[Haemophilus influenzae vaccines].

The haemophilus vaccines are made of the type b capsular polysaccharide of haemophilus influenzae, polyribosylribitol phosphate (PRP), which is responsive of pathogenic power. However the responsiveness of children to PRP vaccine was found to be very poor in infants. The concept of conjugating carbohydrate with a carrier protein increasing immunogenicity of the PRP led to 4 PRP--protein conjugate vaccines: PRP-diphtheria toxoid conjugated vaccine (PRP-D), outer-membrane protein of Neisseria meningitidis conjugated (PRP-OMP), cross-reacting mutant diphtheria protein (PRP-HbOC) and tetanus-Toxoi

Haemophilus influenzae type b conjugate vaccines.

In summary, all of the Hib conjugate vaccines are highly immunogenic and efficacious in children older than 12-15 months of age, and HbOC, PRP-OMPC, and PRP-T are highly immunogenic and demonstrated to be efficacious in infants as young as 2 months old. HbOC, PRP-OMPC, and PRP-T have been licensed in numerous countries for infants and are recommended for infant immunization. However, perhaps the greatest tribute one can pay to all four Hib vaccines described in this review is to note the dramatic decrease in the incidence of Hib disease that has occurred since their introduction. In fact, acco

Research progress on Haemophilus parasuis vaccines.

Haemophilus parasuis (HPS) is the causative agent of porcine Glässer's disease, which has become prevalent in China in recent years. It is characterized by fibrinous polyserositis, arthritis, and meningitis, but often shows mixed infection with other upper respiratory tract pathogens, causing heavy economic losses to the pig industry. Vaccination is an important means to prevent and control HPS infection, and the currently available vaccines are mainly the inactivated type or subunit vaccines containing immunogenic HPS proteins. This study reviews recent advances in HPS vaccines, analyzes the

[Haemophilus vaccines. Their importance in tropical pediatrics].

Immunization with Haemophilus vaccine is undertaken to prevent invasive infections due to Haemophilus influenzae type b. Purulent meningitis is the most frequent chiefly in less than 2 years infants. Vaccine is composed of purified PRP which is poorly immunogenic and protective before two years. Actual vaccines are conjugate with protein which give them a protective and immunogenic power in very long young infants. They must be included in the schedule of infants immunizations, and fitting to other immunizations (DTP-Polio) is the best. The frequency of H. influenzae meningitis is high in trop

Vaccines for Nontypeable Haemophilus influenzae: the Future Is Now.

Infections due to nontypeable Haemophilus influenzae result in enormous global morbidity in two clinical settings: otitis media in children and respiratory tract infections in adults with chronic obstructive pulmonary disease (COPD). Recurrent otitis media affects up to 20% of children and results in hearing loss, delays in speech and language development and, in developing countries, chronic suppurative otitis media. Infections in people with COPD result in clinic and emergency room visits, hospital admissions, and respiratory failure. An effective vaccine would prevent morbidity, help contro

Update on Haemophilus influenzae serotype b and meningococcal vaccines.

Use of conjugate Haemophilus influenzae type b (Hib) vaccines has resulted in the near elimination of Hib invasive disease among infants in the United States in only 10 years, which places this intervention among the most notable public health achievements of the past decade. This has radically altered our perception of the major causes of bacterial meningitis and invasive bacterial disease among children, increasing the prominence of meningococcal disease as an important cause of childhood and adult meningitis and leading researchers to apply the same conjugate technology to the development o

Stability testing in Haemophilus influenzae type b conjugate vaccines.

Coupling T-cell-independent antigens such as the capsular-polysaccharide (PRP) of H. influenzae type b (HIB) to a T-cell-dependent protein-carrier enhances the immune response, particularly in individuals who are poor responders to such antigens. An adequate immunogenicity is dependent upon the characteristics of the PRP antigen, the protein carrier, the efficiency of conjugation and the stability of the conjugate. Since adequate immunogenicity tests are not available, stability studies in such products are essentially confined to molecular size distribution and determination of the degree of

Conjugate Haemophilus influenzae type b vaccines for sickle cell disease.

BACKGROUND: People affected with sickle cell disease are at high risk of infection from Haemophilus influenzae type b. Before the implementation of Haemophilus influenzae type b conjugate vaccination in high-income countries, this was responsible for a high mortality rate in children under five years of age. In African countries, where coverage of this vaccination is still extremely low, Haemophilus influenzae type b remains one of the most common cause of bacteraemias in children with sickle cell disease. The increased uptake of this conjugate vaccination may substantially improve the surviva

Haemophilus influenzae type b conjugate vaccines - a South African perspective.

