Microinjection of kainic acid into the rat hippocampus.

The effects of unilateral injection of kainic acid into the rate hippocampus have been examined in terms of morphologic, neurochemical and behavioral sequelae. Infusion of 10 nmoles if kainate causes a rapid and complete degeneration of neuronal perikarya in the entire hippocampal formation followed by gliosis and atrophy of the region. This unilateral neuronal loss is accompanied by a 50% decrease in the specific activity of the biochemical markers for GABAergic neurons including glutamic acid decarboxylase, endogenous GABA and synaptosomal uptake of [3H]GABA. The extrinsic hippocampal cholin

Polarization anatomy of a kainic acid seizure.

Slow voltage-sensitive dyes work by accumulating in brain tissue and report the average membrane potential of neurons and glia. The voltage-sensitive dye diO-C2-5 was used to monitor the polarization state of 27 brain structures in the rat during a systemically induced, behaviorally mild, kainic acid seizure using a 20 s recording period. The effects of the anesthetic agent used in the experiment were minimized by delaying the dye injection and seizure mapping for one day. Eleven areas were depolarized during the seizure, but 16 other areas did not change their polarization state compared to c

Kainic acid pig model of hippocampal epilepsy.

Translational large-animal models that can accommodate human-scale implantable devices are essential for advancing neuromodulation therapies in epilepsy. This study establishes a kainic acid (KA)-induced porcine model of mesial temporal lobe epilepsy (mTLE) using clinical imaging, stereotactic surgery, and a fully implantable neural stimulator-recorder (INSR) device designed for humans. Seven pigs (six KA-treated and one saline control) underwent MRI-guided stereotactic implantation of electrodes targeting bilateral hippocampus (HPC) and anterior thalamus (ANT), followed by intra-hippocampal K

Kainic acid-induced seizures in the common marmoset.

Treatment of epilepsy remains difficult because patients suffer from pharmacoresistant forms of the disease and drug side-effects. Thus, there is an urgent need to identify not only new antiepileptic drug candidates but also novel epileptic animal models. Here, we characterize seizures induced with kainic acid (KA) in the common marmoset (Callithrix jacchus). Adult marmosets received 0.1, 1, or 10 mg/kg of KA subcutaneously. All animals exhibited early convulsive behavior (seizure scores of I and II on the Racine scale). Seizure scores were low at lower KA doses, but the highest dose of KA tes

Kainic acid-induced decrease in hippocampal corticosteroid receptors.

The potential role of excitatory amino acids in the regulation of brain corticosteroid receptors was examined using systemic administration of kainic acid. Administration of kainic acid (5, 10, and 15 mg/kg) to 24-h adrenalectomized rats that were killed 3 h later produced large, dose-related decreases in glucocorticoid receptors (GR) in hippocampus (23-63%), frontal cortex (22-76%), and striatum (41-49%). Kainic acid did not decrease hypothalamic GR. Hippocampal mineralocorticoid receptors (MR) were also markedly decreased (50-71%) by kainic acid. Significant decreases in corticosteroid recep

Regulatory changes following kainic acid-induced striatal lesions.

Kainic acid-induced lesions (KAL) of the striatum produce body weight and regulatory deficits in the rat. Unlike lateral hypothalamic rats. KAL rats drink more during food deprivation and eat more afterwards as compared to both baseline conditions and control rats. The present study investigated these effects further. As in previous studies, food deprivation was found to cause polydipsia and increased postdeprivational food intake in the KAL animal. Urination and defecation, often used as an index of emotionality, were also found to increase under these conditions. When the antidiuretic hormon

The Kainic Acid Models of Temporal Lobe Epilepsy.

Experimental models of epilepsy are useful to identify potential mechanisms of epileptogenesis, seizure genesis, comorbidities, and treatment efficacy. The kainic acid (KA) model is one of the most commonly used. Several modes of administration of KA exist, each producing different effects in a strain-, species-, gender-, and age-dependent manner. In this review, we discuss the advantages and limitations of the various forms of KA administration (systemic, intrahippocampal, and intranasal), as well as the histologic, electrophysiological, and behavioral outcomes in different strains and specie

Kainic acid-induced neurodegenerative model: potentials and limitations.

