[Hiccup and dysfunction of the inferior olivary complex].

Two patients with chronic hiccup and MRI demonstrated lesions at the level of the inferior olivary complex are here reported. Whereas the neurological examination was normal in case 1, case 2 had the syndrome of palatal myoclonus and progressive ataxia. In the case 1, extensive electrophysiological studies were consistent with hiccup, in the absence of any other disorder (in particular, there was no evidence of palatal myoclonus). These findings suggest that the dysfunction of the inferior olivary complex could be implicated in the pathogenesis of hiccup.

On the question of nomenclature of homologous subdivisions of the inferior olivary complex.

The wide variety of terms, or other designations, that have been applied to the mammalian IO have been described above and are summarized in Tables I-IV. Although divergent in their general character there are some common threads that can be used to weave an acceptable and appropriate set of terms that can be applied to most, if not all, mammals studied to date. Concerning the MAO it is clear that there are six principal subdivisions of this part of IO (Table II A, II B). In addition to individual descriptive terms (such as 'ventral part') they have been designated as "subgroups", "cell groups

Drug-induced rhythmical activity in the inferior olivary complex of the rat.

Experiments have been performed on pentobarbitone anesthetized or decerebrated rats. The nature of the synchronous rhythmical activity which occurs in the inferior olive following the electrophoretic or systemic administration of harmaline, harmine, dihydro-beta-erythroidine and various other compounds, is described. Harmine was shown to reduce the late phase of biphasic unitary action potentials and to evoke massed synchronous rhythmical activity on which the units were superimposed. The beta-carboline was more effective than ACh or DL-homocysteate (DLH) in increasing cell discharge rates. Sy

Inferior olivary nuclear complex of the rat: morphology and comments on the principles of organization within the olivocerebellar system.

This study was undertaken (1) to provide a description of the normal morphology and anatomical interrelationships within the inferior olivary cell groups and (2) to determine the topographical organization of projections from this nuclear complex to the cerebellum. Conventional histological methods and the technique of retrograde transport of horseradish peroxidase (HRP) and WGA-HRP in conjunction with the sensitive chromogen tetramethyl benzidine were used in this series of experiments. In common with that of other animals, the inferior olivary nucleus of the rat consists of three subdivision

Patterns of convergence in the central nucleus of the inferior colliculus of the Mongolian gerbil: organization of inputs from the superior olivary complex in the low frequency representation.

Projections to the inferior colliculus (IC) from the lateral and medial superior olivary nuclei (LSO and MSO) were studied in the gerbil (Meriones unguiculatus) with neuroanatomical tract-tracing methods. The terminal fields of projecting axons were labeled via anterograde transport of biotinylated dextran amine (BDA) and were localized on series of horizontal sections through the IC. In addition, to make the results easier to visualize in three dimensions and to facilitate comparisons among cases, the data were also reconstructed into the transverse plane. The results show that the terminal f

The origin of the reticulo-olivary projection in the rat: a retrograde horseradish peroxidase study.

Electrophoretic injections of horseradish peroxidase were made in different parts of the rat inferior olivary complex using a ventral approach. Data from these injections provide anatomical evidence for the existence of a projection to the inferior olive which takes origin from reticular nuclei in the brainstem. The majority of reticulo-olivary neurons are located in the nucleus raphe obscurus and nucleus raphe pallidus. Other reticular nuclei which contribute to this projection include the nucleus reticularis ventralis and nucleus reticularis gigantocellularis. Analysis of injections confined

Disynaptic Inhibitory Cerebellar Control Over Caudal Medial Accessory Olive.

The olivocerebellar system, which is critical for sensorimotor performance and learning, functions through modules with feedback loops. The main feedback to the inferior olive comes from the cerebellar nuclei (CN), which are predominantly GABAergic and contralateral. However, for the subnucleus d of the caudomedial accessory olive (cdMAO), a crucial region for oculomotor and upper body movements, the source of GABAergic input has yet to be identified. Here, we demonstrate the existence of a disynaptic inhibitory projection from the medial CN (MCN) to the cdMAO via the superior colliculus (SC)

A Transient Feature of the Inferior Olive Supports the Development of Cerebellar Internal Models.

