Genetic variability of enterobacterial resistant integrons.

Sulfamide-resistant (SulR) natural strains of enterobacteria (11.9% of a total of 797 cultures, isolated with enteric microflora of honeybee gut, bee-fermented pollen, and plant issues) were tested for class 1 antibiotic resistance integrons (MRI). Only 5.3% of SulR strains were MRI-positive. Mutability of weak (wild type) MRI promotor has been shown. A more active hybrid promotor type has been amplified in isolated Klebsiella oxytoca strains. Regulatory genetic modifications in MRI are fraught with the development of multiple drug resistance of opportunistic strains.

Integrons in Enterobacteriaceae: diversity, distribution and epidemiology.

Integrons are versatile gene acquisition systems that allow efficient capturing of exogenous genes and ensure their expression. Various classes of integrons possessing a wide variety of gene cassettes are ubiquitously distributed in enteric bacteria worldwide. The epidemiology of integrons associated multidrug resistance in Enterobacteriaceae is rapidly evolving. In the past two decades, the incidence of integrons in enteric bacteria has increased drastically with evolution of multiple gene cassettes, novel gene arrangements and complex chromosomal integrons such as Salmonella genomic islands.

ESBL and Integron Profiles in Pediatric EPEC/STEC.

BACKGROUND: Diarrheagenic Escherichia coli (DEC) is a major cause of pediatric diarrhea, particularly in low- and middle-income countries. Antimicrobial resistance, including extended-spectrum β-lactamase (ESBL) production, further complicates clinical management. This study investigated DEC pathotypes, resistance patterns, ESBL prevalence and integron carriage among children with acute diarrhea in Mansoura, Egypt. METHODS: A total of 120 children (median age 6 years; 50.8% male) presenting with acute diarrhea were enrolled. Clinical data were recorded, and stool samples were cultured for E.

Integron activity accelerates the evolution of antibiotic resistance.

Mobile integrons are widespread genetic platforms that allow bacteria to modulate the expression of antibiotic resistance cassettes by shuffling their position from a common promoter. Antibiotic stress induces the expression of an integrase that excises and integrates cassettes, and this unique recombination and expression system is thought to allow bacteria to 'evolve on demand' in response to antibiotic pressure. To test this hypothesis, we inserted a custom three-cassette integron into Pseudomonas aeruginosa and used experimental evolution to measure the impact of integrase activity on adap

Integron integrase binds to bulged hairpin DNA.

Gene cassettes are short, monogenic DNA elements that translocate between integrons through site-specific excision and integration. These events require that an integron-coded tyrosine recombinase forms a reactive complex with two sites, at least one of which belongs to the attC class. An attC site can be divided into two pairs of short repeats flanking a palindromic central region. The nucleotide sequence of attC among different cassettes varies extensively, implying that the site contains a structural recognition determinant with low sequence constraints. Oligonucleotides representing many d

Competence-mediated DNA uptake diversifies Vibrio cholerae sedentary chromosomal integrons.

Bacteria often survive viral attack and environmental stress by sharing genes that enhance their defenses. The cholera pathogen Vibrio cholerae carries a sedentary chromosomal integron (SCI), a genetic element containing hundreds of mostly promoterless gene cassettes, about 10% of which encode antiviral systems. Cassettes are thought to reshuffle under stress to the favorable first array position, yet the SCI in pandemic V. cholerae has remained static for more than 60 years. In this study, we show that SCI diversification efficiently occurs by horizontal transfer linked to the genus's aquatic

Sedentary chromosomal integrons as biobanks of bacterial antiphage defense systems.

Integrons are genetic systems that drive bacterial adaptation by acquiring, expressing, and shuffling gene cassettes. While mobile integrons are well known for spreading antibiotic resistance genes, the functions of the hundreds of cassettes carried by sedentary integrons remain largely unexplored. We show that many of these cassettes encode small variants of known antiphage systems that favor their inclusion in the integron. We also demonstrate that nearly 10% of the integron cassettes in the pandemic Vibrio cholerae strain encode novel antiphage functions. Most of these novel systems have li

A comparative study of class 1 integrons in Acinetobacter baumannii.

