An Uncommon Presentation of Human Otoacariasis by Haemaphysalis longicornis.

Ticks are obligate parasites on animals and sometimes humans. They usually suck the blood of the hosts and can carry various infectious diseases as a vector. Otoacariasis is the presence of ticks and mites within the ear canal and relatively common in domestic and wild animals. However, tick infestations of human ear canals are rarely reported in the scientific literature and hardly occur in developed countries. Herein, we report a rare case of otoaracariasis involving Haemaphysalis longicornis . A 9-year-old girl living in a suburb presented with otalgia of left ear for 1 day. Otoscopic exami

Epidemiology of toxoplasmosis: role of the tick Haemaphysalis longicornis.

BACKGROUND: Toxoplasma gondii infection is mainly caused by ingestion of water or food that is contaminated with oocysts excreted by cats, or by eating raw meat containing T. gondii tissue cysts. However, oral transmission does not explain the common occurrence of toxoplasmosis in a variety of hosts, such as herbivorous animals, birds, and wild rodents. Little information exists on the maintenance of T. gondii parasites in nature and routes of transmission to domestic and wild animal hosts. Therefore, this study evaluated the role of Haemaphysalis longicornis ticks in the epidemiology of toxop

Molecular evidence of Babesia equi transmission in Haemaphysalis longicornis.

We studied the tick, Haemaphysalis longicornis, to determine the possibility of both transovarial and transstadial transmission of Babesia equi. We also studied the usefulness of the needle injection method for pathogenic tick-transmitted organisms including Babesia parasites. Erythrocytes infected with B. equi were injected into the midgut of engorged adults or nymphs using a hypodermic needle passed through the integument. DNA of B. equi in ticks was detected using nested polymerase chain reaction (PCR). B. equi DNA was present in adults, eggs, and larvae, indicating that transovarial transm

Draft genome sequence data of Haemaphysalis longicornis Oita strain.

Haemaphysalis longicornis Neumann, 1901 is one of the most well-known hard ticks because of its medical and veterinary importance. Haemaphysalis longicornis transmit a wide range of pathogens among vertebrates, affecting humans and animals in Asia and Oceania. In Japan, the tick species is a major pest of cattle because it can spread a protozoan parasite Theileria orientalis, which causes theileriosis and produces economic losses to the livestock industry (Yokoyama et al. 2012 [1]). Apart from bovine theileriosis, H. longicornis is a vector of bovine babesiosis caused by Babesia ovata, canine

Anaplasma species detected in Haemaphysalis longicornis tick from China.

Anaplasma are tick-borne obligatory intracellular bacteria, which infect humans and other animals. The Anaplasma species carried by ticks in China are not well studied. We collected 3145 questing Haemaphysalis longicornis ticks including 120 larvae, 2460 nymphs and 565 adults from vegetation in Jiaonan County, Shandong Province, China from 2013 to 2015. All ticks were examined for the presence of Anaplasma species by nested PCR amplification of the 16S rRNA gene. For further differentiation of A. capra from A. centrale, gltA and msp2 genes were sequenced for A. capra. Three Anaplasma species w

Characterization of acid phosphatase from the tick Haemaphysalis longicornis.

The full-length cDNA encoding acid phosphatase (HL-3) from Haemaphysalis longicornis was obtained by 5' rapid amplification of cDNA ends (RACE). The cDNA contained a 1137 bp open reading frame (ORF) coding for 356 amino acids with a predicted theoretical isoelectric point (pI) of 6.35 and molecular weight of 41.0 kDa. The recombinant protein was expressed in Escherichia coli. The enzyme could hydrolyze para-nitrophenyl phosphate (pNPP) substrate at an optimum pH of 5.0. Real-time RT-PCR analysis showed that the HL-3 transcripts were expressed in various stages of unfed ticks and were significa

Haemaphysalis longicornis and Carvacrol as Acaricide: Efficacy and Mechanism of Action.