The inferior olive (IO) supports motor learning by supplying the cerebellum with critical sensory and motor input. In adult rats, that input includes externally generated limb stimulation. In contrast, the IO of Postnatal Day 8 (P8) rats does not exhibit responses to external stimuli. Instead, IO activity primarily reflects corollary discharges associated with the production of self-generated limb twitches during active (REM) sleep. Because corollary discharges are necessary for the computation of internal models, we tested the hypothesis that IO-related corollary discharge is necessary for th

Kainic acid lesions of the superior olivary complex: a horseradish peroxidase study of surviving brain-stem projections.

Lesions of the superior olivary complex of the adult ferret were made by pressure injection of kainic acid (5 nM/microliters in Locke's solution) through a glass micropipette inserted into the lower brain-stem. Small injections of kainic acid (1.5 microliters) produced a localized loss of nerve cells in the superior olivary complex in the vicinity of the pipette tip without apparent damage to fibers of passage in the trapezoid body. The extent of neural damage was determined by the absence of cell bodies in Nissl-stained sections. Integrity of fibers in the trapezoid body and other decussating

Olivocerebellar circuit development as a substrate for dystonia pathogenesis.

Despite the progress in identifying genetic and molecular contributors to dystonia, the synaptic and circuit mechanisms that disrupt motor maturation and serve as the substrate for disease pathogenesis are only beginning to emerge. In this chapter, we highlight evidence implicating the olivocerebellar node as a key site of pathogenesis in the dystonia network. As an example of aberrant olivocerebellar circuit activity leading to dystonia, here we examine a mouse model in which excitatory neurotransmission is silenced at inferior olive to cerebellum synapses upon Vglut2 mediated conditional com

Structural arrangement of auditory brainstem nuclei in the bats Phyllostomus discolor and Carollia perspicillata.

The structure of the mammalian auditory brainstem is evolutionarily highly plastic, and distinct nuclei arrange in a species-dependent manner. Such anatomical variability is present in the superior olivary complex (SOC) and the nuclei of the lateral lemniscus (LL). Due to the structure-function relationship in the auditory brainstem, the identification of individual nuclei supports the understanding of sound processing. Here, we comparatively describe the nucleus arrangement and the expression of functional markers in the auditory brainstem of the two bat species Phyllostomus discolor and Caro

Topographical Relationship in the Mesodiencephalic Projection to the Inferior Olive Linked With Cerebellar Hemispheric Lobules in Mice.

The mesodiencephalic junction (MDJ) is located in a caudorostrally elongated, column-shaped region encompassing the medial accessory oculomotor nucleus and perirubral area. It relays cerebral projections to the rostral subnuclei of the inferior olive (IO), which, in turn, send climbing fibers to the cerebellar hemisphere and receive nucleo-olivary projections from the cerebellar nuclei (CN), forming a parallel, loop-shaped, modular circuitry. However, the topographical organization of the MDJ-IO projection, which may contribute to functional localization within the cerebellar hemisphere, has n

The rheumatoid hand in the light of fluorescence: a diagnostic technique of the future?

The inferior olive provides the climbing fibers to Purkinje cells in the cerebellar cortex, where they elicit all-or-none complex spikes and control major forms of plasticity. Given their important role in both short-term and long-term coordination of cerebellum-dependent behaviors, it is paramount to understand the factors that determine the output of olivary neurons. Here, we use mouse models to investigate how the inhibitory and excitatory inputs to the olivary neurons interact with each other, generating spiking patterns of olivary neurons that align with their intrinsic oscillations. Anatomy of superior olivary complex and lateral lemniscus in Etruscan shrew.

