Comparative genomics and metabolic model analysis of primary and secondary endosymbionts present in different species of whitefly.

Candidatus Portiera aleyrodidarum (hereafter Portiera) is the primary endosymbiont of whiteflies (Hemiptera: Aleyrodidae), residing in specialized bacteriocytes that also co-localize diverse secondary endosymbionts. This study aimed to carry out in-depth genomic analysis of different strains of Portiera and secondary endosymbionts namely Arsenophonus, Cardinium, Hamiltonella, Rickettsia, and Wolbachia obtained from different whitefly species (data obtained from NCBI). Interestingly, hierarchical clustering of the amino acid identity data resulted in a dendrogram that resolved the different str

Avocado seed waste bioconversion into poly(3-hydroxybutyrate) by using Cobetia amphilecti and ethyl levulinate as a green extractant.

The avocado processing industry produces up to 1.3M tons of agro-waste annually. Chemical analysis of avocado seed waste (ASW) revealed that it is rich in carbohydrates (464.7 ± 21.4 g kg-1) and proteins (37.2 ± 1.5 g kg-1). Optimized microbial cultivation of Cobetia amphilecti using an acid hydrolysate of ASW, generated poly(3-hydroxybutyrate) (PHB) in a 2.1 ± 0.1 g L-1 concentration. The PHB productivity of C. amphilecti cultivated on ASW extract was 17.5 mg L-1 h-1. The process in which a novel ASW substrate was utilized has been further augmented by using ethyl levulinate as a sustainable

Characterization and bioactivities of glutaminase-free L-asparaginase from Salinicola acorporae S4-41.

BACKGROUND: The medical community has long searched for a safer and more effective cancer treatment method. Clinically, L-asparaginase derived from bacteria has shown promising results. However, it carries the risk of immunological and hypersensitive reactions due to its interaction with glutaminase. Therefore, finding a new source of glutaminase-free L-asparaginase (ASNase) is important. RESULTS: In this study, 35 bacterial isolates from 12 saline environmental samples were tested for ASNase, and the most promising bacterium was identified as Salinicola acroporae S4-41. This bacterium produce

Catalase-peroxidase StKatG is a bacterial manganese oxidase from endophytic Salinicola tamaricis.

Manganese (Mn) oxides in iron/manganese plaques are widely distributed in the rhizosphere of wetland plants and contribute significantly to elemental cycling and pollutant removal. Mn oxides are primarily produced by bacterial processes using Mn oxidases. However, the molecular mechanism underlying the formation of rhizosphere Mn oxides is still largely unknown. This study identified a manganese-oxidizing enzyme, the catalase-peroxidase StKatG, from an endophytic bacterium Salinicola tamaricis from the wetland plant. The gene encoding StKatG was cloned and overexpressed in Escherichia coli. Th

An engineered bacterial symbiont maps micron-scale sugar gradients in the honeybee gut.

The honeybee gut microbiota plays a key role in shaping host health and susceptibility to disease. Yet, the nutrient environment it experiences within the gut remains poorly characterized. In particular, little is known about the spatial distribution of nutrients across the microbial community, as resolving such fine gradients in vivo has been technically challenging. Here, we engineer the native honeybee symbiont Snodgrassella alvi as a living biosensor to quantify the bioavailability of the dietary sugar arabinose within the gut. By expanding the genetic toolkit for S. alvi through chromosom

Portiera Gets Wild: Genome Instability Provides Insights into the Evolution of Both Whiteflies and Their Endosymbionts.

Whiteflies (Hemiptera: Sternorrhyncha: Aleyrodidae) are a superfamily of small phloem-feeding insects. They rely on their primary endosymbionts "Candidatus Portiera aleyrodidarum" to produce essential amino acids not present in their diet. Portiera has been codiverging with whiteflies since their origin and therefore reflects its host's evolutionary history. Like in most primary endosymbionts, the genome of Portiera stays stable across the Aleyrodidae superfamily after millions of years of codivergence. However, Portiera of the whitefly Bemisia tabaci has lost the ancestral genome order, refle

Analysis of the Composition and Functions of the Microbiome in Diabetic Foot Osteomyelitis Based on 16S rRNA and Metagenome Sequencing Technology.