Introduction of Hib vaccine is known to positively impact on reduction of both morbidity and mortality in children less than 5 years of age. Incorporation of this vaccine into a National EPI, however, does come at a significant cost, which is especially important in non-GAVI funded countries. Compounded reduction in response in certain patient populations and possible indication of booster doses further impacts on cost-benefit analyses. Despite these issues, South Africa has supplied Hib vaccine as part of the National EPI in the form of a combination vaccine, Pentaxim, which combines Hib with

Haemophilus influenzae type b polysaccharide-protein conjugate vaccines in children.

Prevention of invasive infections caused by Haemophilus influenzae type b (Hib) bacteria has become possible with new vaccines. Covalent coupling to a protein carrier increases the immunogenicity of Hib capsular polysaccharide, and changes it to a T-cell dependent antigen. In clinical use, the conjugate vaccines are immunogenic in infancy, induce immunologic memory, and protect efficiently from invasive infections. They even seem to reduce oropharyngeal carriage of Hib bacteria. Wide-scale use of Hib conjugate vaccines has demonstrated their safety and low reactogenicity. Studies to combine th

Hib Vaccines: Their Impact on Haemophilus influenzae Type b Disease.

Haemophilus influenzae serotype b (Hib) is an important cause of serious, invasive infections, particularly in young children. Since 1985, a series of vaccines composed of the type b capsular polysaccharide polyribosylribitol phosphate (PRP), followed by PRP conjugated to various proteins, have been licensed for use in the United States and worldwide. The conjugated vaccines offer increased immunogenicity and prolonged durability of immune protection compared to the plain PRP vaccine and increasingly are combined with other childhood vaccines for decreased cost and increased ease of vaccinatio

Combination vaccines for diphtheria, tetanus, pertussis, and Haemophilus influenzae type b.

The ability to combine the standard DTP and Haemophilus b conjugate vaccine considerably simplifies the childhood immunization schedule and process. In addition to reducing the number of immunizations by half, the combination product reduces administrative aspects associated with vaccination including tracking. Simplification of the immunization process should have a positive impact on the vaccine delivery and utilization. Perhaps more importantly, an ability to create combination vaccines will be critical for inclusion of new antigens appropriate for infant vaccines. The combination of DTP an

Efficacy and Prolonged Safety of Haemophilus influenzae Type b Conjugate Vaccines.

OBJECTIVE: The purpose of this study was to find data proving the influence of the Haemophilus influenzae type b (Hib) conjugate vaccination on the frequency of invasive Hib illness. METHODOLOGY: A systematic literature search was conducted on the PubMed database to identify peerreviewed publications pertaining to the epidemiology of Haemophilus influenzae meningitis, both before and after the introduction of Haemophilus influenzae type b (Hib) conjugate vaccines. The search query employed a combination of relevant keywords, including "invasive," "Haemophilus," "influenzae," "meningitis," and

Challenges for the development of vaccines against Haemophilus influenzae and Neisseria meningitidis.

The development of protein-polysaccharide conjugate vaccines has had a major impact on Haemophilus influenzae type b disease. The application of this technology to Neisseria meningitidis is also striking, particularly for serogroup C. However, significant challenges exist for the development of vaccines against non-typeable H. influenzae and against N. meningitidis serogroup B. Issues such as non-vaccine-strain replacement and correlates of protection need to be addressed as well as the longer-term implications of vaccination against what are essentially 'normal' microflora.

Siblings of patients with Haemophilus meningitis have impaired anticapsular antibody responses to Haemophilus vaccine.

Siblings of patients with type b Haemophilus influenzae meningitis are at increased risk of developing Haemophilus disease. We immunized 26 healthy siblings and 25 control subjects using a vaccine containing the type b polysaccharide capsule (10 micrograms PRP) and pertussis vaccine (4 opacity units) (Lederle Laboratories) to determine whether siblings of patients with Haemophilus meningitis had an impaired antibody response to PRP. Using two intramuscular injections one month apart, we found that the siblings had a lower response to PRP. One month after the second injection, 12 of 24 of the s

Long Term Impact of Conjugate Vaccines on Haemophilus influenzae Meningitis: Narrative Review.

H. influenzae serotype b (Hib) used to be the commonest cause of bacterial meningitis in young children. The widespread use of Hib conjugate vaccine has profoundly altered the epidemiology of H. influenzae meningitis. This short review reports on the spectrum of H. influenzae meningitis thirty years after Hib conjugate vaccine was first introduced into a National Immunization Program (NIP). Hib meningitis is now uncommon, but meningitis caused by other capsulated serotypes of H. influenzae and non-typeable strains (NTHi) should be considered. H. influenzae serotype a (Hia) has emerged as a sig
