The Flaviviridae include almost 70 viruses, nearly half of which have been associated with human disease. These viruses are among the most important arthropod-borne viruses worldwide and include dengue, yellow fever, and Japanese encephalitis viruses. Morbidity and mortality caused by these viruses vary, but collectively they account for millions of encephalitis, hemorrhagic fever, arthralgia, rash, and fever cases per year. Most of the members of this family are transmitted between vertebrate hosts by arthropod vectors, most commonly mosquitoes or ticks. Transmission cycles can be simple or c

Salivary gland hypertrophy viruses (SGHVs) are a unique, unclassified group of entomopathogenic, double-stranded DNA viruses that have been reported from three genera of Diptera. These viruses replicate in nuclei of salivary gland cells in adult flies, inducing gland enlargement with little obvious external disease symptoms. Viral infection inhibits reproduction by suppressing vitellogenesis, causing testicular aberrations, and/or disrupting mating behavior. Historical and present research findings support a recent proposal of a new virus family, the Hytrosaviridae. This review describes the d

The effective management of emerging insect-borne disease is dependent on the use of safe and efficacious chemical insecticides. Given the inherent ability of insects to develop resistance, it is essential to propose innovative strategies because insecticides remain the most important element of integrated approaches to vector control. Recently, intracellular phosphorylation and dephosphorylation of membrane receptors and ion channels targeted by insecticides have been described as new processes for increasing the sensitivity of insecticides. An efficient method might be to infect host insects

Several flaviviruses are important pathogens for humans and animals (Dengue viruses, Japanese encephalitis virus, Yellow-fever virus, Tick-borne encephalitis virus, West Nile virus). In recent years, numerous novel and related flaviviruses without known pathogenic capacity have been isolated worldwide in the natural mosquito population. However, phylogenetic studies have shown that genomic sequences of these viruses diverge from other flaviviruses. Moreover, these viruses seem to be exclusive of insects (they do not seem to grow on vertebrate cell lines), and were already defined as mosquito-o

Mosquitoes are known to vector arthropod-borne viruses (arboviruses) that pose a global public health issue in the form of mosquito-borne viral diseases such as chikungunya fever, dengue fever, Japanese encephalitis, yellow fever, and Zika. Besides, mosquitoes may also carry insect-specific viruses (ISVs), which are evolutionarily alike arboviruses yet do not infect vertebrates. These ISVs have been shown to affect the ability of mosquitoes to transmit arboviruses, as well as potentially inhibit arbovirus infections in vertebrate hosts. Yet, ISVs still constitute a relatively new and little-re

Sf9, the insect cell line commonly used for gene expression by recombinant baculovirus (BV), can be infected by St. Louis encephalitis (SLE) virus, a flavivirus, resulting in a persistent, productive, and cytopathic infection, while retaining the ability to be infected with a recombinant baculovirus (rBV). We now demonstrate using double immunofluorescence that single cells are dually infected with SLE virus and rBV. Fourteen additional viruses including additional flaviviruses, other arbovirus classes, vesicular stomatitis virus (VSV), and herpes simplex virus, type 1 (HSV-1) failed to produc

Aedes aegypti and A. albopictus mosquitoes are the main vectors for dengue virus (DENV) and other arboviruses, including Zika virus (ZIKV). Understanding the factors that affect transmission of arboviruses from mosquitoes to humans is a priority because it could inform public health and targeted interventions. Reasoning that interactions among viruses in the vector insect might affect transmission, we analysed the viromes of 815 urban Aedes mosquitoes collected from 12 countries worldwide. Two mosquito-specific viruses, Phasi Charoen-like virus (PCLV) and Humaita Tubiacanga virus (HTV), were t

Insects, like most animals, have intimate interactions with microorganisms that can influence the insect host's lipid metabolism. In this chapter, we describe what is known so far about the role prokaryotic microorganisms play in insect lipid metabolism. We start exploring microbe-insect lipid interactions focusing on endosymbionts, and more specifically the gut microbiota that has been predominantly studied in Drosophila melanogaster. We then move on to an overview of the work done on the common and well-studied endosymbiont Wolbachia pipientis, also in interaction with other microbes. Taking

UNLABELLED: Systemic viral infection of insects typically begins with the primary infection of midgut epithelial cells (enterocytes) and subsequent transit of the progeny virus in an apical-to-basal orientation into the hemocoel. For insect-vectored viruses, an oppositely oriented process (basal-to-apical transit) occurs upon secondary infection of salivary glands and is necessary for virus transmission to non-insect hosts. To examine this inversely oriented virus transit in these polarized tissues, we assessed the intracellular trafficking of two model viral envelope proteins (baculovirus GP6

Viruses were identified from male anthropophilic mosquitoes from Mato Grosso (MT) State, Midwest Brazil from February 2017 to January 2018. Mosquitoes tested included Aedes (Stegomyia) aegypti (1139 males; 84 pools), Culex quinquefasciatus (9426 males; 179 pools), Culex sp. (3 males; 3 pools) and Psorophora albigenu (1 male; 1 pool) collected from four cities of MT. Pools were subjected to viral RNA extraction followed by RT-PCRs specific for ten flaviviruses, five alphaviruses and Simbu serogroup of orthobunyaviruses. Positive pools were passaged three times in VERO cells (alphavirus and orth

