Maternal-fetal histocompatibility in intrauterine growth retarded and normal weight babies.

PROBLEM: To determine whether risk for intrauterine growth retardation (IUGR) is increased in HLA-DQA1 compatible pregnancies. METHOD: Paired maternal and cord blood samples were obtained from 30 IUGR and 31 non-IUGR pregnancies delivered at a university hospital. Samples were typed for eight HLA-DQA1 alleles using 10 sequence-specific oligonucleotides probes. Associations between IUGR and HLA-DQ compatibility status, and other risk factors were examined using logistic regression analysis. RESULTS: HLA-DQ compatibility and history of spontaneous abortion were not individually significant ris

Localization of fetal major histocompatibility complex antigens and maternal leukocytes in murine placenta. Implications for maternal-fetal immunological relationship.

An immunohistochemical study of the days 14-19 murine placenta and uterus using a panel of antibodies specific for maternal and paternal class I major histocompatibility complex (MHC) antigens, specific leukocyte subsets, and other lineage-specific markers was performed to elucidate the nature of the maternal tolerance of the fetal implant in the uterus. Fetally derived trophoblast lining maternal blood spaces lacked MHC antigens, but other interstitial trophoblasts expressed fetally derived polymorphic class I MHC determinants. The latter class I MHC-bearing trophoblasts were most prominent i

Histopathology of bile ducts in F1 rats hybridized from parents with different major histocompatibility antigens, undergoing inoculation with maternal strain lymphocytes during the fetal period.

In 1979, Seemayer suggested that biliary atresia (BA) might represent maternal or postnatally induced graft-versus-host reactions in which the liver is the principal target for aggressor lymphocytes. We attempted to produce perinatal graft-versus-host disease (GVHD) models to clarify the causal relationship between BA and GVHD. F1 rats were hybridized from parents with different major histocompatibility antigens: inbred strain maternal Lewis (LEW) and paternal Brown Norway (BN). F1 hybrid rats between 16 and 18 days' gestational age (term: 21 days) were given a single intrahepatic inoculation

Fetal-maternal HLA-C mismatch is associated with decidual T cell activation and induction of functional T regulatory cells.

Human leukocyte antigen-C (HLA-C) is the only polymorphic classical histocompatibility antigen expressed by fetal trophoblasts at the fetal-maternal interface. Interactions between HLA-C and decidual natural killer (NK) cells may facilitate trophoblast invasion into maternal tissue. Thus far no evidence has been provided that decidual T cells specifically recognize and respond to fetal alloantigens at the fetal-maternal interface. In this study, we show that pregnancies containing a HLA-C mismatched child induce an increased percentage of CD4(+)CD25(dim) activated T cells in decidual tissue. I

Nonclassical HLA and pseudogenes in maternal-fetal tolerance and cancer.

The immunological tolerance protecting the fetus from maternal rejection during pregnancy involves nonclassical human leukocyte antigen (HLA) class I molecules (HLA-G, HLA-E, HLA-F) interacting with maternal immune-inhibitory receptors. Cancers similarly exploit these molecules to evade immune detection and promote tumor progression. Pseudogenes within the major histocompatibility complex may modulate these pathways via noncoding RNA, gene conversion, or protein interactions, although their precise roles remain unclear. Furthermore, fetal-maternal microchimerism potentially reinforces maternal

Immunologic and neuroendocrine interactions in pregnancy.

The neuroendocrine system helps in the success of the fetal graft by suppressing maternal cellular immune response against the foreign paternal histocompatibility (HLA) antigens. In addition, placenta absorbs the antibodies directed against the paternal HLA antigens, thus inhibiting the humoral rejection of the fetal graft. In contrast, neuropeptides released in the maternal blood stream under adverse mental states may stimulate lymphocyte blastogenesis and natural killer (NK) cell activity resulting in premature loss of the fetus. Further, homozygosity of a lethal gene which is in linkage dis

Fetal microchimerism and the two-stage model of preeclampsia.

