B-cell growth and differentiation factors.

The number of recombinant factors which have been shown to regulate B cell activation, proliferation, and differentiation now stands at nine (IL-1, IL-2, IL-4, IL-5, IL-6 (BSF-2), BCGFLOW, IFN-alpha and gamma, and TNF). Several others have been described but are not yet fully characterized or available as recombinant gene products and will not be discussed. Recent work with these factors, especially IL-4, has revealed a remarkable diversity of function. Most of them seem able to act at more than one stage of B cell activation, proliferation and differentiation, thus invalidating the concept of

[Differentiation of the neural crest. Influence of growth factors].

The peripheral nervous system (PNS) is derived from a transitory embryonic structure, the neural crest with an additional contribution of the cephalic ectodermal placodes to the sensory ganglia of certain cranial nerves. In vivo grafting experiments, taking advantage of the quail/chick marker system, allowed the fate map of the neural crest to be constructed and the precise level of origin along the neuraxis of the component cells of PNS ganglia to be defined. It appeared that the neural crest is regionalized in different areas from which a limited number of phenotypes arise. However, if the p

Platelet-Released Growth Factors Induce Differentiation of Primary Keratinocytes.

Autologous thrombocyte concentrate lysates, for example, platelet-released growth factors, (PRGFs) or their clinically related formulations (e.g., Vivostat PRF®) came recently into the physicians' focus as they revealed promising effects in regenerative and reparative medicine such as the support of healing of chronic wounds. To elucidate the underlying mechanisms, we analyzed the influence of PRGF and Vivostat PRF on human keratinocyte differentiation in vitro and on epidermal differentiation status of skin wounds in vivo. Therefore, we investigated the expression of early (keratin 1 and kera

Dendritic cells: expansion and differentiation with hematopoietic growth factors.

Dendritic cells are critical initiators of immune responses mediated by T and B lymphocytes. Dendritic cells process antigens captured in the periphery, then emigrate to lymphoid organs. There they complete their maturation by upregulating important accessory molecules and secreting cytokines, all of which support potent stimulation of antigen-specific lymphocytes. These lymphocytes return to the periphery to complete the immune response. Investigators have discovered culture systems that use exogenous hematopoietic cytokines to support the growth, differentiation, and maturation of dendritic

Differential expression of growth differentiation factor-9 in keloids.

OBJECTIVE: The mechanisms of keloid invasion are largely unknown. This study aims to analyze the differentially expressed genes between keloid peripheral and central areas and thus to define the molecule that might be responsible for keloid invasion. METHODS: The gene chip of transforming growth factor-β (TGF-β) superfamily signaling pathway was used to analyze differentially expressed genes of the fibroblasts derived from peripheral area and central area of 3 keloids. The differential expression of growth differentiation factor-9 (GDF-9) was also confirmed by quantitative PCR and Western-blo

Gene regulatory roles of growth and differentiation factors in retinal development.

Retinal ganglion cell (RGC) differentiation is tightly controlled by extrinsic and intrinsic factors. Growth and differentiation factor 15 (GDF-15) promotes RGC differentiation, opposite to GDF-11 which inhibits RGC differentiation, both in the mouse retina and in human stem cells. To deepen our understanding of how these two closely related molecules confer opposing effects on retinal development, here we assess the transcriptional profiles of mouse retinal progenitors exposed to exogenous GDF-11 or -15. We find a dichotomous effect of GDF-15 on RGC differentiation, decreasing RGCs expressing

Metabolic Messenger: growth differentiation factor 15.

Growth differentiation factor 15 (GDF15; also known as macrophage-inhibitory cytokine-1) is a stress-responsive cytokine that is overexpressed under a broad range of conditions. It has a role in regulating appetite and body weight and is an aetiological factor in anorexia-cachexia syndromes, as well as nausea and vomiting during pregnancy. Long after its original cloning, its receptor was identified as GFRAL, a distant member of the GDNF receptor family within the TGFβ superfamily, with RET as its co-receptor. Both of these are highly localized to specific hindbrain regions. Although many of G

Dopamine as a growth differentiation factor in the mammalian brain.

The catecholamine, dopamine, plays an important role in the central nervous system of mammals, including executive functions, motor control, motivation, arousal, reinforcement, and reward. Dysfunctions of the dopaminergic system lead to diseases of the brains, such as Parkinson's disease, Tourette's syndrome, and schizophrenia. In addition to its fundamental role as a neurotransmitter, there is evidence for a role as a growth differentiation factor during development. Recent studies suggest that dopamine regulates the development of γ-aminobutyric acidergic interneurons of the cerebral cortex.

Nerve growth factor promotes human hemopoietic colony growth and differentiation.

Nerve growth factor (NGF) is a neurotropic polypeptide necessary for the survival and growth of some central neurons, as well as sensory afferent and sympathetic neurons. Much is now known of the structural and functional characteristics of NGF, whose gene has recently been cloned. Since it is synthesized in largest amounts by the male mouse submandibular gland, its role exclusively in nerve growth is questionable. NGF also causes histamine release from rat peritoneal mast cells in vitro, and we have shown elsewhere that it causes significant, dose-dependent, generalized mast cell proliferatio

Influence of growth factors on growth and differentiation of 3T3-L1 preadipocytes in serum-free conditions.

