A new species of Haliclona (Demospongiae: Haplosclerida: Chalinidae) from southeastern Brazil and the first record of Haliclona vansoesti from the Brazilian coast.

In this paper we describe two species of the cosmopolitan sponge genus Haliclona from Rio de Janeiro State, SE Brazil, one of which is new to science and the other a new record to Brazil. Haliclona (Rhizoniera) fugidia sp. nov. is brownish-pink, salmon or cream, thickly encrusting, without oscular tubes and tangential ectosomal reticulation. Choanosomal skeleton is a mostly unispicular ladder-like reticulation of oxeas, very organized near the sponge surface and denser and more disorganized in the interior of the sponge. Haliclona (Halichoclona) vansoesti de Weerdt et al., 1999 was originally

Percutaneous extraction using sponge spicules for disease diagnosis and therapeutic drug monitoring.

Diagnostic and therapeutic drug monitoring are necessary means of precision medicine. Our previous study demonstrated that the sponge Haliclona sp. spicules (SHS) can be employed as a new class of microneedles, capable of forming a substantial number of microchannels within the skin. In this study, dermal interstitial fluid (ISF) was obtained via percutaneous extraction using sponge Haliclona sp. spicules (SHS) to explore the correlation between the concentration of target compounds in the extract and their content in the skin or blood. Methodologically, we compared SHS with dermaroller, ultra

Marine Natural Compound (Neviotin A) Displays Anticancer Efficacy by Triggering Transcriptomic Alterations and Cell Death in MCF-7 Cells.

We investigated the anticancer mechanism of a chloroform extract of marine sponge (Haliclona fascigera) (sample C) in human breast adenocarcinoma (MCF-7) cells. Viability analysis using MTT and neutral red uptake (NRU) assays showed that sample C exposure decreased the proliferation of cells. Flow cytometric data exhibited reactive oxygen species (ROS), nitric oxide (NO), dysfunction of mitochondrial potential, and apoptosis in sample C-treated MCF-7 cells. A qPCR array of sample C-treated MCF-7 cells showed crosstalk between different pathways of apoptosis, especially BIRC5, BCL2L2, and TNFRS

Enhanced skin delivery of biomacromolecules using hollow sponge spicules.

This study introduces hollow sponge Haliclona sp. spicules (oSHS) as an innovative microneedle technology for enhancing the skin delivery of biomacromolecules. oSHS were developed by etching SHS in 2 mol/L NaOH for 48 h, optimizing hollow channel formation while preserving structural integrity. In vitro experiments demonstrated that oSHS significantly enhanced drug absorption efficiency compared to SHS, microneedles, roller microneedles, and iontophoresis alone. Combining oSHS with iontophoresis yielded a synergistic effect, increasing biomacromolecule delivery by up to 35.09% (p <0.05). In vi

The topical application of Sphistin12-38 in combination with sponge spicules for the acne treatment.

We demonstrated for the first time that a marine-derived antimicrobial peptide (AMP), Sph12-38, exhibit high antimicrobial activity against P. acnes with a minimum bactericidal concentration (MBC) value of 7 μM. Meanwhile, Sph12-38 has no significant cytotoxicity to human keratinocytes (HKs) at its high concentration (33.5 μM). The topical application of sponge Haliclona sp. spicules (SHS) dramatically enhanced the skin penetration of Sph12-38 up to 40.9 ± 5.9% (p < 0.01), which was 6.1 ± 0.9-fold higher than that of Sph12-38 alone. Further, SHS resulted in the accumulation of most Sph12-38 in

Marine Organisms from the Yucatan Peninsula (Mexico) as a Potential Natural Source of Antibacterial Compounds.

A total of 51 sponges (Porifera) and 13 ascidians (Chordata) were collected on the coast of the Yucatan Peninsula (Mexico) and extracted with organic solvents. The resulting extracts were screened for antibacterial activity against four multidrug-resistant (MDR) bacterial pathogens: the Gram-negative Acinetobacter baumannii, Klebsiella pneumoniae, and Pseudomonas aeruginosa and the Gram-positive Staphylococcus aureus. The minimum inhibitory concentrations (MICs) of the organic extracts of each marine organism were determined using a broth microdilution assay. Extracts of eight of the species,

Unlocking Marine Microbial Metabolites of Sponges and Sediments as a Source of Anti-Trypanosoma cruzi Compounds.

Marine microbial metabolites have long served as valuable sources for drug discovery, exhibiting diverse biological activities, including antiparasitic, antimicrobial, and antiviral effects. Chagas disease (CD), a neglected tropical disease caused by Trypanosoma cruzi, affects approximately 8 million people worldwide. Currently, only two drugs are available for treatment, both associated with significant toxicity, underscoring the urgent need for new therapeutic alternatives. In this context, and considering the remarkable chemical diversity of marine bacteria, the present study investigated t

Sponge cell reaggregation: Cellular structure and morphogenetic potencies of multicellular aggregates.

Sponges (phylum Porifera) are one of the most ancient extant multicellular animals and can provide valuable insights into origin and early evolution of Metazoa. High plasticity of cell differentiations and anatomical structure is characteristic feature of sponges. Present study deals with sponge cell reaggregation after dissociation as the most outstanding case of sponge plasticity. Dynamic of cell reaggregation and structure of multicellular aggregates of three demosponge species (Halichondria panicea (Pallas, 1766), Haliclona aquaeductus (Sсhmidt, 1862), and Halisarca dujardinii Johnston, 18

Coated sponge spicules for remodeling skin immune homeostasis in psoriasis treatment by releasing self-assembled phospholipid complex nanoparticles.

Psoriasis is an autoimmune skin disease where dendritic cells play a crucial role. In psoriasis topical treatment, pathologically-induced thickening of the stratum corneum and epidermal hyperplasia severely hinder the penetration of drugs into the deep skin, thereby reducing therapeutic efficacy. Here, we developed the curcumin-POPG phospholipid complex coated Sponge Haliclona sp. Spicules (CUR-Ph-SHS) with the supercritical antisolvent-fluidized bed coating (SAS-FB) technology. This innovative formulation was designed to enhance skin permeation and suppress dendritic cell activation in the ps

Marine Natural Products and Drug Resistance in Latent Tuberculosis.

Pyrazinamide (PZA) is the only drug for the elimination of latent Mycobacterium tuberculosis (MTB) isolates. However, due to the increased number of PZA-resistance, the chances of the success of global TB elimination seems to be more prolonged. Recently, marine natural products (MNPs) as an anti-TB agent have received much attention, where some compounds extracted from marine sponge, Haliclona sp. exhibited strong activity under aerobic and hypoxic conditions. In this study, we screened articles from 1994 to 2019 related to marine natural products (MNPs) active against latent MTB isolates. The