Carvacrol derived from Origanum vulgare oil was evaluated for its acaricidal efficacy and mechanism of action against Haemaphysalis longicornis, a primary vector of severe fever with a thrombocytopenia syndrome. Essential oils extracted from O. vulgare leaves cultivated in Germany, Albania, and Iran were analyzed. Among them, the German oil exhibited the highest acaricidal potency due to its elevated carvacrol content (83.38%). Carvacrol was isolated and its identity was confirmed using GC/MS, NMR, and FT-IR analyses. Carvacrol demonstrated significant toxicity across all developmental stages

Comparative Analysis of Bisexual and Parthenogenetic Populations in Haemaphysalis Longicornis.

Haemaphysalis longicornis, a three-host tick with a wide host range, is widely distributed in different countries and regions. It stands out among ticks due to its unique feature of having both parthenogenetic and bisexual populations. Despite their morphological resemblance, the characteristics of the parthenogenetic population have been overlooked. In this comprehensive study, we systematically compared the similarities and differences between these two populations. Our investigation revealed that the parthenogenetic H. longicornis, widely distributed in China, was found in ten provinces, su

Gut microbiota landscape of Haemaphysalis longicornis under Borrelia burgdorferi infection.

This study aimed to explore the reason for Haemaphysalis longicornis restricting Borrelia burgdorferi colonization and transmission from the perspective of gut microbiota, and to investigate the impact of different infection statuses on the Haemaphysalis longicornis microbiota and its potential role in pathogen transmission. Pathogen-free Haemaphysalis longicornis ticks and IFNAR1-/- mice were used to establish infection models. Ticks fed on pathogen-infected mice, and their midguts were analyzed at day 4 and 10 post-feeding. DNA was extracted from the midguts, and the V3-V4 region of bacteria

Baseline susceptibility of Haemaphysalis longicornis to organophosphate, carbamate, and pyrethroid acaricides.

BACKGROUND: The Asian longhorned tick, Haemaphysalis longicornis, continues to expand its range in North America, and synthetic acaricides are likely to play an increasing role in managing this species. Acaricide resistance is common in some tick species that infest livestock. However, baseline acaricide susceptibility has not previously been examined in this invasive tick. RESULTS: We used a standard larval packet test to evaluate the susceptibility of the Asian longhorned tick to acaricides currently or formerly used in tick control: propoxur, carbaryl, bifenthrin, permethrin, and coumaphos

Laboratory Evidence for Transovarial Transmission of Babesia gibsoni by Haemaphysalis longicornis.

Babesia gibsoni is a globally distributed tick-borne apicomplexan parasite that threatens dog health, yet its transmission dynamics in Haemaphysalis longicornis remain incompletely defined. Under controlled laboratory conditions, a Babesia-free parthenogenetic H. longicornis colony was used to evaluate transovarial transmission, transmission competence of the resulting F1 stages, retention of transmission competence after feeding on non-susceptible rabbits, and post-molt transmission after larval or nymphal acquisition feeding. Tick samples from the corresponding experimental lineages were scr

Localization of secreted ferritin (FER2) in the embryos of the tick Haemaphysalis longicornis.

Despite the absence of a blood meal, embryogenesis involves many processes that require nutrients and other essential elements, including iron. Due to the lack of an external source of these nutrients, these requirements are acquired maternally. Because of the toxic nature of iron, they are transferred through iron transport molecules such as secreted ferritin (FER2). Here we tried to follow the trail of the FER2 through indirect immunofluorescence, and we observed an apparent shift of FER2 from the germ layer at the early part of development to the appendages during the late stage of embryoge

Investigation of the presence of Ochrobactrum spp. and Brucella spp. in Haemaphysalis longicornis.

