A genomic DNA library of Haemophilus paragallinarum strain Modesto was created. Screening of this library identified four clones that reacted specifically with all 56 isolates of H. paragallinarum tested and failed to react with 24 closely related bacteria from the genera Pasteurella and Actinobacillus. All four clones also failed to react with DNA extracted from one field isolate each of Mycoplasma gallisepticum and Mycoplasma synoviae. The probes based on these four clones were approximately 1.8, 2.3, 3.5, and 5.5 kb in size. The four probes were able to detect between 7.8 and 31.25 ng of pu

The efficacy of the antimicrobial drug norfloxacin for treating infectious coryza was examined in 26-week-old male broiler breeders. Chickens were inoculated in the infraorbital sinus with the causal organism, Haemophilus paragallinarum. Four experimental groups were set up: control uninfected chickens, infected untreated chickens, and infected chickens treated for 5 days with either 20 mg norfloxacin/kg body weight or 40 mg norfloxacin/kg body weight. The first clinical signs were seen 24 hr postinfection. Of the observed clinical signs, sinus edema was ameliorated by the treatment, and the p

The enterobacterial repetitive intergenic consensus polymerase chain reaction (ERIC-PCR) technique was used for fingerprinting of reference strains and Mexican isolates of Haemophilus paragallinarum. A total of nine ERIC patterns were given by the nine serovar reference strains of this bacteria. Two Modesto (C-2) reference strains from different sources showed the same ERIC pattern. Seventeen ERIC patterns were obtained among 29 Mexican isolates included in the study, belonging to serovars prevalent in Mexico (A-1, A-2, B-1, and C-2). Obtained results indicate that the ERIC-PCR technique could

Avibacterium paragallinarum is the pathogen involved in infectious coryza (IC), an acute infectious upper respiratory disease in chickens. The prevalence of IC has increased in China in recent years. There is a lack of reliable and effective procedures for gene manipulation, which has limited the research on the bacterial genetics and pathogenesis of A. paragallinarum. Natural transformation has been developed as a method of gene manipulation in Pasteurellaceae by the introduction of foreign genes or DNA fragments into bacterial cells, but there has been no report on natural transformation in

We report Avibacterium paragallinarum and Klebsiella pneumoniae coinfection in a grey crowned crane (Balearica regulorum). The crane was recovered from illegal captivity and released at a grey crowned crane (GCC) rehabilitation facility located at Akagera National Park in Rwanda. One year after being transferred, the bird presented with clinical signs suggesting a respiratory disease. Those signs included severe dyspnoea with mouth breathing, sneezing and nasal discharge. The crane was put on a 3-day treatment with antibiotics (ceftiofur 200 mg/ml at 50 mg/kg intramuscularly) and anti-inflamma

The bacterium Avibacterium paragallinarum is the etiologic agent of infectious coryza of chickens. There are nine serovars of A. paragallinarum , and serovar C-1 has emerged in outbreaks of infectious coryza in layer hens in the Americas, with all isolates having been obtained from infectious coryza-vaccinated chickens. In the current study, the clinical and histopathologic outcomes of experimental infections in chickens with A. paragallinarum of serovar C-1 were investigated. The Japanese serovar reference strain, H-18, and a Mexican isolate, ESV-135, were included in the study. No difference

The resistance to anti-microbial agents of bacteria isolated from pathological conditions of birds in Victoria, 1978 to 1983, was determined for isolates of Escherichia coli, Salmonella species, Staphylococcus aureus, Pasteurella multocida, P. anatipestifer, Yersinia pseudotuberculosis and Haemophilus paragallinarum. The isolates of E. coli had a high prevalence of resistance to tetracycline and sulphonamides, and a lower prevalence of resistance to furazolidone and sulphamethoxazole-trimethoprim. The isolates of Salmonella spp commonly had resistance to tetracycline, sulphonamides, furazolido

Infectious coryza (IC) is a respiratory disease of chickens, including pullets, layers, and broilers, caused by the bacteria Avibacterium paragallinarum (AP), which was previously known as Hemophilus gallinarum. IC classically causes production decreases and mortality in chickens, frequently paired with swelling of the sinuses, mucoid nasal discharge, and respiratory rales. Although IC is considered an endemic disease of chickens in California, it has been unusual to rare in commercial chickens in Pennsylvania. The last reported IC case in Pennsylvania was in 2002, involving broiler breeders.

