Slow volume transients in amphibian skeletal muscle fibres studied in hypotonic solutions.

The influence of extracellular hypotonicity on the relationship between cell volume (V(c)) and resting membrane potential (E(m)) was investigated in Rana temporaria skeletal muscle. V(c) was measured by confocal microscope imaging of fibres through their transverse (xz) planes, and E(m) was determined using standard microelectrode techniques. Hypotonic solutions first elicited a rapid increase in fibre volume, DeltaV(R+) that fulfilled expectations of simple osmotic behaviour described in earlier reports. However, this was consistently followed by a slow increase in V(c) (DeltaV(S+)) to 10-15%

The hemodynamic and cardiovascular effects of near-drowning in hypotonic, isotonic, or hypertonic solutions.

It has been postulated that near-drowning in fresh water may cause hemodilution and hypervolemia due to the hypotonicity of the aspirated water. In contrast, near-drowning in seawater, because of its hypertonicity, may lead to hypovolemic shock. We evaluated the hemodynamic effects of the instillation of 20 mL/kg of solutions of various tonicities (sterile water, 0.225% sodium chloride, 0.45% sodium chloride, normal saline, 2% sodium chloride, and 3% sodium chloride) into the lungs of anesthetized dogs and compared the results with those for control animals who were made anoxic for five minute

Epithelium-on Corneal Collagen Cross-Linking with Hypotonic Riboflavin Solution in Progressive Keratoconus.

INTRODUCTION: Epithelium-off cross-linking (epi-off CXL) has long been established as the gold standard treatment for progressive keratoconus. Several protocols for epithelium-on (epi-on) CXL have been proposed to help reduce post-operative pain and facilitate visual recovery, but there is no epi-on treatment approach that is currently approved in the United States. The hydrophilic and macromolecular characteristics of conventional epi-off riboflavin formulations may create clinical challenges for absorption through an intact epithelium. This study investigates the clinical efficacy of a dextr

Differential effects of isotonic and hypotonic 4% albumin solution on intracranial pressure and renal perfusion and function.

OBJECTIVES: Albumin is used to resuscitate trauma patients but may increase intracranial pressure (ICP). Its effects on renal blood flow and function are unknown. Our aim was to examine the effects of hypertonic albumin on ICP and renal function, and if any effects are due to the hypotonicity of the solution containing albumin or to albumin itself. DESIGN, SETTING AND SUBJECTS: Cross-over, randomised controlled experimental study of six adult Merino ewes in the animal facility of a research institute. METHOD: Sheep were implanted with flow probes around the pulmonary and renal arteries and a

Hypotonic, Acidic Oxidizing Solution Containing Hypochlorous Acid (HClO) as a Potential Treatment of Hailey-Hailey Disease.

Hailey-Hailey disease (HHD) is a rare, chronic and recurrent blistering disorder, characterized by erosions occurring primarily in intertriginous regions and histologically by suprabasal acantholysis. Mutation of the Golgi Ca2+-ATPase ATP2C1 has been identified as having a causative role in Hailey-Hailey disease. HHD-derived keratinocytes have increased oxidative-stress that is associated with impaired proliferation and differentiation. Additionally, HHD is characterized by skin lesions that do not heal and by recurrent skin infections, indicating that HHD keratinocytes might not respond well

Impact of hypotonic solutions on the stromal swelling, lactate dehydrogenase and aldehyde dehydrogenase activity of the keratocytes of the bovine cornea.

The present studies were designed to explore the relationship between the swelling-related changes of the collagen-cell (keratocyte) matrix of the corneal stroma, and the integrity of the cells. From recent postmortem eyes of adult cattle, complete stroma preparations were dissected out and allowed to swell in solution (free swelling) or enclosed within a 12 kDa cut-off dialysis membrane with or without spacers. The swelling was at 4 degrees C with either water, a hypotonic phosphate-buffered saline (PBS, pH 7.0), a hypotonic mixed salt (MS) solution (pH 7.5), or an isotonic mixed salt solutio

Na(+)-K(+)-2Cl(-) cotransporter-mediated fluid secretion increases under hypotonic osmolarity in the mouse submandibular salivary gland.

Water-handling epithelia are sensitive to the osmotic environment. In this study, the effects of a hypo-osmotic challenge on carbachol (CCh)-induced fluid secretion was investigated using an ex vivo submandibular gland perfusion technique and intracellular pH and Ca(2+) measurements. The osmolality of the perfusion solution was altered to examine the response of the gland to a hypotonic challenge. The flow rate was increased by 34% with a 30% hypotonic solution (225 mosmol/kgH2O), although the Ca(2+) response was unchanged. The lowering of the external Cl(-) by 50% abolished this increase in t

Effect of corneal cross-linking on biomechanical properties of swollen rabbit corneas.

Corneal cross-linking (CXL) is an effective method to prevent the progression of keratoconus. CXL combined with hypotonic riboflavin solution is a modified treatment for thin corneas, which are deemed to be below the safe thickness threshold. In this study, rabbit corneas were subjected to different hydration levels using different osmolarity of riboflavin dextran solutions before CXL. Inflation testing was performed to evaluate the corneal biomechanical stiffening effect of hypotonic riboflavin solutions crosslinking. One-month post-CXL, the stromal demarcation line depth (DLD) and the biomec

Electrolyte vs. glucose-electrolyte isotonic solutions for oral rehydration therapy in horses.

An isotonic electrolyte solution with a composition similar to equine sweat was compared to an isotonic glucose-glycine-electrolyte solution for oral rehydration therapy in exercising horses. Ten horses were dehydrated by using frusemide and allocated randomly to receive 4 different oral solutions: isotonic sweat-like electrolyte solution, half-strength hypotonic electrolyte solution, isotonic glucose-glycine-electrolyte solution, and plain water. Solutions were given by nasogastric tube using the same volume as the bodyweight lost by each horse. Blood samples were collected before and through

Effect of isotonic versus hypotonic maintenance fluid therapy on urine output, fluid balance, and electrolyte homeostasis: a crossover study in fasting adult volunteers.

BACKGROUND.: Daily and globally, millions of adult hospitalized patients are exposed to maintenance i.v. fluid solutions supported by limited scientific evidence. In particular, it remains unclear whether fluid tonicity contributes to the recently established detrimental effects of fluid, sodium, and chloride overload. METHODS.: This crossover study consisted of two 48 h study periods, during which 12 fasting healthy adults were treated with a frequently prescribed solution (NaCl 0.9% in glucose 5% supplemented by 40 mmol litre -1 of potassium chloride) and a premixed hypotonic fluid (NaCl 0.

Tear diluents in the treatment of keratoconjunctivitis sicca.

To determine the optimum solution concentration for lowering elevated tear film osmolarity in keratoconjunctivitis sicca (KCS), tear osmolarity was measured in four KCS patients before and after instillation of either an isotonic saline solution or one of four hypotonic saline solutions (range, 75-225 mOsm/L). Average tear osmolarity one minute after instillation was significantly lower with the hypotonic solutions than with the isotonic saline (mean +/- SEM, 290 +/- 3 mOsm/L vs. 317 +/- 1 mOsm/L, P less than 0.0005). Solutions 150 mOsm/L or less were most effective in lowering osmolarity; the
