Anticancer drug idarubicin - derivative of doxorubicin - is commonly used in treatment of numerous cancer types. However, in contrast to doxorubicin, its biophysical properties are not well established yet. Additionally, potential direct interactions of idarubicin with other biologically active aromatic compounds, such as pentoxifylline - representative of methylxanthines - were not studied at all. Potential formation of such hetero-aggregates may result in sequestration of the anticancer drug and, in consequence, reduction of its biological activity. This work provide description of the idaru

An early phase II study of Idarubicin was performed in patients with acute leukemia. The dosages administered were 10 mg/m2, 12 mg/m2, or 15 mg/m2 by iv bolus, once daily for 3 consecutive days. The treatment was given to 47 patients who were in relapse or whose diseases had been refractory to remission induction therapy. Of the 47 patients, 35 were evaluable for response. The patients who showed a response (complete or partial remission) were 9 of 14 patients (64.3%) in the 10 mg/m2 group, 1 of 12 patients (8.3%) in the 12 mg/m2 group, and 3 of 9 patients (33.3%) in the 15 mg/m2 group, respec

BACKGROUND: The myelodysplastic syndromes (MDS) are a group of haemopoietic stem cell disorders of unknown cause, occurring predominantly in elderly patients. They have a high mortality due to deaths from complications of cytopenias or transformation to leukaemia. No effective treatment is known. Preliminary studies have identified idarubicin (IDA), an anthracycline drug that can be administered orally, as an agent with possible activity in MDS.
METHODS: In a phase I-II study we administered oral IDA to 14 predominantly elderly patients (age range 40-79, median 74) with advanced forms of MDS,

A phase I trial of a new anthracycline derivative, idarubicin, was conducted in 16 patients with various advanced solid tumors. Doses administered were 20, 30 and 40 mg/m2 with single oral doses. Dose-limiting factors were both leukopenia and thrombocytopenia reaching a nadir about 2 weeks after the start of treatment and requiring 7 to 10 days for recovery. Other toxicities were anorexia, nausea and vomiting, but alopecia did not occur. The maximum tolerated dose was judged to be 40 mg/m2 and the recommended dose for phase II trials was determined to be 30-35 mg/m2.

BACKGROUND: Blastic crisis (BC) is the terminal event in the natural history of most chronic myelogenous leukemia (CML) patients. Depletion of the normal stem cell compartment, as well as the proliferative advantage and frequent pharmacoresistance of the blastic clone, contribute to the poor prognosis of CML patients in this phase. Recent clinical trials have shown that idarubicin (IDR) in combination with cytosine arabinoside (ARA-C) is more active than daunorubicin at comparable doses in acute myelogenous leukemia (AML). Furthermore, IDR alone also exhibits antitumoral activity in the BC of

BACKGROUND AND OBJECTIVE: Idarubicin is an effective drug in acute leukemia but its use in non-Hodgkin lymphomas (NHLs) is not yet well established. We evaluated its efficacy in patients with diffuse large cell lymphoma (DLCL) by means of a randomized trial comparing two 12-week regimens (VACOP-B and VICOP-B) which differed only in the anthracycline drug used (doxorubicin vs idarubicin).
METHODS: From January 1992 to December 1994, 104 patients aged less than 65 years with de novo advanced stage DLCL were enrolled. Fifty-two patients were treated with VACOP-B (doxorubicin 50 mg/sqm) and 52 wi

We investigated if the antileukemic drug idarubicin induces autophagy, a process of programmed cellular self-digestion, in leukemic cell lines and primary leukemic cells. Transmission electron microscopy and acridine orange staining demonstrated the presence of autophagic vesicles and intracellular acidification, respectively, in idarubicin-treated REH leukemic cell line. Idarubicin increased punctuation/aggregation of microtubule-associated light chain 3B (LC3B), enhanced the conversion of LC3B-I to autophagosome-associated LC3B-II in the presence of proteolysis inhibitors, and promoted the d

Acute myeloblastic leukemia, subtype M1, was diagnosed in a 39-year-old G2P1 Japanese woman at 21 weeks' gestation. Remission-induction polychemotherapy, including daunorubicin, performed for one cycle, did not lead to remission. Second-line chemotherapy, including idarubicin, performed for one cycle, was administrated during the early third trimester of pregnancy. Septic shock occurred due to severe myelosuppression. An emergent cesarean section was performed in response to a nonreassuring pattern of the fetal heart rate and a rapid decrease in fetal amniotic fluid. The neonate delivered at 3

Radiation recall reactions are uncommon delayed tissue reactions seen in previously irradiated sites following treatment with cytotoxic agents. We evaluated a 64-year-old who developed two episodes of acute vulvitis and vaginal necrosis after receiving idarubicin therapy for acute myelogenous leukemia. Three years earlier she had undergone successful radiotherapeutic treatment of a stage I squamous cell carcinoma of the vagina. Her symptoms and examination findings resolved with local therapy and discontinuation of idarubicin. Recall reactions have been associated with the antitumor antibiotic