Ticks are common vectors of human and animal diseases. Ochrobactrum spp. belong to the Brucellaceae family and have recently been recognized as emerging human pathogens. The ability of Haemaphysalis longicornis ticks to carry Ochrobactrum spp. remains uncertain. During June and July 2018, 686 ticks were collected from 11 sites in Pingdingshan Henan province in central China. We extracted 169 DNA samples for Brucellaceae 16S rRNA nested PCR and sequenced them in order to identify Ochrobactrum spp. The data sequences were aligned with NCBI BLAST program and phylogenetic tree was constructed usin

Development of Babesia ovata in the ovary and eggs of the tick, Haemaphysalis longicornis.

The ovary and eggs of adult tick Haemaphysalis longicornis (H. longicornis) infected with Babesia ovata (B. ovata), were examined at regular intervals of time. As a result kinetes could be detected in the ovary for the first time 6 days after engorgement. The kinetics could be classified into three types on the basis of their morphological characteristics. On the 2nd day after oviposition, round-formed protozoa were found in the ooplasm of the egg. In 5 days after oviposition, these round-formed bodies were transformed into club-forms. On the 9th day after oviposition, the kinetes were observe

Babesia sp. BQ1 (Lintan): molecular evidence of experimental transmission to sheep by Haemaphysalis qinghaiensis and Haemaphysalis longicornis.

Ovine babesiosis is an economically important disease induced by tick transmitted haemoparasites throughout the world. In China, several ovine Babesia strains have been isolated from field-collected ticks or sheep blood during the last two decades but little is known about the vector ticks and transmission pattern. Babesia sp. BQ1 (Lintan) is a Babesia strain infective for sheep and goats, isolated from blood of sheep experimentally infested with Haemaphysalis qinghaiensis collected in field. In the present study, we explored the experimental transmission of Babesia sp. BQ1 (Lintan) to sheep b

Transcriptome of Coxiella-like endosymbionts in Haemaphysalis longicornis during blood feeding.

Coxiella-like endosymbionts (CLEs) are obligate nutritional mutualists of ticks that are thought to supplement B vitamins and other essential metabolites that are deficient in the tick's blood meal. Extensive colonization of CLEs has been observed in the ovaries and Malpighian tubules of several tick species, including Haemaphysalis longicornis. Despite experimental evidence linking symbionts to host fitness, their roles in blood feeding remain poorly understood. Here, we conducted a comparative transcriptomic analysis of CLE genes in adult female H. longicornis ticks before and two days after

Synchronous Langat Virus Infection of Haemaphysalis longicornis Using Anal Pore Microinjection.

The tick-borne encephalitis virus (TBEV) serocomplex of flaviviruses consists of arboviruses that cause important diseases in animals and humans. The transmission of this group of viruses is commonly associated with tick species such as Ixodes spp., Dermacentor spp., and Hyalomma spp. In the case of Haemaphysalis longicornis, the detection and isolation of flaviviruses have been previously reported. However, studies showing survival dynamics of any tick-borne flavivirus in H. longicornis are still lacking. In this study, an anal pore microinjection method was used to infect adult H. longicorni

RNA Virome Analysis of Haemaphysalis longicornis Ticks Collected in Hyogo, Japan.

Tick-borne viruses are primarily transmitted to vertebrates by infected ticks during blood feeding and cause various diseases in humans and animals. Haemaphysalis longicornis is one of the main tick species responsible for human tick bites and is thought to be the primary vector of severe fever with thrombocytopenia syndrome, an important tick-borne viral disease in Japan. Although H. longicornis ticks pose a potential risk to humans in Japan, studies on tick-borne viral prevalence of this tick species in Japan are limited. In this study, we conducted RNA virome analysis of H. longicornis tick

Acaricidal effects of diatomaceous earth on Haemaphysalis longicornis under laboratory conditions.

Ticks transmit various diseases to humans and animals, posing a significant global threat to public health. The primary method currently employed to control tick infestations is the use of chemical acaricides. However, improper use leads to environmental contamination, residue in meat and milk, and the development of acaricide-resistant ticks. Therefore, alternative tick control methods that are safe for humans, animals, and the environment and that do not induce resistance are urgently needed. Diatomaceous earth (DE), a naturally occurring physical insecticide, has been proven to effectively
