[Immunogenic cell death in hematological malignancies].

Anticancer therapies using cytotoxic drugs, which had been considered to be immunosuppressive, could induce immunogenic cell death (ICD) in cancer cells. Anticancer drugs, tumor-lytic viruses, and radiation therapies can establish long-term immune memory via emmision of damage-associated molecular pattern from cancer cells, activating the acquired immunity. This phenomenon could prolong the survival of patients with cancer. ICD was originally reported in solid tumors; however, recent studies have shown that it also occurs in hematological malignancy. Several studies have been trying to combine

Patritumab deruxtecan induces immunogenic cell death.

Antibody-drug conjugates (ADCs) enable targeted delivery of cytotoxic payload to cancer cells. Here, we characterized the mode of action of the ADC patritumab deruxtecan, which is a monoclonal antibody specific for Erb-B2 Receptor Tyrosine Kinase 3 (ERBB3, best known as HER3) coupled to the topoisomerase-I inhibitor DXd. Patritumab deruxtecan decreased viability and induced the relocation of calreticulin fused to green fluorescent protein (CALR-GFP) to the periphery of human osteosarcoma U2OS cells engineered to express HER3 but not in their parental counterparts only expressing the CALR-GFP b

Cancer cell death: Cell-autonomous and immunogenic dimensions.

Regulated cell death (RCD) shapes neoplastic transformation, tumor progression, and response to treatment. While apoptosis was long viewed as the only RCD variant, additional modalities, including necroptosis, pyroptosis, and ferroptosis, have been characterized. These interconnected pathways operate in a context-dependent manner to influence the dynamic interplay between malignant and non-malignant cells that governs disease progression or regression, both naturally and during therapy. Major advances stemmed from recognizing immunogenic cell death (ICD) as an RCD type defined by the emission

Lysosomal lipid peroxidation mediates immunogenic cell death.

Cancer cells rely on lysosome-dependent degradation to recycle nutrients that serve their energetic and biosynthetic needs. Despite great interest in repurposing the antimalarial hydroxychloroquine as a lysosomal inhibitor in clinical oncology trials, the mechanisms by which hydroxychloroquine and other lysosomal inhibitors induce tumor-cell cytotoxicity remain unclear. In this issue of the JCI, Bhardwaj et al. demonstrate that DC661, a dimeric form of chloroquine that inhibits palmitoyl-protein thioesterase 1 (PPT1), promoted lysosomal lipid peroxidation, resulting in lysosomal membrane perme

Natural compound inducers of immunogenic cell death.

Accumulating evidence shows that the anti-cancer potential of the immune response that can be activated by modulation of the immunogenicity of dying cancer cells. This regulated cell death process is called immunogenic cell death (ICD) and constitutes a new innovating anti-cancer strategy with immune-modulatory potential thanks to the release of damage-associated molecular patterns (DAMPs). Some conventional clinically-used chemotherapeutic drugs, as well as preclinically-investigated compounds of natural origins such as anthracyclines, microtubule-destabilizing agents, cardiac glycosides or h

Autophagy Modulates Immunogenic Cell Death in Cancer.

Immunogenic cell death (ICD) is a subtype of regulated cell death characterized by the spatiotemporally coordinated emission of damage-associated molecular patterns (DAMPs), such as calreticulin (CALR), ATP, and high-mobility group box-1 (HMGB1), which collectively prime tumor-specific T-cell responses. Autophagy, a lysosome-dependent catabolic process, is increasingly recognized as a key modifier of antitumor immunity and the tumor microenvironment (TME). In preclinical models, autophagy can not only promote ICD by sustaining endoplasmic reticulum (ER) stress, eukaryotic translation initiatio

Datopotamab deruxtecan induces hallmarks of immunogenic cell death.

Antibody-drug conjugates (ADCs) offer a strategy for targeted delivery of cytotoxic agents to cancer cells. In this study, we investigated the mechanism of action of datopotamab deruxtecan, an ADC composed of a monoclonal antibody targeting tumor-associated calcium signal transducer 2 (TACSTD2, also known as trophoblast cell-surface antigen-2 (TROP2)) conjugated to the topoisomerase I inhibitor DXd. Datopotamab deruxtecan reduced the viability of human osteosarcoma U2OS cells engineered to express TROP2, but had no effect on their parental counterparts, which only expressed the CALR-GFP biosen

Targeting Lysosomal Thiols for Immunogenic Cancer Cell Death.

The number and stability of lysosomes (LYs) are different in cancer and healthy cells that makes them a possible target for cancer specific therapy. However, no LY-targeting drug is clinically approved yet. We describe in this paper a new therapeutic approach based on alkylation of lysosomal thiols in cancer cells by reversible thiol binder 11. The treatment with 11 increases the level of lysosomal reactive oxygen species leading to their destabilization, disruption and immunogenic cancer cell death. These effects are not observed in healthy cells. In murine sarcoma Nemeth-Kellner (NK)/Ly-RB m

Assessment of eIF2α phosphorylation during immunogenic cell death.

Immunogenic cell death (ICD) is a modality of cellular demise that when it is induced by certain anticancer treatments can ignite an adaptive anticancer immune response. ICD is characterized by the emission of a specific set of danger-associated molecular patterns (DAMPs) including calreticulin exposure at the plasma membrane, ATP liberation, HMGB1 exodus and type-I IFN release. The apical signaling triggering the appearance of these hallmarks involves the phosphorylation on serine 51 of the α-subunit of eukaryotic initiation factor 2 (EIF2), a key protein in the orchestration of endoplasmic r

Extracorporeal photochemotherapy induces bona fide immunogenic cell death.

