Living with Keratinocytes.

A feature distinguishing human hematopoietic and epithelial stem cells from other equally fascinating stem cells is perhaps their easier translation into a clinical setting. We have devoted nearly our entire scientific career in trying to turn our understanding of epithelial stem cell biology into something that could help people suffering from virtually untreatable diseases of squamous epithelia. We have done that as a team, together with our numerous students, postdocs, technicians and valuable collaborators, clinicians, regulators, and, lately, industrial partners. We had rewarding successe

Updated Perspectives on Keratinocytes and Psoriasis: Keratinocytes are More Than Innocent Bystanders.

Psoriasis is a complex disease triggered by genetic, immunologic, and environmental stimuli. Many genes have been linked to psoriasis, like the psoriasis susceptibility genes, some of which are critical in keratinocyte biology and epidermal barrier function. Still, the exact pathogenesis of psoriasis is unknown. In the disease, the balance between the proliferative and differentiative processes of keratinocytes becomes altered. Multiple studies have highlighted the role of dysregulated immune cells in provoking the inflammatory responses seen in psoriasis. In addition to immune cells, accumula

[Keratinocyte cancer].

Keratinocyte cancer (KC) cases have risen by 172% in Denmark over the last decade and the lifetime risk of developing KC is at least 22%. This highlights the need for innovative strategies in KC prevention, diagnosis, and treatment. To reduce incidence, new prevention methods are crucial. New diagnostics using non-invasive scanning can streamline patient care, especially for low-risk tumours and patients with multiple tumours. New treatments can be more effective and with fewer side effects. Together, these initiatives can improve the patient journey and ease the burden on healthcare resources

Adhesion molecules in keratinocytes.

Adhesion molecules are proteins on the cell surface that are involved in the interactions between lymphocytes and antigen-presenting cells, especially in inflammatory skin diseases and autoimmune bullous disorders. Adhesion molecules include cadherins (subgroups E, N, P, M), integrins, selectins, and the immunoglobulin gene family. Cadherins E in the epidermis including desmocollin 1 and 2 and desmoglein 1 and 3 are essential transmembrane components of desmosome glycoproteins, which play a major role in bullous diseases, pemphigus in particular. Also important are integrin beta 1 alpha 1 and

Adhesion molecules. I: Keratinocyte-keratinocyte interactions; cadherins and pemphigus.

During the last few years, considerable progress has been made in our understanding of the structure and function of cadherins and of the pathophysiology of pemphigus. Cadherins are a multiple gene family of Ca(++)-dependent cell adhesion molecules with a typical single-spanning transmembrane structure. Cadherins have two major subfamilies, classic cadherin and desmosomal cadherin. Classic cadherins, including E-, P-, and N-cadherins, are characterized by a homophilic binding specificity. They localize at adherens junctions and mediate physiologic interaction with the involvement of cytoplasmi

Effect of chemical modification of keratinocytes and collagen in keratinocyte-collagen interactions.

Neonatal and adult keratinocytes isolated from thin sections of split-thickness skin by trypsin-release show a preferential and strong attachment to collagen when compared to plastic, fibronectin-coated plastic, glass, or agar gels. We have investigated the reactive groups of keratinocytes and collagen required for this interaction and have determined the kinetics of attachment. At 37 degrees C both neonatal and adult keratinocytes show a rapid and irreversible attachment to collagen, reaching a plateau phase at 30-60 minutes. The cells cannot be replaced from the gel by extensive washing or b

Beta adrenergic receptors in keratinocytes.

Beta2 adrenergic receptors were identified in keratinocytes more than 30 years ago, but their function in the epidermis continues to be elucidated. Abnormalities in their expression, signaling pathway, or in the generation of endogenous catecholamine agonists by keratinocytes have been implicated in the pathogenesis of cutaneous diseases such as atopic dermatitis, vitiligo, and psoriasis. New studies also indicate that the beta2AR also modulates keratinocyte migration, and thus can function to regulate wound reepithelialization. This review focuses on the function of these receptors in keratin

Keratinocytes and cytokine/growth factors.

Cytokines are polypeptide growth factors produced by most nucleated cells in the body, including epithelial cells, keratinocytes, and Langerhans cells in the skin. Cytokines can be classified into interleukins, tumor necrosis factors, chemokines, colony-stimulating factor, interferons, and growth factors. Like classic hormones, cytokines bind to specific receptors to transmit their messages to target cells. Cytokine receptors can be divided into three cytokine receptor superfamilies: the immunoglobulin superfamily, the hematopoietin family, and the tumor necrosis factor family. Following cytok

Revisiting oxytocin generation in keratinocytes.

Some evidence suggests that oxytocin, which is a neuropeptide conventionally thought to be synthesized in the hypothalamus and released by the posterior pituitary, is generated in peripheral keratinocytes, but the details are lacking and the mRNA analysis is further required. Oxytocin and neurophysin I are generated together as cleavage products after splitting the precursor molecule, preprooxyphysin. To confirm that oxytocin and neurophysin I are also generated in the peripheral keratinocytes, it must first be clarified that these molecules contained in peripheral keratinocytes did not origin

Keratinocytes in allergic skin diseases.

