[G-proteins, their role in the transduction of the hormonal message and their pathology].

Heterotrimeric G proteins belong to the superfamily of proteins which bind and hydrolyse GTP. Each of these GTPases acts as a molecular switch whose "on" and "off" states are triggered by binding and hydrolysis of GTP. Heterotrimeric G proteins (alpha beta gamma complex) transduce hormonal and sensory stimuli across the plasma membrane. The alpha chain contains the site of GTP binding and hydrolysis and the domains of interactions with receptors, effectors and beta gamma complex. GTP binding site is composed of five distinct domains denoted G1-5. Specific aminoacidic substitution or alteration

Localization of G-protein-coupled receptors in health and disease.

G-protein-coupled receptors (GPCRs) represent a superfamily of proteins, characterized by seven transmembrane alpha-helices, that signal through interactions with a family of heterotrimeric GTP-binding proteins, referred to as G proteins. The broad range of physiological functions associated with GPCRs indicates that a better understanding of these receptors and their regulation can provide a solid foundation for novel pharmacological interventions in a variety of disease states.

Characterization of the GTPγS release function of a G protein-coupled receptor.

G protein-coupled receptor (GPCR) signaling is one of the most ubiquitous and sensitive forms of cell surface reception. GPCRs stabilize the nucleotide-free state of heterotrimeric guanine-nucleotide binding proteins (G proteins); however, this state is produced at the cost of relieving the G protein of its stable inhibitor, GDP. Upon agonist binding to receptor, the G protein binds GTP and signal transduction ensues. Herein we demonstrate that the agonist can also stimulate the release of GTP. This receptor-mediated mechanism permits dissociation and reassociation of the G protein as the rece

Innovative Approach Reveals How Receptors Control G Protein State Equilibria.

G protein-coupled receptors (GPCRs) constitute the largest family of drug targets, acting through heterotrimeric G proteins. Agonist bound GPCRs promote GDP release from G proteins allowing their activation upon GTP binding. Here, we analyze this process with innovative tools that detect the main states of native Gi proteins: the GDP-bound, the open nucleotide-free and the active GTP-bound states. We show that receptor activation increases the amount of open-state Gi proteins, consistent with GDP release from inactive Gi proteins. We also observe that the active receptor can release GTP from t

Activation of pertussis toxin-sensitive G-proteins in membranes of SH-SY5Y human neuroblastoma cells and bovine transducin by ethanol.

Effects of ethanol on signal transduction in neuronal membranes are supposed to occur by the interaction with heterotrimeric guanine nucleotide-binding proteins (G-proteins). Several substances affect signal transduction by activation of G-proteins directly independent of receptors. We show that similar to those substances, ethanol stimulates high-affinity guanosine triphosphate (GTP)-hydrolysis in SH-SY5Y membranes at concentrations of 50 mM and higher in a pertussis toxin-sensitive manner. Compared with ethanol at a concentration of 170 mM, other alcohols were without or with respect to meth

Disease-Causing Mutations in the G Protein Gαs Subvert the Roles of GDP and GTP.

The single most frequent cancer-causing mutation across all heterotrimeric G proteins is R201C in Gαs. The current model explaining the gain-of-function activity of the R201 mutations is through the loss of GTPase activity and resulting inability to switch off to the GDP state. Here, we find that the R201C mutation can bypass the need for GTP binding by directly activating GDP-bound Gαs through stabilization of an intramolecular hydrogen bond network. Having found that a gain-of-function mutation can convert GDP into an activator, we postulated that a reciprocal mutation might disrupt the norm

Didemnin binds to the protein palmitoyl thioesterase responsible for infantile neuronal ceroid lipofuscinosis.

The marine natural product didemnin B, currently in clinical trials as an antitumor agent, has several potent biological activities apparently mediated by distinct mechanisms. Our initial investigation of didemnin B resulted in the discovery of its GTP-dependent binding of the translation elongation factor EF1 alpha. This finding is consistent with the protein synthesis inhibitory activity of didemnin B observed at intermediate concentrations. To begin to dissect the mechanisms involved in the cytostatic and immunosuppressive activities of didemnin B, observed at low concentrations, additional

Fluoride activation of the Rho family GTP-binding protein Cdc42Hs.

