Hypergravity studies in the Netherlands.

It looks like that with the utilization phase of the International Space Station (ISS) scientists will have the possibility to perform long duration and more sophisticated microgravity experiments than could be performed previously. In preparation for these spaceflight studies, ground based experiment tools for simulated (or real) microgravity and hypergravity are important. To provide the infrastructure and user support necessary to perform these ground based studies we have setup the Dutch Experiment Support Center, DESC. This paper will focus on the three Dutch centrifuge facilities. It is

Hypergravity effects on pregnancy and parturition.

The events of mammalian parturition (labor and birth) were analyzed in Norway rats exposed to either 1.0-g, 1.5-g or 2.0-g hypergravity (hg) beginning at the approximate midpoint of their pregnancies. Pregnant dams were 24h timelapse videorecorded during parturition (days 22-23). Compared with 1-g controls, hg dams displayed one-half as many lordosis contractions, the predominant labor contraction type in rats. The timing of birth, duration of delivery and number of neonates born was comparable in hg and 1-g control dams. Results are discussed in relation to earlier findings that, following sp

Hypergravity conditioning on ileal movements in rats.

INTRODUCTION: Stress is a common trigger for various physiological disturbances, including feeding disorders. One of the possible mechanisms for feeding disorders may be linked to changes in intestinal movement caused by stress. Therefore, here we investigated in vitro stress-induced changes in ileal movement. METHODS: Rats (female Wistar, SPF) were divided into Control (1 G) and hypergravity (hyperG) groups. HyperG stress was applied daily for 10 min by a centrifugal apparatus for 1 to 30 d. Under barbiturate anesthesia, a 1-cm long section of the ileum was isolated and fixed in a Magnus-typ

Effects of microgravity and hypergravity on platelet functions.

Many serious thrombotic and haemorrhagic diseases or fatalities have been documented in human being exposed to microgravity or hypergravity environments, such as crewmen in space, roller coaster riders, and aircrew subjected to high-G training. Some possible related organs have been examined to explore the mechanisms underlying these gravity change-related diseases. However, the role of platelets which are the primary players in both thrombosis and haemostasis is unknown. Here we show that platelet aggregation induced by ristocetin or collagen and platelet adhesion to von Willebrand factor (VW

Simulated Hypergravity Activates Hemostasis in Healthy Volunteers.

Background Hypergravity may promote human hemostasis thereby increasing thrombotic risk. Future touristic suborbital spaceflight will expose older individuals with chronic medical conditions, who are at much higher thromboembolic risk compared with professional astronauts, to hypergravity. Therefore, we tested the impact of hypergravity on hemostasis in healthy volunteers undergoing centrifugation. Methods and Results We studied 20 healthy seated men before and after 15 minutes under 3 Gz hypergravity on a long-arm centrifuge. We obtained blood samples for hemostasis testing before, immediatel

[Effect of hypergravity on the oxidative-antioxidative status of rats].

The adaptogenic and antioxidant properties of new Ukrainian plant drug "Poliphytolum" was investigated on the model of stress, which was caused of hypergravitation 2 and 5 N.m2/kg2 action. It was established that hypergravitation makes typical stressory disturbances in oxidative homeostasis. The normalization action of "Polyphytolum" on lipid peroxidation is more strong in liver, spleen and blood, and less more--in brain. It may be caused of low level the drug in the brain tissue.

Responses of the photosynthetic flagellate, Euglena gracilis, to hypergravity.

Motility and orientation has been studied in the unicellular photosynthetic flagellate, Euglena gracilis, using real time image analysis capable of tracking up to 200 cells simultaneously in the slow rotating centrifuge microscope (NIZEMI) which allows one to observe the cells' swimming behavior during centrifugation accelerations between 1 g and 5 g. At 1 g the cells show a weak negative gravitaxis, which increases significantly at higher accelerations up to about 3 g. Though most cells were capable of swimming even against an acceleration of 4.5 g, the degree of gravitaxis decreased and some

The effect of hypergravity in intestinal permeability of nanoformulations and molecules.

The oral administration of drugs remains a challenge due to rapid enzymatic degradation and minimal absorption in the gastrointestinal tract. Mechanical forces, namely hypergravity, can interfere with cellular integrity and drug absorption, and there is no study describing its influence in the intestinal permeability. In this work, it was studied the effect of hypergravity on intestinal Caco-2 cells and its influence in the intestinal permeability of different nanoformulations and molecules. It was shown that the cellular metabolic activity and integrity were maintained after exposure to diffe

Neurobehavioural effects of hypergravity conditions in the adult mouse.

