[Mycocin sensitivity patterns of the species Kluyveromyces: Kluyveromyces sensu lato vs. Kluyveromyces sensu stricto].

The Kluyveromyces species reassigned to the genera Lachancea and Vanderwaltozyma are insensitive to five mycocins secreted by Pichiamembranifaciens. The remaining Kluyveromyces species including species transferred to the genera Kazachstania, Nakaseomyces, and Tetrapisispora are sensitive to them. Only the neotype strain is insensitive to mycocins among Kluyveromyceslactis cultures.

Inhibitory action of antibiotics on Kluyveromyces marxianus.

A bioassay containing Kluyveromyces marxianus in microtiter plates was used to determine the inhibitory action of 28 antibiotics (aminoglycosides, beta-lactams, macrolides, quinolones, tetracyclines and sulfonamides) against this yeast in whey. For this purpose, the dose-response curve for each antibiotic was constructed using 16 replicates of 12 different concentrations of the antibiotic. The plates were incubated at 40°C until the negative samples exhibited their indicator (5-7h). Subsequently, the absorbances of the yeast cells in each plate were measured by the turbidimetric method (λ=600n

Deletions and rearrangements in Kluyveromyces lactis mitochondrial DNA.

Three classes of respiratory deficient mutants have been isolated from a fusant between Kluyveromyces lactis and Saccharomyces cerevisiae that contains only K. lactis mtDNA. One class (15 isolates), resemble rho 0 mutants of S. cerevisiae as they lack detectable mtDNA. A second class (16 isolates), resemble point mutations (mit-) or nuclear lesions (pet-) of S. cerevisiae as no detectable change is found in their mtDNA. The third class (five isolates), with deletions and rearrangements in their mtDNA are comparable to S. cerevisiae petite (rho-) mutants. Surprisingly, three of the five deletio

Microbial Competition and Nutrient Limitation Remodel the Volatilome of Kluyveromyces marxianus.

The use of Kluyveromyces marxianus in mixed cultures for fermentation processes has become increasingly relevant. This yeast is characterized by rapid growth, thermotolerance, broad sugar utilization, and the ability to produce aroma-active compounds. In this study, we evaluated changes in the growth and volatilome of a K. marxianus strain isolated from agave fermentation under microbial competition induced by co-cultivation interactions and nutritional limitation induced by a nutrient-deficient medium. The results indicate that these stress factors are significant drivers of metabolic changes

Unlocking the potential of Kluyveromyces marxianus in the definition of aroma composition of cheeses.

INTRODUCTION: The cheese microbiota is very complex and is made up of technologically-relevant, spoilage, opportunistic and pathogenic microorganisms. Among them lactic acid bacteria and yeasts are the main ones. One of the most interesting dairy yeasts is Kluyveromyces marxianus because of its technological properties including the ability to produce aroma compounds. METHODS: This study investigated the contribution of Kluyveromyces marxianus to the gross composition and aroma profile of cow cheeses. Experimental cheeses were prepared by inoculating a co-culture of K. marxianus FM09 and a co

Exploring the phenotypic diversity of oenological traits in Kluyveromyces marxianus strains.

The use of non-Saccharomyces yeasts in the winemaking process may have several positive outcomes. Kluyveromyces marxianus has recently been revealed as a promising species for this industry. While the majority of studies mention the use of K. marxianus in various industries including food production (e.g. dairy and cocoa), recent studies have also shown that its aroma and pectinase production make it a suitable yeast for the wine industry. Nevertheless, only particular strain, IWBT Y885, was investigated. In this study, five different K. marxianus strains as well as one protoplast fusant (BF20

Autoactivated KlPDR1 gene in the control of multidrug resistance in Kluyveromyces lactis.

The KlPDR1 gene encodes a zinc finger transcription factor that has recently been shown to be involved in the control of multidrug resistance of Kluyveromyces lactis . In this work, we provide evidence that the K. lactis KlPDR1 gene is under positive autoregulation by KlPdr1p, which plays a role in the activation of the main multidrug resistance transporter gene KlPDR5. Electrophoretic mobility shift assays, as well as the use of gusA reporter constructs, enabled us to identify the 5'-tataTCCGGGTAactt-3' sequence motif in the KlPDR1 promoter (in the position -326 to -319 bp) as the PDRE (pleio

Reduction of Lactose in Milk by Purified Lactase Produced by Kluyveromyces lactis.

Lactase preparations were purified from cell-free extracts of Kluyveromyces lactis to homogeneity, as determined by disc SDS polyacrylamide gel electrophoresis. A combination of techniques including ammonium sulfate or acetone precipitation and hydroxyapatite chromatography was used for the purification of the enzyme. The enzyme has a pH optimum of 6.5-7.0 with Kms' of 1.25 and 28mM for the substrates O-nitrophenylgalactopyranoside and lactose respectively. Activation energies for denaturation of enzyme and conversion of substrate to product were determined to be 15.1 and 10.0 Kcal/mol, respec

New generation probiotics: combining probiotic and biotechnological potential of Kluyveromyces marxianus.

