Carbon Partitioning and Lipid Remodeling During Phosphorus and Nitrogen Starvation in the Marine Microalga Diacronema lutheri (Haptophyta).

The domesticated marine microalga Diacronema lutheri is of great interest for producing various highly valuable molecules like lipids, particularly long-chain polyunsaturated fatty acids (LC-PUFA). In this study, we investigated the impact of phosphorus (P) and nitrogen (N) starvation on growth, carbon fixation (photosynthetic activity) and partitioning, and membrane lipid remodeling in this alga during batch culture. Our results show that the photosynthetic machinery was similarly affected by P and N stress. Under N starvation, we observed a much lower photosynthetic rate and biomass producti

Assessment of the marine microalga Chrysochromulina rotalis as bioactive feedstock cultured in an easy-to-deploy light-emitting-diode-based tubular photobioreactor.

Marine microalgae have potential to be low-cost raw materials. This depends on the exploitation of different biomass fractions for high-value products, including unique compounds. Chrysochromulina rotalis, an under-explored haptophyte with promising properties, was the focus of this study. For the first time, C. rotalis was successfully cultivated in an 80 L tubular photobioreactor, illuminated by an easy-to-use light-emitting-diode-based system. C. rotalis grew without certain trace elements and showed adaptability to different phosphorus sources, allowing a significant reduction in the N:P r

Mixotrophic lifestyle of the ichthyotoxic haptophyte, Prymnesium parvum, offered different sources of phosphorus.

Many harmful algae are mixoplanktonic, i.e. they combine phototrophy and phagotrophy, and this ability may explain their ecological success, especially when environmental conditions are not optimal for autotrophic growth. In this study, laboratory experiments were conducted with the mixotrophic and ichthyotoxic haptophyte Prymnesium parvum (strain CCAP 946/6) to test the effects of phosphorus (P) sufficiency and deficiency on its growth rate, phagotrophic and lytic activities. P-deficient P. parvum cultures were grown without or with addition of P in the form of inorganic phosphorus (nutrients

Metabarcoding Reveals Diversity of Potentially Toxic Algae in Papeete Port (Tahiti).

Harmful algae are transported in various compartments of maritime vessels, making ports with heavy maritime traffic potential hotspots for their introduction and spread. In this study, we investigate the port of Papeete (Tahiti, French Polynesia), a key hub for numerous South Pacific shipping routes. Using metabarcoding on DNA extracted from water samples (environmental DNA, eDNA) we identified 21 species of harmful algae comprising to Bacillariophyceae (4), Dinophyceae (14), and Haptophyta (3 species). Three of those species are directly associated with fish mortality events without recognize

Deciphering coccolith formation: advanced microscopy insights from the biomineralization of Gephyrocapsa huxleyi.

Coccolithophores are unicellular marine phytoplankton that produce complex and intricately shaped mineralized scales called coccoliths. Coccoliths are produced in an intracellular vesicle where crystal nucleation occurs, from which several individual calcite units develop with anisotropic crystallographic facets, prompting studies into the cellular mechanisms that control crystal growth within the cell. Here, we characterize those morphological developments in three dimensions that occur during the formation of coccoliths by the species Gephyrocapsa huxleyi using cryo-ptychographic X-ray compu

A Polinton-like Virus of C. parva Inhibits the Population Growth of a Newly Isolated Relative of Tethysvirus ontarioense.

The previous discovery of genomes of Polinton-like viruses (PLVs) associated with viruses of Chrysochromulina parva stimulated this research to determine the biological nature of these putative viral hyperparasites. Purification of C. parva viruses to enable co-infection experiments led to the discovery of a previously unknown virus, CpV-BQ3, which, based on sequence information and electron microscopy, is a species of Tethysvirus, a genus within the Megaviricetes. Purification and TEM imaging of CpV-PLV Moe revealed naked icosahedral particles morphologically similar to other cultivated virop

Characterization of Delta-7 Alkenone Desaturase in Haptophyte Gephyrocapsa huxleyi Through Heterologous Expression in Tisochrysis lutea.

The marine haptophyte Gephyrocapsa huxleyi is an ecologically and geochemically important phytoplankton due to its contribution to the global carbon cycle and its ability to biosynthesize certain alkenones. These alkenones are long-chain alkyl ketones with two to four trans-type double bonds. The genes encoding alkenone desaturase in G. huxleyi have not been experimentally characterized so far, partly due to the difficulty of inducing genetic transformation in G. huxleyi. Therefore, we introduced the putative alkenone delta-7 desaturase of G. huxleyi (designated "DesT") to the transformable an

Low-dose stimulation of growth of the harmful alga, Prymnesium parvum, by glyphosate and glyphosate-based herbicides.

Glyphosate-based herbicides (GBH) are widely used around the globe. While generally toxic to phototrophs, organic phosphorus in glyphosate can become available to glyphosate-resistant phytoplankton and contribute to algal bloom development. Few studies have examined the effects of GBH on growth of eukaryotic microalgae and information for the toxic bloom-forming haptophyte, Prymnesium parvum, is limited. Using a batch-culture system, this study examined the effects on P. parvum growth of a single application of Roundup Weed and Grass Killer Super Concentrate Plus® (Roundup SC), Roundup Weed an

New genomic insights into long-chain alkenone biosynthesis by the coccolithophorid marine alga Gephyrocapsa huxleyi.