Aluminum tetrafluoride (AlF4-) activation of heterotrimeric G-protein alpha-subunits is a well established aspect of the biochemistry of these proteins; however, until recently it has been thought that AlF4- does not mediate effects on the Ras superfamily of low molecular weight GTP-binding proteins. Recent work demonstrating aluminum fluoride-induced complex formation between Ras and its GTPase-activating proteins (RasGAP and NF1) has provided important insights into the mechanism of GAP-stimulated GTP hydrolysis. We have characterized the AlF4--induced complex formation between the GDP-bound

GTP hydrolysis is modulated by Arg34 in the RASopathy-associated KRASP34R.

RAS proteins are commonly mutated in cancerous tumors, but germline RAS mutations are also found in RASopathy syndromes such as Noonan syndrome (NS) and cardiofaciocutaneous (CFC) syndrome. Activating RAS mutations can be subclassified based on their activating mechanisms. Understanding the structural basis for these mechanisms may provide clues for how to manage associated health conditions. We determined high-resolution X-ray structures of the RASopathy mutant KRASP34R seen in NS and CFCS. GTP and GDP-bound KRASP34R crystallized in multiple forms, with each lattice consisting of multiple pro

Activation of phospholipase D by ADP-ribosylation factor in rat submandibular acinar cells.

The hydrolysis of membrane phosphatidylcholine by the enzyme phospholipase D is a key initial step in the intracellular release of the signalling molecules phosphatidic acid, diacylglycerol and arachidonic acid. Guanine nucleotide-dependent pathway leading to PLD activation were investigated in enzymatically dispersed rat submandibular acinar cells. Guanosine 5'-O-[gamma-thio]triphosphate (GTP gamma S) caused the time- and concentration-dependent stimulation of PLD in permeabilized cells. This effect was lost in prepermeabilized cells, from which cytosolic components had been allowed to leak,

Inhibition of neutrophil function by aspirin-like drugs (NSAIDS): requirement for assembly of heterotrimeric G proteins in bilayer phospholipid.

Non-steroidal anti-inflammatory drugs (NSAIDs) inhibit neutrophil functions via mechanisms that are independent of their effects on prostaglandin biosynthesis. We examined the effects of sodium salicylate and piroxicam on GTP/GDP exchange by a regulatory G protein (G alpha i). Plasma membrane and cytosol of human neutrophils were prepared by nitrogen cavitation and discontinuous sucrose density centrifugation. Salicylate (3 mM) and piroxicam (50 microM) reduced [35S]GTP gamma S binding to purified plasma membranes [65 +/- 3.7 and 75 +/- 5.3% (P < 0.003) of control, respectively]. Membrane-asso

Characterization of Heterotrimeric G Protein γ4 Subunit in Rice.

Heterotrimeric G proteins are the molecule switch that transmits information from external signals to intracellular target proteins in mammals and yeast cells. In higher plants, heterotrimeric G proteins regulate plant architecture. Rice harbors one canonical α subunit gene (RGA1), four extra-large GTP-binding protein genes (XLGs), one canonical β-subunit gene (RGB1), and five γ-subunit genes (tentatively designated RGG1, RGG2, RGG3/GS3/Mi/OsGGC1, RGG4/DEP1/DN1/qPE9-1/OsGGC3, and RGG5/OsGGC2) as components of the heterotrimeric G protein complex. Among the five γ-subunit genes, RGG1 encodes th

The effects of interfering with GTP-binding proteins on the activation mechanism of calcium release-activated calcium current.

In electrically non-excitable cells, Ca2+ entry is mediated predominantly by the store-operated Ca2+ influx pathway, which is activated by emptying the intracellular Ca2+ stores. Just how the Ca2+ content of the stores is communicated to the activity of store-operated Ca2+ channels in the plasma membrane is unclear. It has been suggested that, in some cell types, the link is accomplished by either a small or a heterotrimeric GTP-binding protein, which is inhibited by guanosine 5'-O-(3-thiotriphosphate) (GTP[gamma-S]) and, in some cases, pertussis toxin. Using the whole-cell patch-clamp techniq

The biochemistry and physiology of metallic fluoride: action, mechanism, and implications.

