ACTIONS OF HEMICHOLINIUM (HC-3) ON NEUROMUSCULAR TRANSMISSION.

Hemicholinium No. 3 (HC-3; alpha,alpha'-dimethylaminoethanol-4, 4'-biacetophenone) produced neuromuscular block of the tibialis anterior muscle-sciatic nerve preparation of the anaesthetized cat which was of slow onset, of long duration and dependent on the nerve stimulus frequency. The failure of the compound to modify the response of the tibialis muscle to close-arterial injections of acetylcholine suggested that the sensitivity of the motor endplate was not changed by it. The neuromuscular block produced by hemicholinium was antagonized by choline but only partly relieved by anticholinester

Hemicholinium-3 impairs spatial learning and the deficit is reversed by cholinomimetics.

The effects of hemicholinium-3 (HC-3) on spatial discrimination learning were studied. Rats were equipped with indwelling cannulae in the right lateral ventricle and, following recovery, were trained on a two platform spatial discrimination task in a water maze. In this task a visible escape platform remains in a fixed position in the pool during a single training session, whilst the location of an identical "float" (which affords no escape) is randomly varied. For each session the location of the fixed escape platform was changed and the rats were retrained to criterion following pretreatment

Prevention and reversal of tolerance to barbiturates by intraventricular injection of hemicholinium-3.

Intracerebroventricular (i.c.v.) injection of phenobarbital, 800 mug, or intraperitoneal (i.p.) injection of hexobarbital, 100 mg/kg, into rats resulted in a loss of righting reflex lasting 15.4 +/- 0.2 min and 20.7 +/- 0.7 min, respectively. A 40-60% reduction in this response was obtained following administration of phenobarbital either i.c.v. (800 mug 4 times daily) or i.p. (80 mg/kg/day) for 4 days. Although these treatments also increased hepatic mixed function oxidase activity, this enzyme induction was shown to be unrelated to the development of tolerance to loss of righting reflex. I.c

The effect of hemicholinium-3 on choline and acetylcholine levels in a sympathetic ganglion.

Pregangliaaonic stimulation of the cat's superior cervical ganglion in the presence of hemicholinium-3 (HC-3) produced the expected depletion of acetylcholine (ACh) stores, but failed to cause a corresponding reduction in the choline content. These results suggest that either HC-3 possesses an intracellular site of action or that in lower doses it selectively inhibits a specialized choline transport system in cholinergic nerves. At a dose of 2 mg/kg, HC-3 probably blocked ACh synthesis completely in ganglia stimulated at 20 Hz. Under these conditions, there was a rapid depletion of ACh to abou

Effects of hemicholinium-3 and choline on hippocampal electrical activity during immobility vs. movement.

Earlier studies have shown that the hippocampal rhythmical slow activity (RSA or theta) which may occur during behavioral immobility (IRSA) is abolished by systemically administered atropine (is atropine-sensitive), although the RSA which accompanies movements, such as walking, running or swimming (MRSA) is atropine-resistant. This study was designed to manipulate brain cholinergic activity in ways other than through the use of postsynaptic receptor antagonists, and to determine the effects of such manipulations on IRSA and MRSA. The IRSA elicited by electrical stimulation of the reticular for

Strain differences in the inhibition by hemicholinium-3 of the pressor response to intraventricular injection of neostigmine in rats.

The pressor response to intracerebroventricular (i.c.v.) injections of neostigmine was examined in three groups of Sprague-Dawley rats (MFS, CS and CFE) obtained from different breeders and in Long-Evans rats (MLE) obtained from one of the same breeders. All rats responded with a rise in blood pressure to i.c.v. injection of neostigmine, although the CFE strain was 6-fold less sensitive to the drug than were the other strains.Prior i.c.v. injection of hemicholinium-3 (HC-3; 0.4 mug) reduced by more than 50% the pressor response in the two strains obtained from the same breeder (MFS and MLE) an

Characterization of [3H]hemicholinium-3 binding associated with neuronal choline uptake sites in rat brain membranes.

