A comparison of the brains of lampreys and hagfishes is carried out in an attempt to reconstruct the anatomy of the brain of the last common ancestor of craniates: i.e., the morphotype of the craniate brain. This brain consisted of tel-, di-, mes-, and rhombencephalic divisions; the presence of a metencephalic/cerebellar division is questionable. All major sensory and motor systems (with the possible exception of the oculomotor system) that are typical of craniates were present in the morphotype. There were extensive bilateral secondary olfactory projections to the telencephalic pallium, as we

As the only surviving lineages of jawless fishes, hagfishes and lampreys provide a critical window into early vertebrate evolution. Here, we investigate the complex history, timing, and functional role of genome-wide duplications in vertebrates in the light of a chromosome-scale genome of the brown hagfish Eptatretus atami. Using robust chromosome-scale (paralogon-based) phylogenetic methods, we confirm the monophyly of cyclostomes, document an auto-tetraploidization (1RV) that predated the origin of crown group vertebrates ~517 Mya, and establish the timing of subsequent independent duplicati

The discovery of an albino specimen of the hagfish, Eptatretus stoutii (Lockington), is reported, together with general notes regarding collection of this species for research purposes. The extreme abundance of hagfish in the area described refutes the generally accepted notion that they are of infrequent occurrence in southern California waters.

Myxine limosa is a burrowing species of hagfish that occurs in the western North Atlantic in areas with muddy substrate and at depths generally greater than 100 meters. Burrowing of M. limosa has been observed from submersibles, but little is known about the behavior of these animals within the substrate or the biomechanical mechanisms involved. Here, we investigated burrowing in M. limosa by observing individuals as they burrowed through transparent gelatin. A photoelastic setup using crossed polarizers allowed us to visualize stress development in the gelatin as the hagfish moved through it.

When attacked, hagfishes produce a soft, fibrous defensive slime within a fraction of a second by ejecting mucus and threads into seawater. The rapid setup and remarkable expansion of the slime make it a highly effective and unique form of defense. How this biomaterial evolved is unknown, although circumstantial evidence points to the epidermis as the origin of the thread- and mucus-producing cells in the slime glands. Here, we describe large intracellular threads within a putatively homologous cell type from hagfish epidermis. These epidermal threads averaged ~2 mm in length and ~0.5 μm in di

Hagfish are the most primitive craniates and scavengers, feeding on dead organisms as well as fish and invertebrates. Hagfish play an important ecological role in recycling nutrients, helping to recycle biomass from the upper water column. We investigated 265 specimens of four hagfish species, including Eptatretus burgeri, Eptatretus yangi, Eptatretus sheni and Eptatretus taiwanae from northeastern Taiwanese waters of the northwestern Pacific from November 2013 to June 2014. Eight species of Anisakid nematodes were identified: Anisakis pegreffii, Anisakis simplex s.s., a recombinant genotype o

The hypothalamic-pituitary system is considered to be a seminal event that emerged prior to or during the differentiation of the ancestral agnathans (jawless vertebrates). Hagfishes as one of the only two extant members of the class of agnathans are considered the most primitive vertebrates known, living or extinct. Accordingly, studies on their reproduction are important for understanding the evolution and phylogenetic aspects of the vertebrate reproductive endocrine system. In gnathostomes (jawed vertebrates), the hormones of the hypothalamus and pituitary have been extensively studied and s

Hagfish produce extraordinary slime as a defense mechanism, releasing exudate from glands that rapidly form a fibrous, soft, ultra-dilute, water-capturing network upon contact with seawater (up to 10 000 times its original volume). The gland thread cell (GTC) produces high-strength protein threads (filament diameter df = 1-3 µm) meticulously coiled into skeins (coil diameter Do ∼ 150 µm) that rapidly unravel upon deployment to reveal their hidden length (Lf = 15 cm), forming a cohesive fibrous slime network through interaction with mucin vesicles and seawater. To date, no engineered material i

The pituitary gland is present in all vertebrates, from agnathans (jawless vertebrates) to mammals, but not in invertebrates. Reproduction in gnathostomes (jawed vertebrates) is controlled by two pituitary gonadotropins (GTHs), luteinizing hormone and follicle-stimulating hormone, which are part of the pituitary glycoprotein hormone (GPH) family. Hagfishes, which lack both jaws and vertebrae, are considered the most primitive vertebrate known, living or extinct. Accordingly, they are of particular importance in understanding the evolution of the pituitary GPHs and their functions related to ve

