High seroprevalence of viral hepatitis among animal handlers in Abeokuta, Ogun State, Nigeria.

Viral hepatitis is a deadly disease which can manifest as acute, chronic, hepatocellular carcinoma, and liver failure. Information about hepatitis is scarce among animal handlers. Due to Federal Government of Nigeria diversification programmes, many people are now involved in animal farming which can make them susceptible to viral hepatitis. This study aimed at determining the prevalence of Hepatitis B, C, and E viruses among animal handlers in Abeokuta, southwestern Nigerian. A total of 156 subjects were recruited for the study. Sociodemographic and risks factors data were fetched from subjec

Liver cell transplantation -- novel animal model for human hepatic viral infections.

AIM: To generate a model of human hepatitis B infection (HBV) in immunocompetent rats with chimeric human liver. METHODS AND RESULTS: Normal rats were tolerized to human hepatocytes by exposure to human hepatocytes at day 17 of gestation. We transplanted human hepatocytes and inoculated HBV into the rats after birth. Mixed lymphocytes assay, a measure for tolerance, indicated that animals fetally exposed to human hepatocytes developed tolerance to human hepatocytes. Spleen lymphocytes from tolerized animals did not proliferate when challenged with donor human hepatocytes. In contrast, control

Establishment of enterically transmitted hepatitis virus animal models using lipid nanoparticle-based full-length viral genome RNA delivery system.

BACKGROUND: Enterically transmitted hepatitis viruses, such as hepatitis A virus (HAV) and hepatitis E virus (HEV), remain notable threats to public health. However, stable and reliable animal models of HAV and HEV infection are lacking. OBJECTIVE: This study aimed to establish HAV and HEV infections in multiple small animals by intravenously injecting lipid nanoparticle (LNP)-encapsulated full-length viral RNAs (LNP-vRNA). DESIGN: In vitro transcribed and capped full-length HAV RNA was encapsulated into LNP and was intravenously inoculated to Ifnar-/- mice, and HEV RNA to rabbits and gerbil

A mouse with humanized liver as an animal model for predicting drug effects and for studying hepatic viral infection: where to next?

INTRODUCTION: The mouse is a common model used in evaluating drug metabolism and hepatitis infectivity. However, these models have limited value due to species difference in hepatic functions, leading to the creation of the chimeric mouse 12 years ago. These models were unique in that their hepatocytes had been replaced with human (hu) hepatocytes (dubbed the 'first-generation chimeric mouse'). Since then, the chimeric mouse has become a practical tool for this area of studies. However, some shortcomings have also been recognized. One major shortcoming is that the mouse cannot mimic hu-liver d

Hepatitis A virus infection in a chimpanzee: duration of viremia and detection of virus in saliva and throat swabs.

The pathogenesis of hepatitis A virus was studied by orally inoculating a chimpanzee with wild-type virus. The virus was initially detected in the animal's serum (day 14 after inoculation); then viral antigen was detected in the tonsils (day 16), virus in the saliva and throat swabs (day 18), and viral antigen in the liver (day 21). The animal was viremic for 2 w. While this study involved a single animal, the presence of hepatitis A virus in saliva and throat swabs suggests a possible oropharyngeal site for viral replication.

A small animal model of chronic hepatitis E infection using immunocompromised rats.

BACKGROUND & AIMS: HEV variants such as swine genotypes within Paslahepevirus species balayani (HEV-A) and rat HEV (Rocahepevirus ratti; HEV-C1) cause chronic hepatitis E in immunocompromised individuals. There are few reliable and accessible small animal models that accurately reflect chronic HEV infection. We aimed to develop an immunocompromised rat model of chronic hepatitis E infection. METHODS: In this animal model infection study, rats were immunosuppressed with a drug combination (prednisolone, tacrolimus, and mycophenolate mofetil) commonly taken by transplant recipients. Rats were c

Physiomimetic In Vitro Human Models for Viral Infection in the Liver.

Viral hepatitis is a leading cause of liver morbidity and mortality globally. The mechanisms underlying acute infection and clearance, versus the development of chronic infection, are poorly understood. In vitro models of viral hepatitis circumvent the high costs and ethical considerations of animal models, which also translate poorly to studying the human-specific hepatitis viruses. However, significant challenges are associated with modeling long-term infection in vitro. Differentiated hepatocytes are best able to sustain chronic viral hepatitis infection, but standard two-dimensional models

Advances and perspectives in animal models of human hepatitis A virus.

Hepatitis A virus (HAV) is an important pathogen that has continuously posed a threat to global public health for over 5000 years. The development of accessible and reliable small animal models, especially murine models, is essential for elucidating HAV pathogenesis and advancing preventive and therapeutic strategies. The first mouse model for HAV infection was established using human-liver chimeric mice, which supported robust viral replication and high viral loads. Later, an Ifnar1-/- mice model was established with a specific mouse-adapted strain, recapitulating key clinical features of hum

Beyond the liver: the broadened tissue tropism of hepatitis E virus.