Seventeen complicated outbreaks of infectious coryza in layer, broiler-breeder, and broiler flocks were studied. In the layer flock outbreaks, drops in egg production of up to 35% were seen. In the broiler flocks and several of the layer flocks, losses due to persistent mortality and/or culling varied between 2 and 5%. Signs of infectious coryza in both layers and broiler-breeders were typical; in broilers, however, swollen head-like syndrome was seen. Except in one flock, no viral diseases were clinically or serologically detected. Excluding broiler-breeders, birds from most other flocks were

Infectious coryza (IC) is an acute infectious upper respiratory disease in chickens. Recently, the prevalence of IC has increased in China. In this study, to clarify the pathogenic mechanism and innate immune response of Avibacterium paragallinarum (A. paragallinarum), an infection experiment with A. paragallinarum was conducted. Our results showed that the whole course of IC was approximately 7 d. The clinical signs score was highest at 3 dpi and decreased from 5 dpi. A large amount of mucus and exudates was found in the infraorbital sinuses and nasal cavity. The A. paragallinarum contents in

Infectious coryza (IC) caused by Avibacterium paragallinarum (AP) has risen in importance as a poultry disease over the past several years because of its increased incidence in commercial poultry in both Europe and the United States. Because of this rise in importance, more attention has been focused on diagnosis, isolation, and surveillance of this bacterial pathogen. As a result, new knowledge has been produced and published. This review was compiled with the main purpose of summarizing and presenting the updated knowledge available about AP. However, the new knowledge can only be understood

Avibacterium paragallinarum (Av. paragallinarum), the causative agent of infectious coryza, is a significant pathogen responsible for substantial economic losses in the poultry industry. Current preventive strategies rely primarily on inactivated vaccines, which have limitations such as vaccine failure and limited cross-protection between serotypes. This study aimed to develop an attenuated strain of Av. paragallinarum as a potential live vaccine candidate. Using the Tn5-Kan transposon, we constructed a transposon mutant library and identified a mutant strain, designated 2019/HB64-40, which ha

Infectious coryza (IC) is a chicken respiratory disease caused by Avibacterium paragallinarum (AP). Recently, multiple naïve-healthy layer (NHL) flocks tested positive for AP by both qPCR and bacterial isolation without clinical signs. Therefore, acquired AP isolates from these NHL flocks were dubbed non-pathogenic AP (npAP). The aim of this study was to investigate the pathogenicity of npAP isolates (Phase I) and their ability to confer protection against IC (Phase II). Seven groups (G1-G7) of 26-week-old laying hens were challenged via intra-sinus injection with seven npAP isolates obtained

The diagnosis of a concurrent infection of Avibacterium paragallinarum and fowl adenovirus (FAdV) in an infectious coryza-like outbreak in the outskirt of Beijing is reported. The primary signs of the infection were acute respiratory signs, a drop in egg production, and the presence of hydropericardium-hepatitis syndrome-like gross lesions. Laboratory examination confirmed the presence of A. paragallinarum by bacterial isolation and a species-specific PCR test. In addition, conventional serotyping identified the isolates as Page serovar A. Fowl adenovirus was isolated from chicken liver specim

Infectious coryza (IC), caused by Avibacterium paragallinarum (Av. paragallinarum), imposes a substantial economic burden on global poultry production. This study established and standardized comparative challenge models employing infraorbital sinus injection and intranasal inoculation to evaluate the pathogenicity of Av. paragallinarum serovars A, B, and C in SPF chickens. Infraorbital sinus injection leads to rapid onset, stable incidence, and more severe disease progression. The clinical symptom scores at 1 days post-infection (dpi) were higher in the infraorbital sinus injection group comp

Inactivated infectious coryza vaccines containing different adjuvants were administered to 6-week-old chickens as a single dose containing 10(8) colony-forming units of Haemophilus paragallinarum HP31. After 3 weeks, all chickens were challenged by intrasinus inoculation of HP31. Two vaccines, one containing an aluminum-hydroxide adjuvant and the other a combined aluminum-hydroxide + avridine (a lipoidal amine) adjuvant, were effective. The three remaining vaccines--containing mineral-oil double-emulsion, avridine, or a combination of mineral-oil double-emulsion + avridine--gave much lower lev

Two isolates of haemophilic bacteria originally isolated in the 1980s from chickens were re-examined. The addition of a 10% sterile filtrate from an overnight culture of Staphylococcus epidermidis allowed growth of both isolates in solid and liquid media that were otherwise not capable of supporting the growth of these isolates. Using the modified media, genotypic and serotypic studies were performed, which confirmed both isolates to be Avibacterium paragallinarum, with one isolate being serovar A and the other serovar C. The unusual growth requirements of these two isolates reinforces the nee
