Extracellular activation of fluorinated aziridinylbenzoquinone in HT29 cells EPR studies.

The plasma membrane of HT29 human colon carcinoma cells was characterized by EPR spectroscopy as the site for redox activation of 3,6-difluoro-2,5-bis(aziridinyl)-1,4-benzoquinone (F-DZQ). Supplementation of HT29 cells with F-DZQ yielded an EPR signal ascribed to the semiquinone species; the hyperfine splitting constants of the 11-line spectrum were 1.4 and 1.35 G for aN and aF, respectively. The intensity of the EPR signal was inhibited competitively by potassium ferricyanide, a compound which has no access to the intracellular milieu and used to evaluate transmembrane NADH-ferricyanide reduc

cAMP-dependent activation of small-conductance Cl- channels in HT29 colon carcinoma cells.

The present study was performed to examine the conductance properties in the colon carcinoma cell line HT29 and the activation of Cl- channels by cAMP. A modified cell-attached nystatin patch-clamp technique was used, allowing for the simultaneous recording of the cell membrane potential (PD) and the conductance properties of the cell-attached membrane. In resting cells, PD was -56 +/- 0.4 mV (n = 294). Changing the respective ion concentrations in the bath indicate that these cells possess a dominating K+ conductance and a smaller Cl- conductance. A significant non-selective cation conductanc

Mechanism of Resveratrol on LPS/ATP-induced pyroptosis and inflammatory response in HT29 cells.

Pyroptosis plays an important role in maintenance of intestinal homeostasis, the abnormal activation of NOD-like receptor thermal protein domain-associated protein 3 (NLRP3) inflammasome can promote the event and development of ulcerative colitis (UC). Its protective effects such as inhibiting pyroptosis in various inflammation-related diseases have been demonstrated, but whether resveratrol (RES) can also alleviate the progression of the disease by inhibiting pyroptosis in UC and the mechanism have rarely been studied. In this study, lipopolysaccharide (LPS) combined with adenosine triphospha

Entamoeba histolytica induces cell death of HT29 colonic epithelial cells via NOX1-derived ROS.

Entamoeba histolytica, which causes amoebic colitis and occasionally liver abscess in humans, is able to induce host cell death. However, signaling mechanisms of colon cell death induced by E. histolytica are not fully elucidated. In this study, we investigated the signaling role of NOX in cell death of HT29 colonic epithelial cells induced by E. histolytica. Incubation of HT29 cells with amoebic trophozoites resulted in DNA fragmentation that is a hallmark of apoptotic cell death. In addition, E. histolytica generate intracellular reactive oxygen species (ROS) in a contact-dependent manner. I

Response of HT29 cells to butyrate depends on time of exposure and glucose deprivation.

The high level of alkaline phosphatase activity in HT29 cells induced after 2 or 5 days of butyrate treatment was decreased during their prolonged exposure (about 30 days) to this agent together with a decrease of sensitivity to apoptosis. However, an enormous additive effect on alkaline phosphatase activity was found after butyrate treatment of glucose-starved cells. In concert with this finding, the substructural analysis revealed a dense brush border, tendency to polarization and morphologically normal mitochondria. It can be concluded that prolonged butyrate treatment of HT29 cells attenua

Cellular chloride depletion inhibits cAMP-activated electrogenic chloride fluxes in HT29-18-C1 cells.

Cyclic AMP-activated chloride fluxes have been analyzed in HT29-18-C1 cells (a clonal cell line derived from a human colon carcinoma) using measurements of cell volume (electronic cell sizing), cell chloride content (chloride titrator) and intracellular chloride activity (6-methoxy-N-(3-sulfopropyl)quinolinium; SPQ). HT29-18-C1 was shown to mediate polarized chloride transport. In unstimulated cells, the apical membrane was impermeable to chloride and net chloride flux was mediated by basolateral furosemide-sensitive transport. Forskolin (10 microM) increased furosemide-insensitive chloride pe

Effect of alkalinization of cytosolic pH by amines on intracellular Ca2+ activity in HT29 cells.

The effect of secondary, tertiary and quaternary methyl- and ethylamines on intracellular pH (pHi) and intracellular Ca2+ activity ([Ca2+]i) of HT29 cells was investigated microspectrofluorimetrically using pH- and Ca2+- sensitive fluorescent indicators, [i.e. 2', 7'-biscarboxyethyl-5(6)-carboxyfluorescein (BCECF) and fura-2 respectively]. Membrane voltage (Vm) was studied by the patch-clamp technique. Secondary and tertiary amines led to a rapid and stable concentration-dependent alkalinization which was independent of their pKa value. Trimethylamine (20 mmol/l) increased pHi by 0.78 +/- 0.03

Toxoplasma gondii: changes of transepithelial ion transport in infected HT29/B6 cell monolayers.

An in vitro system was established to study the effect of coccidian parasites on ion transport systems in epithelial tissues using HT29/B6, a human colon carcinoma cell line, and Toxoplasma gondii as a model parasite. Ion transport was measured in perfusion chambers 5, 10 and 15 h post-infection using monolayers in which approximately 30% of the cells were parasitized. The infection had rapid effects on the conductance and unidirectional chloride fluxes of infected cell monolayers, which were two to three times higher than those of uninfected HT29/B6 cell monolayers throughout the observation

Intestinal transport of methylmercury and inorganic mercury in various models of Caco-2 and HT29-MTX cells.

Food is the main pathway of exposure to mercury for most of the population. In food, mercury is generally present as inorganic mercury [Hg(II)] or methylmercury [MeHg]. Both chemical forms have some degree of toxicity, especially MeHg, which is considered a powerful neurotoxicant during development and is classified as a possible human carcinogen. Since the main exposure pathway is oral, gastrointestinal absorption is a decisive step in the process by which mercury reaches the systemic circulation. However, there are few studies that characterize this absorption process. The present work evalu

Caco-2/HT29-MTX co-cultured cells as a model for studying physiological properties and toxin-induced effects on intestinal cells.

Infectious gastrointestinal diseases are frequently caused by toxins secreted by pathogens which may impair physiological functions of the intestines, for instance by cholera toxin or by heat-labile enterotoxin. To obtain a functional model of the human intestinal epithelium for studying toxin-induced disease mechanisms, differentiated enterocyte-like Caco-2 cells were co-cultured with goblet cell-like HT29-MTX cells. These co-cultures formed a functional epithelial barrier, as characterized by a high electrical resistance and the presence of physiological intestinal properties such as glucose

Isolation and characterization of a differentiation-dependent gene in the human colonic cell line HT29-18.

The human colonic epithelial cell line HT29, and its clonal derivatives HT29-18 and HT-29-18-C1, differentiate in vitro. Differential screening of a subtraction cDNA library enriched for sequences unique to HT29-18-C1, a highly differentiated subclone of HT29-18, resulted in the isolation of a differentiation-dependent cDNA clone, A4. A full-length clone encoding A4 was obtained and sequenced to its entirety. It is 945 bp in length and contains an open reading frame (ORF) of 456 bp. The amino acid sequence deduced from the ORF reveals a polypeptide of 152 amino acids with a predicted molecular
