Genotyping of Canadian field strains of infectious bursal disease virus.

For this retrospective study, infectious bursal disease virus (IBDV) was detected in 134 bursal samples that originated from flocks with conditions such as airsacculitis, tracheitis, pneumonia, septicaemia, inclusion body hepatitis, coccidiosis, and/or a history of production problems without overt clinical symptoms. Samples were from seven Canadian provinces: Ontario, Quebec, Manitoba, British Columbia, Nova Scotia, Alberta, and Newfoundland and Labrador. Viral RNA was identified in bursae with moderate to severe and acute to chronic bursal damage. The ages of the flocks from which samples we

Reassortment in infectious bursal disease virus (IBDV) and related birnaviruses.

The Birnaviridae family consists of non-enveloped viruses with a single capsid that have a double-stranded RNA genome divided into two segments. Because they infect birds, fish, rotifers, marine mollusks, and insects, these viruses are of major economic significance to the global poultry and aquaculture industries. In the last decade, reassortment events have led to the emergence and spread of new strains of infectious bursal disease virus (IBDV) in poultry that could complicate control efforts. This review summarizes the literature regarding the molecular epidemiology and pathogenesis of reas

MARK1 suppresses infectious bursal disease virus replication via phosphorylating VP3.

Infectious bursal disease virus (IBDV) of the Birnaviridae family is a non-envelope, double-stranded RNA virus that encodes a VP3 protein with multiple functions, which controls viral genome replication, IFN-β production, and virus traffic in infected cells. Posttranslational modifications (PTMs), such as ubiquitination, of VP3 have been demonstrated for affecting its function and stability. To clarify the mechanism by which VP3 is regulated in IBDV infected cells, we focused on the phosphorylation of VP3. Mass spectrometry analysis identified that microtubule-affinity regulating kinases 1 (MA

Modeling Infectious Bursal Disease Virus (IBDV) Antigenic Drift In Vitro.

Infectious bursal disease virus (IBDV) vaccines do not induce sterilizing immunity, and vaccinated birds can become infected with field strains. Vaccine-induced immune selection pressure drives the evolution of antigenic drift variants that accumulate amino acid changes in the hypervariable region (HVR) of the VP2 capsid, which may lead to vaccine failures. However, there is a lack of information regarding how quickly mutations arise, and the relative contribution different residues make to immune escape. To model IBDV antigenic drift in vitro, we serially passaged a classical field strain bel

Evaluating how infectious bursal disease virus influences influenza A virus transmission in chickens.

Infectious bursal disease virus (IBDV) is a major cause of immunosuppression in chickens that has been shown to affect influenza A virus (avian influenza virus (AIV)) pathogenicity and shedding patterns. In this study, we compared the shedding of a low passage duck origin AIV in two groups of specific- pathogen free chickens: one group that was IBDV-inoculated and one sham-inoculated group. At 3 weeks of age, the birds were divided and intranasally given one of three doses (102, 103, 105) of A/mallard/OH/421/1987 (H7N8) low pathogenicity AIV. Although the bursae of the IBDV-infected chicks had

Infectious Bursal Disease Virus Genotypic Diversity from Poultry in Latin America.

Infectious bursal disease virus (IBDV) is a pathogen that causes Gumboro disease in young chickens. Vaccine strains and field IBDV genotypes are disseminated in chickens from commercial poultry farms worldwide. This study aimed to detect the field IBDV genotypic diversity in poultry farms in Latin America, mainly in Brazil. Bursal samples from 69 broiler flocks in eleven Latin American countries were obtained between 2015 and 2025. All 69 samples tested were positive for IBDV; the VP2 (segment A) and VP1 (segment B) genes were sequenced. Phylogenetic and amino acid substitution analyses were p

Response of embryonic chicken lymphoid cells to infectious bursal disease virus.

We exposed chicken embryos at embryonation day 18 (ED18) to a classical virulent infectious bursal disease virus (IBDV; cIBDV) and an attenuated strain of IBDV (aIBDV) and examined the response of embryonic lymphoid cells to these viruses. Embryos responded much more vigorously to cIBDV than to aIBDV. Following cIBDV exposure, embryonic thymus and bursa showed cellular destruction, enhanced rate of apoptosis and presence of viral proteins detectable by immunohistochemistry. At ED21, thymocytes from cIBDV-exposed embryos were severely deficient (P<0.05) in responding to stimulation in vitro wit

Mice as potential carriers of infectious bursal disease virus in chickens.

To investigate the role of mice as potential carriers of infectious bursal disease virus (IBDV), three mice were inoculated with a very virulent strain of IBDV and allowed to have contact with three uninoculated mice. Faeces, intestine and pooled liver and spleen collected from inoculated mice 12 and 24 h post-inoculation were positive for IBDV by reverse transcriptase-polymerase chain reaction (PCR)-nested PCR (RT-PCR-nPCR). IBDV was detected by RT-PCR-nPCR in 3/3 samples of intestine and 2/3 samples of pooled liver and spleen from uninoculated in-contact mice at 24 h after exposure. Chickens

Molecular Epidemiology, Genogroup Classification and Diversity of Infectious Bursal Disease Virus.

Viral diseases are a major cause of economic losses in the poultry industry. Among these is infectious bursal disease (IBD), which poses a threat to global poultry production. The epidemiology of IBD is changing, while very virulent IBDV (vvIBDV) remains endemic and represents the dominant clinical threat in regions such as Africa and parts of South America, the regional transition towards novel variants (nVarIBDVs) and reassortants (A3B1) is particularly pronounced in Asian and European poultry production, respectively. This review synthesises global IBD epidemiological data reported up to De

Novel variant strains of infectious bursal disease virus isolated in China.

