Morphological studies suggest that there are several types of decapod hemocytes; however, distinguishing criteria based on conventional staining techniques are often subtle or ambiguous. Cytochemical features of ridgeback prawn (Penaeidae: Sicyonia ingentis) hemocytes were studied using specific stains for lysosomes, cytoplasmic contents, and granule enzymes. This approach facilitates the differentiation of cell types in the ridgeback prawn and provides information on the functions of and relation ships among different cell types. Agranular hemocytes and a subgroup of small granule hemocytes c

Hemocytes play several key roles in the mosquito's immune response. Despite most of our understanding regarding their immunological role concerns their responses against bacteria, fungi, and Plasmodium, our knowledge of hemocyte's role in antiviral defense is poorly understood. We performed a comprehensive comparative transcriptomic analysis between the dengue vector Aedes aegypti's two major immune cell types, hemocytes and fat body, revealing a plethora of differentially expressed immune genes that indicates a high level of functional specialization as well as complementation between the two

Hemocytes are the primary immune cells of crustaceans. Few comparison studies have been done among different crustaceans and some key parameters of circulating hemocytes have not been investigated. Here, we compared the circulating hemocytes in six decapod crustaceans, Cherax quadrinatus, Procambarus clarkii, Penaeus vannamei, Penaeus monodon, Eriocheir sinensis, and Scylla paramamosain. Although the hemocytes of different species vary in size, they share common morphological characteristics. Based on their morphological features, circulating hemocytes can be basically classified into granular

This study aimed to evaluate the diagnostic potential of hemocyte parameters and procalcitonin (PCT) in detecting bloodstream infections (BSI) in neonates and explore the contribution of hemocyte activation-related genes to pediatric sepsis through bioinformatics analysis. A cohort of 419 neonates, categorized as BSI (positive blood culture) and control, underwent comparative analysis of PCT and hemocyte parameters. A predictive model for neonatal BSI was established, demonstrating an impressive area under the receiver ROC curve of 0.968 with remarkable sensitivity (92%) and specificity (87.3%

Hemocytes of freshwater bivalves are an important target model for evaluating copper (Cu) toxicity in vitro, with excess Cu causing adverse responses in these organisms. Despite this, the mechanisms underlying cytotoxicity remain poorly understood. The freshwater bivalve Anodonta woodiana, employed as a model organism in freshwater environments, was utilized in this study. Hemocytes of A. woodiana were exposed to various aqueous Cu treatments (0.001, 0.01, 0.1, 1, and 10 mg/L), and a control group (no Cu added) for 3 h to investigate the cytotoxic mechanisms of Cu. The results showed a signifi

Stem cells are essential for the proper functioning of tissues, replacing damaged, senescent cells to ensure tissue regeneration. However, as age advances, the number of these stem cells can change, and their self-renewal abilities can become impaired, leading to disruption of homeostasis, loss of regenerative capacity, and, ultimately, deterioration of tissue function. In Drosophila testis, in addition to the germline and somatic cells involved in spermatogenesis, there are immune cells (hemocytes) with macrophage function. In our study, we aimed to investigate the role of hemocytes in mainta

Snails are the intermediate host of Schistosoma and they play an important role in the transmission of schistosomiasis. The snails in different areas present various susceptibilities, which are related to the different hemocytes in the snails. There is not a common standard in the classification of hemocytes, and generally the hemocytes are divided into two categories: a granulocyte and a hyalinocyte. The granulocyte plays an important role in immunization, while hyalinocyte shows a less effect. Besides, soluble factors may also play a role in immunization.

Human infection by Schistosoma mansoni affects more than 100 million people worldwide, most often in populations of developing countries of Africa, Asia, and Latin America. The transmission of S. mansoni in human populations depends on the presence of some species of Biomphalaria that act as an intermediate host. The compatibility between S. mansoni and its intermediate host is influenced by behavioral, physiological, and genetical factors of the mollusc and the parasite. The susceptibility level of the mollusc has been attributed to the capacity of internal defense system (IDS)-hemocytes and

Mosquitoes are the most important arthropod disease vectors, transmitting a broad range of pathogens that cause diseases such as malaria, lymphatic filariasis, and yellow fever. Mosquitoes and other insects are able to mount powerful cellular and humoral immune responses against invading pathogens. To date, most studies have concentrated on the humoral response. In the current study we describe the hemocytes (blood cells) of the yellow fever mosquito, Aedes aegypti, by means of morphology, lectin binding, and enzyme activity and immunocytochemistry. Our light and electron microscopic studies s

Nodulation, the lepidopteran insect immune response to large numbers of microbes in the blood (hemolymph) consists of the coordination of the blood cell (hemocyte) types the granular cells and plasmatocytes in terms of granular cell-bacteria adhesion and hemocyte-hemocyte adhesion (microaggregation). Hemocyte-microbe adhesion is influenced by the secondary messenger, cAMP, and cAMP-dependent protein kinase A. In the present study, cholera toxin, an AB5 protein known to indirectly stimulate adenylate cyclase, is used to examine the hemocyte responses to glass, bacteria and hemocyte-hemocyte mic

