Melatonin and healthy aging.

Preservation of a robust circadian rhythmicity (particulsarly of the sleep/wake cycle), a proper nutrition and adequate physical exercise are key elements for healthy aging. Aging comes along with circadian alteration, e.g. a disrupted sleep and inflammation, that leads to metabolic disorders. In turn, sleep cycle disturbances cause numerous pathophysiological changes that accelerates the aging process. In the central nervous system, sleep disruption impairs several functions, among them, the clearance of waste molecules. The decrease of plasma melatonin, a molecule of unusual phylogenetic con

Copeptin kinetics in healthy aging.

CONTEXT: The risk for disturbances in water homeostasis-resulting in hypo- or hypernatremia-increases with age. Age-related changes in arginine vasopressin (AVP) kinetics have been suggested, but findings are inconclusive due to challenges in quantifying AVP. Copeptin is a stable surrogate marker for AVP secretion, yet no data on age-related changes of copeptin kinetics are available. OBJECTIVE: We investigated copeptin kinetics in healthy ageing, hypothesizing a reduced within-subject range of copeptin in older individuals. METHODS: For this randomized controlled cross-over trial, 16 health

[Nutrition, frailty and healthy aging].

Introduction: frailty is a common clinical syndrome in older adults, associated with functional decline, increased risk of dependency, and a greater healthcare burden. Malnutrition, frequently observed in frail individuals, is linked to higher morbidity and mortality, particularly in critical situations such as hip fractures. Early diagnosis and comprehensive management of frail elderly, including nutritional intervention and promotion of physical activity, can reverse or slow down its progression. Objective: this manuscript reviews the impact of both interventions with a particular focus on p

Brainstem Dysfunction in Healthy Aging.

The brainstem controls sub-cortical and cortical activity and influences the processing of incoming information. The goal of this study was to characterize age related alterations of brainstem-brain interactions during different brain states detected by dynamic analysis of task-free fMRI. 79 young (20-40 years) and 51 older adults (55-80 years) were studied. Internal brainstem structures were segmented using a new multi-contrast segmentation approach. Brain and brainstem gray matter segmentations were warped onto a population template. The ICV-corrected Jacobian determinants were converted int

Vision through Healthy Aging Eyes.

As life expectancy grows, so too will the number of people adversely affected by age. Although it is acknowledged that many conditions and diseases are associated with age, this mini-review will present a current update of the various visual changes that generally occur in healthy individuals disregarding the possible effects of illness. These alterations influence how the world is perceived and in turn can affect efficiency or the ability to perform ordinary daily tasks such as driving or reading. The most common physical developments include a decreased pupil size and retinal luminance as we

Phosphate and Kidney Healthy Aging.

BACKGROUND: The aging population is increasing rapidly, much faster than our understanding on how to promote healthy aging free of multimorbidities. The aging kidney shows a decline in its function. Whether this decline is preventable or physiological is still debated. Main risks factors for developing CKD are aging common comorbidites, such as hypertension, diabetes, and obesity. Phosphate is vital for our organism, but it is also present in a great variety of food products as food additive and preservative. Due to the higher consumption of processed food in the last century, concern has aris

Predictors of healthy brain aging.

To determine if superior health at old age protects against cognitive impairment (CI) and Alzheimer's disease (AD), we prospectively studied 100 optimally healthy oldest-old (> or =85 years) individuals. Initially, subjects represented the top 3% of the oldest old for health. During 5.6 +/- 0.3 years of follow-up, 34 subjects developed CI, and 23 progressed to AD. By age 100, probability of CI and AD were.65 +/-.09 and.49 +/-.10. Median onset age was 97 years for CI and 100 for AD. Clearly, superior health at old age does not guarantee protection against cognitive decline. Lifetime risks were

Lipidomics in longevity and healthy aging.

The role of classical lipids in aging diseases and human longevity has been widely acknowledged. Triglyceride and cholesterol concentrations are clinically assessed to infer the risk of cardiovascular disease while larger lipoprotein particle size and low triglyceride levels have been identified as markers of human longevity. The rise of lipidomics as a branch of metabolomics has provided an additional layer of accuracy to pinpoint specific lipids and its association with aging diseases and longevity. The molecular composition and concentration of lipid species determine their cellular localiz

Genetics of healthy aging and longevity.

