Hindlimb suspension reduces muscle regeneration.

Exposure of juvenile skeletal muscle to a weightless environment reduces growth and satellite cell mitotic activity. However, the effect of a weightless environment on the satellite cell population during muscle repair remains unknown. Muscle injury was induced in rat soleus muscles using the myotoxic snake venom, notexin. Rats were placed into hindlimb-suspended or weightbearing groups for 10 days following injury. Cellular proliferation during regeneration was evaluated using 5-bromo-2'-deoxyuridine (BrdU) immunohistochemistry and image analysis. Hindlimb suspension reduced (P < 0.05) regene

Spaceflight and hindlimb suspension disuse models in mice.

Osteoporosis is a disease characterized by low bone mass and structural deterioration of bone tissue, leading to bone fragility and increased susceptibility to fractures. The microgravity of space creates an extreme environment that provides a model for osteoporosis in humans. This greatly accelerated form of osteopenia results in a 0.5-2% loss of bone mass per month. Rat models for this osteoporosis have been examined on many occasions, but STS-108 was the first Space Shuttle flight to use mice. Data reported to date indicate that spaceflight experiments with mice hold promise in predicting s

Myosin heavy chain composition of regenerated soleus muscles during hindlimb suspension.

The aim of this study was to determine the MHC profile of regenerated soleus muscles in control (C, n = 8) and hindlimb suspended rats (HS, n = 8). After muscle degeneration was induced by injection of snake venom containing notexin, male rats were either tail suspended for 21 days or submitted to normal weight-bearing activity. Separation and detection of MHCs by sodium dodecyl sulphate-polyarcylamide gel electrophoresis (SDS-PAGE) showed that regenerated soleus muscles from C rats contained only type I and type IIa MHCs. The relative amount of type I MHC was higher in regenerated (93.9 +/- 1

Long-term (6-wk) hindlimb suspension inhibits spermatogenesis in adult male rats.

The International Space Station will allow extended habitation in space and long-term exposure to microgravity (microG). A concern is the impact of long-term microG exposure on the ability of species to reproduce. The model often used to simulate microG is rat hindlimb suspension (HLS), where the hindlimbs are elevated above the cage floor with a tail harness. Experiments described here are the first to examine the effect of long-term HLS on testicular function in adult male rats. Free-roaming (controls), animals with only the tail harnessed but hindlimbs in contact with the cage floor (TO), a

Effects of Hindlimb Suspension on the Development of Hip Bone Morphologies in Growing Rats.

Abnormal hip bone morphologies are associated with various diseases of the hip joint. Weight bearing, especially during growth, may be important to achieve normal acetabulum development. This study aimed to investigate whether hip bone morphologies were affected by hindlimb suspension (HS) in 4 week-old rats. In HS groups, tail suspension was applied for 0, 2, 4, and 8 weeks. Age-matched rats were used as controls. The complex of hip bones with lumbar and sacral vertebrae were assessed based on morphological indexes using three-dimensional reconstructed images from X-ray computed tomography. A

Influence of one-week hindlimb suspension and intermittent high load exercise on rat muscles.

Hindlimb suspension (HS) is a ground-based simulation model for spaceflight commonly used to study the effects of unloading on skeletal muscle. The purpose of the present study was to determine the effects of a daily short-duration, high-load exercise training program in ameliorating the atrophic response and associated adaptations in the mechanical properties of the soleus (SOL) and medial gastrocnemius (MG) muscles to HS. Male Sprague-Dawley rats (mean body weight, approximately 250 g) were assigned randomly to one of three groups: control (CON, n = 7), HS, (n = 7), and HS plus exercise (HS-

Gait analysis for functional evaluation in a surgical hindlimb suspension model of muscle atrophy.

Disuse-induced skeletal muscle atrophy, commonly resulting from bedrest, immobilisation or spaceflight, leads to rapid loss of muscle mass and impaired mobility. Although muscle mass and contractile force are standard assessments in experimental models, these measures often fail to capture neuromuscular co-ordination deficits essential for effective movement. To better characterise these deficits, we employed a mouse hindlimb suspension (HLS) model for 14 days to induce disuse atrophy, confirmed by reductions in muscle mass, fibre type remodelling and satellite cell depletion, all of which wer

The analysis of antioxidant expression during muscle atrophy induced by hindlimb suspension in mice.

Oxidative stress contributes to acceleration of muscle atrophy. However, it is still not completely understood what triggers the production of reactive oxygen species (ROS) during muscle atrophy. The objective of this study was to investigate redox balance during muscle atrophy. ROS generators and antioxidants were analyzed in atrophied soleus muscles after 2 weeks of hindlimb suspension (HLS) in mice. The HLS group showed an increase in lipid peroxidation, upregulated NOX1 and NOXO1, and downregulated mitochondrial complex I subunits NDUFS5 and NDUFV2. Additionally, HLS mice demonstrated a de

Effects of therapeutic magnetic stimulation on acute muscle atrophy in rats after hindlimb suspension.

In most subjects with spinal cord injury, the spinal neurons below the level of injury are spared. Therefore, it is conceivable that the skeletal muscles innervated by these spinal nerves can be activated by applying therapeutic magnetic stimulation along the dorsal spine. The purpose of this study was to evaluate the ability of magnetic stimulation to prevent acute muscle atrophy in rats after hindlimb suspension. Forty adult male Wistar rats were randomly assigned to stimulated and non-stimulated (control) groups. Their hindlimbs were unweighted using a suspension method, causing muscle atro

Animal model of simulated microgravity: a comparative study of hindlimb unloading via tail versus pelvic suspension.

