Staphylococcus epidermidis,

Rabbit CAP18 derived peptides inhibit gram negative and gram positive bacteria.

CAP18 (cationic antimicrobial protein of 18 kDa) was originally isolated from rabbit granulocytes using as an assay the agglutination of Re-lipopolysaccharide coated erythrocytes. The C-terminal 37 amino acids of CAP18 comprise the LPS-binding domain called RNIP, reactive nitrogen inhibitory peptide. Synthetic RNIP has broad antimicrobial activity versus both gram positive [IC50 = 130-200 nM] and gram negative bacteria [IC50 = 20-100 nM). Susceptible strains include Staphylococcus aureus, Klebsiella pneumoniae, Streptococcus pneumoniae, Escherichia coli, Pseudomonas aeruginosa, and Streptococc
