Development of gravity-sensing organs in altered gravity.

Experiments are described in which the development of the gravity-sensing organs was studied in newt larvae reared in microgravity on the IML-2 mission and in Aplysia embryos and larvae reared on a centrifuge at 1 to 5 g. In Aplysia embryos, the statolith (single dense mass on which gravity and linear acceleration act) was reduced in size in a graded fashion at increasing g. In early post-metamorphic Aplysia or even in isolated statocysts from such animals, the number of statoconia produced is reduced at high g. Newt larvae launched before any of the otoconia were formed and reared for 15 days

Neurobiology of fish under altered gravity conditions.

In vertebrates (including man), an altered gravitational environment such as weightlessness can induce malfunction of the inner ear, based on an irregular dislocation of the otoliths from the corresponding sensory epithelia. This dislocation leads to an illusionary tilt, since the otolithic inputs are not in register with other sensory organs. This results in an intersensory conflict. Vertebrates in orbit therefore face severe orientation problems. In humans, the intersensory conflict may additionally lead to a malaise, commonly referred to as space motion sickness (SMS). During the first days

Visually simulated altered gravity modulates perceived object weight.

Gravity is a fundamental force acting on all objects on Earth. It is therefore unsurprising that the brain has internalized regularities associated with terrestrial gravity. An internal model of gravity is thought to arise from the integration of vestibular, visual, proprioceptive and visceral sensory cues. However, it remains unclear whether this internal representation of gravity contributes to the perception of physical object properties. Object weight is determined by the product of its mass and gravitational acceleration. Although previous studies have examined how an object's visual prop

Effect of altered gravity on the neurobiology of fish.

In vertebrates (including humans) altered gravitational environments such as weightlessness can induce malfunction of the inner ears due to a mismatch between canal and statolith afferents. This leads to an illusionary tilt because the inputs from the inner ear are not confirmed by the other sensory organs, which then results in intersensory conflict. Vertebrates in orbit therefore face severe orientation problems. In humans the intersensory conflict may additionally lead to a malaise commonly referred to as space motion sickness (SMS). After the initial days of weightlessness the orientation

Neurobehavioral coping to altered gravity: endogenous responses of neurotrophins.

An altered gravitational environment represents a unique challenge for biological systems that have evolved against gravitational background. Ground-based and space research indicates that the developing nervous system is potentially affected by exposure to hyper/microgravity. With the construction of the orbiting International Space Station long-term research on the nervous system will be possible. With this perspective, we started ground-based studies to characterize mouse behavioral responses to rotation-induced 2 g hypergravity, using a custom-made centrifuge device. Brain levels of nerve

Effects of altered gravity on the swimming behaviour of fish.

Humans taking part in parabolic aircraft flights (PAFs) may suffer from space motion sickness-phenomena (SMS, a kinetosis). It has been argued that SMS during PAFs might not be based on microgravity alone but rather on changing accelerations from 0 g to 2 g. We test here the hypothesis that PAF-induced kinetosis is based on asymmetric statoliths (i.e., differently weighed statoliths on the right and the left side of the head), with asymmetric inputs to the brain being disclosed at microgravity. Since fish frequently reveal kinetotic behaviour during PAFs (especially so-called spinning movement

Neurophysiology of developing fish at altered gravity: background--facts--perspectives.

During the entire evolution of life on Earth, the phylogenetic as well as the individual development of all organisms took place under constant gravity conditions, against which they achieved specific countermeasures for compensation and adaptation. On the one side, gravity represents a factor of physical restriction, which compelled the ancestors of all extant living beings to develop basic achievements to counter the gravitational force (e.g., elements of statics like any kind of skeleton--from actin to bone--to overcome gravity enforced size limits or to keep form). On the other side, alrea

Promethazine Effects on Motion Sickness During Altered Gravity Induced by Parabolic Flight.

INTRODUCTION: Astronauts commonly experience space motion sickness, which can impair astronaut performance and safety. Pharmaceuticals are frequently used to reduce space motion sickness symptoms but have not been extensively studied in partial gravity. In this research effort, we investigated the impact of oral promethazine on motion sickness during parabolic flight. METHODS: We collected motion sickness scores from 12 subjects (6 women) during parabolic flight. Each subject participated in two flights-one 0-G flight and one partial g flight-experiencing 10 parabolas at 0 g, 0.25 g, 0.50 g,

Binocular misalignments elicited by altered gravity provide evidence for nonlinear central compensation.

Increased ocular positioning misalignments upon exposure to altered gravity levels (g-levels) have been strongly correlated with space motion sickness (SMS) severity, possibly due to underlying otolith asymmetries uncompensated in novel gravitational environments. We investigated vertical and torsional ocular positioning misalignments elicited by the 0 and 1.8 g g-levels of parabolic flight and used these data to develop a computational model to describe how such misalignments might arise. Ocular misalignments were inferred through two perceptual nulling tasks: Vertical Alignment Nulling (VAN)

Toll mediated infection response is altered by gravity and spaceflight in Drosophila.

Space travel presents unlimited opportunities for exploration and discovery, but requires better understanding of the biological consequences of long-term exposure to spaceflight. Immune function in particular is relevant for space travel. Human immune responses are weakened in space, with increased vulnerability to opportunistic infections and immune-related conditions. In addition, microorganisms can become more virulent in space, causing further challenges to health. To understand these issues better and to contribute to design of effective countermeasures, we used the Drosophila model of i

A mouse model of neurobehavioural response to altered gravity conditions: an ontogenetical study.