Extracorporeal photochemotherapy (ECP) is employed for the management of cutaneous T cell lymphoma (CTCL). ECP involves the extracorporeal exposure of white blood cells (WBCs) to a photosensitizer, 8-methoxypsoralen (8-MOP), in the context of ultraviolet A (UVA) radiation, followed by WBC reinfusion. Historically, the therapeutic activity of ECP has been attributed to selective cytotoxicity on circulating CTCL cells. However, only a fraction of WBCs is exposed to ECP, and 8-MOP is inactive in the absence of UVA light, implying that other mechanisms underlie the anticancer effects of ECP. Recen

Radiation-Induced Immunogenic Cell Death for Cancer Radioimmunotherapy.

In cancer treatment, traditional radiotherapy (RT) is routinely conducted in local tumors and limited by radioresistance, while recently emerged immunotherapy is confronted with low response rate, high cost, and cytokine release syndrome. Logically, the combination of the two therapeutic modalities into radioimmunotherapy is promising to complement with each other for systemic elimination of cancer cells with high specificity, efficiency, and safety. Especially, RT-induced immunogenic cell death (ICD) plays an imperative role in radioimmunotherapy to evoke systemic immune response against canc

AMPK activation induces immunogenic cell death in AML.

Survival of patients with acute myeloid leukemia (AML) can be improved by allogeneic hematopoietic stem cell transplantation (allo-HSCT) because of the antileukemic activity of T and natural killer cells from the donor. However, the use of allo-HSCT is limited by donor availability, recipient age, and potential severe side effects. Similarly, the efficacy of immunotherapies directing autologous T cells against tumor cells, including T-cell recruiting antibodies, chimeric antigen receptor T-cell therapy, and immune checkpoint inhibitors are limited in AML because of multiple mechanisms of leuke

Immunogenic cell death and tertiary lymphoid structures: bridging tumour cell death and immune organization.

Recent studies suggest that the formation of tertiary lymphoid structures (TLSs) could be associated with better survival from certain cancers. In this Perspective, we propose that immunogenic cell death (ICD) can act as an upstream organizer of tumour-associated TLSs by coordinating vascular, stromal and immune reprogramming in the tumour microenvironment. ICD leads to the release of tumour antigens and damage-associated molecular patterns that activate innate pathways, such as cGAS-STING signalling, in dendritic cells, myeloid cells, endothelial cells and stromal fibroblasts. These activated

DAMPs in the immunogenicity of cell death.

Damage-associated molecular patterns (DAMPs) are endogenous molecules-such as proteins, lipids, and nucleic acids-released or exposed during cellular injury or stress, which shape immune responses by engaging danger sensors on the cell surface or within the cell interior. Recent advances have elucidated molecular links between distinct cell death pathways-apoptosis, necroptosis, pyroptosis, and ferroptosis-and DAMP release, emphasizing how biochemical features (e.g., posttranslational modifications) and temporal dynamics influence immunogenic versus tolerogenic outcomes. Here, we summarize the

Curcumin-mediated modulation of immunogenic cancer cell death pathways.

Immunogenic cell death (ICD) is a regulated form of cell death characterized by the release of danger-associated molecular patterns (DAMPs) such as calreticulin, ATP, and HMGB1, which activate dendritic cells and cytotoxic T lymphocytes to promote antitumor immunity. This review focuses on the molecular mechanisms underlying DAMP release and associated signaling pathways, with an emphasis on how curcumin modulate ICD. Curcumin serves as a representative compound, exhibiting the ability to trigger ICD through endoplasmic reticulum stress, oxidative stress modulation, calcium homeostasis disrupt

FDA-approved phensuximide inhibits RIPK1-dependent immunogenic cell death.

Receptor-interacting serine/threonine kinase 1 (RIPK1) is a pivotal protein controlling cell death and inflammation. RIPK1 is an attractive therapeutic target, given that the inhibition of RIPK1 kinase activity has been shown to be effective in animal models of human diseases such as autoimmune and neurodegenerative diseases. Here, we screened a collection of drugs with structural similarity to necrostatin-1 (Nec-1), an inhibitor of RIPK1, to assess their abilities to regulate RIPK1-mediated immunogenic cell death. Through this small-scale screening of drugs from ongoing clinical trials and FD

Targeting immunogenic cell stress and death for cancer therapy.

Immunogenic cell death (ICD), which results from insufficient cellular adaptation to specific stressors, occupies a central position in the development of novel anticancer treatments. Several therapeutic strategies to elicit ICD - either as standalone approaches or as means to convert immunologically cold tumours that are insensitive to immunotherapy into hot and immunotherapy-sensitive lesions - are being actively pursued. However, the development of ICD-inducing treatments is hindered by various obstacles. Some of these relate to the intrinsic complexity of cancer cell biology, whereas other

Immunogenic cell death in cancer: concept and therapeutic implications.

Mammalian cells responding to specific perturbations of homeostasis can undergo a regulated variant of cell death that elicits adaptive immune responses. As immunogenic cell death (ICD) can only occur in a precise cellular and organismal context, it should be conceptually differentiated from instances of immunostimulation or inflammatory responses that do not mechanistically depend on cellular demise. Here, we critically discuss key conceptual and mechanistic aspects of ICD and its implications for cancer (immuno)therapy.
