Origin of cooperativity in hydrogen bonding.

The origin of non-additivity in hydrogen bonds (H-bonds), usually termed as H-bond cooperativity, is investigated in H-bonded linear chains. It is shown that H-bond cooperativity originates solely from classical electrostatics. The latter is corroborated by comparing the H-bond cooperativity in infinitely-long H-bonded hydrogen cyanide, 4-pyridone and formamide chains, assessed using density functional theory (DFT), against the strengthening of the dipole-dipole interaction upon the formation of an infinite chain of effective point-dipoles. It is found that the magnitude of these effective poi

Intramolecular Hydrogen Bonding in Substituted Aminoalcohols.

The qualifying features of a hydrogen bond can be contentious, particularly where the hydrogen bond is due to a constrained intramolecular interaction. Indeed there is disagreement within the literature whether it is even possible for an intramolecular hydrogen bond to form between functional groups on adjacent carbon atoms. This work considers the nature of the intramolecular interaction between the OH (donor) and NH2 (acceptor) groups of 2-aminoethanol, with varying substitution at the OH carbon. Gas-phase vibrational spectra of 1-amino-2-methyl-2-propanol (BMAE) and 1-amino-2,2-bis(trifluor

Improvement of dentin bonding via adhesive monomers with multiple hydrogen bonding moieties.

Despite advancements in bonding techniques, the resin-dentin interface remains the weakest point in dental restorations, susceptible to collagen degradation and methacrylate hydrolysis. One strategy to enhance the resin-dentin interface is to incorporate hydrogen-bonding-rich functional groups into dental adhesive resins, such as 2-ureido-4[1H]-pyrimidinone (UPy). These hydrogen bonds may bridge the adhesive resin and dentin substrate, which contains collagen and hydroxyapatite, as well as form non-covalent crosslinks within the resin. Here, we utilize UPy-functionalized methacrylamides modifi

Hydrogen bonding in water under extreme confinement.

Fluids under extreme confinement or near interfaces exhibit molecular structures and intermolecular bonding distinct from their bulk analogues. Optical vibrational studies have shown that interfacial and confined water can exhibit altered intramolecular O-H stretching frequencies, a sensitive spectral signature of changes in intermolecular hydrogen bonding1-4. Investigating confined water experimentally at the length scale of intermolecular and surface forces has, however, remained a challenge. Here we report direct molecular-level observations of hydrogen bonding in water confined inside indi

Hydrogen bonding triggered tunable high temperature phosphorescence.

Due to the luminescence quenching at elevated temperature, the development of high temperature phosphorescent (HTP) materials with tunable emission remains a formidable challenge. Herein, a hydrogen bonding (HB) triggered strategy to achieve color-tunable and thermally robust room temperature phosphorescence (RTP) was demonstrated. Using a triazine derivative (TRZ) as a luminescent core and HB acceptor and a series of acids, o-pyridinesulfonic acid (OPS), ethanesulfonic acid (ESA), or trifluoroacetic acid (TFA), as HB donors, a series of stable HB luminescent systems were constructed, which no

Hydrogen Bonding-Driven Adaptive Coacervates as Protocells.

Coacervation based on liquid-liquid phase separation (LLPS) has been widely used for the preparation of artificial protocells and to mimic the dynamic organization of membrane-free organelles. Most complex synthetic coacervates are formed through electrostatic interactions but cannot withstand high ionic strength conditions (>0.1 M). Alternative components and driving forces are highly desired for the formation of natural organelles to overcome the drawbacks of traditional coacervates. Herein, hydrogen bonding-driven adaptive coacervates are reported via the complexation of poly(ethylene glyco

Hydrogen Bonding (Base Pairing) in Antiviral Activity.

Base pairing based on hydrogen bonding has, since its inception, been crucial in the antiviral activity of arabinosyladenine, 2'-deoxyuridines (i.e., IDU, TFT, BVDU), acyclic nucleoside analogues (i.e., acyclovir) and nucleoside reverse transcriptase inhibitors (NRTIs). Base pairing based on hydrogen bonding also plays a key role in the mechanism of action of various acyclic nucleoside phosphonates (ANPs) such as adefovir, tenofovir, cidofovir and O-DAPYs, thus explaining their activity against a wide array of DNA viruses (human hepatitis B virus (HBV), human immunodeficiency (HIV) and human h

Hydrogen bonding properties of non-polar solvents.

A combination of high-throughput NMR titration experiments, UV-Vis absorption titrations and data collected from the literature on 1:1 H-bonded complexes has been used to characterise the H-bond properties of non-polar organic solvents: alkanes, perfluorocarbons, aromatic and halogenated organic solvents. The results are analysed in the context of the electrostatic solvent competition model, which assumes that solvent effects on intermolecular interactions can be interpreted based on the exchange of specific functional group contacts, with minimal involvement of the bulk solvent. For solvents

Hydrogen bonding in salicylsalicylic acid (salsalate) crystals.

An X-ray crystallographic study of the drug salsalicylic acid (salsalate) has been performed. Crystal formation of the drug is influenced by both inter- and intra-molecular hydrogen bonding. In addition an OH group in salsalate can occupy alternate ortho positions resulting in two hydrogen bonding motifs within a single crystal.