The aim of this study was to compare physiological effects of hindlimb suspension (HLS) in tail- and pelvic-HLS rat models to determine if severe stretch in the tail-HLS rats lumbosacral skeleton may contribute to the changes traditionally attributed to simulated microgravity and musculoskeletal disuse in the tail-HLS model. Adult male Sprague-Dawley rats divided into suspended and control-nonsuspended groups were subjected to two separate methods of suspension and maintained with regular food and water for 2 weeks. Body weights, food and water consumption, soleus muscle weight, tibial bone mi

Combination of hindlimb suspension and immobilization by casting exaggerates sarcopenia by stimulating autophagy but does not worsen osteopenia.

Astronauts in space experience a unique environment that causes the concomitant loss of bone and muscle. However, the interaction between these tissues and how osteopenia and sarcopenia affect each other is unclear. We explored this relationship by exaggerating unloading-induced muscle loss using a unilateral casting model in conjunction with hindlimb suspension (HLS). Five-month-old, male C57Bl/6J mice subjected to HLS for 2 weeks displayed a significant decrease in gastrocnemius and quadriceps weight (-9-10%), with a two-fold greater decrease in muscle mass observed in the HLS + casted limb.

Three-dimensional analysis of locomotion patterns after hindlimb suspension and subsequent long-term reloading in growing rats.

The long-term effects of insufficient weight loading during growth on locomotion patterns are not fully understood. The purpose of this study was to determine 1) the effects of hindlimb suspension (HS) in skeletally immature rats on locomotion patterns using a treadmill and a three-dimensional (3D) motion analysis system, and 2) the relationships between locomotion patterns and femoral morphologies, which were reconstructed from 3D computed tomography images taken at 54 weeks old. Four-week-old female rats were subjected to HS four or eight weeks, followed by reloading for until reaching up to

Effect of solar particle event radiation and hindlimb suspension on gastrointestinal tract bacterial translocation and immune activation.

The environmental conditions that could lead to an increased risk for the development of an infection during prolonged space flight include: microgravity, stress, radiation, disturbance of circadian rhythms, and altered nutritional intake. A large body of literature exists on the impairment of the immune system by space flight. With the advent of missions outside the Earth's magnetic field, the increased risk of adverse effects due to exposure to radiation from a solar particle event (SPE) needs to be considered. Using models of reduced gravity and SPE radiation, we identify that either 2 Gy o

The impact of hindlimb suspension on gastrointestinal integrity in aged rats: Segment-specific effects on inflammation and oxidative stress.

BACKGROUND: Aging compromises gastrointestinal integrity and increases inflammation, making the elderly more vulnerable to physiological stressors. Prolonged immobility due to bedrest, frailty, or simulated microgravity, may exacerbate these age-related effects. Despite growing concerns, the impact of immobility on the aging gastrointestinal tract remains largely unexplored. METHODS: Aged (24 month-old) male Sprague-Dawley rats (n = 8 per group) were assigned to control or 14-day hindlimb suspension (HLS) conditions to simulate immobility. Histological and molecular analyses were conducted on

The impact of mitokine MOTS-c administration on the soleus muscle of rats subjected to a 7-day hindlimb suspension.

The aim of the study was to investigate the effect of MOTS-c on the key functional alterations in the rat soleus muscle during 7-day unloading - the transformation of slow fibers into fast ones, atrophy and increased fatigue. We daily intraperitoneally injected male Wistar rats with a short mitochondrial peptide MOTS-c during 7-day unloading of their hind limbs. After the end of the experiment, we conducted an ex vivo fatigue test of soleus muscle and showed that the MOTS-c administration prevents increased fatigue during 7-day hind limb unloading. Also, using immunohistochemical analysis, we

Preventive effects of transcutaneous CO2 application on disuse osteoporosis and muscle atrophy in a rat hindlimb suspension model.

We previously demonstrated that transcutaneous CO2 application promotes muscle fiber-type switching, fracture healing, and osteogenesis by increasing blood flow and angiogenesis. Here, we aimed to investigate the preventive effects of transcutaneous CO2 application on disuse osteoporosis and muscle atrophy in a rat hindlimb suspension model. Eleven-week-old male Sprague-Dawley rats were divided into hindlimb suspension (HS), HS with transcutaneous CO2 application (HSCO2), and control groups. HSCO2 rats were administered transcutaneous 100 % CO2 gas in their bilateral hindlimbs, five times a we

Heat treatment combined with hybrid exercises retraining mitigates cellular markers of protein turnover after hindlimb suspension in male mice: A pilot study.

Recent evidence suggests that heat treatment (HT) and resistance training can limit skeletal muscle mass loss during immobilization. However, the effects of repeated HT sessions combined with hybrid exercises (EX), which promote both endurance and resistance responses, on muscle protein turnover following hindlimb unloading (HU) remain unexplored. This study investigated the effects of reloading strategies using HT and/or EX on protein synthesis, ribosome biogenesis and autophagic markers. Eight-week-old male C57BL/6J mice underwent HU (3 weeks), HU+HT (1 week, 5 sessions, 30 min, 40°C), HU+EX
