Proteomic Characterization of Spodoptera frugiperda granulovirus Occlusion Bodies.

Lepidopteran-infecting baculoviruses represent one of the most promising alternatives for pest biocontrol. In particular, Spodoptera frugiperda granulovirus (SfGV) infects the fall armyworm Spodoptera frugiperda, an increasingly important pest for maize and other crops around the world. SfGV has been investigated due to its capacity to enhance other baculovirus infectivity and as a biopesticide itself. Although the proteome of SfGV occlusion bodies (OB) is fundamental for their infectivity properties, we still lack a detailed understanding of its protein components and how they compare with ot

The Cryptophlebia leucotreta granulovirus-10 years of commercial field use.

In the last 15 years, extensive work on the Cryptophlebia leucotreta granulovirus (CrleGV) has been conducted in South Africa, initially in the laboratory, but subsequently also in the field. This culminated in the registration of the first CrleGV-based biopesticide in 2004 (hence, the 10 years of commercial use in the field) and the second one three years later. Since 2000, more than 50 field trials have been conducted with CrleGV against the false codling moth, Thaumatotibia leucotreta, on citrus in South Africa. In a representative sample of 13 field trials reported over this period, effica

Selection for and Analysis of UV-Resistant Cryptophlebia Leucotreta Granulovirus-SA as a Biopesticide for Thaumatotibia leucotreta.

Cryptophlebia leucotreta granulovirus-SA (CrleGV-SA) is used as a commercial biopesticide for the false codling moth, Thaumatotibia leucotreta, in citrus and other crops. The virus is sensitive to UV irradiation from sunlight, which reduces its efficacy as a biopesticide in the field. We selected a UV-resistant CrleGV-SA isolate, with more than a thousand-fold improved virulence compared to the wild-type isolate, measured by comparing LC50 values. CrleGV-SA purified from infected T. leucotreta larvae was exposed to UV irradiation under controlled laboratory conditions in a climate chamber mimi

Genomic Structure Predicts Granulovirus Resistance Without a Detectable Life-History Trade-Off in Plodia interpunctella.

Understanding the genetic architecture of pathogen resistance and its relationship to host life-history traits is a fundamental problem in evolutionary biology, yet the conditions under which resistance costs manifest as detectable trade-offs remain poorly understood. Here, we examine resistance in Plodia interpunctella (Indian meal moth) to its granulovirus (PiGV) across 12 inbred lines maintained without deliberate pathogen exposure, alongside an independently assembled outbred stock population as a genomic reference. We quantify resistance using LD50 values from dose-response assays, summar

Evaluation of spray-dried lignin-based formulations and adjuvants as solar protectants for the granulovirus of the codling moth, Cydia pomonella (L).

Commercial formulations of the codling moth, Cydia pomonella L., granulovirus (CpGV) are limited by their short residual activity under orchard conditions in the Pacific Northwest. We evaluated spray-dried lignin-encapsulated formulations of CpGV for improved solar stability based on laboratory bioassays with a solar simulator and in field tests in an infested apple orchard. In laboratory tests, aqueous lignin formulations containing a high dosage of 3 x 10(10) occlusion bodies (OB)/L, with and without the additives titanium dioxide (TiO(2)) and sugar, provided significant solar protection of

Genome sequence analysis of baculoviruses from the noctuid moths Eudryas unio and Autographa gamma indicates that they are isolates of Betabaculovirus xecnigri, a species of slow-killing granuloviruses.

Complete genome sequences were determined for three uncharacterized betabaculoviruses (Baculoviridae: Lefavirales) from noctuid moth species. These viruses included Eudryas unio granulovirus 1112 (EuunGV-1112), Eudryas unio granulovirus 1229 (EuunGV-1229), and Autographa gamma granulovirus Darmstadt (AugaGV-Darmstadt). Analysis of the features and open reading frame (ORF) contents of the genome sequences indicated that all three viruses were closely related to Helicoverpa armigera granulovirus (HearGV; Betabaculovirus helarmigerae) and Xestia c-nigrum granulovirus (XecnGV; Betabaculovirus xecn

Genome sequence determination and analysis of a betabaculovirus from the moth Glena bisulca, a defoliating pest of pines.

A baculovirus isolated from the moth Glena bisulca was characterized by electron microscopy of its occlusion bodies (OBs) and sequencing of its genome. The OBs of this virus, Glena bisulca granulovirus isolate 1002 (GlbiGV-1002), were ovocylindrical with a single virion, as described for other betabaculoviruses. The 104,702 bp circular genome contained six direct repeat (dr) regions and 121 open reading frames. A core gene phylogeny placed GlbiGV-1002 in betabaculovirus clade b with Cnaphalocrocis medinalis granulovirus (CnmeGV) as the nearest relative. Kimura-2-parameter nucleotide distance e

Metagenomic Analysis of Viromes of Aedes Mosquitoes across India.