Infectious bursal disease (IBD) is one of the most important immunosuppressive diseases that seriously threaten poultry farming and food safety worldwide. The variant strain of infectious bursal disease virus (IBDV) has been greatly neglected for more than 30 years. Recently, the subclinical infection of suspected IBD, causing considerable economic losses, occurred in the main chicken-farming regions of China. Through RT-PCR, sequencing, and phylogenic analyses, novel variant IBDVs were first identified in six provinces of eastern China. Immunological detection further confirmed the antigenic

Characteristics of a very virulent infectious bursal disease virus from California.

An outbreak of infectious bursal disease (IBD) in two California layer flocks resulted in the isolation of two infectious bursal disease viruses designated rA and rB. Increased mortality plus gross and histopathology in the layer flocks suggested rA and rB could be very virulent infectious bursal disease virus (vvIBDV). Preliminary studies indicated that high mortality resulted when bursa homogenates from the layer farms were used to inoculate specific-pathogen-free (SPF) chicks. In addition, rA and rB contained VP2 amino acid sequences typically seen in vvIBDV. Molecular and in vivo studies w

Chicken recombinant antibodies specific for very virulent infectious bursal disease virus.

A phage-displayed single chain variable fragment (scFv) antibody library was constructed from the immune spleen cells of chickens immunized with very virulent infectious bursal disease virus (vvIBDV) strain CS89. A library consisting of around 9.2 x 10(7) clones was subjected to 3 rounds of panning against captured CS89 virus. Analysis of individual clones by nucleotide sequencing revealed at least 22 unique scFv antibodies binding to vvIBDV in ELISA. Testing of the scFv antibody panel in ELISA against classical, variant or vaccine strains and a wide variety of vvIBDV isolates from the UK, Chi

Comparative bursal cytokine gene expression and apoptosis in vaccinated chickens following virulent infectious bursal disease virus challenge.

The bursal cytokine gene expression and apoptosis were compared in vaccinated chickens with either live or immune-complex infectious bursal disease virus (IBDV) vaccines with or without virulent IBDV challenge. The cytokine gene expressions were evaluated at 5 and 12 day-post-challenge (DPC). The apoptotic marker Caspase-3 was determined by IHC on collected bursae, thymus, spleen, and kidneys at 12 DPC. A significantly decreased bursal cytokine levels were observed in the all-vaccinated birds except for IL-6 in the classic IBD vaccines at 5DPC. A significant upregulation of the IL-2 was observ

Glycoprotein Production by Bursal Secretory Dendritic Cells in Normal, Vaccinated, and Infectious Bursal Disease Virus (IBDV)-Infected Chickens.

The aim of this study is to follow the gp production in IBDV-vaccinated and challenged birds. The progress of IBDV infection was monitored using anti-VP2 immunocytochemistry, light and transmission electron microscopy. In the medulla of the bursal follicle, the Movat pentachrome staining discovered an extracellular glycoprotein (gp) produced by bursal secretory dendritic cells (BSDCs). The secretory granules of BSDCs either discharge resulting in extracellular gp or fuse together forming intracellular corpuscles. The double fate of granules suggests a dual function of BSDCs: (a.) Infectious bursal disease virus (IBDV) as a novel oncolytic virotherapy in glioblastoma.

BACKGROUND: Glioblastoma (GBM) is the most aggressive form of cancer of the central nervous system. Despite advances in immunotherapies and standard-of-care treatments for GBMs, clinical outcomes remain limited-owing to the immunosuppressive tumor microenvironment and the intrinsic resistance of GBM to conventional approaches. As a result, there is growing interest in rational combination strategies, particularly those pairing oncolytic viruses with immune-based therapies or established treatment modalities. Oncolytic viruses, by displaying conditionally enabled tumor cell-restricted replicati

Characterization and pathogenicity of a novel variant infectious bursal disease virus in China.

Infectious bursal disease (IBD) is a highly epidemic and immunosuppressive disease of 3- to 6-week-old chicks caused by infectious bursal disease virus (IBDV). Since 2017, there has been a notable increase in the isolation rates of novel variant IBDV strains in China, of which characteristic amino acid residues were different from those of early antigen variants. In this study, one IBDV strain was isolated from a farm with suspected IBD outbreak in Shandong Province, China, which was designated LY21/2. The strain LY21/2 could replicate in MC38 cells with previous culture adaption in SPF chick

A Naturally Occurring Novel Reassortant Strain of Infectious Bursal Disease Virus from Quails.

Infectious bursal disease virus (IBDV) primarily triggers severe immunosuppression in birds and has resulted in substantial economic losses to the poultry industry globally. Here, a novel naturally reassortant IBDV strain, designated IBDV_G3, was isolated from quails during a disease outbreak in Jiangsu Province, China. Genome sequencing revealed that IBDV_G3 carried segment A derived from the serotype-2-related A0 lineage, while segment B clustered within the B1a classical-like lineage, and this genomic constellation was further confirmed in the original uncultured quail tissues. IBDV_G3 repl

Phylogenetic and pathogenic analyses of circulating infectious bursal disease virus strains in China.

Infectious bursal disease (IBD) remains one of the most important immunosuppressive diseases in poultry and has been endemic in China for over four decades. In recent years, outbreaks characterized by markedly reduced mortality have emerged. To investigate the current prevalence and pathogenicity of infectious bursal disease virus (IBDV), strains were isolated from vaccinated poultry farms in central and eastern China, and representative isolates of distinct genotypes were evaluated for pathogenicity. Phylogenetic analysis of segment A (HVR) and segment B (B-marker) classified segment A into e
