Actions of cholinergic drugs on cells in the interpeduncular nucleus.

The muscarinic versus nicotinic effects of acetylcholine (ACh) on the activity of cells in the interpeduncular nucleus (IPN) of guinea pig brain slices were investigated using electrophysiologic techniques. Nicotinic and muscarinic drugs acted postsynaptically to increase the firing rate of cells in the IPN. The excitatory actions of ACh were blocked both by atropine and by d-tubocurarine. These results indicated that cholinergic receptors of IPN cells seem to have both nicotinic and muscarinic characteristics.

Norepinephrine in the interpeduncular nucleus of the rat: normal distribution and the effects of deafferentation.

We used correlative biochemical and histochemical methods to examine (1) the norepinephrine (NE) projection from the paired locus coeruleus (LC) to the midline interpeduncular nucleus (IPN) of the adult rat and (2) the ability of the LC to respond to denervation of their target following removal of noradrenergic afferents (6-hydroxydopamine lesions of the LC) or non-noradrenergic afferents (lesion of the paired fasciculi retroflexi(FR]. Histofluorescence revealed that the NE innervation from the two LC to the IPN is symmetric and overlapping. This projection is confined to rostral, central, an

Substance P in the interpeduncular nucleus of the rat: normal distribution and the effects of deafferentation.

The interpeduncular nucleus (IPN) is a midbrain structure that receives its major afferents from the medial habenulae via the fasciculi retroflexi. Among the axons projecting to the IPN is a population of substance P (SP)-containing axons. The IPN has been subdivided into the central, dorsal, intermediate, rostral, and lateral subnuclei using cytoarchitectonic criteria. The distribution of SP among these subnuclei was determined by using Sternberger's ('79) peroxidase-antiperoxidase technique. In the normal IPN the rostral subnucleus can be subdivided into two sectors on the basis of SP conten

Effects of nicotine, methamphetamine and cocaine on extracellular levels of acetylcholine in the interpeduncular nucleus of rats.

There is increasing evidence that the cholinergic habenulo-interpeduncular pathway and the dopaminergic mesolimbic pathway may jointly mediate the reinforcing properties of addictive drugs. However, the effects of addictive drug on the functioning of the habenulo-interpeduncular pathway have not been well-characterized. Thus, several drugs of abuse (i.e., nicotine, cocaine, amphetamine) have been shown to alter the morphology of the habenulo-interpeduncular pathway, causing selective degeneration of the cholinergic neurons in this area. On the other hand, morphine was shown to alter the neuroc

Ultrastructural and neurochemical effects of the presumed cholinergic toxin AF64A in the rat interpeduncular nucleus.

We have studied the effect of the presumptive cholinergic neurotoxin, ethylcholine mustard aziridinium ion (compound AF64A), on ultrastructure and neurochemical markers in the rat interpeduncular nucleus (IPN). Stereotaxic injections of 1 nmol of AF64A resulted in extensive degeneration of synaptic terminals within 40 h. Ultrastructural damage to neuronal cell bodies, dendrites and axons was also sometimes observed at this stage. Five days after the injection, more severe degenerative changes were observed in a larger number of neuronal cell bodies, axons and dendrites. High affinity uptake of

An NTS-expressing inhibitory circuit from interpeduncular nucleus to dorsal raphe nucleus controls pain and comorbid depression in male mice.

Chronic pain often co-occurs with depression; however, the underlying neural mechanisms remain unclear. This study explored a specific inhibitory pathway-interpeduncular nucleus (IPN) neurotensin (NTS)-expressing neurons projecting to the dorsal raphe nucleus (DRN)-in mediating pain and depression comorbidity in male mice. We demonstrate that complete Freund's adjuvant (CFA) mice show increased IPNNTS neuronal activity. Optogenetically activating IPNNTS neurons or their projections to DRN in naive mice reliably induces pain- and depressive-like behaviors. Conversely, optogenetic inhibition of

Heparan sulfate endosulfatases, Sulf1 and Sulf2, are required for formation of the medial habenula-interpeduncular nucleus circuit.