PURPOSE OF REVIEW: The aim of the review is to provide up-to-date information on the multiple roles of epidermal keratinocytes in the immune reactions associated with allergic contact dermatitis and atopic dermatitis skin diseases. RECENT FINDINGS: In the last two decades, it has become clear that keratinocytes are highly active immunological cells, with major control over the acute and the chronic phases of skin inflammation by means of cytokine/chemokine production and surface molecule expression. Keratinocyte responses in skin allergic reactions are rather disease-specific and keratinocyte

Human keratinocytes express fractalkine/CX3CL1.

BACKGROUND: fractalkine/CX3CL1 is a unique chemokine that has properties of both chemoattractants and adhesion molecules. The major source of this chemokine in the skin is still controversial. OBJECTIVE: studies were undertaken to determine the expression of fractalkine in human skin. METHODS: RT-PCR, Western blotting, and immunostaining were performed with normal human epidermal keratinocytes (NHEK) and HaCaT cells, human keratinocyte cell line, for the presence of fractalkine. Biopsy specimens of normal and diseased skin were also investigated. RESULTS: we identified that NHEK and HaCaT c

Keratinocyte dysfunction in vitiligo epidermis: cytokine microenvironment and correlation to keratinocyte apoptosis.

Vitiligo is a skin disorder characterized by loss of functional melanocytes. Keratinocytes contribute to melanocyte homeostasis, and keratinocyte alteration may play a role in melanocyte dysfunction in vitiligo. In particular, the release of melanogenic mediators and the level of functioning keratinocytes may affect melanocyte dysfunction in vitiligo epidermis. Keratinocyte-derived mediators involved in pigmentation, analysed by in situ hybridization, and epidermal apoptosis, detected by TUNEL assay and electron microscopy, were evaluated in lesional and perilesional skin biopsies from 15 pati

Expression of Fas antigen on keratinocytes in vivo and induction of apoptosis in cultured keratinocytes.

Fas antigen, which belongs to a nerve growth factor/tumor necrosis factor receptor superfamily, is a membrane protein that induces apoptosis. In humans, distribution of Fas antigen has been reported on cell lines and lymphocytes. Immunohistochemical studies revealed Fas antigen on the keratinocytes of lesional epidermis in lichenoid drug eruption, erythema multiforme, contact dermatitis, bullous pemphigoid, pemphigus vulgaris, and herpes zoster; it is co-expressed with intercellular adhesion molecule-1. Cultured keratinocytes expressing Fas antigen increased from 8.4% to 34.6% after stimulatio

Keratinocyte gene therapy.

BACKGROUND: Gene therapy is currently being used in clinical trials to treat a variety of diseases. In keratinocyte gene therapy, the gene that will correct the disease by expressing the normal protein or enzyme is inserted and expressed in keratinocytes. Keratinocytes have significant potential, as a target cell of gene therapy, in the treatment of both systemic diseases as well as skin diseases caused by a genetic defect in keratinocytes. OBSERVATIONS: Although keratinocyte gene therapy is not yet being tested in clinical trials, animal models do exist where keratinocytes are being used to

Keratinocyte Carcinoma: A Review.

IMPORTANCE: Keratinocyte carcinomas are skin cancers that arise from keratinocytes and are composed of basal cell carcinomas (BCCs) and cutaneous squamous cell carcinomas (cSCCs). Keratinocyte carcinomas are common in North America, Australia, New Zealand, and Europe. Approximately 5.4 million keratinocyte carcinomas are diagnosed in the US annually. OBSERVATIONS: Keratinocyte carcinomas are primarily located on the head and neck (40%-64% of BCCs; 35%-45% of cSCCs). BCC typically presents as a pink, smooth, raised lesion or a pink to red, flat lesion. cSCC typically presents as a red, scaly,

Premalignant keratinocytic neoplasms.

Premalignant keratinocytic keratoses are common, especially in pale-complected persons in whom they appear most often as an actinic keratosis. Although the actinic keratosis has a very low malignant potential, arsenic, tar, thermal, scar, reactional, and radiation keratoses may be more clinically aggressive. This article discusses these premalignant keratinocytic neoplasms.

Basal keratinocyte herniation.

Basal cell herniations, i.e. basal keratinocyte processes protruding through basal lamina gaps were observed in psoriasis, circinate balanitis, pityriasis rubra pilaris, patch tests, gold dermatitis and conjunctivitis. This indicates that they are not specific to psoriasis and tumours as has been reported.

The ciliated human keratinocyte.

Human keratinocytes were investigated for the presence of single cilia. Almost all basal keratinocytes were found to carry a single cilium in normal, occluded, and psoriatic skin. The ciliary structure was progressively reduced in keratinocytes approaching the surface. No remnants of the ciliary apparatus were found in the granular layer. In one case of nickel-allergic dermatitis (patch test), the keratinocytes had lost their cilia; the significance of this surprising finding remains to be elucidated.
