Advances in protein chemistry and structural biology2026eng
The rapid growth of global population is intensifying the demand for higher protein production. To satisfy the protein demand edible insects are emerging as a promising alternative protein source. Insect proteins exhibit high nutritional quality, favorable amino acid profiles, and significant functional versatility. However, their incorporation in the food requires a lot of processing to break it in to powder or other forms such as protein concentrate and isolate. The processing often applies thermal and non-thermal treatments which bring many structural modifications and alters the secondary,

The global demand for sustainable protein sources has positioned insects as a viable alternative to traditional livestock because of their impressive molecular efficiency and minimal ecological footprint. This chapter presents a detailed overview of biochemical design, nutritional, and industrial potential of protein sourced from insects, including high digestibility of amino acids, and rich content of bioactive peptides with antioxidant, antimicrobial, and anti-inflammatory properties. We highlight the efficiency of species such as Hermetia illucens, Tenebrio molitor, and Acheta domesticus to

Studying the structure and function of eukaryotic membrane proteins requires obtaining monodisperse, active proteins that can be used for further analysis. Eukaryotic expression systems are preferred because they provide the essential posttranslational modifications needed for proper folding and localization. Here, we describe effective approaches for the initial protein expression screening in insect cells of the human membrane protein PIGF, an adaptor protein involved in the biosynthesis of glycosylphosphatidylinositol anchors. The methodologies include construct design and cloning, followed

The survival of insects depends on their ability to detect molecules present in their environment. Odorant-binding proteins (OBPs) form a family of proteins involved in chemoreception. While OBPs were initially found in olfactory appendages, recently these proteins were discovered in other chemosensory and non-chemosensory organs. OBPs can bind, solubilize and transport hydrophobic stimuli to chemoreceptors across the aqueous sensilla lymph. In addition to this broadly accepted "transporter role", OBPs can also buffer sudden changes in odorant levels and are involved in hygro-reception. The ph

UNLABELLED: Systemic viral infection of insects typically begins with the primary infection of midgut epithelial cells (enterocytes) and subsequent transit of the progeny virus in an apical-to-basal orientation into the hemocoel. For insect-vectored viruses, an oppositely oriented process (basal-to-apical transit) occurs upon secondary infection of salivary glands and is necessary for virus transmission to non-insect hosts. To examine this inversely oriented virus transit in these polarized tissues, we assessed the intracellular trafficking of two model viral envelope proteins (baculovirus GP6

Sucking pests pose a serious agricultural challenge, as available transgenic technologies such as Bacillus thuringiensis crystal toxins (Bt) are not effective against them. One approach is to produce fusion protein toxins for the control of these pests. Two protein toxins, Hvt (ω-atracotoxin from Hadronyche versuta) and onion leaf lectin, were translationally fused to evaluate the negative effects of fusion proteins on Phenacoccus solenopsis (mealybug), a phloem-feeding insect pest. Hvt was cloned both N-terminally (HL) and then C-terminally (LH) in the fusion protein constructs, which were ex

Immune responses have been described for many different insect species. However, it is generally acknowledged that immune systems must therefore differ from those of vertebrates. An effective humoral immune response has been found in pupae of the cecropia moth, Hyalophora cecropia. The expression of this multicomponent system requires de novo synthesis of RNA and proteins and its broad antibacterial activity is due to at least three independent mechanisms, the most well known of which is the insect lysozyme. However, this enzyme is bactericidal for only a limited number of Gram-positive bacter

BACKGROUND: Serum protein electrophoresis (SPE) has emerged as a potentially valuable diagnostic tool in avian medicine; yet, there is limited information regarding SPE in cranes. Since 2000, 20 cases of unilateral periocular or facial soft tissue swelling, blepharitis, feather loss, and ocular or nasal discharge attributed to insect bite hypersensitivity were observed in cranes from a captive breeding center. SPE may be useful for evaluating these lesions.
OBJECTIVE: The aim of this study was to characterize the inflammatory response in cranes with hypersensitivity reactions using SPE.

In the course of this study more than 20 proteins have been isolated from the larval cuticle of Manduca sexta. Synthesis, secretion, transport and accumulation of four particular proteins, representative members of four characteristic groups, were followed during metamorphosis by immunoblot and immuncytochemical methods and are described in detail in this paper. We established that only some of the proteins of the soft cuticle of Lepidopteran larvae are synthesized in epidermal cells at the beginning of the larval stages and are digested during the moulting period (MsCP29). Other proteins (MsC

CRAL_TRIO domain proteins are known to bind small lipophilic molecules such as retinal, inositol and Vitamin E and include such gene family members as PINTA, α-tocopherol transfer (ATT) proteins, retinoid binding proteins, and clavesins. In insects, very little is known about either the molecular evolution of this family of proteins or their ligand specificity. Here we characterize insect CRAL_TRIO domain proteins and present the first insect CRAL_TRIO protein phylogeny constructed by performing reciprocal BLAST searches of the reference genomes of Drosophila melanogaster, Anopheles gambiae, A