Hemicholinium-3 (HCh-3) is a potent and specific inhibitor of the high-affinity choline transport process (HAChT) localized on cholinergic neurons. In this study, the specific binding of [3H]HCh-3 (120 Ci/mmol) was characterized in crude synaptic membranes prepared from rat brain. The binding of [3H]HCh-3 to forebrain membranes was saturable, reversible and specific with an apparent Kd under optimal conditions of 35 nM and a Bmax of 56 fmol/mg protein. The potency of various HAChT inhibitors correlated with their apparent affinities for the specific [3H]HCh-3 binding site. The specific binding

Selective cortical decrease of high-affinity choline uptake carrier in Alzheimer's disease: an autoradiographic study using 3H-hemicholinium-3.

3H-hemicholinium-3 (3H-HC-3) binding, a marker of the presynaptic high-affinity choline uptake carrier (HACU), was measured by autoradiography in several brain regions of 17 Alzheimer's disease (AD) patients and of 11 matched controls. A significant decrease in the density of 3H-HC-3 binding sites was found in entorhinal cortex, hippocampus and layers I-III of the frontal cortex. By contrast, in the caudate-putamen the number of 3H-HC-3 binding sites in AD cases was comparable to that of control striata. These data concur with previous results using classical presynaptic markers and reflect th

Further studies on the biochemical characterization and autoradiographic distribution of [3H]hemicholinium-3 binding sites in rat brain: a presynaptic cholinergic marker.

Hemicholinium-3 (HC-3) is a potent inhibitor of the high-affinity choline uptake system (HACU). Here we report on the biochemical characterization and autoradiographic distribution of [3H]hemicholinium-3 binding sites in rat brain, confirming and expanding results from previous studies. The binding of [3H]HC-3 to striatal membranes was specific, to a single site, sodium-dependent, saturable, and of high-affinity, Kd values being about 3 nM for striatum, 5 nM for the hippocampus and 12 nM for neocortex. [3H]HC-3 specific binding exhibited a pharmacological profile suggestive of physiologically

Quantal mechanism of transmitter release during progressive depletion of the presynaptic stores at a ganglionic synapse. The action of hemicholinium-3 and thiamine deprivation.

In the present experiments we interfered with the mechanism of acetylcholine (ACh) synthesis in the rat superior cervical ganglion by impairing the supply of either the choline group (hemicholinium no. 3 [HC-3]treatment) or the acetyl group (thiamine deprivation). Under both conditions stimulation causes in the ganglion a progressive decline in ACh output associated with a depletion of transmitter tissue content. ACh release from the terminals of a single preganglionic fiber was estimated from the quantum content value of the evoked excitatory postsynaptic potentials (EPSP's) recorded intracel

Alteration of retinal choline metabolism in an experimental model for photoreceptor cell degeneration.

The choline analog hemicholinium-3 causes selective degeneration of the cones and rods in the rabbit retina. These experiments examine some biochemical effects of hemicholinium-3 on metabolic pathways involved in choline metabolism. No effect of the drug on several important photoreceptor biosynthetic functions was found: leucine incorporation into TCA-precipitable material, glycerol incorporation into phospholipids, and phosphatidylcholine formation by either base exchange or choline phosphotransferase activity. However, hemicholinium-3 does selectively affect free choline use: high-affinity

The contribution of cholinergic postganglionic neurotransmission to contractions of rabbit detrusor.

Field stimulation was used to elicit a contractile response in muscle strips from rabbit detrusor. The blockade of this response by tetrodotoxin (1 X 10(-7) M) ranged from 100% at 1 Hz to 86% at 40 Hz. At concentrations which produced strictly muscarinic antagonism (up to 4 X 10(-7) M) atropine depressed the frequency-response curve by about 42% at maximum but was much less effective at frequencies below 10 Hz. Similarly, treatment of the strips with hemicholinium-3 (5.2 X 10(-4) M) for 90 minutes in the presence of field stimulation at 60 Hz, depressed the frequency-response curve by 52% at m

Inhibition of high affinity choline uptake in the rat brain by neurotoxins: effect of monosialoganglioside GM1.

Mustard derivatives of ethyl-choline and hemicholinium-3 have been suggested as possible specific cholinergic neurotoxins. Free choline was determined by bioassay. 2 The respiration of animals given 0.35 mumol/kg of HC-3 was hardly affected; however, 4 out of 6 rabbits given the same dose of TPHC-3 exhibited varying degrees of respiratory impairment. All animals that received 2 injections (1 h apart) of either HC-3 (1.4 mumol/kg) or TPHC-3 (0.7 mumol/kg) developed respiratory difficulty about 1 h after the second injection. 3