The role of platelet-derived growth factor (PDGF) and epidermal growth factor (EGF) in the adipose differentiation of 3T3-L1 preadipocytes was investigated in a chemically defined culture system. To eliminate the influence of unidentified adipogenic compounds present in serum or serum-derived medium additives, cells from a stock culture were grown to confluence under serum-free conditions in the presence of recombinant PDGF and insulin. To trigger differentiation of these cells, the usual stimuli for adipogenesis such as corticosterone and insulin at supraphysiological concentrations together

Role of growth factors and their interplay during oligodendroglial differentiation and maturation.

Oligodendrocytes (OLs) facilitate the myelination within the central nervous system (CNS). Neuronal axons are originally devoid of myelin and acquire myelination during development. OLs are derived from oligodendrocyte precursor cells (OPCs) during development, and this ability to differentiate persists throughout maturity. The abnormal differentiation of oligodendrocytes leads to various neurological disorders and neurodegenerative diseases characterized by disruptions in myelin formation, which is crucial for proper nerve function. In order to develop suitable strategies for their treatments

Growth differentiation factor 11 is a protective factor for osteoblastogenesis by targeting PPARgamma.

Growth and differentiation factor 11 (GDF11) is a TGF-β family member that is expressed widely and plays multiple roles, including regulating axial skeletal patterning during development. However, the function of GDF11 in the bone is barely understood. Here we identify GDF11 as a potent regulator of bone homeostasis. GDF11 level is significantly decreased in the plasma and bone marrow of aging mice and mice with osteoporosis. Pharmacologic GDF11 gain of function leads to a striking increase in osteoblastogenesis and inhibits adipogenesis from bone marrow mesenchymal stem cells by altering the

Growth differentiation factor 6 as a putative risk factor in neuromuscular degeneration.

Mutation of Glass bottom boat, the Drosophila homologue of the bone morphogenetic protein or growth/differentiation factor (BMP/GDF) family of genes in vertebrates, has been shown to disrupt development of neuromuscular junctions (NMJ). Here we tested whether this same conclusion can be broadened to vertebrate BMP/GDF genes. This analysis was also extended to consider whether such genes are required for NMJ maintenance in post-larval stages, as this would argue that BMP genes are viable candidates for analysis in progressive neuromuscular disease. Zebrafish mutants harboring homozygous null mu

Growth differentiation factor 15 (GDF-15) in endocrinology.

Human growth differentiation factor 15 (GDF-15) is a widely distributed protein that has shown to play multiple roles in both physiological and pathological conditions. In healthy individuals, GDF-15 is mainly expressed in the placenta, followed by the prostate, although low levels of expression have also been detected in different organs. GDF-15 acts through a recently identified receptor called glial-derived neurotrophic factor (GDNF) receptor alpha-like (GFRAL) which signals through the rearranged during transfection (RET) tyrosine kinase receptor. The effects of GDF-15 are pleiotropic and

Nerve growth factor: role in growth, differentiation and controlling cancer cell development.

Recent progress in the Nerve Growth Factor (NGF) research has shown that this factor acts not only outside its classical domain of the peripheral and central nervous system, but also on non-neuronal and cancer cells. This latter observation has led to divergent hypothesis about the role of NGF, its specific distribution pattern within the tissues and its implication in induction as well as progression of carcinogenesis. Moreover, other recent studies have shown that NGF has direct clinical relevance in certain human brain neuron degeneration and a number of human ocular disorders. These studie

Growth and differentiation factor 15 as a biomarker for mitochondrial myopathy.

OBJECTIVE: We investigated if Growth and Differentiation Factor 15 (GDF-15) can be used as a biomarker to distinguish patients with mitochondrial myopathy from patients with other myopathies. METHODS: Serum GDF-15 was measured in 28 patients with mitochondrial disease, 24 with metabolic myopathies, 27 with muscular dystrophy and 21 healthy controls. RESULTS AND CONCLUSIONS: Our findings indicate that elevated GDF-15 can distinguish patients with mitochondrial myopathy from other myopathies, including metabolic myopathies. This suggests that increases in GDF-15 is specific to respiratory chai

Growth differentiation factor 15 in erythroid health and disease.

PURPOSE OF REVIEW: Growth differentiation factor 15 (GDF15) was identified as a hepcidin-suppression factor that is expressed at high levels in patients with ineffective erythropoiesis. This review addresses the regulation, expression and potential functions of GDF15 in the context of erythroid biology. RECENT FINDINGS: GDF15 expression during late erythroid differentiation was discovered as part of an erythroblast transcriptome project. As GDF15 expression is associated with cellular stress or apoptosis, further investigation of the cytokine was focused upon its involvement in ineffective er