Based on the auditory periphery and the small head size, Etruscan shrews (Suncus etruscus) approximate ancestral mammalian conditions. The auditory brainstem in this insectivore has not been investigated. Using labelling techniques, we assessed the structures of their superior olivary complex (SOC) and the nuclei of the lateral lemniscus (NLL). There, we identified the position of the major nuclei, their input pattern, transmitter content, expression of calcium binding proteins (CaBPs) and two voltage-gated ion channels. The most prominent SOC structures were the medial nucleus of the trapezoi

Multifaceted Mesodiencephalic Triangles: Insights into Hypertrophic Olivary Degeneration and Oculopalatal Tremor Pathophysiology.

Oculopalatal tremor is a rare neurological disorder characterized by rhythmic oscillations of ocular and palatal muscles. This phenomenon is commonly associated with hypertrophic degeneration of the inferior olive due to loss of GABAergic cerebello-olivary fibers. Oculopalatal tremor highlights the complex interplay between cerebellar, mesodiencephalic, and olivary networks. The principal, medial accessory, and dorsal accessory subnuclei of the inferior olive exhibit rhythmic subthreshold oscillations that are hypothesized to play an important role in oculopalatal tremor. However, delayed trem

A disynaptic basal ganglia connection to the inferior olive: potential for basal ganglia influence on cerebellar learning.

Recent studies have shown that the cerebellum and the basal ganglia are interconnected at subcortical levels. However, a subcortical basal ganglia connection to the inferior olive (IO), being the source of the olivocerebellar climbing fiber system, is not known. We have used classical tracing with CTb, retrograde transneuronal infection with wildtype rabies virus, conditional tracing with genetically modified rabies virus, and examination of material made available by the Allen Brain Institute, to study potential basal ganglia connections to the inferior olive in rats and mice. We show in both

Tonotopic distribution and inferior colliculus projection pattern of inhibitory and excitatory cell types in the lateral superior olive of Mongolian gerbils.

Sound localization critically relies on brainstem neurons that compare information from the two ears. The conventional role of the lateral superior olive (LSO) is extraction of intensity differences; however, it is increasingly clear that relative timing, especially of transients, is also an important function. Cellular diversity within the LSO that is not well understood may underlie its multiple roles. There are glycinergic inhibitory and glutamatergic excitatory principal neurons in the LSO, however, there is some disagreement regarding their relative distribution and projection pattern. He

Excitatory nucleo-olivary pathway shapes cerebellar outputs for motor control.

The brain generates predictive motor commands to control the spatiotemporal precision of high-velocity movements. Yet, how the brain organizes automated internal feedback to coordinate the kinematics of such fast movements is unclear. Here we unveil a unique nucleo-olivary loop in the cerebellum and its involvement in coordinating high-velocity movements. Activating the excitatory nucleo-olivary pathway induces well-timed internal feedback complex spike signals in Purkinje cells to shape cerebellar outputs. Anatomical tracing reveals extensive axonal collaterals from the excitatory nucleo-oliv

Hypertrophy of the inferior olivary nucleus in corticobasal degeneration: A neuropathological study.

AIMS: Corticobasal degeneration (CBD) is a rare neurodegenerative disorder. The status of the inferior olivary nucleus (ION) in CBD has been inadequately investigated. In this study, we conducted a pathological investigation of the ION in CBD. MATERIALS AND METHODS: We reviewed the data of Japanese patients with pathologically confirmed CBD who underwent consecutive autopsies between 1985 and 2020 at our institute. We retrospectively examined clinical data from medical records and clinicopathological conferences and semi-quantitatively assessed the ION, central tegmental tract, superior cereb

Microlesions of the inferior olive reduce vestibular modulation of Purkinje cell complex and simple spikes in mouse cerebellum.

Cerebellar Purkinje cells have two distinct action potentials: complex spikes (CSs) are evoked by single climbing fibers that originate from the contralateral inferior olive. Simple spikes (SSs) are often ascribed to mossy fiber-granule cell-parallel fiber inputs to Purkinje cells. Although generally accepted, this view lacks experimental support. Vestibular stimulation independently activates primary afferent mossy fibers and tertiary afferent climbing fibers that project to the uvula-nodulus (folia 8-10). CSs and SSs normally discharge antiphasically during sinusoidal roll-tilt. When CSs inc