Hagfishes may encounter low dissolved oxygen in their natural habitats, a consequence of association with hypoxic sediments and their feeding behaviour. In teleost fish, hypoxia exposure decreases ion uptake, speculated to be a mechanism for energy conservation. Although hagfishes osmoconform, they do regulate extracellular fluid concentrations of divalent cations such as calcium. The current study hypothesised that exposure of hagfish to hypoxia (0.4 kPA, 24 h) would reduce calcium uptake (determined via in vitro isolated skin and gut epithelial transport assays) and calcium accumulation (det

Modern views of agnathan phylogeny consider Petromyzoniformes and Myxiniformes to belong to distinct classes that diverged from a common ancestor at a remote period, perhaps in the lower Cambrian, greater than 600 million years ago. Both are more primitive than elasmobranchs, holocephalans and bony fishes. Myelin is well developed in elasmobranchs and other fishes but was reported to be lacking in the spinal cord of lampreys. In order to search further for possible early myelin in some part of the nervous system of one of the agnathan stems, or for further evidence that it first appeared in ch

Hagfishes are unique to the vertebrate lineage in that they acquire dissolved nutrients across multiple epithelia including the intestine, gill, and skin. This feat has been attributed to their immersive feeding behavior that likely simultaneously provides benefits (nutrient rich) and potentially adverse (hypercapnia, hypoxia, high environmental ammonia) physiological effects. Examinations have been conducted of the ex vivo transport capabilities of specific nutrients as well as in vivo effects of the hypothesized feeding environments, yet the physiological effects of feeding itself have never

The capillary tuft of glomeruli of the hagfish mesonephros contains both 'low'-pressure and 'high'-pressure glomerular vessels (LPGVs and HPGVs). The existence of the HPGV raised the possibility that pressure filtration could occur in the hagfish kidney when the blood pressure was sufficiently high. Therefore, measurements of glomerular capillary pressure were made in HPGVs and LPGVs whilst single glomeruli were perfused with hagfish Ringer's solution that contained the colloid Ficoll 70. Calculations of the effective colloid osmotic pressure in perfused capillaries were made; these showed tha

By recording the locomotor activity rhythm of hagfish, Eptatretus burgeri, in which surgical lesions of the preoptic nucleus (PON) were made, we searched for the location of the circadian pacemaker in the hypothalamus. The characteristic rhythms were lost in animals lacking the anterior part of the hypothalamus, which includes the PON. Cuts in front of the PON did not affect the rhythm, but cuts behind the PON caused the animals to loose their rhythms. Destruction of the PON with a high-frequency lesion generator also caused loss of rhythms. These findings indicate that the pacemaker may be lo

The brain of the Pacific hagfish, Eptatretus stouti, was studied immunocytochemically using antisera against somatostatin (SRIH), arginine vasopressin (AVP), and adrenocorticotropic hormone (ACTH). SRIH-immunoreactive perikarya were distributed bilaterally in the postoptic nucleus and in the hypothalamic nucleus. Although several short, stained fibers were observed in the vicinity of the perikarya, SRIH-immunoreactivity was not found in the neurohypophysis, nor in other parts of the brain. On the other hand, presumed arginine vasotocin (AVT) perikarya were distributed in an arc-shaped region e

Acidification is a key component of digestion throughout metazoans. The gut digestive fluid of many invertebrates is acidified by the vesicular-type H+-ATPase (VHA). In contrast, vertebrates generate acidic gut fluids using the gastric H+/K+-ATPase (HKA), an evolutionary innovation linked with the appearance of a true stomach that greatly improves digestion, absorption and immune function. Hagfishes are the most basal extant vertebrates, and their mechanism of digestive acidification remains unclear. Herein, we report that the stomachless Pacific hagfish (Eptatretus stoutii) acidify their gut

Prolactin-releasing peptide2 (PrRP2) belongs to the RFamide peptide group and is a paralog of prolactin-releasing peptide (PrRP). Recent studies demonstrated that PrRP2, but not PrRP, regulates prolactin release in teleosts. The evolutionary origin of PrRP and PrRP2 dates back to at least early vertebrates because homologs of PrRP/PrRP2 were identified in lampreys, one of the earliest branch of vertebrates class Agnatha. However, PrRP/PrRP2 remains to be identified in hagfish, another representative species of class Agnatha. Here, we examined the distribution of PrRP2 in the brain and pituitar
