Differentiation of intestinal smooth muscle relaxation caused by drugs that inhibit phosphodiesterase.

In this study a number of chemically unrelated smooth muscle relaxants were tested: a) for potency of phosphodiesterase (PDE)-inhibition, using guinea pig colon-PDE and rat erythrocyte-PDE, b) for potency and duration of relaxation of the isolated guinea-pig colon, c) for their interaction with NH4Cl and orciprenaline as well as with "high calcium". Compared with the spasmolytic effect inhibition of guinea pig colon-PDE or rat erythrocyte-PDE was strong for papaverine and some other relaxants but was low or virtually absent with verapamil, hexobendine and bencyclane. The spasmolytic effect of

Subtypes of nucleoside transport inhibitory sites in heart: a quantitative autoradiographical analysis.

We evaluated the interaction of several nucleoside transport inhibitors and substrates with the binding of [3H]nitrobenzylthioinosine ([3H]NBMPR) to nucleoside transport sites in guinea pig cardiac sections. Using quantitative autoradiography, we determined inhibition constants for inhibition of [3H]NBMPR binding to both coronary endothelial cells and cardiac myocytes. We studied the interactions of NBMPR, nitrobenzylthioguanosine, dipyridamole, dilazep, hexobendine, lidoflazine, mioflazine, soluflazine, adenosine, inosine and uridine for these two cell types. Of the compounds tested in this s

Limiting solubilities and ionization constants of sparingly soluble compounds: determination from aqueous potentiometric titration data only.

A new method is described for the concomitant determination of limiting solubilities and ionization constants of sparingly soluble compounds, i.e., drugs. Aqueous potentiometric titration data were recorded both before and after precipitation of the compound and subjected to computer-assisted analysis. Limiting solubilities and ionization constants were obtained for nucleoside transport inhibitors, viz., dilazep, soluflazine, and hexobendine. The method was validated by comparison of titration results for known antidepressants with data from the literature. The procedure was found to be rapid

Assessment of pharmacological effects on cerebral blood flow.

The effect of various drugs on hemispheric and regional cerebral blood flow (CBF) was investigated in a total of 410 patients. While a few drugs (midodrin, proxazole, vincamine, hexobendine, extract of ginkgo biloba, dextran and ouabain) were able to improve hemispheric CBF, only ephedrine combined with xanthines decreased CBF. For vincamine the dependency of the effect on certain plasma levels was established. Only ouabain of the tested cardiac glycosides effected CBF; their similar hemodynamic actions suggest here an influence of ouabain on cerebral vessels. For the evaluation of drug effect

[Pharmacologic modification of the circulation in the brain].

Hemispheric and regional cerebral blood flow were measured in 448 patients before and after application of various drugs. Of the tested substances, only a few affected hCBF: ephedrine-xanthines decreased hCBF, and midodrine, proxazole, vincamine, hexobendine, extract of ginkgo biloba, dextran, dextran-sorbitol, carboanhydrase inhibitor, and ouabain incresaed hCBF. In the rCBF maps different reaction patterns of regional flow were observed which were statistically tested by regression analysis. Homogeneous responses were seen as diffuse increases or decreases of rCBF. Heterogeneous responses oc

[Instenon in the treatment of acute disorders of the cerebral circulation].

Effectiveness of Instenon (Alkaloid Pharm. Co.), the preparation, containing hexobendin, hydroxytheophyllin, etamivan was investigated in treatment of patients with acute cerebrovascular disorders. Instenon was applied in acute phase of ischemic stroke both in form of ampules and dragee as well as for intravenous injections too. Marked positive influence of Instenon was observed on motor, sensory, autonomic and psychic functions. Instenon in form of ampules had advantage compared to dragee form in terms of faster positive alterations onset. Application of Instenon in form of ampules was more e

Adenine nucleotides and synaptic transmission in the in vitro rat hippocampus.

1 The effects of adenosine and various derivatives were examined in the in vitro hippocampal slice preparation from rat.2 The amplitudes of extracellularly recorded field potentials from the CA1 region were depressed by adenosine, and this effect could be antagonized by methylxanthines. Because presynaptic field potentials were unaffected by adenosine, while the field e.p.s.p. was depressed, adenosine would appear to act at a synaptic site to depress transmission.3 Adenosine deaminase, which breaks down adenosine to inosine, increased the amplitude of synaptic responses, while hexobendine, whi

[Effect of instenon on the uterine contractile activity].

The effect of instenon upon the uterine contractile activity was studied in pregnant rats. The drug administration at a dose resulted in significant suppression of the myometrium contractility. The effect was not decreased when instenon was injected against the background of oxytocin. On the other hand, the concurrent injection of oxytocin upon a single or long-term administration of instenon produced a pronounced stimulating action on the myometrium contractility.

[The use of instenon in children with minimal brain dysfunction].

Minimal brain dysfunction (MBD) represents the most common type of neuropsychic disorders in childhood. Resulting in focal damages, underdevelopment and dysfunction of one or another cortical regions of brain hemispheres, MBD manifested in children as movement and speech disorders, dysgraphy, dyslexia, dyscalculia and attention deficit hyperactivity disorder. In the open controlled study, an efficacy of the complex nootropic medication "Instenon" was evaluated in the treatment of 59 MBD patients, aged 4-12 years. Control group included 27 children with MBD assigned to low polyvitamin (Sana Sol

[Use of instenon in complex treatment of patients with optic nerve atrophy caused by craniocerebral trauma].

Instenon, a combined nootropic agent, was used on the step-type basis within a complex treatment of optic nerve atrophy caused by craniocerebral trauma. Our study revealed that instenon, when used within a complex treatment of a partial optic nerve atrophy caused by craniocerebral trauma, improved the visual acuity in 85.7% of patients and it expanded the total visual field in 76.2% of patients, whereas the routine therapy ensured the related improvements in 58.9% and 61.5% of patients, respectively. Besides, the drug provided for a better retinal electric sensitivity and for a better optic ne