Excitotoxicity is considered to be an important mechanism involved in various neurodegenerative diseases in the central nervous system (CNS) such as Alzheimer's disease (AD). However, the mechanism by which excitotoxicity is implicated in neurodegenerative disorders remains unclear. Kainic acid (KA) is an epileptogenic and neuroexcitotoxic agent by acting on specific kainate receptors (KARs) in the CNS. KA has been extensively used as a specific agonist for ionotrophic glutamate receptors (iGluRs), for example, KARs, to mimic glutamate excitotoxicity in neurodegenerative models as well as to d

Selective effects of kainic acid on diencephalic neurons.

Injections of kainic acid (KA) into the lateral hypothalamus (LH) produce neuronal loss in this region without apparent damage to medial forebrain bundle fibers passing through the area. Cellular destruction is not limited to the LH; the neuronal loss in the thalamic reticular nucleus, the subthalamic nucleus and zona incerta is more extensive than that in the LH. Since all of the nuclei of ventral thalamic origin except the ventral lateral geniculate nucleus (VLGN) were destroyed by LH injections of KA, we sought to determine whether this nucleus also is sensitive to KA. Injections directly i

Protective effects of lycopene on kainic acid-induced seizures.

Lycopene (LCP) is a carotenoid that protects against many diseases by alleviating oxidative stress. However, the effect of LCP on epileptic seizures has not been examined well in previous studies. In the current work, we employed kainic acid (KA) to induce experimental epileptic seizures in mice, and investigated the function of LCP during this process. We found that the onset and extent of KA-induced seizures were alleviated in LCP-pretreated mice. Nissl staining of hippocampus showed that the granule cell dispersion lesion induced by KA was improved by the LCP treatment. Additionally, we ana

Alpha 2-adrenoceptors modulate kainic acid-induced limbic seizures.

We have tested several compounds interfering with the brain monoamine (noradrenaline, dopamine, serotonin) and acetylcholine systems for their effects on limbic seizures produced by systemically (s.c.) injected kainic acid as well as on neurochemical changes in amygdala/pyriform cortex resulting from the kainic acid treatment. The characteristic neurochemical changes induced by s.c. kainic acid were a decrease in noradrenaline and an increase in 5-hydroxyindoleacetic acid in the acute (3 h after kainic acid injection) suggesting strongly increased neurotransmitter turnover in noradrenergic and

Ido1 and Ido2 deficiencies attenuate kainic acid-induced ictogenesis.

Insults to the brain in the form of injury, infection and exposure to seizurogenic chemicals can induce seizures (ictogenesis). Seizure episodes during any of these events is one of the biggest risk factors for the development of treatment-resistant epilepsy, making the acute ictogenic period a critical time for intervention. Indoleamine-2,3-dioxygenases (Ido1 & Ido2) modulate systemic and central neuroprotective and inflammatory responses through both enzymatic and non-enzymatic mechanisms. We have previously found that Ido1-/- (knockout) mice have increased seizure incidence following viral

Intrinsic brain network stability during kainic acid-induced epileptogenesis.

OBJECTIVE: Altered intrinsic brain networks have been revealed in patients with epilepsy and are strongly associated with network reorganization in the latent period. However, the development and reliability of intrinsic brain networks in the early period of epileptogenesis are not well understood. The current study aims to fill this gap by investigating the test-retest reliability of intrinsic brain networks in the early stage of epileptogenesis. METHODS: We used the rat intrahippocampal kainic acid model of mesial temporal lobe epilepsy. Three sessions of resting-state functional magnetic r

Anticonvulsant effect of gentamicin on the seizures induced by kainic acid.