To determine the influence of gravity during critical periods of development is important in the perspective of long-term spaceflight and exploration, data coming from this kind of studies providing insight into basical biological phenomena underlying the development of the nervous system and its plasticity. Aim of the present study was to evaluate neurobehavioural responses to hypergravity exposure in CD-1 mice at different stage of development. Early adolescent (postnatal day 28, PND 28), adolescent (PND 42) and young-adult (PND 60) male and female mice were exposed to acute 2g rotational-ge

Evaluating the Suitability of Perfusion-Based PD Probes for Use in Altered Gravity Environments.

Measurable changes in electrophysiology have been documented in spaceflight, creating a pathway for disease genesis and progression in astronauts. These electrophysiology changes can be measured using potential difference (PD). A probe to measure PD was developed and is used clinically on Earth; this probe relies on fluid perfusion to establish an electrical connection to make PD measurements. The changes to fluid behavior in microgravity and partial gravity (including lunar and Martian gravity) drives the need to test this probe in a spaceflight environment. Here, we test the PD probe in a no

Modeling orientation perception adaptation to altered gravity environments with memory of past sensorimotor states.

Transitioning between gravitational environments results in a central reinterpretation of sensory information, producing an adapted sensorimotor state suitable for motor actions and perceptions in the new environment. Critically, this central adaptation is not instantaneous, and complete adaptation may require weeks of prolonged exposure to novel environments. To mitigate risks associated with the lagging time course of adaptation (e.g., spatial orientation misperceptions, alterations in locomotor and postural control, and motion sickness), it is critical that we better understand sensorimotor

Artificial gravity during a spaceflight analog alters brain sensory connectivity.

Spaceflight has numerous untoward effects on human physiology. Various countermeasures are under investigation including artificial gravity (AG). Here, we investigated whether AG alters resting-state brain functional connectivity changes during head-down tilt bed rest (HDBR), a spaceflight analog. Participants underwent 60 days of HDBR. Two groups received daily AG administered either continuously (cAG) or intermittently (iAG). A control group received no AG. We assessed resting-state functional connectivity before, during, and after HDBR. We also measured balance and mobility changes from pre

"Critical periods" in vestibular development or adaptation of gravity sensory systems to altered gravitational conditions?

1. A feature of sensory, neuronal and motor systems is the existence of a critical period during their development. Modification of environmental conditions during this specific period of life affects development in a long-term manner, or even irreversibly. Deprivation is the prefered approach to study the existence and duration of critical periods. For gravity sensory systems, space flights offer the only opportunity for deprivation conditions. 2. Studies in a fish (Oreochromis mossambicus) and an amphibian (Xenopus laevis) revealed a significant sensitivity of their roll-induced static vesti

Rapid Downregulation of H3K4me3 Binding to Immunoregulatory Genes in Altered Gravity in Primary Human M1 Macrophages.

The sensitivity of human immune system cells to gravity changes has been investigated in numerous studies. Human macrophages mediate innate and thus rapid immune defense on the one hand and activate T- and B-cell-based adaptive immune response on the other hand. In this process they finally act as immunoeffector cells, and are essential for tissue regeneration and remodeling. Recently, we demonstrated in the human Jurkat T cell line that genes are differentially regulated in cluster structures under altered gravity. In order to study an in vivo near system of immunologically relevant human cel

Gravity-awareness: a deep learning and LLM framework for predicting human adaptation to altered gravitational environments.

Earth's gravity fundamentally shapes human behavior. The brain encodes this force as an internal model of gravity, enabling the prediction and interpretation of gravitational effects during perception and action. Understanding how this model adapts to altered gravity is critical for predicting human performance in spaceflight. We present a computational framework for modelling neurophysiological adaptation across diverse gravitational environments. The framework has two components trained on open-access data from altered-gravity studies, particularly parabolic flights. The first component (Cor

Expression of hypoxia-inducible genes is suppressed in altered gravity due to impaired nuclear HIF1α accumulation.

Extravehicular activities, the backbone of manned space exploration programs, set astronauts into mild hypoxia. Unfortunately, microgravity aggravates threatening symptoms of hypoxia such as vision impairment and brain edema. Hypoxia-inducible factors (HIFs) sense cellular hypoxia and, subsequently, change the cells' expression profile instantaneously by rapidly translocating-most likely cytoskeleton-dependently-into the nucleus and subsequently forming transcription complexes with other proteins. We tested the hypothesis that this fundamental process could be altered by sudden changes in grav

Enhancing gait training with anti-gravity treadmill 'Alter-G' in patients with Parkinson's disease.

BACKGROUND: Freezing of Gait (FoG) is a debilitating motor symptom affecting nearly half of individuals with Parkinson's disease (PD), increasing fall risk and reducing independence. Despite pharmacological and neuromodulation therapies, residual symptoms often persist, underscoring the need for complementary rehabilitation strategies. OBJECTIVE: To evaluate the effects of body weight-supported treadmill training using differential air pressure on gait and mobility in PD patients. METHODS: Forty patients with idiopathic PD were randomly assigned to an experimental group (EG, n = 20), who rec