Metagenomic analysis of Aedes aegypti and Ae. albopictus mosquitoes from diverse geographical regions of India revealed the presence of several insect viruses of human interest. Most abundant reads found in Ae. aegypti mosquitoes were of Phasi Charoen-like virus (PCLV), Choristoneura fumiferana granulovirus (CfGV), Cell fusing agent virus (CFAV), and Wenzhou sobemo-like virus 4 (WSLV4), whereas WSLV4 and CfGV constituted the highest percentage of reads in Ae. albopictus viromes. Other reads that were of low percentage included Hubei mosquito virus 2 (HMV2), Porcine astrovirus 4 (PAstV4), and W

Rodent Gut Bacteria Coexisting with an Insect Gut Virus in Tapeworm Parasitic Cysts: Metagenomic Evidence of Microbial Selection in Extra-Intestinal Clinical Niches.

In medicine, parasitic cysts (e.g., brain cysticerci) are believed to be sterile, and are primarily treated with antiparasitic medications, not antibiotics, which could prevent abscess formation and localized inflammation. This study quantified the microbial composition of parasitic cysts in a wild rodent, using multi-kingdom metagenomics to comprehensively assess if parasitic cysts are sterile, and further understand gut microbial translocation and adaptation in wildlife confined environments, outside the gut. Analysis was conducted on DNA from two hepatic parasitic cysts from a feline tapewo

Analysis of Coinfection Pathogens From Foot-and-Mouth Disease Virus Persistently Infected Cattle Using Oxford Nanopore Sequencing.

The persistent infection caused by foot-and-mouth disease virus (FMDV) still lacks a reliable explanation, as its etiology and maintenance are intricate and potentially involve concurrent infections with multiple pathogens. In this study, we utilized the nanopore platform for direct sequencing of clinical samples obtained from cattle persistently infected with FMDV serotype O and investigated the distribution characteristics of coinfecting pathogens in their pharyngeal region. Briefly, we exploited Oxford Nanopore sequencing technology to generate high-quality and sufficient sequence data for

Characterization of a newly established potato tuber moth (Phthorimaea operculella Zeller) cell line.

BACKGROUND & OBJECTIVE: Potato tuber moth (PTM), Phthorimaea operculella Zeller is a widely distributed, devastating pest of potatoes attacking the foliage and infest the tubers in both field and store causing serious economic damage. As application of PTM granulovirus (PTM-GV) has shown significant reduction in damage, attempts were made to develop a new cell line from this insect to grow PTM-GV for use as a biopesticide. METHODS: Approximately 100 mg of insect eggs were collected, surface sterilized and crushed gently in a boiling tube aseptically. The tissues were washed with physiological

Bionomic responses of Spodoptera frugiperda (J. E. Smith) to lethal and sublethal concentrations of selected insecticides.

Since 2016, the invasive insect Spodoptera frugiperda (J. E. Smith) (Lepidoptera: Noctuidae) from the Americas has made maize production unattainable without pesticides in parts of Sub-Saharan Africa and Asia. To counteract this pest, farmers often resort to the use hazardous pesticides. This study aimed to investigate botanicals, microbials, and semi-synthetic insecticides in Ghana for pest control without harming local ecosystems. Under laboratory and on-station conditions, the present study evaluated the acute and sublethal responses of S. frugiperda to: (i) Pieris rapae Granulovirus (PrGV)

Comparative genome sequence analysis of Choristoneura occidentalis Freeman and C. rosaceana Harris (Lepidoptera: Tortricidae) alphabaculoviruses.

The complete genome sequences of Choristoneura occidentalis and C. rosaceana nucleopolyhedroviruses (ChocNPV and ChroNPV, respectively) (Baculoviridae: Alphabaculovirus) were determined and compared with each other and with those of other baculoviruses, including the genome of the closely related C. fumiferana NPV (CfMNPV). The ChocNPV genome was 128,446 bp in length (1147 bp smaller than that of CfMNPV), had a G+C content of 50.1%, and contained 148 open reading frames (ORFs). In comparison, the ChroNPV genome was 129,052 bp in length, had a G+C content of 48.6% and contained 149 ORFs. ChocNP

Rodent Gut Bacteria Coexisting with an Insect Gut Virus in Parasitic Cysts: Metagenomic Evidence of Microbial Translocation and Co-adaptation in Spatially-Confined Niches.

In medicine, parasitic cysts or cysticerci (fluid-filled cysts, larval stage of tapeworms) are believed to be sterile (no bacteria), and therein, the treatment of cysticerci infestations of deep extra-intestinal tissues (e.g., brain) relies almost exclusively on the use of antiparasitic medications, and rarely antibiotics. To date, however, it is unclear why common post-treatment complications include abscessation. This study quantified the microbial composition of parasitic cyst contents in a higher-order rodent host, using multi-kingdom shotgun metagenomics, to improve our understanding of g

Conserved HA-peptides expressed along with flagellin in Trichoplusia ni larvae protects chicken against intranasal H7N1 HPAIV challenge.

The immunization of poultry where H5 and H7 influenza viruses (IVs) are endemic is one of the strategies to prevent unexpected zoonoses. Our group has been focused on conserved HA-epitopes as potential vaccine candidates to obtain multivalent immune responses against distinct IV subtypes. In this study, two conserved epitopes (NG-34 and CS-17) fused to flagellin were produced in a Baculovirus platform based on Trichoplusia ni larvae as living biofactories. Soluble extracts obtained from larvae expressing "flagellin-NG34/CS17 antigen" were used to immunize chickens and the efficacy of the vacci