To evaluate the behavioural response to a hypergravity condition in CD-1 mice, young adult subjects of both sexes were exposed to 2 g for a single 60 min rotational session. Motion sickness (MS) and ethological-type scoring of different activities were used to evaluate the behavioural response. Nerve growth factor (NGF) and brain derived neurotrophic factor (BDNF) levels were also assessed. Behavioural scores indicated a transient mild sickness associated with hypergravity, with reduction in spontaneous activity. In males kaolin consumption (a MS index) increased following rotation while femal

Impaired spatial learning after hypergravity exposure in rats.

Most astronauts experience spatial disorientation after exposure to weightlessness, indicating that constant gravity is utilized as a stable external reference during spatial cognition. We attempted to elucidate the role of constant gravity in spatial learning using a radial arm maze test on rats housed in a hypergravity environment (HG) produced by a centrifuge device. Male Wistar rats were kept in 2G linear acceleration for 2 weeks before the spatial learning task, which lasted for 10 days. The control rats were placed close to the centrifuge device but not exposed to hypergravity. Spatial l

Sperm Motility of Mice under Simulated Microgravity and Hypergravity.

For deep space exploration, reproductive health must be maintained to preserve the species. However, the mechanisms underlying the effect of changes in gravity on male germ cells remain poorly understood. The aim of this study was to determine the effect of simulated micro- and hypergravity on mouse sperm motility and the mechanisms of this change. For 1, 3 and 6 h, mouse sperm samples isolated from the caudal epididymis were subjected to simulated microgravity using a random position machine and 2g hypergravity using a centrifuge. The experimental samples were compared with static and dynamic

Neurobehavioural responses to hypergravity environment in the CD-1 mouse.

Behavioural responses of CD-1 mice exposed to 2 g hypergravity (HG; 60 or 120 min) were investigated during adolescence and at adulthood. To characterise motion sickness (MS), pica behaviour, a reliable MS index measured through kaolin consumption, and spontaneous activity were evaluated before, during and after HG exposure. Short- and/or long-lasting effects on emotional responses, exploratory behaviour and spatial learning performances were also investigated and brain levels of nerve growth factor (NGF) and brain derived neurotrophic factor (BDNF) assessed. An increased pica behaviour during

The effect of short-term microgravity and hypergravity on eyelid and brow position.

PURPOSE: Spaceflights induce periocular facial changes which could contribute to ocular conditions which affect astronauts. This study is to validate parabolic flight as a suitable model for studying gravitational facial changes. METHODS: Healthy participants (n = 13; 37 ± 10 years) underwent short-term exposure to microgravity and hypergravity during parabolic flight. Facial images were captured and differences in measurements from pupil center to upper and lower eyelid margins (MRD1 and MRD2), and to the inferior and superior eyebrow margin (PTBi and PTBs) under normogravity, microgravity,

Fos induction in the amygdala by vestibular information during hypergravity stimulation.

Altered gravity environments including both hypo- and hypergravity can elicit motion sickness. Vestibular information is known to be essential for motion sickness, but its other neural substrates are poorly understood. We previously showed that bilateral lesions of the amygdala suppressed hypergravity-induced motion sickness in rats, using pica behavior as an emetic index. We show in the present study that during hypergravity stimulation, vestibular information activated the central nucleus of the amygdala (CeA), as determined by the induction of Fos expression, in comparison between normal an

Effects of hypergravity environment of the ultrastructure of the golden hamster parathyroid gland.

The fine structure of the parathyroid glands of golden hamsters exposed to 2, 5 or 10 g environment for 5 h was studied. In the centrifuged hamsters, many secretory granules are located in a peripheral position just beneath the plasma membrane of chief cells, and the Golgi complexes and cisternae of the granular endoplasmic reticulum are significantly increased compared with those of control animals. There are no significant differences between the control and centrifuged animals with regard to secretory granules, large secretory granules, lysosomes, vacuolar bodies and lipid droplets. These f

Is sleep affected after microgravity and hypergravity exposure? A pilot study.

Sleep is known to be affected in space travel and in residents of the international space station. But little is known about the direct effects of gravity changes on sleep, if other factors, such as sleep conditions, are kept constant. Here, as a first exploration, we investigated sleep before and after exposure to short bouts of microgravity and hypergravity during parabolic flights. Sleep was measured through actigraphy and self-report questionnaires in 20 healthy men and women before and after parabolic flight. Higher sleep fragmentation and more awakenings were found in the night after the