Halogen-Bonding-like Interactions Complement Hydrogen Bonding in a Human Flavin-Dependent Dehalogenase.

An accurate description of halogen bonding in biomolecules remains a challenge in modern biochemistry, as exemplified by the flavin-dependent human dehalogenase, iodotyrosine deiodinase. Analyses of the static crystal structure and kinetics data suggested that halogen bonding is absent in the enzyme's active site, which does not preclude its transient formation along the binding or unbinding pathway. No studies have yet explored iodotyrosine deiodinase's dynamic behavior from the perspective of halogen bonding. Here, extensive equilibrium and nonequilibrium molecular dynamics (MD) simulations

Hydrogen-bonding-controlled stereospecific synthesis of Z-enamides.

In this study, a series of Z-enamide compounds were synthesized from α-amino ketones and alkynyl esters. Using triethylamine as the base catalyst, the reaction proceeded smoothly at room temperature to yield stereospecific products with high yields. Two X-ray crystals of products confirmed the crucial role of hydrogen bonding in stereospecificity.

Uptake of Hydrogen Bonding Molecules by Benzene Nanoparticles.

The uptake of molecules on nanometer-size clusters of polyaromatic hydrocarbons (PAHs) is important for the condensation of water on PAH aerosols in the atmosphere and for ice mantle growth on nanoparticles in the interstellar medium. We generate benzene clusters BzN of mean size N̅ ≈ 300 (radius R̅ ≈ 2.2 Å) as a model system for the PAH nanoparticles. Using molecular beams and mass spectrometry detection, we investigate the uptake of water, methanol, and ethanol by these clusters. All picked up molecules are highly mobile on BzN and generate clusters within <3 ms. The relative uptakes for the

Topological analysis of hydrogen bonding in protein structure.

A recent study has shown that topological stereoisomers exist for the polypeptide chain in disulfide-containing proteins that are represented by non-planar graphs. This topological stereochemistry is demonstrated in the structure of variant 3 toxin in the venom of the North American scorpion Centruroides sculpturatus Ewing and the structure of toxin II from the North African scorpion Androctonus australis Hector. In this report, we found that a similar topological analysis can be applied to the hydrogen bonding in alpha-helices and beta-sheets within protein molecules, and we described the top

Stereoretentive Norrish-Yang Photocyclization Mediated by Hydrogen Bonding.

The Norrish-Yang photocyclization of aryl alkyl ketones is a classical photochemical transformation that proceeds through triplet 1,4-diradicals whose conformations govern the fate of the reaction. Directing the reactivity of these diradicals is difficult because they can evolve through competing pathways, including cyclization and Norrish type II fragmentation. Achieving stereocontrol is even more challenging because radicals generated from enantiopure substrates are expected to undergo rapid configurational scrambling. Here we report a stereoretentive Norrish-Yang photocyclization of enantio

Stereoelectronic and hydrogen-bonding effects on hydroxyproline conformation.

Collagen's structural integrity and biological function derive from its characteristic triple helix, governed by a unique amino acid sequence containing the rare residue (2S,4R)-hydroxyproline (4R-Hyp). Substituting 4R-Hyp with its diastereomer 4S-Hyp markedly reduces triple-helix thermal stability and impairs collagen function. The origins of this destabilization have mostly been ascribed to stereoelectronic effects such as the gauche effect and n → π* interactions. Here, we dissect the role of different stabilizing interactions on molecular conformation using linear infrared and two-dimensio

The Long and Short Radii for Hydrogen in Hydrogen Bonded Complexes.

Hydrogen bonding was initially identified by comparing the distance between the heavy atoms D and A in a DH•••A hydrogen bond, to the sum of their van der Waals (vdW) radii as locating the H atom was not possible. In recent years, hydrogen bond radii (HBR) have been defined for typical hydrogen bond donors DH and acceptors, A. However, both these approaches make an unreasonable assumption of treating all the atoms as spheres. This study advances the understanding of hydrogen bond distances by estimating the shape of hydrogen atoms in HD molecules and in A•••HD complexes through electron de

Solid-State Hydrogen-Bond Alterations in a [Co2 Fe2 ] Complex with Bifunctional Hydrogen-Bonding Donors.

Compound 1 solv exhibited a gradual electron-transfer-coupled spin transition (ETCST), and the removal of solvent molecules led to an abrupt thermal ETCST behavior with increased transition temperature. X-ray structural analysis revealed that the modification of ETCST was caused by a significant alteration of a h

Exploring Orthogonality between Halogen and Hydrogen Bonding Involving Benzene.

The concept of orthogonality between halogen and hydrogen bonding, brought out by Ho and coworkers some years ago, has become a widely accepted idea within the chemists' community. While the original work was based on a common carbonyl oxygen as acceptor for both interactions, we explore here, by means of M06-2X, M11, ωB97X, and ωB97XD/aug-cc-PVTZ DFT calculations, the interdependence of halogen and hydrogen bonding with a shared π-electron system of benzene. The donor groups (specifically NCBr and H2O) were placed on either or the same side of the ring, according to a double T-shaped or a per
