Opium induces apoptosis in jurkat cells.

BACKGROUND: The direct effect of some opioids on immune cells has been demonstrated. The aim of this study was to assess the apoptotic effect of opium on Jurkat T lymphocyte cells. METHODS: Different concentrations of opium (2.86 × 10-3 to 2.86 × 10-11 g/ml) were added to 24-well plates containing 5 × 105 Jurkat cells. Apoptotic events were assessed after 6, 24, and 72 hours by flow-cytometric detection of surface phosphatidylserine. FINDINGS: Significant differences in apoptosis of Jurkat cells were seen at 24 and 72 hours in different concentrations of opium (P < 0.05). After 72 hours, sig

[Regulation of Mitochondrial Autophagy by FADD in Jurkat Cells].

OBJECTIVE: To investigate the regulatory effect of Fas-associated death domain protein (FADD) on the number of mitochondria in Jurkat cells. METHODS: The FADD gene in Jurkat cell line was knocked out by lentivirus packaging CRISPR/Cas9-FADD plasmid. The protein levels of FADD and PGC1-α in virus-transfected Jurkat cells and the expression of LC3B/β-actin protein after rapamycin-induced autophagy were detected by Western blot. And the effect of FADD on autophagy was reversely validated through a rescue experiment. qPCR was used for relative quantification of mtCO1/β-globin in rapamycin induced

Minerval induces apoptosis in Jurkat and other cancer cells.

Minerval is an oleic acid synthetic analogue that impairs lung cancer (A549) cell proliferation upon modulation of the plasma membrane lipid structure and subsequent regulation of protein kinase C localization and activity. However, this mechanism does not fully explain the regression of tumours induced by this drug in animal models of cancer. Here we show that Minerval also induced apoptosis in Jurkat T-lymphoblastic leukaemia and other cancer cells. Minerval inhibited proliferation of Jurkat cells, concomitant with a decrease of cyclin D3 and cdk2 (cyclin-dependent kinase2). In addition, the

Calmodulin regulation of Ca2+ entry in Jurkat T cells.

We have previously proposed a role for calmodulin (CaM) in the regulation of initiation of Ca2+ entry in Jurkat T cells, as well as in the regulation of the current that mediates Ca2+ entry, IT. In this report, we provide evidence for the mechanism of CaM action. We have previously shown that activation-induced Ca2+ entry into Jurkat T cells is mediated by a current we have called IT. In the whole cell variation, but not the perforated patch variation, of the patch clamp technique, this current is short-lived (under 6 min) suggesting that the current is under the control of a diffusible compon

Cytolethal distending toxin upregulates RANKL expression in Jurkat T-cells.

Cytolethal distending toxin, a bacterial exotoxin produced by a number of Gram-negative species, causes growth arrest and morphological alterations in host cells. Among these species are Haemophilus ducreyi, the etiological agent of chancroid, and the periodontal pathogen Aggregatibacter actinomycetemcomitans, highly implicated in localized aggressive periodontitis. CDT induces receptor activator of NF-kappaB ligand (RANKL) expression in periodontal fibroblasts, the key bone-resorbing cytokine. T-cells are actively involved in localized inflammation-induced bone destruction, including periodon

Peripheral benzodiazepine receptor: characterization in human T-lymphoma Jurkat cells.

Peripheral benzodiazepine receptor (PBR) has been considered a promising drug target for cancer therapy, and several ligands have been developed for this purpose. Human T-lymphoma Jurkat cells have been considered as lacking PBR and are often used as negative control to prove the specificity of PBR ligands effects. It is surprising that we evidenced PBR protein expression in this cell line by means of Western blotting and immunocytochemistry assays using specific anti-PBR antibodies. PBR intracellular localization was evidenced in mitochondria and nuclei, as demonstrated by confocal and electr

Fas/FasL interaction mediates imbalanced cytokine/cytotoxicity responses of iNKT cells against Jurkat cells.

