Soluble immune checkpoint proteins as predictive biomarkers for lymph node metastases in penile cancer.

BACKGROUND: Penile cancer (PeCa) is a rare but aggressive disease where lymph node metastases (LNM) represent the most significant prognostic factor. Accurate identification of LNM remains a clinical priority, but traditional imaging and clinical parameters often fail to detect occult LNM. Soluble immune checkpoint proteins (sICs) have recently emerged as potential non-invasive biomarkers in various malignancies, although unexplored in PeCa. The primary aim of this study was to explore the value of a panel of 14 sICs for predicting LNM in PeCa. The secondary aim was to compare plasma sIC level

Targeting Immune Checkpoint Proteins in Cancer Therapy and the Potential of RNAi-Based Immunotherapy.

Cancer immunotherapy via targeting immune checkpoint proteins (ICPs) has transformed the treatment of multiple malignancies, offering improved clinical outcomes over conventional therapies. Immune checkpoint inhibitors (ICIs), including FDA-approved monoclonal antibodies against CTLA-4, PD-1, and PD-L1, as well as emerging agents targeting novel ICPs, have demonstrated strong therapeutic efficacy by restoring antitumor immune responses. In parallel, RNA interference (RNAi)-based approaches involving microRNAs (miRNAs) and small interfering RNAs (siRNAs) have emerged as promising alternative st

Mass Spectrometry-Based Multi-Protein Panel Assay for Detecting Immune Checkpoint Proteins in Immune Cells of Oral Cancer Patients.

Cancer immunotherapy represents a transformative approach to cancer treatment, paving the way for personalized and precision medicine strategies. Despite the significant advances in immunotherapy, the treatment response rate remains a major clinical challenge. The stratification of patients into responders and nonresponders is paramount for immunotherapy treatment response, which reduces the likelihood of treatment challenges, expenses, and potentially severe adverse effects in cases where patients are not likely to respond. Therefore, in the present study, we optimized a mass spectrometry-bas

Immune-checkpoint proteins, cytokines, and microbiome impact on patients with cervical insufficiency and preterm birth.

BACKGROUND: Microenvironmental factors, including microbe-induced inflammation and immune-checkpoint proteins that modulate immune cells have been associated with both cervical insufficiency and preterm delivery. These factors are incompletely understood. This study aimed to explore and compare interactions among microbiome and inflammatory factors, such as cytokines and immune-checkpoint proteins, in patients with cervical insufficiency and preterm birth. In particular, factors related to predicting preterm birth were identified and the performance of the combination of these factors was eval

Immune checkpoint protein VSIG4 as a biomarker of aging in murine adipose tissue.

Adipose tissue is recognized as a major source of systemic inflammation with age, driving age-related tissue dysfunction and pathogenesis. Macrophages (Mφ) are central to these changes yet adipose tissue Mφ (ATMs) from aged mice remain poorly characterized. To identify biomarkers underlying changes in aged adipose tissue, we performed an unbiased RNA-seq analysis of ATMs from young (8-week-old) and healthy aged (80-week-old) mice. One of the genes identified, V-set immunoglobulin-domain-containing 4 (VSIG4/CRIg), encodes a Mφ-associated complement receptor and B7 family-related immune checkpoi

Immune checkpoint molecules signal regulatory protein alpha in the development of hepatocellular carcinoma.

Hepatocellular carcinoma (HCC) is a primary malignant tumor of the liver and one of the most common malignant tumors, as well as the third leading cause of cancer-related death. In recent years, immune checkpoint inhibitors have emerged as a key strategy in cancer treatment. However, anti-programmed cell death 1/programmed death ligand 1 therapies, one of the main immunotherapeutic approaches, only elicit a response in only approximately 20% of advanced HCC. This suggests that there may be other immune checkpoints playing important roles in HCC immunotherapy. Recent studies have highlighted Si

Cancer-Intrinsic Protein Neddylation Modulates the Intratumoral Immune Landscape to Constrain Immune Checkpoint Blockade Therapy.

