The species of the genus Odilia Durette-Desset, 1973 (Heligmonellidae, Nippostrongylinae) are re-distributed among eight genera of which five are new. This classification is mainly based on certain characters of the synlophe not previously taken into account at the supraspecific level. These characters mainly include the presence or absence of a careen, the relative size of the ridges forming the careen, the development and position of ridge 1', the development of the left ridge and right ridge, and the distribution of the largest ridges. Eighteen of the 20 known species are rearranged in the

Two new species of heligmosomoid Trichostrongylina nematodes belonging to the genera Neoheligmonella Durette-Desset, 1970 and Heligmonina Baylis, 1928 are described. They are parasitic in the small intestine of three species of Mastomys from Senegal living in sympatry: M. natalensis (Smith, 1834), M. erythroleucus (Temminck, 1853) and M. huberti (Wroughton, 1909). Neoheligmonella granjoni n. sp. is closely related to three species from Senegal. They concern: N. bai Diouf & Durette-Desset, 2002 and N. dielmensis Diouf, Bâ & Durette-Desset, 1998, both parasitic in Arvicanthis niloticus Geoffroy,

A total of 138 nematodes were found in the small intestine of Ctenomys talarum (Octodontidae) from Mar de Cobo, Argentina. A new nematode species, Pudica ctenomydis n. sp., is described. The new species more closely resembles P. pujoli Durette-Desset, 1990, parasite of Microcavia niata Thomas, from Bolivia. It can be distinguished from P. pujoli by the number of ridges and characteristics of the synlophe, the spicular morphology, differences in length between rays 9 and 10, and by the presence of a symmetrical caudal bursa and a cuticular expansion surrounding the body between vulva and anus i

The species described as Longistriata fortuita Freitas, Lent, and Almeida, 1937 is here redescribed from new material collected from the type host, Holochilus chacarius balnearum Thomas, and the type locality, San Martín del Tabacal, Salta, Argentina. Neotypes are designed for the species since the type material deposited by the authors is lost. The original description did not include the synlophe or the female and both are here described. Several characters of the synlophe as the number of ridges (14-19), the ridges continuous and all around body, and the presence of a gradient of size of th

The in vitro nematocide activity of seventeen 6,7-diarylpteridines has been tested using three different experimental models, Caenorhabditis elegans, Nippostrongylus brasiliensis and Heligmosomoides polygyrus. The method of evaluation of inhibition in the secretion of acetylcholinesterase by H. polygyrus seems to be the most indicated to avoid false positives. The in vivo activities, against Trichinella spiralis, of the most in vitro active pteridines have been assayed. All pteridine derivatives bearing 6,7-di-p-bromophenyl substituents have shown in vitro nematocide activities in the three ex

Hassalstrongylus Durette-Desset, 1971 (Nematoda: Heligmonellidae), includes 19 species that are distributed from the southwestern United States to central-western Argentina. Hassalstrongylus aduncus is a parasitic nematode of rodents from the subfamilies Arvicolinae, Murinae, and Sigmodontinae, and has been recorded from southern Virginia and Oklahoma to Costa Rica. This species was described by Chandler in 1932; the morphology of the synlophe was not included. Subsequently, in 1972, Durette-Desset described only the synlophe of the middle region of the body in both sexes. Despite its wide geo

The heligmosomid nematodes Heligmosomum mixtum and Heligmosomoides glareoli are dominant helminths infecting bank voles (Clethrionomys glareolus) in the temperate forests of NE Poland. Both are relatively long-lived species that accumulate in hosts with increasing host age. Based on studies showing that the closely related species, Heligmosomoides bakeri is immunomodulatory in murine hosts, we hypothesized that heligmosomid-infected bank voles should show higher prevalence and abundance with other helminths. To test this hypothesis, we analysed a database containing quantitative data on helmin

The survival of Heligmosomoides polygyrus was monitored during primary infections in female C57Bl10, NIH and BALB/c mice at low and high intensities of infection. Survivorship curves were fitted for each data set and analysed. C57Bl10 mice, given either low or high intensities of infection, harboured parasites for 28-37 weeks, heavier infections surviving marginally but significantly longer. Essentially the survivorship curves of H. polygyrus in C57Bl10 mice could be accounted for by senility, the increased probability of worms with a longer life-span occurring at high infection intensities an

Plant secondary metabolites (PSMs) may improve gastrointestinal health by exerting immunomodulatory, anti-inflammatory and/or antiparasitic effects. Bark extracts from coniferous tree species have previously been shown to reduce the burden of a range of parasite species in the gastrointestinal tract, with condensed tannins as the potential active compounds. In the present study, the impact of an acetone extract of pine bark (Pinus sylvestris) on the resistance, performance and tolerance of genetically diverse mice (Mus musculus) was assessed. Mice able to clear an infection quickly (fast respo

The crystal structure of GST Nu2-2 (HpolGSTN2-2) from the model hookworm nematode Heligmosomoides polygyrus has been solved by the molecular replacement method and refined to a resolution of 1.71 A, providing the first structural data from a class of nematode-specific GSTs. By structural alignment with two Sigma class GSTs, glutathione could be rationally docked into the G-site of the enzyme. By comparing with all mammalian GST classes, a novel, long, and deep cleft was identified at the H-site, providing a potential site for ligand binding. This new GST class may support the establishment of