Metagenome sequencing has not been used in infected bone specimens. This prospective observational study explored the microbiome and its function in patients with diabetic foot osteomyelitis (DFO) and posttraumatic foot osteomyelitis (PFO) based on 16S rRNA sequencing and metagenome sequencing technologies. Spearman analysis was used to explore the correlation between dominant species and clinical indicators of patients with DFO. High-throughput sequencing showed that all the specimens were polymicrobial. The microbial diversity was significantly higher in the DFO group than in the PFO group.

Structure of the Gut and Ovary, with Associated Microbiota Across Life Stages in the Striped Stem Borer Chilo suppressalis (Lepidoptera: Crambidae).

The striped stem borer Chilo suppressalis (Lepidoptera: Crambidae) is one of the most serious pests of water bamboo Zizania latifolia. Microbiota dynamics across the life cycle of C. suppressalis are a prerequisite for comprehending the symbiotic relationship between C. suppressalis and its microbiota. In this study, we characterized the structural features of the gut and ovary, as well as the associated microbiota of C. suppressalis. The gut is anatomically divided into the foregut, midgut and hindgut. Notable structural and ultrastructural differences were observed between adults and larvae,

Cyanide treatment of mining tailings using suspended biomass and moving bed biomass reactors.

Mexico is the top producer of silver and is on the eighth place from producing gold in the world. For instance, the hydrometallurgical extraction process produces wastewater (mining tailing) characterized by being composed with varying concentrations of cyanide and heavy metals. The purpose of this research was to study the biodegradation of cyanide contained in mining tailings by means of a bacterial consortium isolated from a tailings dam. For this purpose, three types of Eckendfelder reactors were used, one with suspended biomass (BS) and two moving bed biofilter reactors, one with biomass

GFAJ-1 is an arsenate-resistant, phosphate-dependent organism.

The bacterial isolate GFAJ-1 has been proposed to substitute arsenic for phosphorus to sustain growth. We have shown that GFAJ-1 is able to grow at low phosphate concentrations (1.7 μM), even in the presence of high concentrations of arsenate (40 mM), but lacks the ability to grow in phosphorus-depleted (<0.3 μM), arsenate-containing medium. High-resolution mass spectrometry analyses revealed that phosphorylated central metabolites and phosphorylated nucleic acids predominated. A few arsenylated compounds, including C6 sugar arsenates, were detected in extracts of GFAJ-1, when GFAJ-1 was incub

Bacterioplankton Dynamics within a Large Anthropogenically Impacted Urban Estuary.

The abundant and diverse microorganisms that inhabit aquatic systems are both determinants and indicators of aquatic health, providing essential ecosystem services such as nutrient cycling but also causing harmful blooms and disease in impacted habitats. Estuaries are among the most urbanized coastal ecosystems and as a consequence experience substantial environmental pressures, providing ideal systems to study the influence of anthropogenic inputs on microbial ecology. Here we use the highly urbanized Sydney Harbor, Australia, as a model system to investigate shifts in microbial community com

Chronic Intermittent Hypoxia Participates in the Pathogenesis of Atherosclerosis and Perturbs the Formation of Intestinal Microbiota.

Chronic intermittent hypoxia (CIH) is the prominent signature of highly prevalent obstructive sleep apnea (OSA) pathophysiology, which leads to increased risk and aggravation of atherosclerotic cardiovascular diseases. However, whether intestinal microbiota is implicated in the mechanisms linking CIH to arteriosclerosis (AS) pathogenesis remains unclear. The association of CIH with the development of altered gut microbiota (GM) may provide the opportunity to develop preventive strategies for atherosclerotic cardiovascular risk reduction. Illegal dumping of oil and gas wastewater alters arid soil microbial communities.

The Permian Basin, underlying southeast New Mexico and west Texas, is one of the most productive oil and gas (OG) provinces in the United States. Oil and gas production yields large volumes of wastewater with complex chemistries, and the environmental health risks posed by these OG wastewaters on sensitive desert ecosystems are poorly understood. Starting in November 2017, 39 illegal dumps, as defined by federal and state regulations, of OG wastewater were identified in southeastern New Mexico, releasing ~600,000 L of fluid onto dryland soils. To evaluate the impacts of these releases, we anal
