Modulation of imatinib cytotoxicity by selenite in HCT116 colorectal cancer cells.

Imatinib is a principal therapeutic agent for targeting colorectal tumours. However, mono-targeting by imatinib does not always achieve complete cancer eradication. Selenite, a well-known chemopreventive agent, is commonly used in cancer patients. In this study, we aimed to explore whether selenite can modulate imatinib cytotoxicity in colorectal cancer cells. HCT116 cells were treated with different concentrations of imatinib and/or selenite for 24, 48 and 72 hr. Imatinib-selenite interaction was analysed using isobologram equation. As indicators of apoptosis, DNA fragmentation, caspase-3 act

Genetic and transcriptomic alterations underlying aggressiveness in 5-fluorouracil-resistant HCT116 cells.

Resistance to 5-fluorouracil (5-FU) remains an obstacle to effective colorectal cancer (CRC) treatment. Genetic alterations play significant roles in the development of drug-induced chemoresistance, leading to tumor recurrence and increased aggressiveness. This study sought to characterize the transcriptomic profiles of 5-FU-resistant CRC cells. HCT116 colon cancer cells were progressively exposed to increasing concentrations of 5-FU to induce resistance. Chemoresistance and aggressiveness were investigated using MTT, clonogenic, and scratch assays. Transcriptomic alterations were analyzed wit

H-RAS V12-induced radioresistance in HCT116 colon carcinoma cells is heregulin dependent.

The abilities of mutated active K-RAS and H-RAS proteins, in an isogenic human carcinoma cell system, to modulate the activity of signaling pathways following exposure to ionizing radiation is unknown. Loss of K-RAS D13 expression in HCT116 colorectal carcinoma cells blunted basal extracellular signal-regulated kinase 1/2 (ERK1/2), AKT, and c-Jun NH2-terminal kinase 1/2 activity. Deletion of the allele to express K-RAS D13 also enhanced expression of ERBB1, ERBB3, and heregulin but nearly abolished radiation-induced activation of all signaling pathways. Expression of H-RAS V12 in HCT116 cells

HCT116 cell line heterogeneity: A useful tool for preclinical research.

Cancer cell heterogeneity is one of the reasons for anticancer chemotherapy failure. Nowadays, this field of research is receiving increasing attention, but mainly as a descriptive characteristic of the tumor. In this study, the property of heterogeneity of tumor colonies is considered as the basis for the identification of effective treatment schemes. A series of sequential sortings of HCT116 cells were performed based on their ability to accumulate Rhodamine 123. Subpopulations were obtained that differ significantly in their ability to accumulate the dye, including side population cells exh

Different DNA lesions trigger distinct cell death responses in HCT116 colon carcinoma cells.

The pleiotrophic cellular response to DNA damage includes activation of cell cycle checkpoints, induction of DNA repair pathways, and initiation of programmed cell death among others. The fate of cells with damaged DNA depends on the coordination of these different responses. The clinical efficacy of genotoxic therapies is influenced by cell fate and thus by how the DNA damage response is coordinated. While a great deal has been learned about how different DNA lesions activate distinct cell cycle checkpoints and DNA repair pathways, less is known about whether the type of DNA lesion influences

Enhancement of Lysosomal Biogenesis in the HCT116 Cells Treated with Doxorubicin under Normoxic Conditions.

Hydrophobic weakly basic drugs, such as doxorubicin and sunitinib, are currently key components of cancer chemotherapy. It has been shown that several of these compounds induce increase in the total lysosomal volume in tumor cells. Moreover, hypoxia, a hallmark of solid tumors in vivo, promotes chemoresistance by sequestering doxorubicin within lysosomes. To enhance efficacy of chemotherapy, various strategies have been proposed, including those aimed at lysosome destabilization. Inhibition of autophagy is widely recognized as a means to reduce chemoresistance. However, it remains unclear whet

Supervillin-Mediated ZO-1 Downregulation Facilitates Migration of Cisplatin-Resistant HCT116 Colorectal Cancer Cells.

