Exploring the brain through posterior hypothalamus surgery for aggressive behavior.

Neurological surgery offers an opportunity to study brain functions, through either resection or implanted neuromodulation devices. Pathological aggressive behavior in patients with intellectual disability is a frequent condition that is difficult to treat using either supportive care or pharmacological therapy. The bulk of the laboratory studies performed throughout the 19th century enabled the formulation of hypotheses on brain circuits involved in the generation of emotions. Aggressive behavior was also studied extensively. Lesional radiofrequency surgery of the posterior hypothalamus, whic

Role of posterior hypothalamus in hypobaric hypoxia induced pulmonary edema.

To investigate the role of posterior hypothalamus and central neurotransmitters in the pulmonary edema due to hypobaric hypoxia, rats were placed in a high altitude simulation chamber (barometric pressure-294.4 mmHg) for 24 h. Exposure to hypobaric hypoxia resulted in increases in mean arterial blood pressure, renal sympathetic nerve activity, right ventricular systolic pressure, lung wet to dry weight ratio and Evans blue dye leakage. There was a significant attenuation in these responses to hypobaric hypoxia (a) after lesioning posterior hypothalamus and (b) after chronic infusion of GABAA r

The Role of the Posterior Hypothalamus in the Modulation and Production of Rhythmic Theta Oscillations.

Theta rhythm recorded as an extracellular synchronous field potential is generated in a number of brain sites including the hippocampus. The physiological occurrence of hippocampal theta rhythm is associated with the activation of a number of structures forming the ascending brainstem-hippocampal synchronizing pathway. Experimental evidence indicates that the supramammillary nucleus and posterior hypothalamic nuclei, considered as the posterior hypothalamic area, comprise a critical node of this ascending pathway. The posterior hypothalamic area plays an important role in movement control, pla

Prolonged hyperglycemia & hyperinsulinemia increases BDNF mRNA expression in the posterior ventromedial hypothalamus and the dorsomedial hypothalamus of fed female rats.

Brain-derived neurotrophic factor (BDNF) plays a key role in neuronal development, synaptic plasticity, and the central control of energy homeostasis. Peripheral metabolic signals such as leptin and glucose regulate hypothalamic BDNF gene expression. However, the effects of long-term hyperglycemia and/or hyperinsulinemia on BDNF mRNA levels in the hypothalamus and other brain regions where BDNF regulates physiological functions have not been investigated. Therefore, using in situ hybridization we examined whether high glucose, high insulin, or both affected BDNF gene expression in vivo. Ovarie

Influence of the posterior hypothalamus on the visual cortex in various states of the reticular formation.

It has been demonstrated in chronic experiments on wakeful rabbits that the stimulation of the posterior hypothalamus by a single electrical stimulus leads to the formation in the visual cortex of a short-latency response which exerts a substantial influence on the formation of the reaction to light stimulation. Depending upon the intervals between the hypothalamic and light stimuli, an initial suppression of the response is observed (1-15 msec), a subsequent selective facilitation of its positive component in the presence of the simultaneous suppression of the negative (20-100 msec), and the

[The influence of the posterior hypothalamus on the visual cortex in different states of the reticular formation].

In alert rabbits, stimulation of posterior hypothalamus induced short-latency responses in the visual cortex and affected the formation of the EPs to light. Depending on the intervals between the stimuli, either initial extinction of the EPs (1-15 msec) or subsequent selective facilitation of its positive component with simultaneous depression of negative (20-100 msec) one, or complete recovery of the response (200-300 msec), were observed. Aminazine and benactizine made the effect of the posterior hypothalamic stimulation on visual EPs to light less obvious and changed its dynamics. Other fin

Lateral hypothalamus CRFR1 regulation of chronic binge drinking: divergence along anterior-posterior axis.

Binge alcohol drinking increases the risk of developing an alcohol use disorder (AUD) and comorbid psychopathology. The lateral hypothalamus (LH) is a brain structure that integrates cognitive and sensory information to tightly regulate motivated behavior, including binge drinking. Importantly, LH function is vulnerable to modulation by the pro-stress neuropeptide corticotropin-releasing factor (CRF), and acute antagonism of CRF receptor 1 (CRFR1) in the LH blunts binge drinking. However, the role of LH CRFR1 in chronic binge drinking is unknown. We used genetically targeted knockdown (KD) of

Orexin receptor 2 expression in the posterior hypothalamus rescues sleepiness in narcoleptic mice.

