Technologies for Conservation of Horseshoe Crabs: A Patent Landscape Analysis.

BACKGROUND: The horseshoe crab, a Xiphosurid species with an ancient lineage that dates back 450 million years, has proven to be a precious asset to the pharmaceutical industry. The blood extracted from these creatures is an irreplaceable component in detecting bacterial endotoxins, crucially important in pharmaceutical and functional settings. Unfortunately, these living ancient organisms are threatened from multiple perspectives and are now considered endangered. While efforts to conserve these creatures are underway, exploring technologies for their conservation can help us understand the l

Metal levels in horseshoe crabs (Limulus polyphemus) from Maine to Florida.

There is considerable concern for the health of spawning populations of horseshoe crabs (Limulus polyphemus) along the east coast of North America because of an increase in the harvest, an apparent decrease in population levels, and the dependence of migrating shorebirds on a superabundant supply of horseshoe crab eggs during their migratory stopover on Delaware Bay. In addition to overfishing, population declines could be caused or recovery slowed, by pollution. In this paper, we examine the levels of metals (arsenic, cadmium, chromium, lead, manganese, mercury, and selenium) in the eggs, leg

A SPECIATIONAL HISTORY OF "LIVING FOSSILS": MOLECULAR EVOLUTIONARY PATTERNS IN HORSESHOE CRABS.

Horseshoe crabs' exceptional morphological conservatism over the past 150 My has led to their reputation as "living fossils," but also has served to obscure phylogenetic relationships within the complex. Here we employ nucleotide sequences from two mitochondrial genes to assess molecular evolutionary rates and patterns among all extant horseshoe crab species. The American species Limulus polyphemus proved to be the sister taxon to a clade composed of the Asiatic species Tachypleus gigas, T. tridentatus, and Carcinoscorpius rotundicauda, whose relationships inter se were not resolved definitive

A horseshoe crab receptor structurally related to Drosophila Toll.

Innate immunity against microbial pathogens relies on the pattern recognition of cell wall components on invading microbes. Recent evidence has shown that a mammalian Toll-like receptor (TLR) is activated by bacterial lipopolysaccharides (LPS). The innate immunity in invertebrates is also triggered by LPS, as seen in the hemolymph coagulation in horseshoe crab. We report the cloning of a TLR from the Japanese horseshoe crab Tachypleus tridentatus. A cDNA coding for Tachypleus Toll was isolated from a hemocyte cDNA library and the open reading frame codes for a proprotein including a signal seq

Traumatic telson avulsion and attempted external stabilization in Atlantic horseshoe crabs (Limulus polyphemus).

Atlantic horseshoe crabs (Limulus polyphemus) are prevalent in public aquarium touch pools. Despite their popularity, the literature concerning medical management under managed care is sparse. Noninfectious conditions include trauma to the exoskeleton and compound eyes; however, injury to the soft tissue connecting the telson and opisthosoma has not been previously reported. This report describes telson avulsion in three Atlantic horseshoe crabs at a public aquarium and attempted external stabilization with a vascular silicone tie in two of three affected animals. The horseshoe crab that recei

Non-specific electrostatic interactions and the conversion of horseshoe crab haemocyanin into a phenoloxidase.

Haemocyanin is a haemolymph (blood)-based protein and the functional equivalent to haemoglobin - supplying tissues with oxygen in decapod crustaceans, chelicerates, and shelled molluscs. In addition to oxygen transport, haemocyanin plays several roles in innate immunity, wound healing, and ecdysis. Under certain conditions in vitro and in vivo, horseshoe crab (Limulus polyphemus) haemocyanin is converted into a phenoloxidase-like enzyme, yet the protein-ligand interactions associated with this conversion remain unclear. Negatively charged ligands, such as phosphatidylserine and sodium dodecyl

The horseshoe crab: a model for gram-negative sepsis in marine organisms and humans.

The roles of the amebocyte in providing hemostasis and controlling infection, and its reaction to endotoxin, suggest that the response of platelets and the blood coagulation system in various mammals to gram-negative infection or endotoxin is an evolutionary remnant of this ancient mechanism. In humans, this mechanism occasionally subverts its presumed protective function by overresponding in a manner that results in pathophysiologic thrombosis or hemorrhage. (In this regard, it is interesting that human platelets are much more resistant to the effects of bacterial endotoxins than are other sp

Toxicity assessment for the horseshoe crab Carcinoscorpius rotundicauda collected from Cambodia.

In this study, we assessed the toxicity of the horseshoe crab Carcinoscorpius rotundicauda collected from Cambodia within two successive months during rainy (April-May) and dry (December-January) seasons, respectively. Toxicity assessments of the collected specimens by standard mouse bioassay showed marked individual variation, and their toxin profiles by liquid chromatography/mass spectrometry (LC/MS) revealed tetrodotoxin (TTX) was the main toxin while no paralytic shellfish toxins (PSTs) were detected. All specimens were toxic and the highest toxicity values were 315, 113, 60, 47, 44 and 38

Spatial control of rhabdom shedding in the lateral eye of the American horseshoe crab, Limulus polyphemus.

Membrane leaves the rhabdom of Limulusphotoreceptors either by transient shedding at dawn or throughout the day by light-driven shedding. We examined whether the light trigger for transient shedding and the light drive for light-driven shedding are localized properties of the illuminated photoreceptors or whether they are an array property of the retina. Four experiments were conducted during which the lateral eye was exposed to one of a variety of different illumination patterns for a day, fixed, dissected and cut into serial frozen sections. Horseshoe crab hemocyte-derived antimicrobial polypeptides, tachystatins, with sequence similarity to spider neurotoxins.

