Comparison of two DNA extraction methods widely used in aquatic microbial ecology.

In recent years, the rapid advances of culture-independent methods and new molecular tools have revolutionized our understanding of microbial biodiversity and ecological functions. DNA extraction from microbial communities is a critical step in this process and several methods have been proposed and used, but the influence of the extraction method on the outcome and ultimately on ecological inferences from the results is not yet precisely determined. Here, we compared two of the most commonly used extraction methods in aquatic microbial ecology, and investigated whether the two methods yielded

[Abundance Dynamic of Oocysts in the Groundwater of Mbankomo, a Sub-urban Area Yaounde: Physico-chemical Factors and Health Risk].

To determine the dynamics of dissemination of the different forms of enteric sporozoa in the groundwater of Mbankomo, a study was carried out from August 2018 to July 2019. The physico-chemical analyzes were carried out both in the field and in the hydrobiology and environment laboratory of the University of Yaoundé I. The observation of protozoan oocysts was carried out after application of the Ziehl-Neelsen staining. Physico-chemical analysis showed slightly acidic water (6.32 ± 0.43 UC), with high turbidity (31.30 ± 23.56 mg/l). Biological analysis indicated the presence of protozoan oocyst

Evaluation of the floating time of a corpse found in a marine environment using the barnacle Lepas anatifera L. (Crustacea: Cirripedia: Pedunculata).

Human activities involving water may result in a crime scene. Typically, death may be due to natural causes, homicide, or mass disasters. Decomposition in water is a complex process where many factors may interplay. Human remains in water are subject to many potential interactions, depending upon the remains themselves, the type of water and the characteristics of the water. A number of studies are focused on the decomposition process of the corpse in water, on the identification of the post mortem submersion interval (PMSI) and on the diagnosis of drowning, but very few studies consider the f

[Mercury pollution in selected food products in Poland in the context of their health safety].

Mercury is a silvery-white chemically active transition metal without known physiological role. It is also a toxin accumulated in living organisms--especially in the aquatic creatures (fish and shellfish). Mercury compounds are involved in biogeochemical cycles (especially atmospheric and hydrobiologic), and thus they become contaminants of food. Such process is facilitated by the increasing environmental chemization. Toxic effects of mercury compounds result from their high affinity to sulfur (sulfhydryl groups of amino acids), accumulation in parenchymal organs, metallic mercury lipophilicit

Exploring global Cryptosporidium emissions to surface water.

The protozoan parasite Cryptosporidium is a major cause of diarrhoea worldwide. This paper presents the first model-based inventory with 0.5 by 0.5 degree resolution of global Cryptosporidium emissions for the year 2000 from humans and animals to surface water. The model is based on nutrient distribution modelling, because the sources and transport of oocysts and nutrients to the surface water are comparable. Total emissions consist of point source emissions from wastewater and nonpoint source emissions by runoff of oocysts in manure from agricultural lands. Results indicate a global emission

Modeling vitamin B1 transfer to consumers in the aquatic food web.

Vitamin B1 is an essential exogenous micronutrient for animals. Mass death and reproductive failure in top aquatic consumers caused by vitamin B1 deficiency is an emerging conservation issue in Northern hemisphere aquatic ecosystems. We present for the first time a model that identifies conditions responsible for the constrained flow of vitamin B1 from unicellular organisms to planktivorous fishes. The flow of vitamin B1 through the food web is constrained under anthropogenic pressures of increased nutrient input and, driven by climatic change, increased light attenuation by dissolved substanc

Emerging human infectious diseases of aquatic origin: a comparative biogeographic approach using Bayesian spatial modelling.

BACKGROUND: With the increase in unprecedented and unpredictable disease outbreaks due to human-driven environmental changes in recent years, we need new analytical tools to map and predict the spatial distribution of emerging infectious diseases and identify the biogeographic drivers underpinning their emergence. The aim of the study was to identify and compare the local and global biogeographic predictors such as landscape and climate that determine the spatial structure of leptospirosis and Buruli Ulcer (BU). METHODS: We obtained 232 hospital-confirmed leptospirosis (2007-2017) cases and 2

[Elucidation of Phenomena Involving Cyanobacteria in Freshwater Ecosystem by Chemically Ecological Approach].

Lakes Sagami and Tsukui are reservoirs constructed by connecting to the Sagami River. Because of eutrophication of the lakes, cyanobacteria have appeared every year. This review deals with phenomena related to occurrence of cyanobacteria that have been observed for 40 years since 1974 at the lakes. These 40 years of observations raised three interesting issues including the retention of cyanobacteria on their surfaces. These phenomena have been attributed to the usual factors, such as illuminance, nutrition and water temperature, but our research results suggested that they cannot be resolved

Hierarchical dynamics influence the distribution of immature black flies (Diptera: Simuliidae).