Fetal cells cross the placenta during pregnancy and some have the ability to persist in maternal organs and circulation long-term, a phenomenon termed fetal microchimerism. These cells often belong to stem cell or immune cell lineages. The long-term effects of fetal microchimerism are likely mixed, potentially depending on the amount of fetal cells transferred, fetal-maternal histocompatibility and fetal cell-specific properties. Both human and animal data indicate that fetal-origin cells partake in tissue repair and may benefit maternal health overall. On the other hand, these cells have been

Human CD8+ T-regulatory cells with low-avidity T-cell receptor specific for minor histocompatibility antigens.

Maternal/fetal microchimerism resulting from cell exchanges during pregnancy constitutes a reservoir of persisting alloantigen in mother and adult offspring. These alloantigens induce minor histocompatibility antigen-specific immune responses in both the mother and her offspring, including CD8(+) T regulatory (T(R)) cells with low T-cell receptor binding to major histocompatibility complex tetramers. Although they bind cognate major histocompatibility complex/peptide relatively poorly, these CD8 T(R) nonetheless inhibit high-avidity, tetramer-bright CD8 T effector responding to the same minor

New insights in understanding biliary atresia from the perspectives on maternal microchimerism.

Biliary atresia (BA) is a fibroinflammatory cholangiopathy and portal venopathy. It is of unknown etiology and is associated with systemic immune dysregulation, in which the first insult begins before birth. Maternal microchimerism is a naturally occurring phenomenon during fetal life in which maternal alloantigens promote the development of tolerogenic fetal regulatory T-cells in utero. However, maternal cells may alter the fetus's response to self-antigens and trigger an autoimmune response under certain histocompatibility combinations between the mother and the fetus. A recent report on a s

Maternal-fetal HLA sharing and preeclampsia: variation in effects by seminal fluid exposure in a case-control study of nulliparous women in Iowa.

Whereas histocompatibility is critical for transplantation, HLA histoincompatibility is associated with successful pregnancy. Literature on HLA sharing and preeclampsia has been inconsistent; most studies focused on maternal-paternal rather than maternal-fetal sharing. This study examines whether maternal-fetal histocompatibility is associated with preeclampsia, and whether effects vary by semen exposure history. This case-control study of nulliparous women was designed to examine associations among HLA sharing, semen exposure, and preeclampsia. 258 preeclampsia cases and 182 normotensive cont

A New Look at Immunogenetics of Pregnancy: Maternal Major Histocompatibility Complex Class I Educates Uterine Natural Killer Cells.

Our incomplete knowledge of maternal-fetal interface (MFI) physiology impedes a better understanding of the pathological mechanisms leading to pregnancy complications, such as pre-eclampsia and fetal growth restriction. At the MFI, uterine natural killer (uNK) cells do not attack fetal cells but engage in crosstalk with both fetal and maternal cells to support feto-placental development. However, mother and fetus are genetically half-mismatched and certain combinations of variable immune genes-human leukocyte antigens (HLAs) and killer-cell immunoglobulin-like receptor (KIR), indeed, the most

Human leucocyte antigen class Ib molecules in pregnancy success and early pregnancy loss.

BACKGROUND The human leucocyte antigen (HLA) class Ib molecules, HLA-E, -F and -G, are expressed at the materno-fetal interface. Because of the apparent immunoregulatory functions of these proteins, they may be involved in successful acceptance of the semi-allogenic fetus during pregnancy. METHODS The literature on polymorphisms of the three genes, expression patterns of the proteins, and interactions with immune cell receptors have been evaluated to elucidate whether HLA-E, -F and -G are involved in the pathogenesis of some cases of recurrent miscarriages and unexplained infertility. How does variability of immune system genes affect placentation?