With the advance of metatranscriptomic sequencing, an increasing number of symbiotic viruses have been recognized in insects. These insect symbiotic viruses not only modulate key aspects of insect biology, including physiology, fecundity, development, and behavior, but also influence the vector competence of arthropods for transmitting arboviruses. The majority of such vectors belong to the orders Hemiptera and Diptera, which transmit a wide spectrum of plant and animal arboviruses. Although insect symbiotic viruses usually replicate exclusively within insect cells, several symbiotic viruses h

The group of Insect-specific viruses (ISVs) includes viruses apparently restricted to insects based on their inability to replicate in the vertebrates. Increasing numbers of ISVs have been discovered and characterized representing a diverse number of viral families. However, most studies have focused on those ISVs belonging to the family Flaviviridae, which highlights the importance of ISV study from other viral families, which allow a better understanding for the mechanisms of transmission and evolution used for this diverse group of viruses. Some ISVs have shown the potential to modulate arb

We collected 8,700 mosquitoes in three sites in China, which belonged to seven species. Their viromes were tested using metagenomic sequencing and bioinformatic analysis. The abundant viral sequences were detected and annotated belonging to more than 50 viral taxonomic families. The results were verified by PCR, followed by phylogenetic analysis. In the present study, we identified partial viral genes of dengue virus (DENV), a novel circovirus (CCV), densovirus (DNV), Japanese encephalitis virus (JEV), and Wuhan mosquito virus (WMV) in mosquitoes. Metagenomic analysis and PCR amplification rev

Plant viruses are evolutionarily endowed with traits that enhance their survival and dissemination attributes both in host plants and insect vectors. This review highlights current understanding of these multifaceted interactions, positioning plant viruses as master regulators of the intricate tripartite interactions. Direct viral effects on vector physiology and behaviour, highlighting evidence that viral infection may contribute to changes in olfactory systems, epigenetic modifications, involvement of salivary effectors, and horizontal gene transfer events that may vary vector competence, we

UNLABELLED: Arthropod-borne virus (arbovirus) infections are increasing globally, and invasive mosquito species are spreading. Since the end of the last century, Aedes japonicus has continued to spread in Europe as well as in North America. Aedes japonicus is known to be able to transmit several viruses, but extensive information about the vector competence of Ae. japonicus for alphaviruses is missing. Therefore, we infected field-caught Ae. japonicus mosquitoes from Germany with different alphaviruses that occur in areas with either tropical or moderate temperatures and are clustered as arthr

Most plant viruses rely on insect vectors for transmission. In general, nonviruliferous vectors acquire viruses from infected plants, whereas viruliferous vectors subsequently move to uninfected plants for virus inoculation. However, they are not merely passive passengers within their vector. Persistent-propagative plant viruses, which invade and replicate in insect vectors, can actively and directly reshape their physiology, morphology, and feeding behavior to facilitate efficient transmission. Recent studies have uncovered both indirect plant-mediated mechanisms and direct vector-targeted me

A comprehensive understanding of the virome in mosquito vectors is crucial for assessing the potential transmission of viral agents, designing effective vector control strategies, and advancing our knowledge of insect-specific viruses (ISVs). In this study, we utilized Oxford Nanopore Technologies metagenomics to characterize the virome of Aedes aegypti mosquitoes collected in various regions of Colombia, a country hyperendemic for dengue virus (DENV). Analyses were conducted on groups of insects with previous natural DENV infection (DENV-1 and DENV-2 serotypes), as well as mosquito samples th

Aedes aegypti and Culex pipiens complex mosquitoes are prolific vectors of arboviruses that are a global threat to human and animal health. Increased globalization and ease of travel have facilitated the worldwide dissemination of these mosquitoes and the viruses they transmit. To assess disease risk, we determined the frequency of arboviruses in western Kenyan counties bordering an area of high arboviral activity. In addition to pathogenic viruses, insect-specific flaviviruses (ISFs), some of which are thought to impair the transmission of specific pathogenic arboviruses, were also evaluated.

Malaria remains a leading cause of morbidity and mortality in large parts of the world. Resistance threatens current preventative and curative strategies, highlighting the need for novel complementary strategies. The opportunities that the mosquito virome may offer to reduce pathogen transmission have not been systematically explored for malaria control. In this study, we assessed whether insect-specific viruses affect Plasmodium development in mosquitoes. A panel of 15 viruses was tested for in vitro and in vivo replication in anopheline cells and mosquitoes. From this panel, the RNA viruses

Most alphaviruses are transmitted by mosquito vectors and infect a wide range of vertebrate hosts, with a few exceptions. Eilat virus (EILV) in this genus is characterized by a host range restricted to mosquitoes. Its chimeric viruses have been developed as safe and effective vaccine candidates and diagnostic tools. Here, we investigated the interactions between these insect-specific viruses (ISVs) and mosquito cells, unveiling their potential roles in determining vector competence and arbovirus transmission. By RNA sequencing, we found that these ISVs profoundly modified host cell gene expres