The beetle Acanthoscelides obtectus (Say) causes severe post-harvest losses in the common bean (Phaseolus vulgaris). Under laboratory conditions, the susceptibility of A. obtectus to five coleopteran-specific Cry toxic proteins from Bacillus thuringiensis (Cry1Ba, Cry1Ia, Cry3Aa, Cry7Ab, and Cry23/37) was evaluated. After 30 days exposure, Cry proteins demonstrated high activity against A. obtectus adults (100% mortality). Proteins showed statistical differences in toxicity parameters compared to the control treatment, but the parameters were similar among them, and indicated that the final to

The introduction of terrestrial invertebrate processed animal proteins (PAPs) from insects in animal feed has introduced challenges for official controls in the European Union. Currently, their detection relies only on light microscopy and is entirely dependent on the expertise of trained operators. Recent studies have highlighted two primary problems regarding insect PAP detection: specificity issues due to the misidentification of non-insect structures as insect derived ones, and the difficulty in categorizing a wide morphological diversity of particles resulting from the grinding of whole i

The mannose-specific snowdrop lectin (Galanthus nivalis agglutinin: GNA), when fed to insects, binds to the gut epithelium and passes into the haemolymph. The ability of GNA to act as a carrier protein to deliver an insecticidal spider venom neurotoxin (Segestria florentina toxin 1: SFI1) to the haemolymph of lepidopteran larvae was investigated. Constructs encoding SFI1 and an SFI1/GNA fusion protein were expressed in Pichia pastoris. The insecticidal activity of purified recombinant proteins on injection was found to be comparable to published values for SfI1 purified from spider venom [Toxi

DJ-1 is a conserved, disease-associated protein that protects against oxidative stress and mitochondrial damage in multiple organisms. Human DJ-1 contains a functionally essential cysteine residue (Cys106) whose oxidation is important for regulating protein function by an unknown mechanism. This residue is well-conserved in other DJ-1 homologues, including two (DJ-1α and DJ-1β) in Drosophila melanogaster. Because D. melanogaster is a powerful model system for studying DJ-1 function, we have determined the crystal structure and impact of cysteine oxidation on Drosophila DJ-1β. The structure of

Insect pest control in crop production incurs substantial economic costs annually on a global scale. Although broad-spectrum chemical pesticides were once considered the most effective solution, their overreliance has led to adverse effects on beneficial insects, human health, and the environment, as well as the development of pesticide-resistant insect populations. Consequently, there is an urgent need for alternative pest management strategies that minimize pesticide use and reduce unintended impacts on natural enemies, thereby maintaining ecological balance. Host plant resistance plays a pi

The full-length putative structural proteins encoded by open reading frame 2 (ORF2) and ORF3 of hepatitis E virus have been cloned and expressed in recombinant baculovirus. Sera obtained from 28 Sudanese pediatric patients with acute hepatitis and 19 pediatric control patients were analyzed for reactivity to hepatitis E virus by using the baculovirus-expressed ORF2 and ORF3 proteins in a Western blot (immunoblot) format. Seventeen of the 18 patients classified as having non-A, non-B hepatitis, without acute antibody markers for hepatitis A, B, or C viruses, Epstein-Barr virus, or cytomegalovir

Several types of proteins are used in athletes' supplementation; nevertheless, given the problem of protein deficiency in the world and the growing need for ecological sources of protein, it is very interesting to study the quality of alternative protein sources, such as insect protein. This study investigated the nutritional value, micronutrient content, amino acid profile, and chemical score of banded cricket protein quality in the form of flour, defatted flour, and a protein preparation, as well as popular commercial protein supplements. In addition, in vitro digestion was performed, and th

Indirect evidence previously suggested that Arabidopsis (Arabidopsis thaliana) vegetative storage protein (VSP) could play a role in defense against herbivorous insects. To test this hypothesis, other AtVSP-like sequences in Arabidopsis were identified through a Basic Local Alignment Search Tool search, and their transcriptional profiles were investigated. In response to methyl jasmonate application or phosphate starvation, AtVSP and AtVSP-like genes exhibited differential expression patterns, suggesting distinct roles played by each member. The rise in the global population and increased awareness of sustainable protein sources have led to a surge in the expansion of edible insects. Edible insects are an eco-friendly, protein-rich substitute for conventional animal proteins. However, the wide acceptance of edible insects remains limited due to numerous concerns about food safety, shelf-life stability, and consumer acceptability. This review explores recent technological advancements in insect processing that address these critical challenges. Novel and advanced food processing approaches such as high-pressure processing, pulsed e