Hepatitis E virus (HEV) is a leading cause of acute viral hepatitis, yet the virus has been associated with extrahepatic complications, including neurological and renal disorders. These extrahepatic manifestations pose challenges for the diagnosis and treatment of HEV and are considered a reservoir for disease relapse. In addition to recent in vivo patient data, several animal and in vitro models have been established in the last few years, confirming the broad HEV tissue tropism. However, it remains unclear whether extrahepatic complications arise from viral replication within the tissue or f

Occurrence of Hepatitis E Virus in Human Patients, Food and Animal Products From Slovakia.

Hepatitis E virus (HEV) is the only zoonotic pathogen that causes human viral hepatitis. Foodborne transmission of this virus is one of the most important transmission routes. In this study, performed in various hospitals between 2019 and 2024, we detected the HEV RNA in samples of 66 suspected patients in Eastern Slovakia. In total, 36 patients were found positive. Phylogenetic analysis of selected sequences (n = 26) showed that the majority (n = 22) clustered to genotype HEV-3 group 1. There were only four isolates that clustered to HEV-3 group 2. Statistically significant risk factors assoc

Hepatitis E virus infection during pregnancy: Advances in animal models.

Hepatitis E virus (HEV) is one of the major pathogens causing acute viral hepatitis worldwide, which usually causes acute self-limited diseases in general individuals. However, it can lead to high mortality and adverse pregnancy outcomes in pregnant women. Due to the lack of effective and stable cell culture models for HEV, the establishment of suitable animal models for HEV infection during pregnancy is necessary. An electronic search of the relevant database was conducted to identify eligible articles. Main animal models for the study of HEV infection during pregnancy include rabbits, swine,

Butchering activity is the main risk factor for hepatitis E virus (Paslahepevirus balayani) infection in southwestern Nigeria: a prospective cohort study.

INTRODUCTION: Paslahepevirus balayani (Hepatitis E virus; HEV) is an emerging virus that poses as a public health threat. The virus is now reported to be the leading cause of acute viral hepatitis, with a unique impact on African settings. Our aim was to evaluate the prevalence and risk factors for HEV infection in three cohorts (animal handlers, villagers, and students). METHODS: A prospective cross-sectional study was carried out on a total of 752 subjects from southwestern Nigeria. In all individuals, anti-HEV IgG and anti-HEV IgM antibodies were evaluated by using ELISA (confirming positi

Puzzles for Hepatitis E Virus.

Hepatitis E virus (HEV) is an important but understudied virus that has been the major cause of acute viral hepatitis worldwide. In recent decades, our understanding of this neglected virus has changed greatly: novel forms of viral proteins and their functions have been discovered; HEV can transmit via blood transfusion and organ transplantation; HEV can infect many animal species and the number is still increasing; HEV can induce chronic hepatitis and extra-hepatic manifestations. However, we are short of effective treatment measures to counter the virus. In this chapter we tend to briefly in

Prospect of acute hepatitis E virus outbreak in the context of the COVID-19 Pandemic in Africa: A contingency plan.

Hepatitis E virus (HEV) is the primary cause of acute viral hepatitis in Sub-Saharan Africa (SSA). It is transmitted by oral-faecal route via contaminated water or animal products, which is more pronounced in SSA, where inadequate hygiene measures, low socioeconomic situation, and frail healthcare system increase HEV susceptibility. During the coronavirus disease-19 (COVID-19), numerous initiatives concerning viral hepatitis relief in SSA are implemented. However, national programs do not support most services and are neglected when national and local attention rests on the COVID-19 pandemic.

Recent Drug Development in the Woodchuck Model of Chronic Hepatitis B.

Infection with hepatitis B virus (HBV) is responsible for the increasing global hepatitis burden, with an estimated 296 million people being carriers and living with the risk of developing chronic liver disease and cancer. While the current treatment options for chronic hepatitis B (CHB), including oral nucleos(t)ide analogs and systemic interferon-alpha, are deemed suboptimal, the path to finding an ultimate cure for this viral disease is rather challenging. The lack of suitable laboratory animal models that support HBV infection and associated liver disease progression is one of the major hu

Hepatitis E virus: host tropism and zoonotic infection.

Hepatitis E virus (HEV), the causative agent of hepatitis E, is an understudied but important pathogen. HEV typically causes self-limiting acute viral hepatitis, however chronic infection with neurological and other extrahepatic manifestations has increasingly become a significant clinical problem. The discovery of swine HEV from pigs and demonstration of its zoonotic potential led to the genetic identification of very diverse HEV strains from more than a dozen other animal species. HEV strains from pig, rabbit, deer, camel, and rat have been shown to cross species barriers and infect humans.