The effects of Heligmosomoides bakeri infection on the course of a concurrent Cryptosporidium parvum infection were studied in C57BL/6 mice. Mice were initially infected with 80 L(3) of H. bakeri and then challenged with 10(4) oocysts of C. parvum, administered during the patent period of the nematode infection (28 day post H. bakeri infection). The number of C. parvum oocysts excreted in the feces and the number of adult H. bakeri in the small intestine were monitored during the experiment. Concurrent H. bakeri infection resulted in a prolonged course of infection with C. parvum. The intensit

Helminths regulate host immune responses to ensure their own long-term survival. Numerous studies have demonstrated that these helminth-induced regulatory mechanisms can also limit host inflammatory responses in several disease models. We used the Heligmosomoides bakeri (Hb) infection model (also known as H. polygyrus or H. polygyrus bakeri in the literature) to test whether such immune regulation affects skin inflammatory responses induced by the model contact sensitiser dibutyl phthalate fluorescein isothiocynate (DBP-FITC). Skin lysates from DBP-FITC-sensitized, Hb-infected mice produced le

Maternal bacterial and viral infections that induce neuroinflammation in the developing brain are associated with impaired cognitive function and increased anxiety in the offspring. In contrast, maternal infection with the immunoregulatory murine gastrointestinal (GI) nematode, Heligmosomoides bakeri, appears to benefit neurodevelopment as juvenile 2- and 3-week-old male and female offspring had enhanced spatial memory, which may be due to a Th2/Treg biased neuroimmune environment. Here, the impact of maternal H. bakeri infection during pregnancy and lactation on the spatial and anxiety-like b

Protein deficiency impairs local and systemic immune responses to Heligmosomoides bakeri infection but little is known about their individual and interactive impacts on tissue architecture of maternal lymphoid (thymus, spleen) and visceral (small intestine, kidney, liver, pancreas) organs during the demanding period of lactation. Using a 2 × 2 factorial design, pregnant CD1 mice were fed a 24% protein sufficient (PS) or a 6% protein deficient (PD) isoenergetic diet beginning on day 14 of pregnancy and were infected with 100 H. bakeri larvae four times or exposed to four sham infections. On day

Nematode infection upregulates interleukin-4 (IL-4) and IL-13 and induces STAT6-dependent changes in gut function that promote worm clearance. IL-4 and IL-13 activate the type 2 IL-4 receptor (IL-4R), which contains the IL-13Rα1 and IL-4Rα chains. We used mice deficient in IL-13Rα1 (IL-13Rα1(-/-)) to examine the contribution of IL-13 acting at the type 2 IL-4R to immune and functional responses to primary (Hb1) and secondary (Hb2) infections with the gastrointestinal nematode parasite Heligmosomoides bakeri There were differences between strains in the IL-4 and IL-13 expression responses to Hb

Chronic primary infections with Heligmosomoides polygyrus (Nematospiroides dubius) are still relatively poorly documented, particularly in relation to the role of host resistance in limiting worm survival. In the present work the duration of infection with H. polygyrus was studied in CFLP mice given doses of infective larvae ranging from 50 to 500 L3. The least heavily infected (50 L3) group ceased egg production earliest (week 36) whereas eggs were still detected in the faeces of mice given 500 larvae in week 42. At autopsy (week 42) mice given 50 larvae had virtually lost their entire worm b

An assay is described for the quantitative detection of excretory/secretory antigens liberated by the intestinal nematode Heligmosomoides polygyrus and voided in the host's faeces. A rabbit polyclonal antiserum to the antigens detected them in a dose-dependent manner and was sufficiently sensitive to recognise infection in mice carrying a mean worm burden of nine worms. The assay was specific, giving higher optical density readings with the faeces of mice infected with H polygyrus than with faeces from mice carrying patent infections with Trichuris muris, Hymenolepsis microstoma or the intesti

The nematocidal activity of a broad-spectrum antiparasitic agent, nitazoxanide [( N-(5-nitrothiazol-2-gammal)salicylamide; NTZ], was evaluated in both in vitro and in vivo models using Caenorhabditis elegans, Heligmosomoides polygyrus and Trichinella spiralis. In vitro, NTZ (100 muM) exhibited a low activity against C. elegans and had no effect on embryonation and hatching of H. polygyrus eggs. At concentrations of 100 and 50 muM, the inhibition of excretion/secretion of acetylcholinesterase and acid phosphatase of adult H. polygyrus by NTZ was variable. The in vitro effects of mebendazole (5

Neonatal immune development begins in pregnancy and continues into lactation and may be affected by maternal diet. We investigated the possibility that maternal protein deficiency (PD) during a chronic gastrointestinal (GI) nematode infection could impair neonatal immune development. Beginning on d 14 of pregnancy, mice were fed protein-sufficient (PS; 24%) or protein-deficient (PD; 6%) isoenergetic diets and were infected weekly with either 0 (sham) or 100 Heligmosomoides bakeri larvae. Pups were killed on d 2, 7, 14, and d 21 and dams on d 20 of lactation. Lymphoid organs were weighed. Cytok

Basophils orchestrate protection against reinfections with gastrointestinal helminths and ticks, but the underlying mechanisms remain elusive. We investigated the role of Fc receptors on basophils, the antibody isotypes IgG1 and IgE, and basophil-derived IL-4/IL-13 during challenge infections with Heligmosomoides polygyrus and Nippostrongylus brasiliensis. Using mixed bone marrow chimeras, we found that activating Fc receptors on basophils were required for protective immunity but not for regulation of basophil homeostasis. Furthermore, rapid worm expulsion was impaired in IgE-deficient but no
