Grid cells: the missing link in understanding Parkinson's disease?

The mechanisms underlying Parkinson's disease (PD) are complex and not fully understood, and the box-and-arrow model among other current models present significant challenges. This paper explores the potential role of the allocentric brain and especially its grid cells in several PD motor symptoms, including bradykinesia, kinesia paradoxa, freezing of gait, the bottleneck phenomenon, and their dependency on cueing. It is argued that central hubs, like the locus coeruleus and the pedunculopontine nucleus, often narrowly interpreted in the context of PD, play an equally important role in governi

Speed modulations in grid cell information geometry.

Grid cells, with hexagonal spatial firing patterns, are thought critical to the brain's spatial representation. High-speed movement challenges accurate localization as self-location constantly changes. Previous studies of speed modulation focus on individual grid cells, yet population-level noise covariance can significantly impact information coding. Here, we introduce a Gaussian Process with Kernel Regression (GKR) method to study neural population representation geometry. We show that increased running speed dilates the grid cell toroidal-like representational manifold and elevates noise st

Fragmentation of grid cell maps in a multicompartment environment.

To determine whether entorhinal spatial representations are continuous or fragmented, we recorded neural activity in grid cells while rats ran through a stack of interconnected, zig-zagged compartments of equal shape and orientation (a hairpin maze). The distribution of spatial firing fields was markedly similar across all compartments in which running occurred in the same direction, implying that the grid representation was fragmented into repeating submaps. Activity at neighboring positions was least correlated at the transitions between different arms, indicating that the map split regularl

Improved Visual Recognition Memory Model Based on Grid Cells for Face Recognition.

Traditional facial recognition methods depend on a large number of training samples due to the massive turning of synaptic weights for low-level feature extractions. In prior work, a brain-inspired model of visual recognition memory suggested that grid cells encode translation saccadic eye movement vectors between salient stimulus features. With a small training set for each recognition type, the relative positions among the selected features for each image were represented using grid and feature label cells in Hebbian learning. However, this model is suitable only for the recognition of famil

Retrospective Grid Cells in the Medial Entorhinal Cortex Encode Past Paths During Spatial Navigation.

Neurons dynamically switch between prospective and retrospective coding depending on task demands. Prospective coding optimizes future actions, while retrospective coding helps learn from past experiences. The grid cells in the medial entorhinal cortex (MEC) play a key role in spatial navigation, with periodic firing fields to encode current position. While their role in prospective coding has been reported recently, whether and how grid cells exhibit retrospective coding (encoding past paths) remains unclear. Here, we combined large-scale two-photon imaging in freely moving mice with well-est

A model of episodic memory: mental time travel along encoded trajectories using grid cells.

The definition of episodic memory includes the concept of mental time travel: the ability to re-experience a previously experienced trajectory through continuous dimensions of space and time, and to recall specific events or stimuli along this trajectory. Lesions of the hippocampus and entorhinal cortex impair human episodic memory function and impair rat performance in tasks that could be solved by retrieval of trajectories. Recent physiological data suggests a novel model for encoding and retrieval of trajectories, and for associating specific stimuli with specific positions along the trajec

Spatial memory deficits in Alzheimer's disease and their connection to cognitive maps' formation by place cells and grid cells.

Whenever we navigate through different contexts, we build a cognitive map: an internal representation of the territory. Spatial navigation is a complex skill that involves multiple types of information processing and integration. Place cells and grid cells, collectively with other hippocampal and medial entorhinal cortex neurons (MEC), form a neural network whose activity is critical for the representation of self-position and orientation along with spatial memory retrieval. Furthermore, this activity generates new representations adapting to changes in the environment. Though there is a norma

Disruption of the grid cell network in a mouse model of early Alzheimer's disease.