The rapid antitumor cytokine production and direct cytotoxicity confer invariant NKT (iNKT) cells ideal candidates for cancer therapy. However, the therapeutic potential of iNKT cells in T-cell malignant diseases remains elusive, as antigen presentation by T cells (T-T presentation) has been suggested to induce hyporesponsiveness of iNKT cells. In this study, we found discrepancies in iNKT cell responses against two T cell-origin cell lines (Jurkat and Molt-4). Human iNKT cells exhibited more intensive cytotoxicity and less efficient cytokine production in response to Fas-bearing Jurkat cells

A genome-wide survey of mutations in the Jurkat cell line.

BACKGROUND: The Jurkat cell line has an extensive history as a model of T cell signaling. But at the turn of the 21st century, some expression irregularities were observed, raising doubts about how closely the cell line paralleled normal human T cells. While numerous expression deficiencies have been described in Jurkat, genetic explanations have only been provided for a handful of defects. RESULTS: Here, we report a comprehensive catolog of genomic variation in the Jurkat cell line based on whole-genome sequencing. With this list of all detectable, non-reference sequences, we prioritize pote

XXYLT1 inhibits NOTCH1 activation in Jurkat cells while promoting cell proliferation.

Glycosylation, a key post-translational modification, regulates protein function in many contexts. Epidermal growth factor-like repeats undergo domain-specific O-glycosylation such as O-glucosylation, O-fucosylation, and O-GlcNAc'ylation. O-Glucose glycans are attached to specific serine residues by the action of protein O-glucosyltransferase 1 (POGLUT1) and can be elongated with two xylose residues by glucoside α1-3xylosyltransferase 1 (GXYLT1) or glucoside α1-3xylosyltransferase 2 (GXYLT2) and xyloside α1-3xylosyltransferase 1 (XXYLT1) in mammals. Chlamydia trachomatis serovar L2 infection model using human lymphoid Jurkat cells.

Chlamydia trachomatis L2 invasively attacks lymphatic and subepithelial tissues of the genital tract during the formation of primary lesions. This subsequently results in lymphadenopathy, and suggests a greater propensity for systemic dissemination. However, whether lymphocytes are a potential vehicle cell for the dissemination of this infection remains unknown. We therefore assessed the growth properties of C. trachomatis L2 in lymphoid Jurkat cells compared with those observed in epithelial HeLa cells. Both cells supported the growth of C. trachomatis with a similar increase in infective pro

Gastrin-CCK-B type receptors on human T lymphoblastoid Jurkat cells.

The presence of specific receptors for gastrointestinal hormones on T cells and their involvement in the immune response are still matters of debate. We reported the effects of gastrin-cholecystokinin (CCK)-related peptides on J.RT3-T3.5 Jurkat cells. A single class of high-affinity binding sites (dissociation constant approximately 0.1 nM) for gastrin and CCK-8 was evident on these cells. These peptides dose-dependently induced a transient increase in intracellular Ca2+ concentration ([Ca2+]i), which was independent of extracellular Ca2-. L-365,260 was 150- to 300-fold more potent than L-364,

Indirubin Inhibits TRAIL-Induced Activation of Death Receptor 5 in Jurkat Cells.

Death receptor 5 (DR5) is an apoptosis-inducing membrane receptor that mediates cell death in several life-threatening conditions. There is a crucial need for the discovery of DR5 antagonists for the therapeutic intervention of conditions in which the overactivation of DR5 underlies the pathophysiology. DR5 activation mediates cell death in non-alcoholic fatty liver disease (NAFLD) and neurodegenerative processes including amyloid-beta (Aβ) accumulation, spinal cord injury (SCI), and brain ischemia. In the current work, we used fluorescence resonance energy transfer (FRET) to monitor the confo

Effect of PEGylated Graphene Oxide Nanoparticles on the Metabolism of Jurkat Cells.

