Immunological characteristics of a recombinant alphaherpesvirus with an envelope-embedded Cap protein of circovirus.

INTRODUCTION: Variant pseudorabies virus (PRV) is a newly emerged zoonotic pathogen that can cause human blindness. PRV can take advantage of its large genome and multiple non-essential genes to construct recombinant attenuated vaccines carrying foreign genes. However, a major problem is that the foreign genes in recombinant PRV are only integrated into the genome for independent expression, rather than assembled on the surface of virion. METHODS: We reported a recombinant PRV with deleted gE/TK genes and an inserted porcine circovirus virus 2 (PCV2) Cap gene into the extracellular domain of

Immunological characteristics of the recombinant pseudorabies virus with chimeric PCV Cap protein in pigs.

Porcine circovirus type 2 (PCV2) is one of the main pathogens causing porcine circovirus-associated diseases (PCVAD). We recently reported the immunogenicity of the recombinant PRV with an envelope-embedded Cap protein of PCV2 (PRV-Cap) in mice. Here, we further evaluated the immunoprotective efficacy of PRV-Cap virus in pigs. Following vaccination, the PRV-Cap stimulated the production of neutralizing antibodies against PRV and PCV2, along with protected piglets from the challenge of the lethal PRV, the virulent PCV2b and PCV2d. Peripheral blood mononuclear cells analysis revealed that PRV-Ca

Preparation and immunological effects of a bivalent subunit candidate vaccine based on the PCV-Cap-2-3 gene engineered from baculovirus.

INTRODUCTION: Porcine circoviruses (PCVs), belonging to the genus Circovirus in the family Circoviridae, are prevalent worldwide and pose a serious threat to the pig industry. Although vaccines against PCV2 are available, PCV3 vaccine development, prevention strategies, and surveillance for co-infections remain limited, leading to delayed outbreak responses. Therefore, the aim of this study was to develop a bivalent subunit vaccine targeting the prevalent strains of PCV2 and PCV3. METHODS: Recombinant baculoviruses, Baculovirus-PCV2-Cap and Baculovirus-PCV3-Cap, were constructed to express th

Dietary capsaicin supplementation attenuates jejunal injury of piglets challenged with lipopolysaccharide.

BACKGROUND: Intensive breeding system makes piglets expose to infectious agents causing immunological stress. Lipopolysaccharide (LPS), a material found in Gram-negative bacteria’s cell walls, can induce immunological stress. Some studies demonstrated that immunological stress was harmful to intestinal functions of piglets. Capsaicin (CAP) has been reported to improve intestinal functions both in vivo and in vitro experiments. Thus, this study’s objective was to look into whether CAP supplementation could be an effective mean to protect jejunum of piglets against immunological stress. RESULTS:

Microtubule assembly and conanavalin A capping in lymphocytes: reappraisal using normal and abnormal human peripheral blood cells.

We have analyzed the assembly of microtubules and the distribution of concanavalin A(Con A)-receptor complexes in the same populations of human peripheral blood T and B lymphocytes. We hoped to resolve the prolonged controversy over the relationship of microtubules to Con A cap formation in lymphocytes and to explain the abnormally high spontaneous and colchicine-induced Con A capping that was observed recently in lymphocytes from a patient with an inherited form of severe combined immunodeficiency disease (SCID) characterized by total immunologic dysfunction despite normal numbers and distrib

[Catastrophic antiphospholipid syndrome after ovulation induction].

INTRODUCTION: The catastrophic antiphospholipid syndrome (CAPS) is a rare expression of the antiphospholipid syndrome (APLS). The all predicting factors of happening of CAPS are not yet discovered. Some women presenting an APLS with infertility have recourse to ovulation induction (OI). EXEGESIS: After an OI, a woman is admitted for acute renal failure, hypertension, seizures, respiratory failure. History and immunologic tests are in favour of APLS. Renal biopsy highlights a thrombotic microangiopathy. Diagnosis of CAPS is made. Patient improves after respiratory assistance, parenteral treatm

Community-acquired pneumonia mimicking tuberculosis in a patient with Down syndrome treated with adjunctive injectable corticosteroids.

Community-acquired pneumonia (CAP) is a frequent cause of morbidity in individuals with Down syndrome (DS) due to their inherent immunological vulnerabilities and anatomical predispositions. We present the case of a young woman in late adolescence with DS who developed severe CAP with radiological findings mimicking pulmonary tuberculosis. Despite initial diagnostic uncertainty, the patient improved remarkably following initiation of antibiotics combined with injectable corticosteroids. This case highlights both the diagnostic challenge of distinguishing CAP from tuberculosis in high-risk popu

[Study of the chronic apical periodontitis influence upon the patient's organism status].

During clinical-immunological study of the patients with chronic apical periodontitis (CAP) some differences were found between the following categories--CAP involving not more than 2 teeth and plural CAP. There were found the changes of both general immunity indices and its local components. Chronic inflammation course in periodontal tissues led to imbalance between cellular and humoral local immune reactions. It was shown that the presence of protracted multiple infectious focuses in periodontal tissues led to organism sensitizing and secondary immune deficiency.