Formation of the placenta is a crucial step in mammalian pregnancy. Apart from its function in ensuring an optimal supply of nutrients and oxygen to the fetus, the placenta is also the interface at which allo-recognition of invading trophoblast cells by the maternal immune system can potentially occur. We summarise here the "state of the art" on how variability of immune system genes that code for major histocompatibility complex (MHC) molecules and natural killer receptors (NKR) may impact on human placentation. MHC and NKR are the most polymorphic human genes. Our recent reports point out th

Parental HLA compatibility, fetal wastage and neural tube defects: evidence for a T/t-like locus in humans.

To test the hypothesis that a locus in or near the human major histocompatibility complex (HLA) contributes to both involuntary fetal loss and neural tube defects (NTD), we evaluated sharing of antigens of the HLA-A, HLA-B, or HLA-DR loci of couples who had three or more first-trimester spontaneous abortions or who had a child with an NTD (myelomeningocele or anencephaly). HLA-A antigen sharing was increased in couples with three or more spontaneous abortions and in couples who had an anencephalic fetus, when compared with couples who had three or more pregnancies and no fetal loss. Increased

Fetal Surgical conditions and the unraveling of maternal-fetal tolerance.

Fetal surgery is a fascinating field that will continue to evolve as we develop a more refined understanding of the underlying biology of various birth defects. Since preterm labor is a frequent outcome of fetal intervention, examining the mechanisms that lead to a breakdown in maternal-fetal tolerance is vital to developing strategies to overcome this limitation. The trafficking of cells between the mother and fetus during pregnancy plays a critical role in the education of the fetal immune system and may have implications for postnatal transplantation tolerance. Maternal cells may also be th

Maternal immunological recognition of pregnancy in equids.

There is little evidence for maternal immunological recognition of pregnancy in most species with the striking exception of the members of the genus Equus. Almost all mares make strong cytotoxic antibody responses to paternally inherited fetal antigens by Day 60 of gestation. Most of these responses are directed against antigens of the Major Histocompatibility Complex (MHC), which constitutes the primary immunogenetic barrier to successful organ transplantation. The source of fetal MHC antigens in the pregnant mare appears to be the specialized trophoblast cells of the chorionic girdle region

Immunotolerant functions of HLA-G.

HLA-G is a nonclassical major histocompatibility complex class I molecule selectively expressed on cytotrophoblasts at the fetal-maternal interface, where it plays a role in materno-fetal tolerance. In contrast to classical HLA-A, -B and -C class I molecules, HLA-G is characterized by (i) a tissue-restricted distribution, (ii) a limited polymorphism and (iii) a transcription of spliced messenger RNAs encoding for at least four membrane-bound and two soluble HLA-G isoforms. Extensive studies over the past few years have identified HLA-G as a molecule involved in immune tolerance. In this review

PBX1 expression in uterine natural killer cells drives fetal growth.

Abundant decidual natural killer (dNK) cells at the maternal-fetal interface are important during early pregnancy. However, functional subsets of dNK cells remain poorly understood. We describe a CD49a+PBX homeobox 1 (PBX1)+ dNK cell subset that promotes fetal development in humans and mice. The expression of PBX1 in dNK cells is up-regulated via the activated AKT1 pathway through the interaction of major histocompatibility complex G with the immunoglobulin-like transcript 2 receptor. PBX1 drives pleiotrophin and osteoglycin transcription in dNK cells, further promoting fetal development. Decr

Trophoblast expression of the minor histocompatibility antigen HA-1 is regulated by oxygen and is increased in placentas from preeclamptic women.

INTRODUCTION: Maternal T-cells reactive towards paternally inherited fetal minor histocompatibility antigens are expanded during pregnancy. Placental trophoblast cells express at least four fetal antigens, including human minor histocompatibility antigen 1 (HA-1). We investigated oxygen as a potential regulator of HA-1 and whether HA-1 expression is altered in preeclamptic placentas. METHODS: Expression and regulation of HA-1 mRNA and protein were examined by qRT-PCR and immunohistochemistry, using first, second, and third trimester placentas, first trimester placental explant cultures, and t
