BACKGROUND: Genomic imprinting is an epigenetic gene regulatory mechanism; disruption of this process during early embryonic development can have major consequences on both fetal and placental development. The periconceptional period and intrauterine life are crucial for determining long-term susceptibility to diseases. Treatments and procedures in assisted reproductive technologies (ART) and adverse in-utero environments may modify the methylation levels of genomic imprinting regions, including insulin-like growth factor 2 (IGF2)/H19, mesoderm-specific transcript (MEST), and paternally expres

Imprinted genes have been posited to have important roles in human brain development and cognition, but their effects in nonclinical populations have yet to be investigated. Single-nucleotide polymorphisms (SNPs) of the imprinted gene LRRTM1 have previously been associated with schizophrenia risk and with handedness in individuals with dyslexia. We tested the hypothesis that genetic variation (SNPs) and epigenetic variation (methylation) in this gene are associated with schizotypy and handedness in a nonclinical population. Risk alleles of the three schizophrenia-linked SNPs were associated wi

We review the hypothesized and observed effects of two of the major forms of genomic conflicts, genomic imprinting and sexual antagonism, on human health. We focus on phenotypes mediated by peptide and steroid hormones (especially oxytocin and testosterone) because such hormones centrally mediate patterns of physical and behavioral resource allocation that underlie both forms of conflict. In early development, a suite of imprinted genes modulates the human oxytocinergic system as predicted from theory, with paternally inherited gene expression associated with higher oxytocin production, and in

In research on animal numerical cognition, newly hatched domestic chicks have been shown to rely on distinct strategies when confronted with quantitative choices. In some conditions, such as after imprinting on a specific set of objects, chicks preferentially approach the larger set of familiar items, indicating sensitivity to magnitude. In other conditions, however, their responses are governed not by magnitude per se, but by the degree of similarity between the test objects and a previously experienced set, where similarity is defined in terms of conformity to specific perceptual constraints

Angelman Syndrome (AS), characterized by mental retardation, absence of speech, seizures and motor dysfunction, is caused by genetic defects leading to loss of expression of the maternal copy of the chromosome 15q11-13 imprinted region. Most cases are sporadic, being caused by de novo deletion of maternal chromosome 15q11-13 (75%) or by paternal uniparental disomy (3-4%). Familial cases can occur, due to mutations in the UBE3A gene or in the imprinting center. We describe the case of a pregnant woman having two nephews with AS caused by a UBE3A mutation; lack of communication within the family

Phenylketonuria (PKU) is a rare genetic disorder caused by a defect in the metabolism of phenylalanine (Phe). Currently, the most commonly used treatment for PKU is dietary Phe restriction. Problems associated with Phe restricted diets include lack of universal availability, high treatment costs, and reduced adherence to continued treatment with age and finally the development of psychological and neurological problems in a significant proportion of patients despite early start of treatment. One possible approach to decreasing blood Phe level, is inhibition of GI tract absorption of this amino

Tourette syndrome has significant genetic determinants. The mode of transmission, while mildly controversial, generally is thought to be due to a single major locus inherited either as an autosomal dominant trait with reduced penetrance, or as a trait with intermediate inheritance in which some heterozygotes manifest the disorder. These is evidence for a Tourette syndrome spectrum of symptoms that includes obsessive-compulsive disorder. Systematic genome linkage studies of Tourette syndrome are progressing, but to date there are no significant linkage findings, although the search has included

Early olfactory imprinting has been proposed as a strategy to enhance the performance of detection dogs, yet empirical evidence from operational contexts remains limited. This study evaluated the effects of early exposure to target odours on four core detection variables: sensitivity under tight-containment conditions, reaction distance, work endurance, and detection reliability. Twenty operational adult dogs served as controls, and four dogs underwent an early-learning protocol beginning during sensitive stages of neurosensory development. All dogs completed parallel test batteries under equi

Ten thousand broiler chicks on commercial farms in Israel were imprinted to blue and red plastic boxes (20 x 20 x 40 cm) containing loud speakers (8 omega, 12 W) equipped with a tape recorder, in which a classical music cassette had been placed. Upon reaching 3 wk of age, an attempt was made to move these chicks from the training area (A) to a new area (B) using the imprinting objects. This experiment was repeated four times: May, August and November of 1985, and in January 1986. During the warm months of May and August 30 to 60% of the experimental chicks followed the imprinting objects towar