OBJECTIVE: Epilepsy is a chronic neurological disorder affecting approximately 0.5-2% of the population worldwide. Gentamicin (GM) is an aminoglycoside antibiotic used to treat several types of bacterial infections. We investigate whether the administration of GM can reduce seizures in temporal lobe epilepsy (TLE). METHODS: The animal model of temporal lobe epilepsy (TLE) was established by kainic acid (KA) intrahippocampal injection. Behavioral test and Electroencephalography (EEG) recordings were performed to detect the effects of GM on the seizures triggered by KA injection in rats. Furthe

Status epilepticus alters hippocampal ultrastructure in kainic acid rat model.

Kainic acid (KA) model of epilepsy is a reliable tool to study temporal lobe epilepsy (TLE), the most common type of partial epilepsy in adults. Substantial body of data suggest that the KA-induced status epilepticus (SE) leads to several molecular and structural changes in the hippocampus, including sclerosis, sprouting of mossy fiber, reorganization of inter-neuronal networks, alterations in neuropeptide signaling, gliosis, and synaptic transmission dysregulation. However, no details on the ultrastructural changes, especially in relationship to synapses are available. This information is imp

Benzothiadiazines derivatives as novel allosteric modulators of kainic acid receptors.

The majority of excitatory neurotransmission in vertebrate CNS is mediated by glutamate binding to different types of receptors. Among them, a-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid receptor (AMPAR) and kainite receptors (KAR) are ionotropic receptors playing important pathophysiological roles. A number of small molecules acting as positive allosteric modulators (PAM) of AMPAR have been proposed as drugs for neurological disorders, however, there is no such abundance of ligands capable of modulating KARs activity. We investigated the ability of IDRA21 and of its derivative, compoun

Reduced Interhemispheric Coherence in Cerebellar Kainic Acid-Induced Lateralized Dystonia.

The execution of voluntary muscular activity is controlled by the primary motor cortex, together with the cerebellum and basal ganglia. The synchronization of neural activity in the intracortical network is crucial for the regulation of movements. In certain motor diseases, such as dystonia, this synchrony can be altered in any node of the cerebello-cortical network. Questions remain about how the cerebellum influences the motor cortex and interhemispheric communication. This research aims to study the interhemispheric cortical communication between the motor cortices during dystonia, a neurol

Overexpression of apolipoprotein E4 increases kainic-acid-induced hippocampal neurodegeneration.

Apolipoprotein E (apoE) has an intricate biological function in modulating immune responses and apoE isoforms exhibit diverse effects on neurodegenerative and neuroinflammatory disorders. In the present study, we investigated the individual roles of apoE isoforms in the kainic acid (KA)-induced hippocampal neurodegeneration with focus on immune response and microglia functions. ApoE2, 3 and 4 transgenic mice as well as wild-type (WT) mice were treated with KA by intranasal route. ApoE4 overexpressing mice revealed several peculiarities as compared with other transgenic mice and WT mice, i.e. (

Prevention of kainic acid-induced seizure-like activity by antiepileptic drugs.

Intracerebroventricular kainic acid (KA) injections produced a seizure-like activity syndrome in mice. It was shown that some antiepileptic drugs (lefadol, depakine, phenobarbital) inhibited this syndrome, diazepam being the most effective in this respect. However, acetazolamide and diphenylhydantoin exerted a rather weak or negligible effect even when given in high doses. Present results may suggest that the effects of intracerebroventicular injections of KA may be employed to test antiepileptic properties of drugs.

Effect of epileptogenesis on hypercapnic cardioventilatory response in kainic acid rats.

OBJECTIVE: Cardioventilatory failure is the leading mechanism proposed to underlie sudden unexpected death in epilepsy (SUDEP), which occurs predominantly at night in patients with generalized tonic-clonic seizures. Interictal hypercapnic cardioventilatory responses are suggested to be involved, as they are ablated in chronically epileptic kainic acid (KA) rats, a temporal lobe epilepsy model with focal to bilateral tonic-clonic seizures. However, how this impairment emerges during epileptogenesis and whether it is influenced by day/night period remain unclear. Here, we aimed to investigate th