Adult black flies (Simuliidae) are medically important insects and they are the sole vector of Onchocerca volvulus. Immature black flies are major components of aquatic macroinvertebrate assemblages in streams and play a vital role in nutrient dynamics. In this study, we examined effect of hierarchical dynamics (spatio-temporal pattern) on the distribution of immature black flies in South Indian streams. The sampling was done in streams of Western Ghats, South India. A total of 16 species belong to two subgenera: Simulium (10 species) and Gowmphostilbia (6 species) of Simulium were observed. A

Severe Hearing Loss in the World's First Successfully Captive-Born Yangtze Finless Porpoise: Impact of High Underwater Sound Exposure and Congenital Hearing Disorders.

Aquariums globally have seen significant growth in recent decades. However, persistent negligence exists concerning underwater sound pollution in aquariums and its impact on cetaceans. Here, the audiogram of Taotao, the world's first successfully captive-born and bred Yangtze finless porpoise, and the underwater sound levels in the Baiji Aquarium at the Institute of Hydrobiology, Chinese Academy of Sciences were examined. In contrast to the previously published U-shaped audiograms of the Yangtze finless porpoise, Taotao's audiogram exhibited a W-shaped pattern. Additionally, the audiogram of T

Optimization of Slash and Clear Community-Directed Control of Simulium damnosum Sensu Stricto in Northern Uganda.

Onchocerciasis, caused by infection with Onchocerca volvulus, has been targeted for elimination by 2030. Currently, onchocerciasis elimination programs rely primarily on mass distribution of ivermectin. However, ivermectin alone may not be sufficient to achieve elimination in some circumstances, and additional tools may be needed. Vector control has been used as a tool to control onchocerciasis, but vector control using insecticides is expensive and ecologically detrimental. Community-directed removal of the trailing vegetation black fly larval attachment sites (slash and clear) has been shown

Nutritional status and prey energy density govern reproductive success in a small cetacean.

A variety of mammals suppress reproduction when they experience poor physical condition or environmental harshness. In many marine mammal species, reproductive impairment has been correlated to polychlorinated biphenyls (PCBs), the most frequently measured chemical pollutants, while the relative importance of other factors remains understudied. We investigate whether reproductively active females abandon investment in their foetus when conditions are poor, exemplified using an extensively studied cetacean species; the harbour porpoise (Phocoena phocoena). Data on disease, fat and muscle mass a

Aeolian dust and hydro-biological characteristics: Decoding dust storm impacts on phytoplankton in the northern Arabian Gulf.

Aeolian dust is an essential source of growth-limiting nutrients for marine phytoplankton. Despite being at the core of the Global Dust Belt, the response of the Arabian Gulf ecosystem to such atmospheric forcing is rarely documented. Here, the hydro-biological effect of mineral dust was studied in the northern Arabian Gulf (NAG) off Kuwait through monthly water sampling (December 2020 to December 2021), dust-storm follow-up sampling, and mineral dust and nutrient addition in-situ experiments. The multivariate analysis of oceanographic data revealed pronounced hydro-biological seasonality. The

Got Glycogen?: Development and Multispecies Validation of the Novel Preserve, Precipitate, Lyse, Precipitate, Purify (PPLPP) Workflow for Environmental DNA Extraction from Longmire's Preserved Water Samples.

Unfiltered and filtered water samples can be used to collect environmental DNA (eDNA). We developed the novel "Preserve, Precipitate, Lyse, Precipitate, Purify" (PPLPP) workflow to efficiently extract eDNA from Longmire's preserved unfiltered and filtered water samples (44-100% recovery). The PPLPP workflow includes initial glycogen-aided isopropanol precipitation, guanidium hypochlorite and Triton X-100-based lysis, terminal glycogen-aided polyethylene glycol precipitation, and inhibitor purification. Three novel eDNA assays that exclusively target species invasive to Australia were also deve

Range-wide Yangtze freshwater dolphin expedition: The last chance to see Baiji?

BACKGROUND, AIM AND SCOPE: There are two species of fresh water cetaceans surviving in the Yangtze River system in China: Baiji (Lipotes vexillifer) and Yangtze finless porpoise (Neophocaena phocaenoides asiaeorientalis). As a result of the expansion of human activities on the river, their distribution ranges appear to be decreasing and in the case of the Baiji, are even being restricted to several sections. The Baiji is the world's most critically endangered cetacean species with a population estimated at only a few tens of individuals. The Yangtze finless porpoise is the world's only freshwa