The effect of graphene oxide (GO) nanoparticles of 100-200 nm in size coated with linear (LP-GO) and branched (BP-GO) polyethylene glycol at concentrations of 5 and 25 μg/mL on the metabolism of Jurkat tumor cells was studied. It was found that LP-GO nanoparticles at a concentration of 25 μg/mL can enhance basal glycolysis of Jurkat T-lymphocyte tumor cell line cells, while LP-GO and BP-GO at the same concentration can reduce the indicators of compensatory glycolysis. Despite this, GO nanoparticles coated with linear and branched PEG at a concentration of 5 μg/mL do not have pronounced effects

Photodynamic responsiveness of human leukemia Jurkat/A4 cells with multidrug resistant phenotype.

UNLABELLED: Photodynamic therapy (PDT) is considered as a possible alternative approach to overcoming multidrug resistance (MDR). Analysis of cross-resistance to PDT in cells with different MDR pathways and resistance levels seems to be advantageous for elucidating the general mechanisms of cancer cell resistance to various treatment modalities. AIM: The aim of the study was to clarify whether the Jurkat/A4 leukemia cells with MDR phenotype are cross-resistant to PDT. METHODS: Human T-cell acute lymphoblastic leukemia line Jurkat and Jurkat/A4 subline with MDR phenotype were used. 5-Aminolev

Dinitrosyl-iron triggers apoptosis in Jurkat cells despite overexpression of Bcl-2.

Cells expressing the cytokine-inducible NO synthase are known to trigger apoptosis in neighboring cells. Paramagnetic dinitrosyl nonheme iron complexes (DNIC) were found in tumor tissue about 40 years ago; however, the role of these NO(+)-bearing species is not completely understood. In the human Jurkat leukemia cell line, the application of the model complex DNIC-thiosulfate (50-200 microM) induced apoptosis (defined by phosphatidylserine externalization) in a concentration- and time-dependent manner. In Jurkat cells, the pan-caspase inhibitor, zVADfmk (50 microM), and/or stable transfection

Intercellular mechanical communication drives the directional migration of Jurkat T immune cells.

Although the mechanisms by which chemokines regulate immune responses have been extensively elucidated, the contributions of biophysical mechanical cues to the modulation of immune cell migratory behaviors remain largely unexplored. In this study, we employed a modular co-culture system in which suspension Jurkat T lymphocytes (with RAW264.7 macrophages serving as a positive migratory control) were paired with two force-generating cell types-human airway smooth muscle cells (hASMCs) and human lung adenocarcinoma A549 cells-cultured on type I collagen hydrogels to investigate the effects of int

Overexpression of MiR-506 in Jurkat (Acute T Cell Leukemia) Cell Line.

BACKGROUND & OBJECTIVE: Acute lymphoblastic leukemia (ALL) is a malignant disease that arises from various mutations in B or T-lymphoid progenitors. MicroRNAs (miRNAs) regulate gene expression by binding to the 3' untranslated region of protein-coding genes. Dysregulation of miRNA expression may result in the development of cancerous phenotypes. Therefore, for the first time in this field, the present study aims to investigate the effect of overexpression of miR-506 in Jurkat (acute T cell leukemia) cell line. METHODS: In this study, Jurkat cell lines were cultured in RPMI-1640 medium. Next,

Electroporation outcomes in human Jurkat cells are affected by exposure to simulated microgravity.

Electroporation (EP) is commonly used to permeabilize cell membranes to deliver nucleic acids and other biomolecules. EP-based methodologies have the potential to support in-space biomanufacturing; however, it is currently unclear whether cellular adaptations to microgravity influence EP outcomes. To investigate this, human T-lymphocyte (Jurkat) cells were exposed to simulated microgravity (SµG) in a rotating wall vessel (RWV) bioreactor prior to EP to deliver exogenous fluorescent molecules or plasmid DNA. We observed that two hours of SµG exposure led to a 20% decrease in fluorophore uptake