We investigated anticancer therapy with a novel combination of D-penicillamine (D-pen) and Idarubicin (Ida) in a synthetic dual drug conjugate (DDC). D-pen and Ida were covalently linked to poly(α)-L-glutamic acid (PGA) via reducible disulfide and acid-sensitive hydrazone bonds, respectively. The DDCs showed cell uptake and sustained release of the bound drugs in conditions mimicking the intracellular release media (10mM glutathione and pH 5.2). The in-vitro cytotoxicity of DDCs was comparable to unconjugated Ida in several sensitive and resistant cancer cell lines and correlated with the rate

PURPOSE: The aim of this study was to evaluate in an upfront phase II study the response to idarubicin in children with extraocular retinoblastoma.
PATIENTS AND METHODS: The starting dose of idarubicin was 15 mg/m(2)/d (days 1 and 2) weeks 0 and 3. After an interim evaluation, the dose was reduced to 10 mg/m(2)/d (days 1 and 2) weeks 0 and 3 because of hematopoietic toxicity. Response was evaluated at week 6.
RESULTS: At the Hospital JP Garrahan (Buenos Aires, Argentina), 10 patients (five bilateral) were entered onto the study from 1995 to 1998. A total of 19 cycles were administered. Extra

Idarubicin, a new analogue of daunorubicin, was administered to 27 patients with advanced breast cancer in a phase II trial. The drug was given orally at a dose of 30-35 mg/m2 every 3 weeks. Twenty-two patients were evaluable for response. All evaluable patients were previously treated with one or more chemotherapeutic regimens, including an anthracycline in more than 50% of the cases. Partial remissions were obtained in 5 patients, for a response rate of 23%. The median duration of response was 191 days. Mild nausea and vomiting were common. Diarrhea, which occurred in less than 50% of the pa

Chemotherapy-induced mucositis, characterized by diarrhoea and villous atrophy, is a severe side effect contributing to reduced quality of life and premature death in cancer patients treated with cytostatics. Despite its high incidence, there is no effective supportive therapy available. The main objective of this study was to determine if the anti-inflammatory drugs anakinra and/or dexamethasone-which have different mechanisms-of-action-might be used to effectively treat idarubicin-induced mucositis in rats. Mucositis was induced through a single injection with 2 mg/kg idarubicin (with saline

Idarubicin (Ida), an analogue of daunomycin, was linked to a mouse monoclonal antibody against the B cell differentiation antigen CD19. Determination of the activity of both the antibody and drug moieties was carried out in vitro using a pre-B cell human acute lymphoblastic leukaemia cell line (NALM-6). A 23% loss in antibody binding and a 20-fold loss in drug activity were observed upon conjugation. Selective cytotoxicity for CD19+ cells, however, was obtained. Measurement of the cytotoxicity, antibody activity and release of the breakdown product, 14-OH-Ida, showed that the conjugates remain

Idarubicin is a new derivative of Daunorubicin which was found to be more potent and more active than Daunorubicin and Doxorubicin in several experimental leukemias. Its antileukemic activity in preclinical models prompted the introduction of Idarubicin into clinical studies. As a single agent, Idarubicin produced complete remission in 20% and 30% of patients with heavily pretreated pediatric and adult acute myeloid leukemia (AML) and acute lymphoblastic leukemia (ALL) respectively. Idarubicin combined with Cytarabine and/or other antileukemic agents produced complete remissions in 46% of pati

6 patients with refractory anaemia with excess blasts (RAEB) were treated with a new oral anthracycline, idarubicin. 3 patients achieved complete remission, and the remaining 3 achieved partial remission. These responses were maintained for 8-60 wk, the longer durations being in the patients who achieved complete remission. Treatment was given on an outpatient basis, and all but 1 patient remained at home for most of the disease course. Unwanted effects were mild. It would thus appear that oral idarubicin is an effective treatment for RAEB, in addition to allowing outpatient management. Larger

Thirty one patients with solid tumors were entered into a phase I trial with idarubicin, a new anthracycline antibiotic with oral antitumor activity in animals. The drug was scheduled to be given for 4 consecutive weeks at doses ranging from 10 to 20 mg/m2. Leukopenia was the dose-limiting toxicity. Thrombocytopenia was occasionally seen. Since several patients could not receive the third and fourth administrations of the drug at 17.5 and 20 mg/m2, higher doses were administered only for 2 consecutive weeks. With this schedule, the maximum tolerated dose was 25 mg/m2 and leukopenia was again t

BACKGROUND: Anthracyclines are key components of induction therapy for acute myelogenous leukemia with well-understood cardiotoxicity. Quizartinib inhibits FLT3 and has been shown to potentiate apoptosis and promote maladaptive remodeling after myocardial infarction in mice; however, the impact of FLT3 inhibition on a heart already exposed to anthracyclines is poorly understood.
CASE SUMMARY: A 64-year-old man with acute myelogenous leukemia developed fever, hypotension, troponin I elevation, and diffuse ST-segment elevation after treatment with cytarabine and idarubicin followed by quizartin