Narcolepsy is caused by a loss of orexin/hypocretin signaling, resulting in chronic sleepiness, fragmented non-rapid eye movement sleep, and cataplexy. To identify the neuronal circuits underlying narcolepsy, we produced a mouse model in which a loxP-flanked gene cassette disrupts production of the orexin receptor type 2 (OX2R; also known as HCRTR2), but normal OX2R expression can be restored by Cre recombinase. Mice lacking OX2R signaling had poor maintenance of wakefulness indicative of sleepiness and fragmented sleep and lacked any electrophysiological response to orexin-A in the wake-promo

Xenon inhalation increases norepinephrine release from the anterior and posterior hypothalamus in rats.

PURPOSE: To investigate the effect of xenon (Xe) and nitrous oxide (N(2)O) on norepinephrinergic neuronal activity in the rat medial preoptic area (mPOA) and posterior hypothalamus (PH) using microdialysis. METHODS: Sixty male Wistar rats were equally allocated to two groups: mPOA and PH. A microdialysis probe was implanted into the mPOA or the PH. In both groups, each animal was exposed to one of the following inhalations: 25% oxygen (control, n=6), 30% Xe (n=6), 60% Xe (n=6), 30% N(2)O (n=6) or 60% N(2)O (n=6). Administration of muscimol into the posterior hypothalamus reduces hyperthermia induced by hippocampal neostigmine injection.

The firing rate of the sympathetic nerves innervating interscapular brown adipose tissue (IBAT), IBAT and colonic temperatures (T(IBAT) and T(C)) and oxygen (O(2)) consumption were monitored in urethane-anesthetized male Sprague-Dawley rats. These variables were measured for 40 min before (baseline values) and 40 min after an injection of neostigmine (5 x 10(-7) mol in 1 microl of saline) into the hippocampus and a bilateral administration of a GABA(a)-agonist, muscimol (28 ng in 0.5 microl of saline, per side) into the posterior hypothalamus. The same variables were recorded in other rats, bu

Modulation of trigeminal neuropathic pain by optogenetic inhibition of posterior hypothalamus in CCI-ION rat.

Posterior hypothalamus (PH), an important part of the descending pain processing pathway, has been found to be activated in trigeminal autonomic cephalalgias. However, there are very few studies conducted and information regarding its implications in trigeminal neuropathic pain (TNP). Therefore, we aimed to ascertain whether optogenetic inhibition of PH could affect the outcomes of a chronic constriction injury in the infraorbital nerve (CCI-ION) rat model. Animals were divided into the TNP animal, sham, and naive-control groups. CCI-ION surgery was performed to mimic TNP symptoms, and the opt

Functional and morphological changes in the hypothalamus-pituitary posterior lobe system after hypophysectomy in the dog.

Acute diabetes insipidus-like symptoms have been reported as a complication after hypophysectomy in dogs. These symptoms are believed to be the consequence of deficiency of arginine vasopressin (AVP) secretion. The symptoms spontaneously resolve within 2 weeks, but the mechanism is unclear. In the present study, AVP secretion related to increases in Na+ concentration and serum osmotic pressure was measured, and immunohistochemical analysis in the paraventricular and supraoptic nuclei was performed after hypophysectomy in normal dog. In the hypertonic saline test, the plasma AVP concentration s

Benzodiazepine receptors in the medial-posterior hypothalamus mediate the reduction of hippocampal theta frequency by chlordiazepoxide.

Separate sub-populations of benzodiazepine receptors are involved in determining hippocampal theta frequency in freely moving rats. Systemic injection of the benzodiazepine chlordiazepoxide (CDP) reduced the frequency of theta elicited by stimulation of the brainstem reticular formation, and theta recorded during fixed interval (FI) bar pressing. Infusion of the benzodiazepine receptor antagonist flumazenil into the supramammillary nucleus (mSuM) or the posterior hypothalamic nucleus (PH) antagonised CDP in both cases. Flumazenil was similarly effective in mSuM and PH, suggesting that these nu

Selective enhancement of shock avoidance responding elicited by GABA blockade in the posterior hypothalamus of rats.

Recent studies have shown that blockade of gamma-aminobutyric acid (GABA) in the posterior hypothalamus in anesthetized rats elicits cardio-respiratory stimulation similar to that seen in emotional defense reactions and, in conscious rats, locomotor arousal suggesting a flight response. The present study was conducted in order to test the hypothesis that the behavioral effects elicited by GABA blockade in the posterior hypothalamus were the results of disinhibiting a mechanism whose activation selectively enhances reactivity to aversive stimuli. Male rats were trained on a Sidman shock avoidan
