Detection of feline kobuvirus in filtered fecal inocula and molecular epidemiological characterization of feline kobuvirus in Japan.

Feline kobuvirus (FKoV) is an enteric virus detected in cats in several countries, but its prevalence and genomic characteristics in Japan remain unclear. Here, we report the detection of FKoV in specific-pathogen-free (SPF) cats inoculated with filtered fecal material that had tested negative for FKoV by an initial RT-PCR assay. The inoculum was derived from a shelter-housed cat positive for a feline norovirus (FNoV) strain (KU22; GVI.1; GenBank accession no. LC935948), which shared 99% VP1 nucleotide identity with the Japanese strain M49-1. FKoV RNA was undetectable in all four inoculated ca

Caprine Kobuvirus VP1 Protein Activates the Mitochondrial Apoptotic Pathway via Interaction with BAD.

Caprine kobuvirus (CKoV) is a member of the Picornaviridae family that causes enteritis in goats and poses a significant health threat, yet its molecular pathogenesis remains largely unknown. Here, we show that experimental infection of goats with CKoV induces apoptosis in the spleen, lungs, and intestine tissues. In vitro, CKoV infection of HEK293T cells triggers apoptosis, with the viral structural protein VP1 identified as sufficient to induce apoptosis. VP1 activates the mitochondrial (intrinsic) apoptotic pathway, as evidenced by increased reactive oxygen species (ROS) production, activat

Detection of Systemic Canine Kobuvirus Infection in Peripheral Tissues and the Central Nervous System of a Fox Infected with Canine Distemper Virus.

Canine kobuvirus (CaKV) is a globally distributed pathogen of dogs and is predominantly associated with infection of the gastrointestinal tract. However, an etiological link to enteric disease has not been established since CaKV has been identified in both asymptomatic dogs and animals with diarrheic symptoms. In this study, an extraintestinal CaKV infection was detected by next-generation sequencing in a fox (Vulpes vulpes) in Germany concomitant with a canine distemper virus (canine morbillivirus; CDV) co-infection. Phylogenetic analysis of the complete coding region sequence showed that thi

Development and validation of a reverse transcription-insulated isothermal PCR assay for detection of caprine kobuvirus and its epidemiological and molecular characterization in China.

Caprine kobuvirus (CKoV) is an emerging diarrheal pathogen in goats. We developed and validated a reverse transcription-insulated isothermal PCR (iiPCR) targeting the conserved 3D gene for detection of CKoV. The assay demonstrated high analytical specificity, a limit of detection of 39.9 copies·μL-1, and good repeatability. A total of 722 goat fecal samples were collected from five farms in Sichuan, Chongqing, Yunnan, and Hunan in China and tested using the newly developed method. The overall CKoV positivity rate was 40.44% (292/722), and the farm-level positivity rate was 100% (5/5), with the

Simultaneous detection of canine astrovirus and canine kobuvirus by duplex qPCR: validation and evidence of extraintestinal viral RNA in naturally infected dogs.

Canine astrovirus (mamastrovirus 5 [MAstV5], formerly CaAstV; family Astroviridae, taxon species Mamastrovirus canis) and canine kobuvirus (aichivirus A2 [AiV-A2], formerly CaKoV; Picornaviridae, Kobuvirus aichi) are emerging enteric viruses increasingly detected in both diarrheic and subclinical dogs. Although primarily associated with gastrointestinal illness, recent evidence suggests a potential for systemic dissemination, which remains insufficiently explored. To improve detection, we developed and validated a duplex quantitative real-time PCR (dqPCR) assay for simultaneous identification

Naturally transmitted mouse viruses highlight the heterogeneity of virus transmission dynamics in the dirty mouse model.

UNLABELLED: Specific-pathogen-free (SPF) mice are widely used in biomedical research to model human infections. However, these animals do not always accurately recapitulate human immune responses. This is due, in part, to their lack of infection history. A growing number of studies show that the host microbiome influences the development, progression, and responses of many diseases. To date, the majority of research on the microbiome has focused on the bacterial populations and less on the eukaryotic virome of the host. Here, we characterize a transmission model where SPF mice are exposed to n

Diversity of picornaviruses in rural Bolivia.

The family Picornaviridae is a large and diverse group of viruses that infect humans and animals. Picornaviruses are among the most common infections of humans and cause a wide spectrum of acute human disease. This study began as an investigation of acute flaccid paralysis (AFP) in a small area of eastern Bolivia, where surveillance had identified a persistently high AFP rate in children. Stools were collected and diagnostic studies ruled out poliovirus. We tested stool specimens from 51 AFP cases and 34 healthy household or community contacts collected during 2002-2003 using real-time and sem

Exploring the Potential of Muridae as Sentinels for Human and Zoonotic Viruses.

In recent years, the transmission of viruses from wildlife to humans has raised significant public health concerns, exemplified by the COVID-19 pandemic caused by the betacoronavirus SARS-CoV-2. Human activities play a substantial role in increasing the risk of zoonotic virus transmission from wildlife to humans. Rats and mice are prevalent in urban environments and may act as reservoirs for various pathogens. This study aimed to evaluate the presence of zoonotic viruses in wild rats and mice in both urban and rural areas, focusing on well-known zoonotic viruses such as betacoronavirus, hantav

A Comprehensive Review on Human Aichi Virus.

Although norovirus, rotavirus, adenovirus and Astrovirus are considered the most important viral agents transmitted by food and water, in recent years other viruses, such as Aichi virus (AiV), have emerged as responsible for gastroenteritis outbreaks associated with different foods. AiV belongs to the genus Kobuvirus of the family Picornaviridae. It is a virus with icosahedral morphology that presents a single stranded RNA genome with positive sense (8280 nucleotides) and a poly (A) chain. AiV was first detected from clinical samples and in recent years has been involved in acute gastroenterit