BACKGROUND: Gephyrocapsa huxleyi is a coccolithophoric haptophyte widespread across most marine ecosystems that plays an important role in the carbon cycle. G. huxleyi is one of the few haptophytes known to produce long-chain alkenones (LCAs), which are lipids composed of C35-C42 n-alkyl chains, with two to four trans-double bonds and a keto group at either the 2nd or 3rd carbon positions, which are widely used as proxies for paleotemperature reconstruction. Despite the biomarker value of LCAs, little is known about their biosynthesis, which severely limits the understanding of the regulation

Spatial Relations between Coccoliths and Their Confining Membrane During Crystal Morphogenesis.

Coccoliths are multicrystalline calcite structures formed by microalgae within an intracellular vesicle. The morphology of each crystal is complex, and recent studies postulate that coccolith morphogenesis is regulated by the bounding membrane of its vesicle. However, the limited information about the native-state organization within the cell makes it difficult to understand which structural aspects of the membrane are responsible for morphogenesis. Here, we examined the vesicular environment during the formation of Calcidiscus leptoporus coccoliths, using advanced cryo-electron microscopy and

Investigating challenges in handling, stability and recovery of prymnesins.

Prymnesium parvum is a globally distributed haptophyte that produces an array of ichthyotoxic compounds known as prymnesins. Prymnesins are large polyethers ranging in size from 1600 to 2300 Da. Initial investigations towards the development of prymnesin reference materials identified significant problems with recovery and stability. A systematic series of studies was conducted to determine the effects of solvent composition, pH, evaporation to dryness and solid-phase extraction (SPE) resin on the stability and recovery of prymnesins. Experiments investigated A- and B-type prymnesins to verify

Human fatalities related to a Phaeocystis harmful algal bloom in the North Sea.

On May 11, 2020, five surfers drowned after asphyxiation in a massive foam bank near the Dutch coast. We present a detailed account of the event and an examination of factors that govern local foam events, that are traditionally caused by Phaeocystis globosa (Prymnesiophyceae). The data support the hypothesis that the foam originated from a Phaeocystis bloom which if correct would make this accident the first report of human fatalities due to a harmful algal bloom in The Netherlands, and the first globally due to a Phaeocystis bloom. The rarity of massive foam banks and the absence of local re

Helicosphaera carteri (Prymnesiophyceae) under high carbon dioxide: An experimental study.

The coccolithophore Helicosphaera carteri is an understudied, yet ecologically and biogeochemically important, marine calcifier. Hence, its response to ocean acidification has implications for ecosystem function and the marine carbon cycle. Here we employed dilute batch cultures featuring a coupled C-system manipulation (295, 444, and 600 μatm CO2) to analyze the response of H. carteri in terms of growth rate and particulate carbon production, two key eco-physiological and biogeochemical parameters. We highlight that both growth rate and organic carbon production are CO2 limited at 295 µatm bu

Infrared gas analyzer as a cutting-edge tool for studying photosynthesis in marine microalgae: a case study on Isochrysis galbana.

Photosynthesis is a fundamental process in microalgae, playing a crucial role in global carbon cycling and biotechnological applications. However, methods for measuring photosynthesis in microalgae remain underdeveloped compared to terrestrial plants. This study explores the application of Infrared Gas Analyzer (IRGA) technology to assess photosynthetic performance in Isochrysis galbana, a commercially valuable marine golden-brown microalga. IRGA enables real-time CO₂ exchange measurements in an open system, providing insights into carbon fixation dynamics when combined with chlorophyll a fluo

The Plant Alkaloid Camptothecin as a Novel Antifouling Compound for Marine Paints: Laboratory Bioassays and Field Trials.

The extensive use of copper and booster biocides in antifouling (AF) paints has raised environmental concerns and the need to develop new AF agents. In the present study, 18 alkaloids derived from terrestrial plants were initially evaluated for AF activity using laboratory bioassays with the bryozoan Bugula neritina and the barnacle Balanus albicostatus. The results showed that 4 of the 18 alkaloids were effective in inhibiting larval settlement of B. neritina, with an EC50 range of 6.18 to 43.11 μM, and 15 of the 18 alkaloids inhibited larval settlement of B. albicostatus, with EC50 values ra

Toxin-assisted micropredation: experimental evidence shows that contact micropredation rather than exotoxicity is the role of Prymnesium toxins.

Blooms of Prymnesium parvum can severely harm fish and zooplankton, presumably through the release of allelopathic exotoxins that offer advantages for Prymnesium in its interactions with competitors and prey. We show that Prymnesium attaches to zooplankton and fish, causing mortality, whereas exposure of these organisms to Prymnesium across a permeable membrane does not cause mortality. We also show that Prymnesium exotoxins are released independently of contact toxicity only in response to experimental procedures or natural causes of stress. Our results are consistent with the idea that toxin

Effect of environmental parameters on growth and calcification in marine coccolithophore Gephyrocapsa huxleyi CCMP371.

The coccolithophore Gephyrocapsa huxleyi is a major producer of CaCO3 in the world's oceans due to its production of coccoliths. Its domesticated cultivation may serve as a sustainable source of biogenic CaCO3 for several industrial applications. Intraspecific variation within G. huxleyi is well recognized, leading to varied physiological responses to environmental drivers. This study optimized the net production of coccoliths in a calcifying strain of G. huxleyi, CCMP371, by serially exploring the impacts of salinity, media nutrient concentration, light intensity, and temperature on growth an

Biomass and coccolith productions in Chrysotila sp. in high density cultivation system.

Coccolithophores are unicellular marine microalgae capable of producing calcium carbonate exoskeletons composed of micrometric scales called coccoliths. Since they require CO₂ for both calcification and biomass production, they are expected to exhibit a potentially higher CO₂ fixation, making them promising candidates for applications in biological carbon capture systems. In addition, coccoliths themselves are considered high-value products due to their potential applications in many fields such as nanobiotechnology. In this study, a high cell density culture of Chrysotila sp. was tested using