Studies of adult female carriers of the fragile X chromosome indicate that certain psychological problems occur with a greater frequency and severity than expected. This study examines the association of parental origin of the fragile X chromosome and of fragility detected in the karyotype with measures of social, educational, and psychological functioning in a group of adult fragile X females of normal intelligence. The results show that, as a group, women who inherit the fragile X chromosome from their mother and who demonstrate positive fragility in the karyotype (MI+ group = [maternal inhe

Filial imprinting in precocial birds provides an unusually tractable example of how a brief early-life experience becomes a durable behavioral commitment. Decades of work in the domestic chick have identified the intermediate medial mesopallium (IMM), thyroid-hormone-dependent regulation of the sensitive period, rapid kinase and translational signaling, receptor and synaptic remodeling, later transcriptional divergence, and distributed retrieval networks. What remains unresolved is how these events relate across scales. Here the ARCH × Φ framework is extended into a recursively coupled executi

Benzodiazepines are widely used anxiolytics and anticonvulsants, and their potent sedative properties are routinely used in presurgical anaesthesia. However, they are also known to induce a strong anterograde amnesia in patients. Specific benzodiazepine antagonists have recently been described, some of which have intrinsic pharmacological properties that are opposite to those of benzodiazepines. These have been called inverse agonists and they have been shown to be proconvulsant or convulsant whereas benzodiazepines are anticonvulsants. Inverse agonists are also anxiogenic rather than anxiolyt

BACKGROUND: Beckwith-Wiedemann syndrome (BWS) is a rare overgrowth syndrome characterized by congenital malformations and predisposition to embryonic tumors. Loss of methylation of imprinting center 2 (IC2) is the most frequent alteration and rarely associated with tumors compared to paternal uniparental disomy of chromosome 11 (UPD(11)pat) and gain of methylation of imprinting center 1.
METHODS: Our study aimed to describe the clinical, histopathological and genetic characteristics of two patients and establish genotype-phenotype correlations. The clinical diagnosis was based on the criteria

We describe a patient who was assigned female at birth because of genital ambiguity without performing further diagnostic procedures and presented at the age of 13-1/2 years because of her strong desire to change her legal sex. Karyotype was 46,XY; clinical, endocrinological, radiological and surgical work-up revealed hypergonadotropic hypogonadism and mixed gonadal dysgenesis. Gender identity reversal was performed after extensive psychological testing and adaptation of living circumstances resulting in a successful integration as a male with normal psychological and social functioning. In se

Affective dependency is characterized by emotional distress (insecure attachment) and dependency to another person with a low self-esteem and reassurance need. The paper proposes a reflection on the definition of emotional dependency and the confusion caused by various denominations. Overprotective and authoritarian parenting, cultural and socio-environmental factors may contribute to the development of dependent personality. Psychological epigenetic factors, such as early socio-emotional trauma could <imprint>> on neuronal circuits in prefronto-limbic regions that are essential for emotional

INTRODUCTION: Early life stress is imprinting regulatory properties with life-long consequences. We investigated heart rate variability in a group of small children with height below the third percentile, who experienced an episode of early life stress due to heart failure or intra uterine growth retardation. These children appear to develop autonomic dysfunction in later life.
RESULTS: Compared to the healthy control group heart rate variability (HRV) is reduced on average in a group of 101 children with short stature. Low HRV correlates to groups of children born small for gestational age (

The distribution of cholinergic receptor binding sites was studied in certain areas of the chick brain, especially that part (IMHV) of hyperstriatum ventrale which has previously been implicated in the imprinting process. There was relatively little binding of [125I]-alpha-bungarotoxin in this region, as shown using an autoradiographic technique. In contrast muscarinic receptor sites, with an affinity for [3H]-quinuclidinyl benzilate (QNB), are present in relatively high concentration in a medial forebrain sample containing IMHV and also in a visual projection area, the hyperstriatum accessori