Immune function in bivalves can be adversely affected by long-term exposure to environmental contaminants. Investigating alterations in immunity can therefore yield relevant information about the relationship between exposure to environmental contaminants and susceptibility to infectious diseases. We have developed a rapid, cost-effective, and miniaturized immunocompetence assay to evaluate the phagocytic activity, viability, and concentration of hemocytes in freshwater and marine bivalves. Preliminary experiments were performed to optimize various aspects of the assay including 1) the time re

We have demonstrated that carpet shell clam (Ruditapes decussatus) hemocytes produce nitric oxide (NO) in response to zymosan or bacterial lipopolysaccharide (LPS). This NO production was partially inhibited by the NO synthase inhibitor, N-omega-nitro-L-arginine (L-NAME). The capability of clam hemocytes to produce NO in response to the bacterial pathogen Vibrio tapetis was also studied. Incubation with bacteria induced a significant NO production by clam hemocytes, even though exogenous NO only slightly decreased the growth of V. tapetis. The effect of exogenous NO on the capability of clam h

Hemocytes are specialized cells found in the circulatory system of invertebrates, such as marine bivalves, and play crucial roles in physiology and health. However, in bivalves, in-depth studies on their in vitro biology beyond one or two days remain scarce, particularly in Crassostrea (Magallana) gigas. In this study, we characterized the hemocytes isolated from triploid C. gigas oysters, maintained in vitro in their natural medium, hemolymph plasma, to assess their morphological, behavioral, and metabolic status over 21 days. A diverse population of hemocyte morphotypes, including granulocyt

Uncovering the complexity of systems in non-model organisms is critical for understanding arthropod immunology. Prior efforts have mostly focused on Dipteran insects, which only account for a subset of existing arthropod species in nature. Here, we describe immune cells or hemocytes from the clinically relevant tick Ixodes scapularis using bulk and single cell RNA sequencing combined with depletion via clodronate liposomes, RNA interference, Clustered Regularly Interspaced Short Palindromic Repeats activation (CRISPRa) and RNA-fluorescence in situ hybridization (FISH). We observe molecular alt

Pediculus humanus humanus is an human ectoparasite which represents a serious public health threat because it is vector for pathogenic bacteria. It is important to understand and identify where bacteria reside in human body lice to define new strategies to counterstroke the capacity of vectorization of the bacterial pathogens by body lice. It is known that phagocytes from vertebrates can be hosts or reservoirs for several microbes. Therefore, we wondered if Pediculus humanus humanus phagocytes could hide pathogens. In this study, we characterized the phagocytes from Pediculus humanus humanus a

The plasma enzyme phenoloxidase plays an important role in host resistance against viral, bacterial, fungal, filarial, and parasitoid challenge. Two Heliothis virescens prophenoloxidase transcripts, HvPPO-1 and HvPPO-2, were assembled from ESTs derived from a hemocyte cDNA library. The 2,363-bp HvPPO-1 contig encoded a 696-amino acid protein. The 3,255-bp HvPPO-2 contig encoded a 684-amino acid protein. Hemocyte and fat body transcript levels of HvPPO-1 were slightly elevated by bacterial infection in 5th instar larvae; however, HvPPO-2 expression was not significantly elevated above controls

An essential component of the insect cellular response is phagocytosis. Analyses of the in vitro phagocytosis could be useful for the studies of the relationship between insects and their pathogens. Fungal metabolites are known to inhibit phagocytosis whereas components of the fungal cell wall stimulate phagocytosis. To achieve a better understanding of fungal pathogenesis in insects, haemocyte populations of two insect species susceptible to Conidiobolus coronatus infection (Galleria mellonella, Dendrolimus pini ) were compared with haemocytes of the resistant species (Calliphora erythrocepha

Hemocytes in the hemolymph of insects perform innate immunity, but systematic studies to compare immunotoxicity of pesticides on hemocytes are still few. In this study, an insect hemocyte system was used to assess the impact of pesticides with different modes of action, which included loss of cell viability, inhibition of hemophagocytosis, and reduction of nitric oxide synthase (NOS) activity. Results showed that piericidin A was the most cytotoxic to hemocytes, chlorfluazuron and hexaflumuron were the next. Also, piericidin A, chlorfenapyr, and fipronil had strong inhibitory effects on hemoph

We have examined the hemocytes of three decapod crustaceans (Homarus americanus, Panulirus interruptus, and Loxorhynchus grandis) and propose a classification of these cells based on morphology, cytochemistry, and studies of cell functions. In all species, hyaline cells and granulocytes were identified. Although we have retained the widely used names for these cells, we show that traditional morphological features alone do not accurately differentiate between these categories. Historically, the term hyaline cell refers to hemocytes that contain no or only a few cytoplasmic granules, whereas gr
