For more than a decade it has been assumed that there is a strong relationship between Ca2+ and gravitropism. There is evidence to suggest that the movement of Ca2+ in the wall might regulate extension growth and that free intracellular Ca2+ might mediate signalling in statocytes. However, it is unlikely that changes in either wall Ca2+ or the concentration of free intracellular calcium, [Ca2+]i, act independently of each other. This results in ambiguity in both experimentation and interpretation of experimental information. It is also uncertain that the observed redistributions of wall Ca2+ a

In higher plants, gravitropism proceeds through three sequential steps in the responding organs: perception of gravity signals, signal transduction and asymmetric cell elongation. Light and temperature also influence the gravitropic orientation of plant organs. A series of Arabidopsis shoot gravitropism (sgr) mutants has been shown to exhibit disturbed shoot gravitropism. SGR5 is functionally distinct from other SGR members in that it mediates the early events of gravitropic responses in inflorescence stems. Here, we demonstrated that SGR5 alternative splicing produces two protein variants (SG

A number of features of the gravitropic response of roots are not readily accounted for by the classical Cholodny-Went theory. These include the observations that (i) in the later stages of the response the growth gradient is reversed with no evident reversal of the auxin gradient; (ii) a major component of the acceleration of growth along the upper side occurs in the distal elongation zone (DEZ), a group of cells located between the meristem and the main elongation, not within the central elongation zone; and (iii) the initiation of differential growth in the DEZ appears to be independent of

Gravitropism is a plant response to gravity that directs organ growth and development, playing a key role in the adaptation of land plants. While its molecular basis has been extensively studied in flowering plants, much less is known about this process in other plant lineages. Here, we investigated the gravitropic response of the liverwort Marchantia polymorpha, a model for early land plant evolution. In darkness, the thallus tips extended upward, forming several straight, narrow structures whose growth direction was consistently opposite to gravity and disrupted by clinostat treatment. These

Polyethersulfone (PES) membranes provide a versatile tool for gravity-related plant studies. Benefits of this system include straightforward setup, no need for specialized equipment, long-term seed viability between plating and hydration/growth, and adaptability to diverse protocols and downstream analyses. Methods outlined here include seed sterilization, planting, growth, and dissection that will transition directly into any RNA extraction protocol.

Phytochromes are a family of photoreceptor molecules, absorbing primarily in red and far-red, that are important in many aspects of plant development. These studies investigated the role of phytochromes in phototropism and gravitropism of seedlings of Arabidopsis thaliana. We used two transgenic lines, one which lacked phytochromes specifically in the roots (M0062/UASBVR) and the other lacked phytochromes in the shoots (CAB3::pBVR). These transgenic plants are deficient in the phytochrome chromophore in specific tissues due the expression of biliverdin IXa reductase (BVR), which binds to precu

Gravitropism is a fundamental adaptive response in plants that enables directional growth to optimize resource acquisition. In this study, we employed forward genetic screening to identify Arabidopsis mutants with defective hypocotyl gravitropism and isolated the short and agravitropic hypocotyl in dark1 (sad1) mutant, which carries a point mutation (G110E) in the SAC1 gene encoding a phosphoinositide phosphatase. Deficiency of SAC1 disrupted gravity-induced polar localization of PIN3 in endodermal cells, impairing auxin redistribution and leading to hypocotyl gravitropism defects. Subcellular

Calmodulin, a primary plant calcium receptor, is known to be intimately involved with gravitropic sensing and transduction. Using the calmodulin-binding inhibitors trifluoperazine, W7 and calmidazolium, gravitropic curvature of Arabidopsis thaliana (L.) Heynh, ecotype Landsberg, roots was separable into two phases. Phase I was detected at very low concentrations (0.01 microM) of trifluoperazine and calmidazolium, did not involve growth changes, accounted for about half the total curvature of the root and may represent the specific contribution of the cap to gravity sensing. Phase II commenced

BACKGROUND: In plants, gravity directs bidirectional growth; it specifies upward growth of shoots and downward growth of roots. Due to gravity, roots establish robust anchorage and shoot, which enables to photosynthesize. It sets optimum posture and develops plant architecture to efficiently use resources like water, nutrients, CO2, and gaseous exchange. Hence, gravitropism is crucial for crop productivity as well as for the growth of plants in challenging climate. Some SGR members are known to affect tiller and shoot angle, organ size, and inflorescence stem in plants.