The medial habenula (MHb), a medial subdivision of the habenula located in the dorsal diencephalon, sends axons towards the interpeduncular nucleus (IPN) through the fasciculus retroflexus. Upon reaching the IPN, MHb axons cross the midline multiple times. Although heparan sulfate proteoglycans have previously been shown to play a role in MHb axon guidance, the role of heparan sulfate 6-O-endosulfatases, Sulfatase 1 (Sulf1) and Sulfatase 2 (Sulf2), has to date not been investigated. We thus examined MHb axon trajectories in Sulf1 and Sulf2 knockout mice and found that Sulf1/2 double knockout (

Dual projections of gonadotropin releasing hormone containing neurons to the interpeduncular nucleus and to the vasculature in the female rat.

Immunofluorescence for gonadotropin releasing hormone (GnRH) in combination with retrograde labeling from the interpeduncular nucleus, as well as from the vasculature confirms that, in the rat, certain GnRH neurons project from the septum-diagonal band to the interpeduncular nucleus. However, about one half of these GnRH neurons also project to fenestrated capillaries as evidenced by uptake and retrograde transport of both peripherally injected Fluoro-Gold and centrally injected rhodamine labeled microspheres. The results indicate that the endocrine effects of GnRH are exerted in part by neuro

Tachykinin binding sites in the interpeduncular nucleus of the rat: normal distribution, postnatal development and the effects of lesions.

Tachykinin binding sites in the basal midbrain were labeled in adult and neonatal rats using 125I-Bolton Hunter (BH) substance P (SP) and 125I-BH eledoisin as ligands. In the adult, binding was very low in the tegmentum and raphe adjacent to the interpeduncular nucleus (IPN). Within the IPN, no binding with either ligand was seen in the target subnuclei of the habenular SP and substance K projections, the lateral subnuclei and the cap of the rostral subnucleus. Labeling with 125I-BH-SP was very light and was restricted primarily to the central subnucleus of the IPN while 125I-BH-eledoisin labe

Genetically Targeted Connectivity Tracing Excludes Dopaminergic Inputs to the Interpeduncular Nucleus from the Ventral Tegmentum and Substantia Nigra.

The "habenulopeduncular system" consists of the medial habenula (MHb) and its principal target of innervation, the interpeduncular nucleus (IP). Neurons in the ventral MHb (MHbV) express acetylcholine along with glutamate, and both the MHb and IP are rich in nicotinic acetylcholine receptors. Much of the work on this system has focused on nicotinic mechanisms and their clinical implications for nicotine use, particularly because the IP expresses the α5 nicotinic receptor subunit, encoded by the CHRNA5 gene, which is genetically linked to smoking risk. A working model has emerged in which nicot

Role of Olfactorily Responsive Neurons in the Right Dorsal Habenula-Ventral Interpeduncular Nucleus Pathway in Food-Seeking Behaviors of Larval Zebrafish.

The habenula (Hb) plays important roles in emotion-related behaviors. Besides receiving inputs from the limbic system and basal ganglia, Hb also gets inputs from multiple sensory modalities. Sensory responses of Hb neurons in zebrafish are asymmetrical: the left dorsal Hb and right dorsal Hb (dHb) preferentially respond to visual and olfactory stimuli, respectively, implying different functions of the left and right dHb. While visual responses of the left dHb (L-dHb) have been implicated in light-preference behavior, the significance of olfactory responses of the right dHb (R-dHb) remains unde

Loss of Goosecoid-like and DiGeorge syndrome critical region 14 in interpeduncular nucleus results in altered regulation of rapid eye movement sleep.

Sleep and wakefulness are regulated primarily by inhibitory interactions between the hypothalamus and brainstem. The expression of the states of rapid eye movement (REM) sleep and non-REM (NREM) sleep also are correlated with the activity of groups of REM-off and REM-on neurons in the dorsal brainstem. However, the contribution of ventral brainstem nuclei to sleep regulation has been little characterized to date. Here we examined sleep and wakefulness in mice deficient in a homeobox transcription factor, Goosecoid-like (Gscl), which is one of the genes deleted in DiGeorge syndrome or 22q11 del

Topography of cholinergic and substance P pathways in the habenulo-interpeduncular system of the rat. An immunocytochemical and microchemical approach.