Impairment of capping in lymphoblastoid cell lines of Duchenne patients indicates an intrinsic cellular defect.

Duchenne muscular dystrophy (DMD) is a lethal sex-linked degenerative disorder of the muscle in man. Generalized cell membrane abnormalities seem to be involved in the pathogenesis of the disease; in particular, the impairment of lymphocyte capping capacities has been repeatedly confirmed. To clarify whether capping impairment is a consequence of factors related to the activity of the disease or an expression of an intrinsic cellular defect, we have investigated the capping capacities of DMD EBV-transformed cell lines. The results indicate a significant impairment of capping capacity in cultur

Coexistence of systemic sclerosis and cryopyrin-associated periodic syndrome.

Systemic sclerosis (SSc) and cryopyrin-associated periodic syndrome (CAPS) are distinct clinical entities belonging to the autoimmune and autoinflammatory diseases, respectively. The coexistence of the two entities has rarely been reported and is poorly characterized. Here, we described a case of a 38-year-old Japanese woman diagnosed with anti-centromere antibody-positive SSc and CAPS carrying the pathogenic mutation in the NLRP3 gene, with a detailed autoantibody profile by a high-throughput comprehensive protein array covering approximately 90% of the human transcriptome. The clinical manif

I and i antigens of human peripheral blood lymphocytes cocap with receptors for concanavalin A.

Surface immunofluorescence experiments using a human anti-i and two anti-I antisera have been performed on human peripheral blood lymphocytes. These are known to contain cold-reactive monoclonal IgM antibodies against the carbohydrate sequence: (formula: see text). A high proportion of B- and T-type lymphocytes express these I and i determinants. In the presence of anti-human immunoglobulin, the cold-reactive membrane-associated complexes of I-anti-I and i-anti-i become stabilized, and redistribution (with patching and capping) can be elicited at 37 degrees C. Dual fluorescence experiments hav

Adaptive immunity in different CT patterns of active tuberculosis and possible variability according to patients' geographic provenience.

BACKGROUND: It is still unclear if low lymphocyte levels are directly related to immunological modifications induced by the TB infection or if they depend on the general pre-existing health impairment of affected patients. Our aim was to detect eventual differences in the immunological status of patients with pulmonary TB compared to an age and sex-matched group of hospitalized patients with other bacterial community-acquired pneumonia (CAP). In addition, we tried to assess an association between alterations in the peripheral lymphocyte subsets and the development of different CT patterns of a

An Engineered Self-biomineralized Oncolytic Adenovirus Induces Effective Antitumor Immunity and Synergizes With Immune Checkpoint Blockade.

Oncolytic adenoviruses (oADV) are promising cancer treatment agents. However, in vivo hepatic sequestration and the host immunologic response against the agents limit the therapeutic potential of oADVs. In this study, we present a combined method with a rational design for improving oADV infection efficiency, immunogenicity, and treatment efficacy by self-biomineralization. We integrated the biomimetic nucleopeptide W6p into the capsid of oADV using reverse genetics, allowing calcium phosphate mineralization to be biologically induced on the surface of oADV under physiologic conditions, result

Abnormal lymphocyte capping in a patient with severe combined immunodeficiency disease.

Despite the presence of normal numbers and distribution of T and B lymphocutes and normal levels of serum immunoglobulins, a five-month-old infant failed to show any evidence of T-cell or B-cell immunity. In trying to identify a specific membrane abnormality as a potential cause of the immunologic dysfunction, we examined the lateral mobility of the cell-surface receptor for concanavalin A. In contrast to normal cells, in which the receptor is distributed uniformly over the cell surface, the patient's lymphocytes showed an unusually high accumulation of concanavalin A receptors in surface caps

Immunogenicity Analysis of PCV3 Capsid Highly Expressed Using Baculovirus.

Porcine circovirus type 3 (PCV3) capsid protein (Cap) is a key antigen for immunological studies and vaccine development. Different optimized PCV3 ORF2 sequences were used to construct six baculovirus transfer plasmids, with the pOET1.1-based design yielding the highest Cap level. Cap expression was confirmed by Western blot, IPMA and IFA. Recombinant baculovirus amplification was optimized, achieving the highest titer at an MOI of 0.1 with a 72 h harvest to 107.5TCID50/0.1 mL, while maximal Cap production was obtained at an MOI of 0.1 with a 96 h harvest. PCV3 Cap virus-like particles (VLPs)

The porcine circovirus 3 humoral response: characterization of maternally derived antibodies and dynamic following experimental infection.

UNLABELLED: Since Porcine Circovirus 3 (PCV3) was first identified in 2016, our understanding of the humoral response is still relatively scarce. Current knowledge of the PCV3 humoral response is primarily based on field studies identifying the seroprevalence of PCV3 Cap-induced antibodies. Studies on the humoral response following experimental PCV3 infection have conflicting results where one study reports the development of the Cap IgG response 7 days postinfection with no concurrent Cap IgM response, while a second study shows a Cap IgM response at the same time point with no detection of C