Early-onset familial Alzheimer's disease (AD) is marked by an aggressive buildup of amyloid beta (Aβ) proteins, yet the neural circuit operations impacted during the initial stages of Aβ pathogenesis remain elusive. Here, we report a coding impairment of the medial entorhinal cortex (MEC) grid cell network in the J20 transgenic mouse model of familial AD that over-expresses Aβ throughout the hippocampus and entorhinal cortex. Grid cells showed reduced spatial periodicity, spatial stability, and synchrony with interneurons and head-direction cells. In contrast, the spatial coding of non-grid ce

Spatially fractionated mini-GRID radiotherapy differentially modulates radiation-induced senescence in murine glioma and normal cells.

BACKGROUND: Spatially fractionated radiotherapy (SFRT) is an emerging therapeutic strategy that enhances tumor control while reducing normal tissue toxicity. Although conventional radiotherapy is known to induce therapy-induced senescence (TIS), its induction by SFRT-particularly mini-GRID radiotherapy-remains poorly characterized. Addressing this question is essential to better understand SFRT-mediated biological responses and optimize its therapeutic potential. METHODS: Two rat glioma cell lines (F98, RG2), immortalized astrocytes (CTX-TNA2), and primary mouse embryonic fibroblasts (MEFs) w

Modeling the influence of optic flow on grid cell firing in the absence of other cues1.

Information from the vestibular, sensorimotor, or visual systems can affect the firing of grid cells recorded in entorhinal cortex of rats. Optic flow provides information about the rat's linear and rotational velocity and, thus, could influence the firing pattern of grid cells. To investigate this possible link, we model parts of the rat's visual system and analyze their capability in estimating linear and rotational velocity. In our model a rat is simulated to move along trajectories recorded from rat's foraging on a circular ground platform. Thus, we preserve the intrinsic statistics of rea

Slow synaptic plasticity from the hippocampus underlies gradual mapping and fragmentation of novel spaces by grid cells.

Animals construct internal "cognitive maps" of the world during navigation in spatial and non-spatial domains, with grid cells in the medial entorhinal cortex (MEC) playing a key role. This requires associating internal position estimates with external cues to reduce spatial uncertainty over time. However, how grid cell representations evolve in novel spaces to support map formation is unclear. To address this question, we longitudinally imaged calcium dynamics of grid cells over 10 days as mice learn operant tasks in novel virtual linear tracks. We observe that spatial tuning of grid cells is

Chromosome 17 In Situ Hybridization Grid-based Analysis in Oral Squamous Cell Carcinoma.

BACKGROUND/AIM: Oral squamous cell carcinoma (OSCC) is an aggressive malignancy due to its increased ability for local metastases and distant lymph node metastases. Extensive cytogenetic analyses have detected chromosome instability (CI) patterns in OSCC including gross chromosome numerical alterations, such as polysomy and sporadically monosomy that negatively affect the biological behaviour of the malignancy. Our aim was to investigate the frequency and impact of chromosome 17 (Chr 17) numerical imbalances in OSCC. MATERIALS AND METHODS: Fifty (n=50) formalin-fixed, paraffin-embedded primar

Exploring the relationship between urban flood risk and resilience at a high-resolution grid cell scale.

The assessment of flood risk and resilience has become increasingly important in recent years for effective urban flood management. While flood resilience and risk are two distinct concepts with unique assessment metrics, there is lack of quantitative analysis and understanding of the relationship between them. This study aims to investigate this relationship at the grid cell level in urban areas. To assess flood resilience for high-resolution grid cells, this study proposes a performance-based flood resilience metric, which is calculated using the system performance curve based on flood durat

Heterojunction Solar Cells With Buried Al Grids and Li3PO4 Electron Selective Contacts Approaching 24% Efficiency.

Dopant-free materials with wide-bandgap are promising alternatives to n-type or p-type amorphous silicon (a-Si) contacts in silicon heterojunction (SHJ) solar cells. They offering strong carrier selectivity through favorable band alignment, negligible parasitic absorption, and full compatibility with low-temperature processing. However, implementing such contacts on the front side remains challenging because optical losses, contact resistivity, and gird patterns should be balanced carefully. Here, these constraints are overcome through an interfacial design that integrates a lithium phosphate

Uncovering 2-D toroidal representations in grid cell ensemble activity during 1-D behavior.