Root gravitropic bending is a complex growth process resulting from differential expansion of cells on the upper and lower sides of a gravistimulated root. In order to genetically dissect the molecular machinery underlying root bending, a thorough understanding of the kinetics and spatial distribution of the growth process is required. We have developed an experimental workflow that enables us to image growing roots at high spatiotemporal resolution and then convert XY-coordinates of root cellular markers into 3D representations of root growth profiles. Here, we present a detailed description

Gravitropism enables plants to adjust growth direction and architecture in response to gravity. The classical Cholodny-Went theory posits that asymmetric auxin redistribution drives differential growth during gravitropism in seed plants, but the nature of the equivalent asymmetric process in moss protonemata is still unclear. Here, through forward genetic screening, we identified GTRD, which encodes a coiled-coil protein whose loss reverses the direction of protonemal gravitropism. We further demonstrated that GTRD forms a complex with the kinesin-14 motor GTRC, and that GTRC drives the minus-

Regulation of the root growth pattern is an important control mechanism during plant growth and propagation. To better understand alterations in root growth direction in response to environmental stimuli, we have characterized an Arabidopsis thaliana mutant, wavy growth 3 (wav3), whose roots show a short-pitch pattern of wavy growth on inclined agar medium. The wav3 mutant shows a greater curvature of root bending in response to gravity, but a smaller curvature in response to light, suggesting that it is a root gravitropism-enhancing mutation. This wav3 phenotype also suggests that enhancement

Root growth and their interactions can provide valuable information for the development of asynchronous logic systems. Here, maize root behavior due to positive gravitropism and nutritropism is evaluated as three-inputs-three-outputs logical gates. Using plant roots as the element for unconventional computing, the Boolean functions of each root tropism were constructed through arithmetic-logical operations. One gravity gate (rGG) and two nutrient gates (rNG1 and rNG2) were fabricated using additive manufacturing. The rGG platform was oriented with roots directly pulled down by gravity which co

Hypocotyls of Arabidopsis thaliana exhibit negative gravitropism in the dark, growing against the gravity vector. The direction of growth is randomized in red light (R). In single mutants lacking either phytochrome A or B randomization of hypocotyl orientation in R is retained. However, a double mutant lacks this response, indicating that either phytochrome A or B is capable of inducing randomization and phytochrome A and B are the only phytochromes involved in this process. The induction of randomization was confirmed using lines that express to different levels PHYA and PHYB cDNAs. Overexpre

The plant hormone auxin plays a critical role in regulating various aspects of plant growth and development, and the spatial accumulation of auxin within organs, which is primarily attributable to local auxin biosynthesis and polar transport, is largely responsible for lateral organ morphogenesis and the establishment of plant architecture. Here, we show that three Arabidopsis INDETERMINATE DOMAIN (IDD) transcription factors, IDD14, IDD15, and IDD16, cooperatively regulate auxin biosynthesis and transport and thus aerial organ morphogenesis and gravitropic responses. Gain-of-function of each I

Multiple Gretchen Hagen 3 (GH3) genes have been implicated in a range of processes in plant growth and development through their roles in maintaining hormonal homeostasis. However, there has only been limited study on the functions of GH3 genes in tomato (Solanum lycopersicum). In this work, we investigated the important function of SlGH3.15, a member of the GH3 gene family in tomato. Overexpression of SlGH3.15 led to severe dwarfism in both the above- and below-ground sections of the plant, accompanied by a substantial decrease in free IAA content and reduction in the expression of SlGH3.9, a

Ratiometric wide-field fluorescence microscopy with 1',7'- bis-(2-carboxyethyl)-5-(and-6)-carboxyfluorescein (BCECF)-dextran demonstrated that gravistimulation leads to rapid changes in cytoplasmic pH (pHc) in columella cells of Arabidopsis roots. The pHc of unstimulated columella cells in tiers 2 and 3, known sites of graviperception (E.B. Blancaflor, J.B. Fasano, S. Gilroy [1998] Plant Physiol 116: 213-222), was 7.22 +/- 0.02 pH units. Following gravistimulation, the magnitude and direction of pHc changes in these cells depended on their location in the columella. Cells in the lower side of

The assumption that the mushroom stem has the ability to undergo autonomic straightening enables a mathematical model to be written that accurately mimics the gravitropic reaction of the stems of Coprinus cinereus. The straightening mechanism is called curvature compensation here, but is equivalent to the 'autotropism' that often accompanies the gravitropic reactions of axial organs in plants. In the consequently revised local curvature distribution model, local bending rate is determined by the difference between the 'bending signal' (generated by gravitropic signal perception systems) and th

BACKGROUND: Gravity is a ubiquitous environmental force that shapes plant architecture. Shoots occupy aerial space to absorb light and carbon dioxide, while roots spread in soil to capture water and nutrients. To ensure plants obtain survival resources, gravity not only promotes vertical growth and thus resists lodging, but also enables plants to stretch horizontally with a certain lateral organ angle and thus affects their high-density planting tolerance. Meanwhile, a large root angle often enhances plant lodging resistance. Further, roots with flexible angles and depth can cope with complex

Angiosperm trees reorient their stems by producing tension wood (TW), a specialised xylem that generates contractile force. TW fibres develop a gelatinous (G) layer rich in cellulose, yet the molecular mechanisms governing G-layer formation remain poorly understood. COBRA-like genes encode glycosylphosphatidylinositol (GPI)-anchored proteins known to regulate cellulose deposition. Here, we functionally characterised PagCOBL5 in hybrid poplar (Populus alba × P. glandulosa) by identifying its upstream regulator and interaction partner. PagCOBL5 was preferentially expressed in mature xylem fibres