The cancer-intrinsic network controlling the response to immune checkpoint blockade therapy remains elusive. Using a published single-cell RNA sequencing (scRNA-seq) dataset from patients with breast cancer, we found that pretherapy mRNA expression of NEDD8, which encodes the regulatory protein essential for neddylation, in cancer cells was negatively associated with T-cell expansion upon pembrolizumab treatment. Genetic ablation of protein neddylation in murine tumors induced a curative response to PD-1 or PD-L1 blockade in mice. Combined analysis of neddylation-deficient murine tumors using

OBJECTIVES: Hepatocellular carcinoma (HCC) is a highly aggressive malignancy with limited treatment options, particularly in advanced stages. Immune checkpoint inhibitors (ICIs) targeting PD-1, PD-L1, and CTLA-4 have shown promise in cancer immunotherapy, but response rates in HCC remain variable. MATERIALS AND METHODS: Hep3B and HepG2 HCC cells were cultured and genetically manipulated to overexpress or deplete LKB1. Western blotting, real-time PCR, and immunofluorescence were used to assess PD-L1 expression at the protein and mRNA levels. The role of Skp2 in PD-L1 regulation was evaluated t

The Interplay of the LncRNA AFAP1-AS1 and the Soluble Immune Checkpoint Proteins sPD-1/sPD-L1 in Breast Cancer: Tumor-Immune Crosstalk.

Breast cancer (BC) remains a major health issue worldwide. Immuno-oncology has recently garnered much attention with unprecedented bench-to-bedside revolutions in cancer diagnosis and treatment. The PD-1/PD-L1 pathway is an adaptive mechanism in cancer immune escape that can be regulated epigenetically via long non-coding RNAs (lncRNAs). In this study, we sought to investigate the potential clinical utility of the circulating lncRNA AFAP1-AS1 and soluble immune checkpoint proteins sPD-1 and sPD-L1 as liquid biopsy biomarkers in BC patients. Serum expression levels of AFAP1-AS1 was quantified v

Immune checkpoint profiling of B7-H proteins predicts survival and treatment response in metastatic clear cell renal cell carcinoma.

The B7-H family of immune checkpoint proteins plays a central role in tumor immune regulation, yet their prognostic and predictive significance in metastatic clear cell renal cell carcinoma (ccRCC) remains incompletely defined. We analyzed a retrospective cohort of primary tumor specimens from 145 patients with synchronous or metachronous metastatic ccRCC to evaluate the expression of B7-H3, B7-H4, B7-H5, and B7-H7 and their association with tumor aggressiveness, treatment response, and long-term survival. Immunohistochemistry revealed marked intratumoral heterogeneity and distinct compartment

Combination immune checkpoint and targeted protein kinase inhibitors for the treatment of renal cell carcinomas.

Kidney cancers comprise about 3% of all new malignancies in the United States. Renal cell carcinomas (RCCs) are the most common type of renal malignancy making up about 85% of kidney cancer cases. Signs and symptoms of renal cell carcinomas can result from local tumor growth, paraneoplastic syndromes, or distant metastases. The classic triad of presentation with flank pain, hematuria, and a palpable abdominal mass occurs in fewer than 10% of patients. Most diagnoses result from incidental imaging findings (ultrasonography or abdominal CT imaging) performed for another reason. Localized disease

Monitoring Circulating Immune Checkpoint Proteins as Predictors of Non-AIDS Morbid Events in People With HIV Initiating Antiretroviral Therapy.

BACKGROUND: Although cell surface immune checkpoint proteins (ICPs) such as PD-1 expressed on T cells are associated with T-cell exhaustion, HIV disease progression, and AIDS events, they have shown limited utility in predicting non-AIDS morbidity. Given that ICPs also exist in soluble forms and are elevated in ART-treated HIV infection, we tested the hypothesis that soluble ICPs may be predictive of non-AIDS events in adults initiating ART. METHODS: Utilizing a nested case-control study from the AIDS Clinical Trials Group ALLRT cohort, we measured plasma levels of 15 soluble inhibitory and a

Protein scaffold engineering for immune checkpoint targeting, tumor microenvironment modulation, and Cancer immunotherapy.

Protein scaffolds are being developed as novel non-antibody-binding platforms to address significant challenges in traditional cancer immunotherapy, including high production costs, limited tumor penetration, immune-related toxicity, and therapeutic resistance. Scaffolds, which consist of Affibodies, DARPins, Anticalins, Monobodies (Adnectin), Knottins, and Avimers, are structurally stable, small in size, and serve as extensively modified proteins, offering higher solubility, rapid clearance from unwanted tissues, and effective recombinant production compared with monoclonal antibodies (mAbs).

Evaluation of affinity matured Affibody molecules for imaging of the immune checkpoint protein B7-H3.

B7-H3 (CD276), an immune checkpoint protein, is a promising molecular target for immune therapy of malignant tumours. Sufficient B7-H3 expression level is a precondition for successful therapy. Radionuclide molecular imaging is a powerful technique for visualization of expression levels of molecular targets in vivo. Use of small radiolabelled targeting proteins would enable high-contrast radionuclide imaging of molecular targets if adequate binding affinity and specificity of an imaging probe could be provided. Affibody molecules, small engineered affinity proteins based on a non-immunoglobuli