Multidrug resistance (MDR) in Acinetobacter baumannii is increasingly reported and has become a significant public concern. The method responsible for the acquisition of resistance genes via integrons from the environment or intra-species in A. baumannii remains to be understood. This study was performed to investigate the transmission route of these integrons using a comparative analysis of published A. baumannii complete genomes. The phylogenetic analysis of A. baumannii type 1 integrases (IntI1) showed that the integrons could be transferred across the two evolutionary lineages, the interna

Role of integrons in antimicrobial susceptibility patterns of Acinetobacter baumannii.

The relationship between the presence and types of integrons and the antimicrobial susceptibility patterns of Acinetobacter baumannii was investigated. A total of 134 non-duplicated A. baumannii isolates, 54.5% (n=73) of which were subsequently found to carry class 1 integrons, were collected from a regional hospital in Taiwan between March and September 2007. Only two types of gene cassette array, aacA4-catB8-aadA1 and aacC1-orfP-orfP-orfQ-aadA1, were identified. Susceptibility data showed that those strains carrying integrons were significantly more resistant to all antibiotics tested except

Genetic architecture and functional dynamics of integrons in Staphylococcus aureus resistance.

Staphylococcus aureus is a major opportunistic pathogen and a leading cause of community and hospital-acquired infections worldwide, with a growing capacity to develop multidrug resistance (MDR). Among the genetic mechanisms driving this resistance, integrons play a critical role by capturing, rearranging, and expressing antimicrobial resistance gene cassettes through site-specific recombination. Although integrons are traditionally associated with Gram-negative bacteria, increasing evidence highlights their significant contribution to resistance dissemination in S. aureus, particularly in met

Microbial pathogenicity and virulence mediated by integrons on Gram-positive microorganisms.

Gram-positive microorganisms are one of leading pathogenic microorganisms in public health, including several typical "Super Bugs" as methicillin-resistant Staphylococcus aureus, Klebsiella pneumoniae carbapenemase and vancomycin-resistant enterococci, which caused a increasement of infections, clinical failures and expenses. Regarded as a common genetic element responsible for horizontal gene transfer, integrons are widely distributed in various pathogens considered as a determinant in the acquisition and evolution of antibiotic resistance. Current investigations mainly focus on the distribut

IS26-associated In4-type integrons forming multiresistance loci in enterobacterial plasmids.

Three distinct multiresistant loci from enterobacterial plasmids each comprised an integron and an IS26-associated sequence. Sequence comparison suggested a common ancestral structure that derived from an IS26 insertion into the 5' conserved segment of an In4-type integron and evolved through acquisition of gene cassettes and IS26-mediated recruitment of additional resistance genes of diverse origin.

The expression of integron arrays is shaped by the translation rate of cassettes.

Integrons are key elements in the rise and spread of multidrug resistance in Gram-negative bacteria. These genetic platforms capture cassettes containing promoterless genes and stockpile them in arrays of variable length. In the current integron model, expression of cassettes is granted by the Pc promoter in the platform and is assumed to decrease as a function of its distance. Here we explored this model using a large collection of 136 antibiotic resistance cassettes and show the effect of distance is in fact negligible. Instead, cassettes have a strong impact in the expression of downstream

A Novel, Integron-Regulated, Class C β-Lactamase.

AmpC-type β-lactamases severely impair treatment of many bacterial infections, due to their broad spectrum (they hydrolyze virtually all β-lactams, except fourth-generation cephalosporins and carbapenems) and the increasing incidence of plasmid-mediated versions. The original chromosomal AmpCs are often tightly regulated, and their expression is induced in response to exposure to β-lactams. Regulation of mobile ampC expression is in many cases less controlled, giving rise to constitutively resistant strains with increased potential for development or acquisition of additional resistances. Antimicrobial Resistance Gene Profiles in Integron-Positive and Integron-Negative Third-Generation Cephalosporin-Resistant E. coli from Human and Animal Sources.

Background/Objectives: Integrons are genetic platforms that allow bacteria to acquire antimicrobial resistance (AMR) genes, making them a focal point for many AMR studies and surveillance programs. This study investigated how the prevalence of integrons (intI and attI genes) in third-generation cephalosporin-resistant E. coli (3GCR-Ec) varied across three different sources (i.e., healthy children, domestic animals and urinary tract infections). The study aimed to determine how different classes of AMR genes vary among 3GCR-Ec with integrons present versus those where integrons are absent. Meth
