Viral hemorrhagic septicemia virus glycoprotein production in tobacco.

Viral hemorrhagic septicemia virus (VHSV) causes mortality in numerous marine and freshwater fish species resulting in heavy losses in fish farming. The glycoprotein gene of VHSV was fused with the cholera toxin B subunit (CTB) and expressed transiently in leaf tissues of Nicotiana benthamiana via the agroinfiltration method. The glycoprotein gene was divided into two parts to improve assembly of CTB fusion proteins (CTB-VHSV99-235 and CTB-VHSV258-417). Production of CTB fusion proteins was confirmed in the agroinfiltrated leaf tissue by western blot analysis. The plant-produced CTB fusion pro

Genetically engineered viral hemorrhagic septicemia virus (VHSV) vaccines.

Viral hemorrhagic septicemia virus (VHSV) has been one of the major causes of mortality in a wide range of freshwater and marine fishes worldwide. Although various types of vaccines have been tried to prevent VHSV disease in cultured fishes, there are still no commercial vaccines. Reverse genetics have made it possible to change a certain regions on viral genome in accordance with the requirements of a research. Various types of VHSV mutants have been generated through the reverse genetic method, and most of them were recovered to investigate the virulence mechanisms of VHSV. In the reverse ge

Viral tropism and pathology associated with viral hemorrhagic septicemia in larval and juvenile Pacific herring.

Viral hemorrhagic septicemia virus (VHSV) genotype IVa causes mass mortality in wild Pacific herring, a species of economic value, in the Northeast Pacific Ocean. Young of the year herring are particularly susceptible and can be carriers of the virus. To understand its pathogenesis, tissue and cellular tropisms of VHSV in larval and juvenile Pacific herring were investigated with immunohistochemistry, transmission electron microscopy, and viral tissue titer. In larval herring, early viral tropism for epithelial tissues (6d post-exposure) was indicated by foci of epidermal thickening that conta

Isolation and characterization of viral hemorrhagic septicemia virus from farmed bastard halibut.

Viral hemorrhagic septicemia virus (VHSV) was detected and identified in healthy bastard halibut Paralichthys olivaceus on the east coast of China through a combination of virus isolation, real-time reverse transcription polymerase chain reaction (RT-PCR) and conventional RT-PCR followed by sequencing. Analysis using MEGA11 software revealed that the isolated VHSV belongs to genotype IVa. Bastard halibut fry were experimentally infected by immersion and gill inoculation with infected cell culture medium and exhibited clinical signs. Cytopathic effects (CPE) were observed again when the epithel

Genome analysis of viral hemorrhagic septicemia virus isolated from Paralichthys olivaceus in China.

This study determined the complete genomic sequence of the viral hemorrhagic septicemia virus (VHSV) strain GY-2307 isolated form farmed paralichthys olivaceus in northeast China. The whole genome length of GY-2307 comprised 11,140 nucleotides (nt), and the overall organization was typical of that observed for all other VHSV strains. The phylogenetic analysis results indicated that the GY-2307 isolate belonged to genotype Ⅳa and significantly related to other Korean and Japanese isolates. The N, P, M, NV and L genes of GY-2307 were compared with the VHSV sequences in GenBank. The results revea

Early-Stage Adhesion Dynamics of Viral Hemorrhagic Septicemia Virus in Olive Flounder, Paralichthys olivaceus.

Viral hemorrhagic septicemia virus (VHSV) is a major pathogen in olive flounder (Paralichthys olivaceus) aquaculture, leading to high mortality rates and significant economic losses. Recurrent outbreaks underscore the need for a deeper understanding of the early-stage adhesion and infection mechanisms of VHSV. To address this gap, this study investigated the adsorption and replication dynamics of VHSV in various olive flounder tissues under different temperature conditions (15°C, 20°C, and 25°C). Viral RNA levels were quantified using RT-PCR, and viral recovery from seawater was assessed using

Early immune responses in rainbow trout liver upon viral hemorrhagic septicemia virus (VHSV) infection.

Among the essential metabolic functions of the liver, in mammals, a role as mediator of systemic and local innate immunity has also been reported. Although the presence of an important leukocyte population in mammalian liver is well documented, the characterization of leukocyte populations in the teleost liver has been only scarcely addressed. In the current work, we have confirmed the presence of IgM+, IgD+, IgT+, CD8α+, CD3+ cells, and cells expressing major histocompatibility complex (MHC-II) in rainbow trout (Oncorhynchus mykiss) liver by flow cytometry and/or immunohistochemistry analysis

Comparison of treatments to inactivate viral hemorrhagic septicemia virus (VHSV-IVb) in frozen baitfish.

Current US state and federal fish health regulations target the spread of viral hemorrhagic septicemia virus-IVb (VHSV-IVb) through movement restrictions of live fish; however, they largely ignore the potential for the virus to be spread through commercial distribution and use of frozen baitfish from VHSV-IVb-positive regions. Some state laws do require treatment of frozen baitfish to inactivate VHSV, and additional methods have been proposed, but few scientific studies have examined the efficacy of these treatments. In this study, bluegills Lepomis macrochirus were challenged with VHSV-IVb an

Application of a multi-epitope DNA vaccine against viral hemorrhagic septicemia virus type Ⅳa.