The topography of cholinergic and substance P containing habenulo-interpeduncular projections has been studied in the rat. The research has been carried out by combining choline acetyltransferase and substance P immunohistochemistry to experimental lesions and biochemical assays in microdissected brain areas. In addition, computer-assisted image analysis has been performed in order to obtain quantification of immunohistochemical data. The results show that cholinergic and substance P containing neurons have a different localization in the medial habenula and project to essentially different ar

The Posterior Perforated Substance: A Brain Mystery Wrapped in an Enigma.

BACKGROUND: There is a dearth of published information on the posterior perforated substance as compared to the anterior perforated substance. We managed to glean facts about the posterior perforated substance that can serve as a landmark for surgical operations in the adjacent regions of the midbrain and the vessels passing through it. Moreover, the posterior perforated substance contains the interpeduncular nucleus responsible for the mental state of the individual. OBJECTIVES: 1) To describe the topography of the blood vessels supplying the posterior perforated substance area from the surg

Atypical Course of the Habenulo-Interpeduncular Tract in Chick Embryos.

Classical studies of the avian diencephalon hardly mention the habenulo-interpeduncular tract (a.k.a. retroflex tract), although both the habenula (HB) (its origin) and the interpeduncular nuclear complex (its target) are present. Retroflex tract fibers were described at early embryonic stages but seem absent in the adult in routine stains. However, this tract is a salient diencephalic landmark in all other vertebrate lineages. It typically emerges out of the caudal HB, courses dorsoventrally across thalamic alar and basal plates just in front of the thalamo-pretectal boundary, and then sharpl

Anatomic comparison of the endonasal and transpetrosal approaches for interpeduncular fossa access.

OBJECT: The interpeduncular cistern, including the retrochiasmatic area, is one of the most challenging regions to approach surgically. Various conventional approaches to this region have been described; however, only the endoscopic endonasal approach via the dorsum sellae and the transpetrosal approach provide ideal exposure with a caudal-cranial view. The authors compared these 2 approaches to clarify their limitations and intrinsic advantages for access to the interpeduncular cistern. METHODS: Four fresh cadaver heads were studied. An endoscopic endonasal approach via the dorsum sellae wit

Distribution of neurons expressing neurokinin B in the rat brain: immunohistochemistry and in situ hybridization.

Neurokinin B (NKB) belongs to the family of neuropeptides named tachykinins. Members of this family such as substance P or neurokinin A have been proposed to function as neurotransmitters or neuromodulators. Searching for possible sites of action of NKB in the central nervous system, we have now investigated its distribution within the rat brain by immunohistochemical techniques and in situ hybridization. For immunohistology two different antisera directed against amino acid sequences within preprotachykinin B were used. One antiserum had been raised against a synthetic derivative of NKB; the

Efferent connections of the habenular nuclei in the rat.

The efferent connections of the medial (MHb) and lateral (LHb) habenular nuclei in the rat were demonstrated autoradiographically following small injections of tritiated amino acids localized within various parts of the habenular complex. Comparison of individual cases led to the following conclusions. MHb efferents form the core portion of the fasciculus retroflexus and pass to the interpeduncular nucleus (IP) in which they terminate in a topographic pattern that refects 90 degrees rotations such that dorsal MHb projects to lateral IP, medial MHb to ventral, and lateral MHb to dorsal IP. A little rein on addiction.

Rewarding and aversive experiences influence emotions, motivate specific behaviors, and modify future action in animals. Multiple conserved vertebrate neural circuits have been discovered that act in a species-specific manner to reinforce behaviors that are rewarding, while attenuating those with an adverse outcome. A growing body of research now suggests that malfunction of the same circuits is an underlying cause for many human disorders and mental ailments. The habenula (Latin for "little rein") complex, an epithalamic structure that regulates midbrain monoaminergic activity has emerged in