Minimal experiments, such as head-fixed wheel-running and sleep, offer experimental advantages but restrict the amount of observable behavior, making it difficult to classify functional cell types. Arguably, the grid cell, and its striking periodicity, would not have been discovered without the perspective provided by free behavior in an open environment. Here, we show that by shifting the focus from single neurons to populations, we change the minimal experimental complexity required. We identify grid cell modules and show that the activity covers a similar, stable toroidal state space during

Grid cell disruption in a mouse model of early Alzheimer's disease reflects reduced integration of self-motion cues.

Converging evidence from human and rodent studies suggests that disrupted grid cell coding in the medial entorhinal cortex (MEC) underlies path integration behavioral deficits during early Alzheimer's disease (AD). However, grid cell firing relies on both self-motion cues and environmental features, and it remains unclear whether disrupted grid coding can account for specific path integration deficits reported during early AD. Here, we report in the J20 transgenic amyloid beta (Aβ) mouse model of early AD that grid cells were spatially unstable toward the center of the arena, had qualitatively

Impact of Chromosome 9 Numerical Imbalances in Oral Squamous Cell Carcinoma: A Pilot Grid-Based Centromere Analysis.

Oral squamous cell carcinoma (OSCC) is considered an aggressive malignancy, mainly due to its increased propensity to provide local and distant lymph node metastases. Gross chromosome instability (CI; polysomy/aneuploidy/monosomy), combined or not with specific gene alterations, is implicated in the development and progression of solid malignancies, including OSCC. In order to further study the relationship between these genetic alterations and the aggressive biological behavior of OSCCs, we investigated the frequency and impact of chromosome 9 numerical imbalances in these tumors. Cyclin D1 Gene Numerical Imbalances in Laryngeal Squamous Cell Carcinoma: A Tissue Microarray Grid Based Analysis.

BACKGROUND: Deregulation of critical proteins involved in cell cycle stability, such as cyclins, is a frequent genetic event in the development and progression of solid malignancies. Concerning laryngeal squamous cell carcinoma (LSCC), cyclin D1 oncogenic transformation leads to an aberrant protein expression and seems to affect the biological behaviour of the neoplasm. The aim of this study was to determine the correlation of cyclin D1 numerical imbalances with the corresponding protein expression levels in LSCCs. MATERIAL AND METHOD: Using tissue microarray (TMA) technology, fifty (n=50) hi

Compromised Grid-Cell-like Representations in Old Age as a Key Mechanism to Explain Age-Related Navigational Deficits.

A progressive loss of navigational abilities in old age has been observed in numerous studies, but we have only limited understanding of the neural mechanisms underlying this decline [1]. A central component of the brain's navigation circuit are grid cells in entorhinal cortex [2], largely thought to support intrinsic self-motion-related computations, such as path integration (i.e., keeping track of one's position by integrating self-motion cues) [3-6]. Given that entorhinal cortex is particularly vulnerable to neurodegenerative processes during aging and Alzheimer's disease [7-14], deficits i

A Grid-Based Nucleus Counting Method for Estimates of the Density of Superficial Conjunctival Cells from Impression Cytology Samples Taken from Normal Healthy Human Eyes.

PURPOSE: To develop and validate an easily procured objective estimate of the cell density of the superficial epithelial cells of the normal bulbar conjunctiva using conjunctival impression cytology (CIC). METHODS: From 20 healthy non-contact lens-wearing young adults, CIC was undertaken off the nasal aspect of the exposed bulbar conjunctiva, the filters stained with Giemsa, and images taken at 200× that only included an all-but-contiguous monolayer of epithelial cells. These images were projected at 1000× magnification and an overlay drawn of the location of the stained nuclei, from which tw