Antimicrobial peptides, named tachystatins A, B, and C, were identified from hemocytes of the horseshoe crab Tachypleus tridentatus. Tachystatins exhibited a broad spectrum of antimicrobial activity against Gram-negative and Gram-positive bacteria and fungi. Of these tachystatins, tachystatin C was most effective. Tachystatin A is homologous to tachystatin B, but tachystatin C has no significant sequence similarity to tachystatins A and B. Tachystatins A and B showed sequence similarity to omega-agatoxin-IVA of funnel web spider venom, a potent blocker of voltage-dependent calcium channels. Ho

[Baculovirus expression system-based expression of horseshoe crab factor C and its activity].

Endotoxins are common exogenous pyrogens. Excessive endotoxins in medical devices and injections can lead to serious consequences such as sepsis, septic shock, and even death. Therefore, endotoxin detection plays a crucial role in medical, pharmaceutical, and food sectors. The wide application of Limulus amebocyte lysate (LAL) has led to a sharp decline in the number of horseshoe crabs. Moreover, the LAL assay has limitations such as interbatch variations and difficulty in quantification. The recombinant factor C (rFC) assay is stable between batches, highly sensitive, and capable of quantitat

Particle size influences physiological responses to chronic exposure to polyethylene particles in juvenile Atlantic horseshoe crabs (Limulus polyphemus).

Plastic pollution poses a growing threat to marine biodiversity, and its biological effects may vary with particle size. Early life stages of the Atlantic horseshoe crab, Limulus polyphemus, inhabit coastal environments that can accumulate plastic particles across a broad size range. This study investigated the effects of small (0.2-9.9 μm) and large (53-63 μm) polyethylene particles on juvenile L. polyphemus exposed for 21 days to 5 μg L-1. Histological characterization of the gastrointestinal tract of the adult horseshoe crab (Limulus polyphemus) with special reference to the stomach.

The American horseshoe crab (Limulus polyphemus) is one of four extant species in the Order Xiphosura, subphylum Chelicerata, and are evolutionarily more closely related to scorpions and spiders, than crabs. The basic structure, function, and physiology of these invertebrates and their internal organs are not well documented in the literature. In this study, the gastrointestinal system, with a focus on the stomach, of adult L. polyphemus were assessed by gross and histologic methods to further characterize the pyloric valve, the lining of the ventricular lumen, and the muscular tunics of the s

Response of moulting genes and gut microbiome to nano-plastics and copper in juvenile horseshoe crab Tachypleus tridentatus.

Nanoplastics (NPs) and heavy metals are typical marine pollutants, affecting the gut microbiota composition and molting rate of marine organisms. Currently, there is a lack of research on the toxicological effects of combined exposure to horseshoe crabs. In this study, we investigated the effects of NPs and copper on the expression of molt-related genes and gut microbiome in juvenile tri-spine horseshoe crabs Tachypleus tridentatus by exposing them to NPs (100 nm, 104 particles L-1) and/or Cu2+ (10 μgL-1) in seawater for 21 days. Compared with the control group, the relative mRNA expression of

Preliminary Reference Interval Estimation for Hemolymph Analysis in Free-Ranging Mangrove Horseshoe Crab (Carcinoscorpius rotundicauda).

BACKGROUND: The mangrove horseshoe crab (Carcinoscorpius rotundicauda), a species thriving in the muddy brackish waters of Cox's Bazar, holds significant ecological and biomedical value. Hematological and biochemical analyses can serve as a valuable diagnostic tool for assessing health; however, reports remain limited for horseshoe crabs. OBJECTIVE: This study investigated hemolymph values to determine preliminary reference intervals (RIs) from free-ranging mangrove horseshoe crabs. METHODS: Forty adult Carcinoscorpius rotundicauda (20 males, 20 females) were collected from intertidal zones

Effects of pristine and UV-aged polystyrene nanoplastics on post-molt horseshoe crabs Tachypleus tridentatus: Aging-mediated divergence in biological effects.

The Chinese horseshoe crab (Tachypleus tridentatus), an intertidal chelicerate with a 400-million-year evolutionary history, is increasingly exposed to anthropogenic pollutants in its coastal nursery habitats. Because of the ecological importance of the species and the vulnerability of the post-molt stage, we investigated the effects of pristine and 7-day UV-aged polystyrene nanoplastics (NPS) on juvenile T. tridentatus at an environmentally relevant nominal concentration of 100 μg/L. Fourier transform infrared spectroscopy confirmed surface oxidation of aged NPS, as reflected by significantly

Responses of growth and hemolymph quality in juvenile Chinese horseshoe crab Tachypleus tridentatus (Xiphosura) to sublethal tributyltin and cadmium.

Responses of growth endpoints and hemolymph constituents in juvenile Chinese horseshoe crab Tachypleus tridentatus under treatments of 0.01 and 0.1 mg/l tributyltin (TBT) and 0.1 and 1 mg/l cadmium (Cd) were examined in a 12-week experiment. A significant decline in final body weight, final prosomal width, percentage of individuals molted and mean molting time was detected under TBT exposures. While morphological abnormalities of the juveniles between TBT treatments and the control were statistically indistinguishable, significantly higher occurrences of carapace erosion and appendage loss wer