Kluyveromyces marxianus has high biotechnological potential, particularly because it can utilize a wide range of substrates and grow faster than other yeasts. It has also attracted interest as a prospective probiotic; however, this application is still not well researched. Two K. marxianus strains with promising biotechnological properties and the proven ability to efficiently produce 2-phenylethanol (2-PE) and ethanol were examined in this study to evaluate their potential as probiotics. Yeasts WUT216 and WUT240 exhibited similar survival rates in a simulated human digestive system as the ref

Extracellular Vesicles from Kluyveromyces marxianus as Potential Postbiotics Against Candida albicans Vaginal Infections.

This study describes extracellular vesicles (EVs) isolated from the culture supernatant of a Kluyveromyces marxianus strain deriving from an artisanal sourdough. Previous work had clearly shown the probiotic properties of the yeast isolate and its antagonistic activities against clinical fluconazole-resistant Candida albicans strains. Characterization of the isolated EVs by nanotracking particle analysis showed they had a mean diameter of 157.7 nm. Proteomic characterization of the purified EVs identified a complex array of 100 proteins. Both C. albicans planktonic growth and biofilm formation

A Novel Polyamine Oxidase from Kluyveromyces marxianus with Potential for Total Polyamine Determination.

Polyamines and their acetylated derivatives are promising biomarkers for noninvasive cancer diagnostics, creating demand for robust enzymatic tools for their detection. In this study, we screened several phylogenetically diverse yeast polyamine oxidases and identified a new enzyme from Kluyveromyces marxianus (KmaPAO) as a promising candidate for analytical development. Among the tested candidates, only enzymes from K. marxianus and Lachancea thermotolerans were obtained in soluble active form, while only KmaPAO could be purified and characterized in detail. KmaPAO was produced in soluble acti

Functional analysis of Mig1 and Rag5 as expressional regulators in thermotolerant yeast Kluyveromyces marxianus.

To analyze the glucose repression mechanism in the thermotolerant yeast Kluyveromyces marxianus, disrupted mutants of genes for Mig1 and Rag5 as orthologs of Mig1 and Hxk2, respectively, in Saccharomyces cerevisiae were constructed, and their characteristics were compared with those of the corresponding mutants of S. cerevisiae. MIG1 mutants of both yeasts exhibited more resistance than the corresponding parental strains to 2-deoxyglucose (2-DOG). Histidine was found to be essential for the growth of Kmmig1, but not that of Kmrag5, suggesting that MIG1 is required for histidine biosynthesis in

Effect of temperature on the lactose hydrolytic capacity of a lactase derived from Kluyveromyces lactis.

In vitro studies of lactose hydrolysis in milk with 20-125 neutral lactase units (NLUs) carried out at 38.0 degrees C for 15 min with a beta-galactosidase derived from Kluyveromyces lactis (Lactaid, Lactaid Inc, Pleasantville, NJ) resulted in 85-95% of the hydrolysis observed with standard incubation conditions (24 h at 4-5 degrees C with 1000 NLU/L). Thirty-three lactose-maldigesting Guatemalan subjects, 16 children and 17 adults, were challenged with oral doses of lactose in milk (children aged less than 12 mo, 2 g/kg body wt; children aged 12-24 mo, 15 g/kg body wt; older children and adult

Extrachromosomal genetics in the yeast Kluyveromyces lactis. Isolation and characterization of antimycin-resistant mutants.

Antimycin-resistant (AR) mutants of the yeast Kluyveromyces lactis, obtained either spontaneously or after manganese treatment, were isolated and genetically characterized. Most of the mutants obtained after manganese mutagenesis and two spontaneous mutants, tolerated high antimycin concentrations (more than 10 micrograms/ml) and were extrachromosomal. One mutant which grew only in low antimycin (1 microgram/ml) showed a Mendelian type of inheritance. The extrachromosomal mutants could be assigned to at least two genetic loci (ARI and ARII). Mutants representative of these two groups showed in

ROS signaling mediates aeration-dependent thermotolerance in Kluyveromyces marxianus for high-temperature xylitol production.

This study uncovers that moderate reactive oxygen species (ROS) signaling mediates aeration-dependent thermotolerance in Kluyveromyces marxianus, challenging the long-held paradigm that ROS function solely as toxic metabolic byproducts. Through integrated transcriptomic profiling and RT-qPCR validation, 19 key transcription factors regulating this adaptive response were identified, and functional assays demonstrated that targeted knockout of GSF2 and RIM101 coupled with MED15 overexpression significantly enhances hypoxic thermotolerance. The ERG6-overexpressing strain YZB559 yields 72.45 g/L x