Fluoride is a well-known G protein activator. Activation of heterotrimeric GTP-binding proteins by fluoride requires trace amounts of Al3+ or Be2+ ions. AlFx mimics a gamma-phosphate at its transition state in a Galpha protein and is therefore able to inhibit its GTPase activity. AlFx also forms complexes with small GTP-binding proteins in the presence of their GTPase-activating proteins (GAP). As phosphate analogs, AlFx or BeFx affect the activity of a variety of phosphoryl transfer enzymes. Most of these enzymes are fundamentally important in cell signal transduction or energy metabolism. Characterization of the intrinsic and TSC2-GAP-regulated GTPase activity of Rheb by real-time NMR.

Tuberous sclerosis complex 2 (TSC2), whose gene is frequently mutated in tuberous sclerosis, increases the guanosine triphosphatase (GTPase) activity of the small heterotrimeric GTP-binding protein (G protein) Rheb, thus resulting in the decreased activity of the mammalian target of rapamycin (mTOR), the master regulator of cell growth. Here, we describe the development of a nuclear magnetic resonance (NMR)-based, quantitative, real-time assay to explore the molecular mechanism of the intrinsic and TSC2-catalyzed GTPase activity of Rheb. We confirmed that TSC2 accelerated GTP hydrolysis by Rhe

Structural and Functional Implication of Natural Variants of Gαs.

Heterotrimeric guanine nucleotide-binding proteins (G proteins) are among the most important cellular signaling components, especially G protein-coupled receptors (GPCRs). G proteins comprise three subunits, Gα, Gβ, and Gγ. Gα is the key subunit, and its structural state regulates the active status of G proteins. Interaction of guanosine diphosphate (GDP) or guanosine triphosphate (GTP) with Gα switches G protein into basal or active states, respectively. Genetic alteration in Gα could be responsible for the development of various diseases due to its critical role in cell signaling. Specifical

Cardiomyopathy in congenital complete lipodystrophy.

Molecular genetic studies have pointed to a relationship between congenital lipodystrophy syndromes and some cardiac disorders. For instance, mutations in LMNA cause either lipodystrophy or cardiomyopathy, indicating that different mutations in the same gene can produce these clinical syndromes. The present authors describe a 10-year-old female with Berardinelli-Seip congenital complete lipodystrophy (MIM 606158) caused by homozygosity for a frameshift mutation in BSCL2. In addition to the typical attributes of complete lipodystrophy, this subject had hypertrophic cardiomyopathy diagnosed in t

Cholera toxin elevates pathogen resistance and induces pathogenesis-related gene expression in tobacco.

In animals, plants and fungi, cholera toxin (CTX) can activate signalling pathways dependent on heterotrimeric GTP binding proteins (G-proteins). We transformed tobacco plants with a chimeric gene encoding the A1 subunit of CTX regulated by a light-inducible wheat Cab-1 promoter. Tissues of transgenic plants expressing CTX showed greatly reduced susceptibility to the bacterial pathogen Pseudomonas tabaci, accumulated high levels of salicylic acid (SA) and constitutively expressed pathogenesis-related (PR) protein genes encoding PR-1 and the class II isoforms of PR-2 and PR-3. Heterotrimeric G proteins containing Gβ1γ2 subunits exhibit subtype-specific mobility differences in live cells.

Heterotrimeric G proteins are key signal transducers in all eukaryotic cells. Anchored to the inner leaflet of the plasma membrane, they transmit and amplify extracellular chemical and physical signals downstream of G protein-coupled receptors. Despite numerous available studies, many biophysical aspects that regulate G protein signaling, including membrane mobility, remain poorly understood. Here, using single-molecule imaging, we show that different subtypes of heterotrimeric G proteins containing Gβ1γ2 subunits exhibit high diversity in their membrane mobility. We demonstrate that the natur