Supervillin (SVIL), the biggest member of the villin/gelsolin superfamily, has recently been reported to promote the metastasis of hepatocellular carcinoma by stimulating epithelial-mesenchymal transition (EMT). However, little is known about the roles of SVIL in the migration of colorectal cancer cells. Here, we investigated the effects of SVIL on the migration of cisplatin-resistant colorectal cancer cells. The model of cisplatin-resistant HCT116 cells (HCT116/DDP) was established. Migration was assessed after SVIL knockdown. Tumor metastasis was assessed using a mouse model with tail vein i

Drug resistance induces the upregulation of H2S-producing enzymes in HCT116 colon cancer cells.

Hydrogen sulfide (H2S) production in colon cancer cells supports cellular bioenergetics and proliferation. The aim of the present study was to investigate the alterations in H2S homeostasis during the development of resistance to 5-fluorouracil (5-FU), a commonly used chemotherapeutic agent. A 5-FU-resistant HCT116 human colon cancer cell line was established by serial passage in the presence of increasing 5-FU concentrations. The 5-FU-resistant cells also demonstrated a partial resistance to an unrelated chemotherapeutic agent, oxaliplatin. Compared to parental cells, the 5-FU-resistant cells

miR-106b fine tunes ATG16L1 expression and autophagic activity in intestinal epithelial HCT116 cells.

BACKGROUND: The microRNAs (miRNAs) regulate gene expression at the posttranscriptional level. ATG16L1, an essential component for autophagy and a risk gene for Crohn's disease, contains two binding sites in the 3'UTR for miR-17 family, including miRs-20a, -93, -106a, and -106b. The purpose of this study was to assess the effects of these miRNAs on ATG16L1 expression and autophagic activity in HCT116 cells. METHODS: The functional binding sites in the ATG16L1 3'UTR were evaluated by transfection of pMIR-GLO vectors bearing the wild type or mutant 3'UTR into cells for luciferase reporter assay.

Exploring cytotoxicity of cordycepin loaded nanovesicles against (HCT116) colon cancer cells: Optimization and cellular evaluation.

Источник пометил публикацию как отозванную

Numerous researchers have investigated cordycepin (COR) as an anti-tumor compound. COR has been documented to have cytotoxic effects on several cancer cells. The current work used a Box-Behnken mathematical design to minimize COR's size. The design incorporated COR concentration, phospholipid concentration and sonication time as variables to minimize the vesicles of COR emulsomes (COR-EMLs). To evaluate degree of improvement of COR cytotoxicity against colorectal cancer (HCT116) cells, cell viability, cell cycle analysis and apoptosis have been assessed. In addition, wound scratching and mitoc

Diphenyl ditelluride anticancer activity and DNA topoisomerase I poisoning in human colon cancer HCT116 cells.

Diphenyl ditelluride (DPDT) is an organotellurium (OT) compound with pharmacological properties, including antioxidant, antigenotoxic and antimutagenic activities when applied at low concentrations. However, DPDT as well as other OT compounds also show cytotoxicity against mammalian cells when treatments occur at higher drug concentrations. Considering that the underlying mechanisms of toxicity of DPDT against tumor cells have been poorly explored, the objective of our study was to investigate the effects of DPDT against both human cancer and non-tumorigenic cells. As a model, we used the colo

Quantitative chemical proteomics reveals anti-cancer targets of Celastrol in HCT116 human colon cancer cells.

BACKGROUND: Celastrol (Cel) is a naturally-derived compound with anti-cancer properties and exerts beneficial effects against various diseases. Although an extensive body of research already exists for Cel, the vast majority are inductive studies with limited validation of specific pathways and functions. The cellular targets that bind to Cel remain poorly characterized, which limits attempts to uncover its mechanism of action. PURPOSE: The present study aims to comprehensively identify the protein targets of Cel in HCT116 cells in an unbiased manner, and elucidate the mechanism of the anti-c

MicroRNA-18a upregulates autophagy and ataxia telangiectasia mutated gene expression in HCT116 colon cancer cells.

Autophagy is an evolutionarily conserved, multi-step lysosomal degradation process in which a cell degrades its own long-lived proteins and damaged organelles. Ataxia telangiectasia mutated (ATM) has recently been shown to upregulate the process of autophagy. Previous studies showed that certain microRNAs, including miR-18a, potentially regulate ATM in cancer cells. However, the mechanisms behind the modulation of ATM by miR-18a remain to be elucidated in colon cancer cells. In the present study, we explored the impact of miR-18a on the autophagy process and ATM expression in HCT116 colon canc