Viral hemorrhagic septicemia virus (VHSV) is a single-stranded RNA virus that infects both freshwater and marine fish. In Korea, VHSV genotype Ⅳa infects cultured olive flounder (Paralichthys olivaceus) annually. As a prevention strategy against infectious viral diseases, this study aimed to develop a multi-epitope DNA vaccine using in silico methods and evaluate its efficacy in vivo in olive flounder. Cytotoxic T-lymphocyte (CTL), helper T-lymphocyte (HTL), and linear B-lymphocyte (LBL) epitopes of the VHSV glycoprotein were predicted using bioinformatic tools and assembled with the linkers A

Identification of rearing temperature-dependent host defense signaling against viral hemorrhagic septicemia virus infection.

Viral hemorrhagic septicemia virus (VHSV) infection is associated with fatal outcomes in the aquaculture production of olive flounder (Paralichthys olivaceus). Olive flounders at low and high temperatures are known to be highly susceptible and resistant to VHSV infection, respectively. To study temperature-dependent innate immune activity, 4-aminobenzoic hydrazide (4-AH), a myeloperoxidase (MPO) inhibitor, was used to treat VHSV-infected olive flounders reared at a high temperature of 20 °C (20VI). Mortality, the MPO transcription, and the proteomic expression pattern of the 20VI group were th

[An experimental contribution to the epizootiology of viral hemorrhagic septicemia of rabbits (rabbit hemorrhagic disease, RHD)--transmission by flies].

In experiments adapted to natural conditions it was established that--a conjunctival infection with about 100 RHD-virus particles was successful--in a milieu without flies transmission over a distance of only 50 cm did not take place--iridescent flies (Phormia spp.) seven hours after contamination with RHD-virus material from died rabbits transmitted RHD to susceptible rabbits in an isolated cage. It ist supposed that transmission in this way will be an epizootiological important one for spreading in narrow distances.

Goldfish Carassius auratus susceptibility to viral hemorrhagic septicemia virus genotype IVb depends on exposure route.

We assessed the susceptibility of goldfish Carassius auratus to infection by genotype IVb of the viral hemorrhagic septicemia virus. Goldfish were infected by intraperitoneal injections of 106 plaque-forming units (pfu) fish-1, single bath exposure of 105 pfu ml-1 for 24 h, or consumption of 0.4 g of commercial fish feed soaked in 107 pfu per 8 fish. The mortality rate of intraperitoneal-infected goldfish was 10 to 32%, although the virus was detected by quantitative RT-PCR in 77% (65/84) of the survivors at the end of the 42 d trial, suggesting a carrier state. Severe gross lesions were obser

Comparative analysis of immunogenicity for viral hemorrhagic septicemia virus (VHSV) vaccines inactivated by different methods.

Viral hemorrhagic septicemia virus (VHSV) belonging to the genus Novirhabdovirus and family Rhabdoviridae is a major viral disease that causes economic loss in the olive flounder (Paralichthys olivaceus). In this study, we produced formalin-, binary ethylenimine (BEI)-, β-propiolactone (BPL)-, and heat-treated, inactivated VHSV vaccines; and observed the glycoprotein changes in the inactivated VHSV vaccines and confirmed their correlation with vaccine efficacy. The control group showed a 100 % cumulative mortality rate, but the groups immunized with formalin- and BPL-inactivated VHSV vaccines

Feedback loop regulation between viperin and viral hemorrhagic septicemia virus through competing protein degradation pathways.

UNLABELLED: Viperin is an antiviral protein that exhibits a remarkably broad spectrum of antiviral activity. Viperin-like proteins are found all kingdoms of life, suggesting it is an ancient component of the innate immune system. However, viruses have developed strategies to counteract viperin's effects. Here, we describe a feedback loop between viperin and viral hemorrhagic septicemia virus (VHSV), a common fish pathogen. We show that Lateolabrax japonicus viperin (Ljviperin) is induced by both IFN-independent and IFN-dependent pathways, with the C-terminal domain of Ljviperin being important

DNA vaccination against viral hemorrhagic septicemia virus (VHSV) affects the capacity of kidney leukocytes to respond to a posterior viral encounter.

Given the lack of antiviral prophylactic measures authorized in aquaculture, vaccination is the most effective method to prevent and control diseases provoked by viral agents. Viral hemorrhagic septicemia virus (VHSV) is an important fish pathogen, known to induce high mortality rates and large economic losses worldwide in many aquaculture fish species, including rainbow trout (Oncorhynchus mykiss). DNA vaccination against VHSV has been proven a very effective method to induce a long-term protection against the virus, yet the mechanisms through which this vaccine induces protection are still u

Development of a neutralization ccombody, synthetic hagfish antibody, for viral hemorrhagic septicemia virus (VHSV) targeting glycoprotein G.

Viral hemorrhagic septicemia virus (VHSV) is a highly contagious fish pathogen affecting a wide range of marine and freshwater fish species worldwide. Its geographic distribution is predicted to expand, increasing the probability of transmission to previously unexposed host species. In the absence of effective control strategies for VHSV, the development of large-scale therapeutic interventions remains a priority for fish farmers to control disease outbreaks. A key target for vaccines and virus-neutralizing agents is the surface glycoprotein G, which plays a crucial role in viral entry and hos