Longevity and healthy aging are among the most complex phenotypes studied to date. The heritability of age at death in adulthood is approximately 25 %. Studies of exceptionally long-lived individuals show that heritability is greatest at the oldest ages. Linkage studies of exceptionally long-lived families now support a longevity locus on chromosome 3; other putative longevity loci differ between studies. Candidate gene studies have identified variants at APOE and FOXO3A associated with longevity; other genes show inconsistent results. Genome-wide association scans (GWAS) of centenarians vs. y

Obesity, lipids, inflammation, and healthy aging.

Cardiovascular disease (CVD) is the leading cause of death and disability globally. Thus, prevention efforts aimed at reducing CVD events allow for greater functional independence and wellbeing over the course of life. Low-density lipoprotein-cholesterol is causally associated with atherosclerosis and is the primary target for atherosclerotic CVD (ASCVD) prevention. More recently, low-grade systemic inflammation has been recognized for its role in atherosclerotic disease initiation and progression. Additionally, there is compelling evidence for combined assessments of lipid and inflammatory bi

How to stay healthy while aging?

BACKGROUND: Aging is characterized by a gradual loss of normal physiological function, culminating in frailty and increased susceptibility to diseases. One plausible candidate for this is the decrease in oxygen supply that occurs with aging. AIMS/OBJECTIVES: The purpose is to compile a literature review to provide well-founded guidelines for healthy aging. MATERIAL AND METHODS: Based on published literature, the significance of changes in oxygen supply and autonomic regulation in aging is analyzed. RESULTS: Both oxygen uptake and its delivery to organs decline with age. This induces low-lev

Healthy aging and chronic kidney disease.

The world population is aging and the prevalence of noncommunicable diseases such as diabetes, hypertension, and chronic kidney disease (CKD) will increase significantly. With advances in medical treatment and public health, the human lifespan continues to outpace the health span in such a way that the last decade of life is generally spent in poor health. In 2015, the World Health Organization defined healthy aging as 'the process of developing and maintaining the functional ability that enables wellbeing in older age.' CKD is increasingly being recognized as a model of accelerated aging and

Neuroendocrine-immune interactions in healthy aging.

AIM: The nervous, endocrine and immune systems are connected by shared neurotransmitters, hormones and cytokines. The function of these systems shows patterns of circadian rhythmicity and a number of age-related changes in the 24-h hormonal and non-hormonal rhythms have been found in older human beings. The aim of this study was to evaluate integration among the nervous, endocrine and immune systems in the elderly. METHODS: Cortisol and melatonin serum levels were measured and lymphocyte subpopulation analyses were performed on blood samples collected every 4 h for 24 h from 15 healthy young-

Hallmarks of healthy cognitive aging: Inter-individual differences in aging trajectories.

Cognitive aging is a highly heterogeneous process, with some individuals preserving stable cognitive performance across the lifespan while others exhibiting pronounced decline. This marked interindividual variability indicates that chronological age alone is a poor predictor of cognitive health. Rather than reflecting uniform degeneration, cognitive aging emerges from divergent biological trajectories spanning molecular, cellular, and network levels. In this review, we synthesize emerging biological hallmarks of healthy cognitive aging, emphasizing studies that characterize longitudinal cognit

Aging Concerns Related to Sexuality and Gender: HIV Prevention and Healthy Aging.

Healthy aging is an important area of research across many populations, but less work has focused on this area among sexual and gender diverse individuals relative to the general population. On the whole, it is known that as the U.S. population ages, increasing attention is needed to understand the intersections between aging, health, and wellbeing. One area of consideration to address in regard to healthy aging is that of HIV prevention, in particular, pre-exposure prophylaxis (PrEP) use. For the current study we assessed these factors in a cross-sectional survey designed to assess disease st

Vascular Contributions to Healthy Aging and Dementia.

Vascular pathologies are among the most common contributors to neurodegenerative changes across the spectrum of normal aging to dementia. Cerebral small vessel disease (SVD) encompasses a wide range of conditions affecting capillaries, small arteries, and arterioles, as well as perivascular spaces and fluid dynamics in the brain, playing a significant role in vascular contributions to cognitive impairment and dementia (VCID). These factors can accelerate the progression of SVD and neuronal degeneration. Since aging is the primary risk factor for Alzheimer's disease (AD) and AD-related dementia
